Abstract:We have constructed cDNA microarrays from the human testis large insert cDNA library, containing 9216 genes, together with several housekeeping genes. The cDNA microarrays were used to identify gene expression differences between human fetal and adult testes. Of >8700 hybridized clones, 731 exhibited significant differential expression characteristics. About 7500 genes were identified when the same cDNA microarrays were used for hybridization with cDNA probes from mouse testis, with 256 genes having significant differential expression between the age of 1-4 weeks. Among these genes, 101 were identified as critically related to testis development and possibly to spermatogenesis since they were found in both human and mouse testes, and expressed differentially at different stages of testis development. Of the 101 development-related genes, 59 full-length cDNAs have been sequenced previously, while the full-length cDNAs of the other 42 genes have not been published. We have obtained 11 full-length sequences of the 42 genes and deposited them in the GenBank. The conserved testis development-related genes found in both human and mouse testes may include genes that are likely to be involved in testicular functions, especially spermatogenesis, thus providing a basis for further functional characterization of the genes in mouse models.

Abstract:Spermatogenesis is a highly coordinated physiological process that requires the correct expression and functions of thousands of developmentally regulated genes. The regulation of spermatogenesis is not well defined, since majority of the related genes have neither been identified nor fully characterized. Hence, it is meaningful to identify and characterize these genes to reveal the mechanism underlying spermatogenesis. In this study, using digital differential display, we identified a novel human testis-specific gene, testis developmental related gene 1 (TDRG1, GenBank DQ168992), via electronic subtraction of human testis UniGene databases from those of non-reproductive tissues. The transcript of the TDRG1 gene has an open-reading frame that encodes 100 amino acids. We next prepared the anti-TDRG1 monoclonal antibody 10B6 and confirmed that it specifically recognizes an 11-kDa protein in the tissue extracts from an adult human testicular sample (age 31 years) by Western blot analysis. RT-PCR coupled with immunohistochemistry of human tissues demonstrated that TDRG1 is exclusively expressed in the testis but not in any other non-reproductive tissues. TDRG1 is mainly located in spermatogenic cells in seminiferous tubules of adult testis. Furthermore, TDRG1 shows the highest expression level in human post-puberty testis, with the expression levels decreasing afterwards with aging. Importantly, TDRG1 mRNA is undetectable in the fetal testis, as judged by RT-PCR. In conclusion, TDRG1 is a developmentally regulated testicular-specific gene. We suggest that TDRG1, a newly identified testis-specific gene, may play important roles in human spermatogenesis.
Character analysis of a novel testis- specific TSC23 gene in mouse and human testis

Abstract:Objective: To identify novel genes involved in spermatogenesis. Methods: Testes cDNA samples from Balb/C mice of different postnatal days were performed with mouse whole genome Affymetrix Genechip to screen novel testis-specific genes. The tissues distribution of the testis-specific genes in mice organs was identified by RT-PCR. The characteristics of selected genes were analyzed by various bioinformatic tools. Results: A novel testis-specific gene (GenBank Accession No:AK016041) with a differential expression in the developmental stages of testis was identified. The full length cDNA of 803bp contained an open reading frame of 606 bp which encoded a putative protein of 201 amino acids, we named this gene TSC23. RT-PCR analysis revealed that TSC23 mRNA was exclusively expressed in mice and human testis. Cellular mapping prediction of TSC23 indicated that its protein would be expressed in nuclear and two putative domains were signal-peptide and transmembrane regions predicted at the protein site 1-18, 36-58 respectivily. Conclusion: The expression profile of TSC23 in human and mice, indicating TSC23 may be involved in testis spermatogenesis.
Identification of A Novel Testis-Specific Gene in Mice and Its Potential Roles in Spermatogenesis

