Abstract:Various geraniol esters act as insect pheromones and display pharmacological activities, especially as neuroprotective agents. Therefore, the search for synthetic strategies alternative to traditional chemical synthesis could help designing ecofriendly routes for the preparation of such bioactive compounds. Hence, this work aims at the microwave-assisted enzymatic synthesis of geranyl esters in solvent-free systems. The process variables were optimized for the synthesis of geranyl acetoacetate, achieving 85% conversion after 60 min using a 1:5 substrates molar ratio (ester to geraniol), 80 °C and 8.4% of Lipozyme 435 lipase without removal of the co-produced methanol. On the other hand, a 95% conversion was reached after 30 min using 1:6 substrates molar ratio, 70 °C and 7% lipase in the presence of 5Å molecular sieves for the methanol capture. In addition, the lipase showed good reusability, maintaining the same activity for five reaction cycles. Finally, under the above optimized conditions, other geraniol esters were successfully synthetized such as the geranyl butyrate (98%), geranyl hexanoate (99%), geranyl octanoate (98%), and geranyl ( R )-3-hydroxybutyrate (56%). These results demonstrate the microwave-assisted lipase-catalyzed transesterification in a solvent-free system as an excellent and sustainable catalytic methodology to produce geraniol esters.

Abstract:A radiochemical, propyl 4-[2-(4,6-dimethoxy-2-pyrimidinyloxy)benzylamino]benzoate with all six benzoic ring carbons labeled by 14 C was made from 4-aminobenzoic acid with all six benzoic ring carbons labeled by C-14 through esterification, condensation, reduction and substitution, and purified by preparative HPLC. Its structure was identified by LC-MS and H-1 NMR. HPLC test showed its chemical purity exceeded 98%. HPLC-LSC and TLC-IIA demonstrated that its radiochemical purity exceeded 98% and specific activity was 1.089 +/- 0.011 mCi/mmol. The total chemical yield and radiochemical yield were all 53%.

Abstract:The efficient asymmetric dihydroxylation of 6,7-epoxygeraniol was performed using a chemoenzymatic process employing a yeast epoxide hydrolase (EH) resolution stereoinversion step, followed by stereoretentive chemical hydrolysis of the remaining epoxide to produce (6 R )-6,7-dihydroxygeraniol at unprecedented high enantiometic excess (ee) (>97.5%) and high isolated yield (72 mol % overall yield over 5 steps from commercially available geraniol). The enzymatic process was completed within 2 h at 250 g/L substrate loading reaching > 49.5 mass % conversion of the racemic epoxide. The reaction was self-limiting and furnished both the homochiral (6 R )-triol and the residual (6 R )-epoxide at > 99% ee, due to enantio-inversion of the (6 S )-epoxide. The (6 R )-epoxide was subsequently chemically hydrolysed, without first needing to separate the epoxide and triol products, to afford the desired (6 R )-triol product in >97.5% ee, at multigram scale. This chemoenzymatic procedure offers an excellent alternative to chemical asymmetric epoxidation or dihydroxylation for the production of enantiopure 6,7-dihydroxygeranyl and 6,7-epoxygeranyl type compounds in general. It exemplifies the benefits of using greener EH bioprocesses to produce such compounds in high ee's and high isolated yields.

Abstract:(R)-Citronellal is a key chiral precursor of high-value chemicals, such as the best-selling flavor compound (-)-menthol; however, the conventional synthesis suffers from low yield and unsatisfactory enantioselectivity. In this study, we developed a highly atom-efficient hydrogen-borrowing cascade for the synthesis of (R)-citronellal from geraniol using alcohol dehydrogenase from Escherichia coli K12 (AdhP) and ene-reductase from Saccharomyces cerevisiae YJM1341 (OYE2p). The key rate-limiting enzyme, AdhP, was subjected to structure-guided semirational engineering, and the triple mutant AdhP260T/284A/268P (M3) was obtained that demonstrated a 1.28-fold improvement in catalytic efficiency (kcat/Km) toward geraniol. After optimization of the reaction conditions, the hydrogen-borrowing cascade system achieved the conversion of 23.14 g/L geraniol into (R)-citronellal at a conversion rate of 98.23% with 96.7% ee. This work represents an alternative approach for the biosynthesis of (R)-citronellal without sacrificing a cosubstrate or additional enzymes.
Using a Novel Data-Driven Combinatorial Mutagenesis Strategy to Engineer an Alcohol Dehydrogenase for Efficient Geraniol Synthesis

