Construction of Recombinant Plasmid Pyyvpr of Gene about Receptor of VT Bacteriophage in E.coli

Abstract:The pJP5603 plasmid is a suicide plasmid and contains the Kan resistance cassette, which was digested with Xma I, Hinc Ⅱand the pUC19phiΦRID plasmid contains the vpr gene, digested with Age Ⅰ, Hinc Ⅱto obtain 778bp piece of vpr aim gene. After cohesive ligation, the recombinant plasmid was transformed into CC118λpir. Colonies were selected on Kan plates. A recombinant plasmid, pYYvpr, was confirmed by PCR to amplify vpr gene and digestion with restriction enzyme AccⅠ. Meanwhile, the recombinant plasmid was not obtained from the blunt ligation of the two plasmid digested with single restriction enzyme Hinc Ⅱ. The recombinant suicide plasmid pYYvpr was transformed into the wild type of E.coli strains, MC1061, which possesses the full length vpr gene. After analysis by PCR, an isogenic knockout mutant, named MC1061△vpr, was confirmed.