Abstract:Aim Identification of a novel gene in mouse testis and its relation to spermatogenesis. Methods Genes expressed during different developmental stages of the mouse testis were screened by DNA microarray. The results of chip analysis were authenticated by reverse transcription-polymerase chain reaction (RT-PCR) technique, as well as the tissue distribution of the selected genes. The characteristics of the selected genes were analyzed by bioinformatics tools. Results A novel gene, TSC77, was identified and located at the mouse chromosome 2G1. The full cDNA length of TSC77 was 2280 bp, with a 2046 bp open reading frame encoding a 681 amino acids protein with a predicted molecular weight of 77.17 kDa. The results of subcellular localization of GFP-TSC77 fusion protein indicated TSC77 protein was located in the nucleus of Cos-7 cells. The analysis of multiple amino acid sequence alignment showed that TSC77 protein was highly homologous with the human CAI40813 (C2orf26 gene, 76%), and rat XP_ 230651 (77%). Three putative domains including nicotine amide dinucleotide phosphate (NAD(P))-nitrite reductase (NirB) domain, uncharacterized NAD flavin adenine dinucleotide (FAD)-dependent dehydrogenases (HcaD) domain, and nicotinamide adenine dinucleotide (NADH) dehydrogenase (Ndh) domain were predicted at the protein site 168-309, 187-245, 181-216, respectively. Gene expression anlysis showed that the mouse TSC77 is preferentially expressed in the mouse testis and its expression increased gradually from the day 9 to day 21. Conclusion Based on its expression during mouse development, TSC77 may play an important role during mouse spermatogenesis.

Abstract:Detailed characterization of the gene expression patterns in spermatogonia and primary spermatocytes is critical to understand the processes which occur prior to meiosis during normal spermatogenesis. The genome-wide expression profiles of mouse type B spermatogonia and primary spermatocytes were investigated using the Solexa/Illumina digital gene expression (DGE) system, a tag based high-throughput transcriptome sequencing method, and the developmental processes which occur during early spermatogenesis were systematically analyzed. Gene expression patterns vary significantly between mouse type B spermatogonia and primary spermatocytes. The functional analysis revealed that genes related to junction assembly, regulation of the actin cytoskeleton and pluripotency were most significantly differently expressed. Pathway analysis indicated that the Wnt non-canonical signaling pathway played a central role and interacted with the actin filament organization pathway during the development of spermatogonia. This study provides a foundation for further analysis of the gene expression patterns and signaling pathways which regulate the molecular mechanisms of early spermatogenesis.
NYD-SP15: A Novel Gene Potentially Involved in Regulating Testicular Development and Spermatogenesis

Abstract:By hybridizing human adult testis cDNA microarrays with human adult and embryo testis cDNA probes, we identified a novel human testis gene, NYD-SP15. NYD-SP15 expression was 3.26-fold higher in adult than in fetal testis; however, there was almost no NYD-SP15 expression in the sperm. NYD-SP15 comprises 3364 base pairs, including a 1545 bp open reading frame encoding a 514 amino acid protein possessing 89% sequence identity with the mouse testis homologous protein. NYD-SP15 is located on human chromosome 13q14.2. The deduced structure of the protein contains two dCMP_cyt_deam domains, indicating a potential functional role for zinc ion binding. The gene is expressed variably in a wide range of tissues, with high expression levels in the testis. Sequence analysis revealed that NYD-SP15 is not a highly conserved protein, with its distribution in high-level species such as vertebrates including Homo, Mus, Rattus, and Canis. The results of semiquantitative polymerase chain reaction in mouse testis representing different developmental stages indicate that NYD-SP15 expression was developmentally regulated. These results suggest the putative NYD-SP15 protein may play an important role in testicular development and spermatogenesis and may be an important factor governing male infertility.
Gene expression profile of 2058 spermatogenesis-related genes in mice.

Abstract:The aim of this study was to characterize the gene expression profiles of mouse testis at different stages by performing large-scale complementary DNA (cDNA) analysis using DNA microarrays. The transcription profile of mouse testis was determined by comparing testis samples of mice aged 4, 9, 18, 35, 54 d and 6 months using Affymetrix Genechip mouse microarrays (430 v.2). Of the six comparative pairs of two neighboring spots examined, lots of differently expressed genes existed on day 18 versus day 9 and on day 35 versus day 18, indicating these genes may be involved in unique events during those periods. In mining the Genechip data, the gene expression profiles of 2058 genes represented in a gradually increased manner in the developmental stages of mice testis, and its validity was confirmed by the data of 21 known spermatogenesis-related testis-specific genes in our chip and the results of 10 random sample from 2058 genes produced by the sub-quantitative RT-PCR. So, we called these 2058 genes spermatogenesis-related genes. By informatics analysis, the gene expression clustering, gene ontology, KEGG pathway and domain regions were predicted for describing the potential roles that may play during mouse spermatogenesis. This study provides a molecular basis for the identity of sperm atogenesis-related gene in mouse testis and leads to the elucidation of the molecular events underlying mammalian male reproduction.
Molecular cloning and character analysis of the TSAF76 gene exclusively expressed in mouse testis