Abstract:The monoterpene geraniol is widely applied in perfume, food, pharmaceutical, and other industries. However, conventional methods of geraniol production, including chemical synthesis and microbial fermentation, are associated with low selectivity and poor yields. Here, we engineered the medium-chain alcohol dehydrogenase AdhP from Escherichia coli K12 as a biocatalyst for the efficient bioconversion of geranial into geraniol. First, a single point mutation library targeting 12 key residues identified based on computational approaches was constructed, and 24 positive mutants with improved reducing activity toward geranial were obtained. Subsequently, we established a data-driven combinatorial mutagenesis strategy for five rounds of engineering with reference to a mathematical "weighting " principle, yielding the six-point mutant M6, which exhibited 8.5-fold higher catalytic efficiency than the wild type. In silico analysis revealed its enhanced structural stability and shortened critical catalytic distances. Recombinant bacterias co-expressing M6 and glucose dehydrogenase were employed as biocatalysts for the reduction of geranial. As a result, 100.0 g/L of geranial was converted to geraniol in 8 h with a space-time yield of up to 291.3 g.L(-1)d(-1). This study provides a multipoint mutant library construction strategy for the accumulation of beneficial mutations to tailor enzymes and provides a potential biocatalyst for efficient geraniol synthesis.

Abstract:Isoprenoids are a large family of natural products with diverse structures, which allow them to play diverse and important roles in the physiology of plants and animals. They also have important commercial uses as pharmaceuticals, flavouring agents, fragrances, and nutritional supplements. Recently, metabolic engineering has been intensively investigated and emerged as the technology of choice for the production of isoprenoids through microbial fermentation. Isoprenoid biosynthesis typically originates in plants from acetyl‐coA in central carbon metabolism, however, a recent study reported an alternative pathway, the Isopentenol Utilization pathway (IUP), that can provide the building blocks of isoprenoid biosynthesis from affordable C5 substrates. In this work, we expressed the IUP in Escherichia coli to efficiently convert isopentenols into geranate, a valuable isoprenoid compound. We first established a geraniol‐producing strain in E. coli that uses the IUP. Then, we extended the geraniol synthesis pathway to produce geranate through two oxidation reactions catalysed by two alcohol/aldehyde dehydrogenases from Castellaniella defragrans. The geranate titer was further increased by optimizing the expression of the two dehydrogenases and also parameters of the fermentation process. The best strain produced 764 mg/L geranate in 24 h from 2 g/L isopentenols (a mixture of isoprenol and prenol). Abstract:Geraniol, an acyclic monoterpenoid of substantial value extracted from the essential oils of various aromatic plants, holds significant commercial and industrial importance in the realms of food, cosmetics, medicine, and bioenergy. Geraniol synthase, which is responsible for geraniol production, has been identified in only several plant species to date. Here, we present the first cloning and characterization of a geraniol synthase (PgfTPS) from Penicillium griseofulvum. This enzyme demonstrates pronounced specificity in catalyzing the conversion of geranyl diphosphate into geraniol. Moreover, through protein modeling and site-directed mutagenesis, we have identified key active-site residues crucial for the catalytic function of PgfTPS. Finally, we utilized engineered Saccharomyces cerevisiae as a host for PgfTPS expression to facilitate geraniol production. Our findings not only advance the development of efficient biocatalysts for geraniol generation but also establish a fundamental basis for further exploration into fungal monoterpene biosynthesis.

Abstract:Geranyl acetate, a significant natural fragrance compound, is typically available only in minute quantities from plant sources. This study focuses on the biosynthesis of this compound through enzymatic esterification of geraniol with acetic acid, utilizing the environmentally benign and generally safe solvent, acetone. The optimization process initially involved key synthesis variables, including acetic acid and geraniol concentrations, temperature, and enzyme loading. Subsequently, a thermodynamic investigation of the biosynthesis was conducted alongside kinetic elucidation, providing insights into the mechanism of the lipase-mediated esterification, which was revealed to follow a Ping-Pong Bi-Bi mechanism with no apparent inhibition by either substrate. The reaction is endothermic, with calculated changes in enthalpy (ΔH), and entropy, (ΔS) at 66 kJ mol–1 and 217 J mol–1 K–1, respectively. The activation energy, Ea, was determined to be 28 kJ mol−1. Esterification was non-spontaneous below 30 °C but shifted to spontaneous at higher temperatures. These findings hold significance for the development of biocatalytic processes.