Abstract:Objective: To identify genes involved in spermatogenesis and/or testicular development. Methods: Hybridization of mouse testicular DNAchips was performed with testicular tissues cDNA microarray to screen the specific gene expressed in testicular tissue. Differences in expression of the screened gene were identified by reverse transcriptase polymerase chain reaction (RT-PCR). Results: By analysing the hybridization signals, a novel gene TSAF76 with a differential expression in 4 days and 18 days mouse testis was identified, and it was registered in GenBank (NO:AF503943).The full length cDNA of 1052 bp contained an open reading frame of 474 bp which encoded a putative protein of 157 amino acids. RT-PCR analyses revealed that TSAF76 mRNA was exclusively expressed in testicular tissues during spermatogenesis. Conclusion: TSAF76 gene functions as a germ cell specific transcription factor that maybe plays important roles in mouse spermatogenesis.
Molecular Cloning and Primary Functional Analysis of a Novel Human Testis-specific Gene

Abstract:In this study,a new data mining tool called Digital Differential Display (DDD) from the NCBI was used to predict testis-specific expressed genes from the expressed sequence tag (EST) database.DDD(digital differential display) was performed between nine testis libraries and seventy libraries derived from other tissues.We identified a new contig of ESTs (HS.326528) which was from testis libraries.To validate the use of bioinformatic approaches in gene discovery,the ESTs (HS.326528),which were predicted to be testis-specific,were chosen for further study.Reverse Transcriptase-Polymerase Chain Reaction (RT-PCR) analysis of matched sets of cDNAs from testis and other tissues indicated that the ESTs were specifically expressed in testis.This result was further validated by multi-tissue Northern blot.The full-length cDNA encompassing the entire open reading frame was cloned and,in view of its apparent specificity to testis,the gene was termed homo sapiens spermatogenesis-related gene 8--SRG8 (GenBank accession number: AY489187).The gene whose full cDNA length is 1 044 bp containing 3 exons and 2 introns is located in human chromosome 15q26.2,the cDNA encodes a novel protein of 105 amino acides with a theoretical molecular weight of 11.7 kD and isoelectric point of 10.09 which shares no significant homology with any known proteins in database.Real time PCR analysis of testis of different developmental periods revealed that SRG8 gene is significantly expressed in adult testis .The green fluorescent protein produced by pEGFP-C3/SRG8 was detected in the nucleus of HeLa cells after 24 h post-transfection.Cell cycle analysis showed that SRG8 can accelerate HeLa cells to traverse the S-phase and enter the G2-phase compared with the control without transfection of SRG8,which suggested that this gene plays an important role in the development of testis.The discovery of SRG8 showed that DDD combined with experiments is a feasible,time-saving strategy to identify new candidate genes for testis-specific development

NYD-SP16, a Novel Gene Associated with Spermatogenesis of Human Testis

Abstract:By hybridizing human adult testis cDNA microarrays with human adult and embryo testis cDNA probes, a novel human testis gene NYD-SP16 was identified. NYD-SP16 expression was 6.44-fold higher in adult testis than in fetal testis. NYD-SP16 contains 1595 base pairs (bp) and a 762-bp open reading frame encoding a 254-amino acid protein with 73% amino acid sequence identity with the mouse testis homologous protein. The NYD-SP16 gene was localized to human chromosome 5q14. The deduced structure of the NYD-SP16 protein contains one transmembrane domain, which was confirmed by GFP/NYD-SP16 fusion protein expression in the cytomembrane of the transfected human choriocarcinoma JAR cells, suggesting that it is a transmembrane protein. Multiple tissue distribution indicated that NYD-SP16 mRNA is highly expressed in the testes and pancreas, with little or no expression elsewhere. Further analysis of abnormal expression in infertile male patients revealed complete absence of NYD-SP16 in the testes of patients with Sertoli-cell-only syndrome and variable expression in patients with spermatogenic arrest. Homologous gene expression in mouse testis was confirmed in spermatogenic cells by in situ hybridization. The results of cDNA microarray, in situ hybridization, and semi-quantitative polymerase chain reaction in mouse testis of different stages indicated that NYD-SP16 expression is developmentally regulated. These results suggest that the putative NYD-SP16 protein may play an important role in testicular development/spermatogenesis and may be an important factor in male infertility.
Analysis of Gene Expression Patterns with Cdna Microarray During Late Stage of Spermatogenesis in Mice

Abstract:The differentiation process of round spermat-ids to spermatozoa during the late stage of spermatogenesis is called spermiogenesis. To explore spermiogenesis-related genes, cDNA microarray was used to study expression patterns of 1176 genes in pachytene spermatocytes, round spermatids and elongating spermatids of Balb/c mice. The results showed that 208 genes were detected in all the three cell types. Most of them were down-regulated from pachytene spermatocytes to round spermatids and elongating spermatids. However, up-regulation of 7 genes expression in round spermatids and 3 genes in elongating spermatids were found. Expression of 7 differentially expressed genes in cDNA arrays was further confirmed by semi-quantitative RT-PCR study. The RT-PCR results indicated that the expression of 6 genes was consistent with that in cDNA arrays, only one gene did not show differential expression by RT-PCR. These results may provide important clues for studying of expression, regulation, and function of spermiogene

Abstract:Objective To analyze the expression characteristics of CatSper gene family in human and mouse tissues.Methods Testis cDNA samples from Balb/C mice of different postnatal days were performed with mouse whole genome Affymetrix Genechip to screen differentially expressed genes.CatSper gene family screened by Genechip were further analysed by RT-PCR for their stage-specific and tissue-specific expression characteristics.Results The distinct expression of CatSper1,CatSper2 and CatSper3 in developmental mouse testis was stage-specific.The abundance of CatSper gene family was higher in testis than any other tissues that we had detected.And CatSper1 and CatSper4 were exclusively expressed in human testis.Conclusion CatSper gene family whose expression were Testis-specific or testis-enriched may play a vital role in human and mouse spermatogenesis.
Identification of a Novel Human Zinc Finger Protein Gene ZNF474 by Digital Differential Display

Abstract:This study was designed to identify a novel testis overexpressed gene related to spermatogenesis by a new strategy of digital differential display (DDD). Based on the generation of expressed sequenced tags ( ESTs), comparing the testis libraries with other tissue or cell line libraries by the DDD program, we identified a new contig of the ESTs which were derived from testis libraries and represented a novel gene. A full-length cDNA sequence of the new gene named ZNF474 ( GeneBank accession number AY461732) by the HUGO Gene Nomenclature Committee in human testis was identified. ZNF474 was 1972 bp in length, located in chromosome 5q23.2. The sequence of the opening reading frame was 377 bp ～ 1471 bp, as was confirmed by RT-PCR and sequencing. The cDNA encodes a novel protein of 364amino acids protein that was 66 % identical to a mouse homologue. Northern blot analyses revealed that ZNF474 mRNA was exclusively expressed in testis and ovary and has one transcript. In situ hybridization revealed that ZNF474 high expressed in adult testis and seminoma. The brown granules of hybridization signal were found in spermatocyte, spermatids and spermatozoa, no expression in sertoli's cells and leydig's cells. We hypothesize that ZNF474 functions as a germ cell-specific transcription factor that plays important roles in spermatid differentiation and oocyte development.
Identification and Expression of a Novel Human Testis-Specific Gene by Digital Differential Display

Abstract:Background Evidence for the importance of genetic factors in male infertility is accumulating. This study was designed to identify a novel testis-specific gene related to spermatogenesis by a new strategy of digital differential display (DDD).Methods Based on the generation of expressed sequenced tags (ESTs), comparing the testis libraries with other tissue or cell line libraries by the DDD program, we identified a new contig of the ESTs which were derived from testis libraries and represented a novel gene. Multi-tissue RT-PCR was performed to analyse its tissue-specific expression. The full-length cDNA of the new gene was obtained using the BLAST program. Sequencing was performed and the result was analysed. Semi-quantitative RT-PCR and Northern blot analyseis of mRNA from differential normal tissues were performed to clarify the expression pattern of the new gene. The sequence of the opening reading frame was integrated into the pQE-30 vector expressed in Escherichia coil strain M15(pREP4). With IPTG induction, the target protein was detected.Conclusions DDD can be confirmed by SPATA12 as a novel computational biology-based approach for identification of the testis-specific expression genes. SPATA12 may function as a testicular germ cell associated gene that plays some roles in spermatogenesis. Moreover, a great amount of SPATA12 protein could be obtained by the gene recombination technique, thus providing a reliable foundation for investigating the biological function of this new protein.