Abstract:Objective To prepare(2E,6E)-octadiene-3,7-dimethyldiol-1acetate.Methods The target compound was synthesized by five steps(acetylating,addition-elimination,replacement,oxidation and rearrangement)with geraniol.Results The desired product was obtained in over all yield of 49.7% and its structure was confirmed by 1H-NMR and IR.Conclusions A new route for the synthesis of (2E,6E)-octadiene-3,7-dimethyldiol-1acetate was explored.
Engineering Escherichia coli for high-yield geraniol production with biotransformation of geranyl acetate to geraniol under fed-batch culture

Abstract:BackgroundGeraniol is an acyclic monoterpene alcohol, which exhibits good prospect as a gasoline alternative. Geraniol is naturally encountered in plants at low concentrations and an attractive target for microbial engineering. Geraniol has been heterologously produced in Escherichia coli, but the low titer hinders its industrial applications. Moreover, bioconversion of geraniol by E. coli remains largely unknown.ResultsRecombinant overexpression of Ocimum basilicum geraniol synthase, Abies grandis geranyl diphosphate synthase, and a heterotic mevalonate pathway in E. coli BL21 (DE3) enabled the production of up to 68.6 ± 3 mg/L geraniol in shake flasks. Initial fed-batch fermentation only increased geraniol production to 78.8 mg/L. To further improve the production yield, the fermentation conditions were optimized. Firstly, 81.4 % of volatile geraniol was lost during the first 5 h of fermentation in a solvent-free system. Hence, isopropyl myristate was added to the culture medium to form an aqueous-organic two-phase culture system, which effectively prevented volatilization of geraniol. Secondly, most of geraniol was eventually biotransformed into geranyl acetate by E. coli, thus decreasing geraniol production. For the first time, we revealed the role of acetylesterase (Aes, EC 3.1.1.6) from E. coli in hydrolyzing geranyl acetate to geraniol, and production of geraniol was successfully increased to 2.0 g/L under controlled fermentation conditions.ConclusionsAn efficient geraniol production platform was established by overexpressing several key pathway proteins in engineered E. coli strain combined with a controlled fermentation system. About 2.0 g/L geraniol was obtained using our controllable aqueous-organic two-phase fermentation system, which is the highest yield to date. In addition, the interconversion between geraniol and geranyl acetate by E. coli was first elucidated. This study provided a new and promising strategy for geraniol biosynthesis, which laid a basis for large-scale industrial application.

Abstract:Geraniol is a rose-scented monoterpene with significant commercial and industrial value in medicine, condiments, cosmetics, and bioenergy. Here, we first targeted geraniol as a reporter metabolite and explored the suitability and potential of Candida glycerinogenes as a heterologous host for monoterpenoid production. Subsequently, dual-pathway engineering was employed to improve the production of geraniol with a geraniol titer of 858.4 mg/L. We then applied a synthetic hybrid promoter approach to develop a decane-responsive hybrid promoter based on the native promoter PGAP derived from C. glycerinogenes itself. The hybrid promoter was able to be induced by n-decane with 3.6 times higher transcriptional intensity than the natural promoter PGAP. In particular, the hybrid promoter effectively reduces the conflict between cell growth and product formation in the production of geraniol. Ultimately, 1194.6 mg/L geraniol was obtained at the shake flask level. The strong and tunable decane-responsive hybrid promoter developed in this study provides an important tool for fine regulation of toxic terpenoid production in cells.