The result of subcellular localization of GFP-TSCPA fusion protein in Cos-7 cells showed that TSCPA protein was expressed in nuclei. RT-PCR analysis revealed that TSCPA was expressed specifically in mouse and human testis. TSCPA gene was expressed weakly in 21-day-old mouse testis and the expression was increased gradually from 38th day to 6th month of mouse testes. No expression of hTSCPA was found in cryptorchidism and Sertoli-cell-only syndrome patients. It was concluded that the expression profile of TSCPA in human and mice indicated that TSCPA might play an important role in spermatogenesis.

Abstract:Testis-specific genes are essential for spermatogenesis in mammalian male reproduction. We have identified a novel gene, Tsc21, exclusively expressed in mice and human testes from the results of the Affymetrix Genechip analysis in the six developmental stages of testis of postnatal Balb/C mice. The full cDNA length of Tsc21 was 810 bp, with a 543 bp open reading frame encoding a 180 amino acids protein with a predicted molecular weight of 21.040 kDa. A Blast search in the mouse genome database localized the Tsc21 gene to mice chromosome 6C3. Multiple amino acid sequence alignment of human, mouse, and rat homologous genes showed that mice Tsc21 protein was highly homologous with the human Tsc21 gene (70%) and rat Tsc21 gene (86%). The results of reverse transcriptase-polymerase chain reaction analysis showed that the mice Tsc21 is exclusively expressed in the testis and epididymis of mice, and its expression is only detected after the mice is 35 days old. Human Tsc21 is also exclusively expressed in testis of human. Considering the expression profile Tsc21 in mice and human, we propose that Tsc21 may play a role during mammalian male spermatogenesis. Our study should be a basis for function characterization of the Tsc21 gene, leading to the elucidation of the molecular events underlying mammalian male reproduction.

Abstract:To identify genes that involved in spermatogenesis. In order to screen the testis-specific genes, testes cDNA samples from BALB/c mice of different postnatal days (days 4, 9, 18, 35, 54 and 6 months) were performed with mouse whole genome Affymetrix chip. The characteristics of the selected gene were analyzed by various bioinformatic tools. The expression profile of the selected gene was identified by RT-PCR. By analyzing the hybridization signals, a gene with a differential expression in the developmental stages of testis was identified. This gene was designated as TSF22.The full length cDNA of 1 597 bp contained an open reading frame of 570 bp which encoded a putative protein of 190 amino acids and a molecular weight of 22.106 kD. RT-PCR analysis revealed that TSF22 mRNA was exclusively expressed in mice testis. TSF22, functions as a testis-specific transcription factor, may play important roles during spermatogenesis.
Gene screening and expressing analysis of 2 058 spermatogenesis-related genes in mice

Abstract:Objective To screen age-dependent mouse spermatogenesis candidate genes at different growth stages of mice. Methods Differential expression profiles of mouse testis genome were comparatively analyzed at aged 4, 9, 18, 35, and 54 days and 6 months using Affymetrix Genechip mouse microarrays. Results There were many differential expression genes between at aged 9 days and at aged 18 days, as well as between at aged18 days and at aged 35 days. By mining the Genechip data, 2 058 genes were found to be gradually increased during the developmental stages of mouse testis. Among these differential genes, 10 differential genes were randomly selected and validated by RT-PCR. Twenty one spermatogenesis-related genes reported in other studies were also found in these 2 058 genes. By bioinformation analysis, gene expression clustering and gene ontology were made to predicte the potential roles of these genes during mouse spermatogenesis. Conelusion The identified spermatogenesis-associated genes will provide some experimental evidences for elucidation of the molecular events mouse spermatogenesis.

Abstract:<h2 class="section-title u-h3 u-margin-l-top u-margin-xs-bottom">Summary</h2><p>Spermatogenesis is a highly regulated process that produces sperm to transmit genetic information to the next generation. Although extensively studied in mice, our current understanding of primate spermatogenesis is limited to populations defined by state-specific markers from rodent data. As between-species differences have been reported in the duration and differentiation hierarchy of this process, it remains unclear how molecular markers and cell states are conserved or have diverged from mice to man. To address this challenge, we employ single-cell RNA sequencing to identify transcriptional signatures of major germ and somatic cell types of the testes in human, macaque, and mice. This approach reveals similarities and differences in expression throughout spermatogenesis, including the stem/progenitor pool of spermatogonia, markers of differentiation, potential regulators of meiosis, RNA turnover during spermatid differentiation, and germ cell-soma communication. These datasets provide a rich foundation for future targeted mechanistic studies of primate germ cell development and <em>in vitro</em> gametogenesis.</p>