Abstract:Geraniol is a valuable monoterpene extensively used in the fragrance, food, and cosmetic industries. Increasing environmental concerns and supply gaps have motivated efforts to advance the microbial production of geraniol from renewable feedstocks. In this study, we first constructed a platform geraniol Escherichia coli strain by bioprospecting the key enzymes geranyl diphosphate synthase (GPPS) and geraniol synthase (GES) and selection of a host cell background. This strategy led to a 46.4-fold increase in geraniol titer to 964.3 mg/L. We propose that the expression level of eukaryotic GES can be further optimized through fusion tag evolution engineering. To this end, we manipulated GES to maximize flux towards the targeted product geraniol from precursor geranyl diphosphate (GPP) via the utilization of fusion tags. Additionally, we developed a high-throughput screening system to monitor fusion tag variants. This common plug-and-play toolbox proved to be a robust approach for systematic modulation of protein expression and can be used to tune biosynthetic metabolic pathways. Finally, by combining a modified E1* fusion tag, we achieved 2124.1 mg/L of geraniol in shake flask cultures, which reached 27.2% of the maximum theoretical yield and was the highest titer ever reported. We propose that this strategy has set a good reference for enhancing a broader range of terpenoid production in microbial cell factories, which might open new possibilities for the bio-production of other valuable chemicals.

Abstract:Natural terpenoids have elaborate structures and various bioactivities, making difficult their synthesis and labeling with isotopes. We report here the enzymatic total synthesis of plant hormone gibberellins (GAs) with recombinant biosynthetic enzymes from stable isotope-labeled acetate. Mevalonate (MVA) is a key intermediate for the terpenoid biosynthetic pathway. 13C-MVA was synthesized from 13C-acetate via acetyl-CoA, using four enzymes or fermentation with a MVA-secreted yeast. The diterpene hydrocarbon, ent-kaurene, was synthesized from 13C-acetate and 13C-MVA with ten and six recombinant enzymes in one test tube, respectively. Four recombinant enzymes, P450 monooxygenases and soluble dioxygenases involved in the GA4 biosynthesis from ent-kaurene via GA12 were prepared in yeast and Escherichia coli. All intermediates and the final product GA4 were uniformly labeled with 13C without dilution by natural abundance when [U-13C2] acetate was used. The 13C-NMR and MS data for [U-13C20] ent-kaurene confirmed 13C-13C coupling, and no dilution with the 12C atom was observed.

Geraniol and geranial dehydrogenases induced in anaerobic monoterpene degradation by Castellaniella defragrans

Abstract:Castellaniella defragrans is a Betaproteobacterium capable of coupling the oxidation of monoterpenes with denitrification. Geraniol dehydrogenase (GeDH) activity was induced during growth with limonene in comparison to growth with acetate. The N-terminal sequence of the purified enzyme directed the cloning of the corresponding open reading frame (ORF), the first bacterial gene for a GeDH (geoA, for geraniol oxidation pathway). The C. defragrans geraniol dehydrogenase is a homodimeric enzyme that affiliates with the zinc-containing benzyl alcohol dehydrogenases in the superfamily of medium-chain-length dehydrogenases/reductases (MDR). The purified enzyme most efficiently catalyzes the oxidation of perillyl alcohol (k(cat)/K(m) = 2.02 × 10(6) M(-1) s(-1)), followed by geraniol (k(cat)/K(m) = 1.57 × 10(6) M(-1) s(-1)). Apparent K(m) values of <10 μM are consistent with an in vivo toxicity of geraniol above 5 μM. In the genetic vicinity of geoA is a putative aldehyde dehydrogenase that was named geoB and identified as a highly abundant protein during growth with phellandrene. Extracts of Escherichia coli expressing geoB demonstrated in vitro a geranial dehydrogenase (GaDH) activity. GaDH activity was independent of coenzyme A. The irreversible formation of geranic acid allows for a metabolic flux from β-myrcene via linalool, geraniol, and geranial to geranic acid.
Clinoptilolite as a natural, active zeolite catalyst for the chemical transformations of geraniol

Abstract:Abstract This work presented the studies with the natural zeolite—clinoptilolite as the catalyst for the isomerization of geraniol. During the research, it turned out that the studied process is much more complicated, and not only isomerization takes place in it, but also dehydration, oxidation, dimerization, cyclization and fragmentation of the carbon chain. Geraniol is an organic raw material which can be obtained not only by a chemical synthesis but also from plants (renewable biomass) by distillation or extraction method, for example a source of geraniol can be a plant—geranium. Before catalytic tests clinoptilolite was characterized by the instrumental methods, such as: XRD, porosity studies—nitrogen adsorption at 77 K, SEM, EDXRF, and FT-IR. Gas chromatography analyses showed that the main products of geraniol isomerization process were 6,11-dimethyl-2,6,10-dodecatrien-1-ol and thumbergol. The selectivity of 6,11-dimethyl-2,6,10-dodecatrien-1-ol and thumbergol depended on the temperature, catalyst content and reaction time. These parameters were changed in the following ranges: 80–150 °C (temperature), 5–15 wt% (catalyst content) and 15–1440 min. (reaction time). The most favorable conditions for 6,11-dimethyl-2,6,10-dodecatrien-1-ol and thumbergol obtaining were: temperature 140 oC, catalyst content 12.5 wt% and reaction time 180 min. At these conditions, the conversion of geraniol amounted to 98 mol%, and the selectivities of 6,11-dimethyl-2,6,10-dodecatrien-1-ol and thumbergol amounted to 14 and 47 mol%, respectively.

Abstract:C-13- and C-14-uniformly labelled catechol Was synthesized from phenol in three steps. Phenol was derivatized with 2-chloro-5-nitrobenzophenone in THF containing NaH, followed by ortho-hydroxylation with 35% aqueous H2O2 in sulphuric acid/glacial acetic acid solution, and by cleavage with piperidine, giving an overall 80% yield with respect to phenol. Quantitative determination of urinary metabolites of geraniol by ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS)

Abstract:Geraniol is a fragrance which occurs in natural terpene oil or is chemically synthesized on a large scale. It is used in a wide variety of consumer products such as perfumes, deodorants, household products and cosmetics. Hence, not only industry workers in the production of geraniol, but also consumers can come into contact with the substance. Human biomonitoring (HBM), i.e. the analytical determination of substances and their metabolites in human biological material, is a key element in the analysis and assessment of the distribution and intensity of occupational and environmental exposure of humans. Therefore, a procedure for the quantitative determination of the urinary metabolites Hildebrandt acid, geranic acid, 3-hydroxycitronellic acid and 8-carboxygeraniol as potential biomarkers of geraniol exposure was developed and validated. The method is based on ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) after enzymatic hydrolysis and liquid-liquid extraction (LLE) of the target analytes. The limit of quantification (LOQ) is 1.5 μg L-1 for 8-carboxygeraniol, 2.7 μg L-1 each for Hildebrandt acid and geranic acid, and 1.8 μg L-1 for 3-hydroxycitronellic acid. The method was applied to urine samples of 41 persons without occupational exposure to geraniol. Hildebrandt acid and geranic acid were detected in all samples, 8-carboxygeraniol in 83% and 3-hydroxycitronellic acid in 81% of the samples. Hildebrandt acid (median: 313 μg L-1, range: 37-1966 μg L-1) was the most abundant metabolite, followed by geranic acid (93 μg L-1; 9-477 μg L-1), 3-hydroxycitronellic acid (18 μg L-1; <LOQ to 70 μg L-1) and 8-carboxygeraniol (9 μg L-1; <LOQ to 46 μg L-1). Hildebrandt acid, geranic acid and 3-hydroxycitronellic acid apparently represent larger relative fractions of the eliminated metabolites, but they are not strictly specific for geraniol since they are metabolites of other terpenes as well, such as citral. In contrast, geraniol seems to be the only parent compound for 8-carboxygeraniol, which makes this metabolite a promising candidate for specific human biomonitoring and risk assessment.

Abstract:Geranylgeraniol (GGOH) is a crucial component in fragrances and essential oils, and a valuable precursor of vitamin E. It is primarily extracted from the oleoresin of Bixa orellana, but is challenged by long plant growth cycles, severe environmental pollution, and low extraction efficiency. Chemically synthesized GGOH typically comprises a mix of isomers, making the separation process both challenging and costly. Advancements in synthetic biology have enabled the construction of microbial cell factories for GGOH production. In this study, Yarrowia lipolytica was engineered to efficiently synthesize GGOH by expressing heterologous phosphatase genes, enhancing precursor supplies of farnesyl diphosphate, geranylgeranyl pyrophosphate, and acetyl-CoA, and downregulating the squalene synthesis pathway by promoter engineering. Additionally, optimizing fermentation conditions and reducing reactive oxygen species significantly increased the GGOH titer to 3346.47 mg/L in a shake flask. To the best of our knowledge, this is the highest reported GGOH titer in shaking flasks to date, setting a new benchmark for terpenoid production.
