Abstract:Advancements in gene transfer technology have indeed opened up exciting possibilities for more effectively manipulating the genetic makeup of live organisms, ranging from microorganisms to plants and animals. Direct and indirect transformations are the two basic types of gene transfer techniques. Indirect method comprises Agrobacterium mediated method as it involves intermediate host between gene of interest and target and this method is most opted one out of all present. Direct gene transformation methods, on the other hand, do not involve the use of an intermediate host organism. Instead, they rely on physical means to transfer genes between cells. Biolistic transformation uses high-velocity particles to deliver DNA into target cells, while microinjection and macroinjection involve the direct injection of DNA into cells. Protoplast fusion combines the genetic material of two different cells by fusing their protoplasts. Natural methods for gene transfer encompass mechanisms that occur naturally in various organism, includes transposition, conjugation, phage and retroviral transductions and bacterial transformation. Chemical techniques utilize chemical agents to facilitate gene transfer, such as calcium phosphate-mediated transformation, polyethylene glycol (PEG)-mediated transformation, DEAE (Diethylethanolamine)-Dextran-mediated transformation. Genes can be also being transferred using electrical techniques such as electroporation and electrofusion. Crop improvement and trait improvement are now being hastened by the fast-rising number of sequenced plant genomes, information from functional genomics data to understand gene function, innovative gene cloning, and tissue culture techniques. Despite being indispensable, its progress is still hindered by the fact that many plant species and agricultural genotypes exhibit low transformability or are resistant to established tissue culture and regeneration conditions. Here, we review the techniques employed in plant transformation and provide a concise overview of their evolution in agricultural crops, from their first inception to present time.

Abstract:Transient gene expression in protoplasts, which has been used in several plant species, is an important and versatile tool for rapid functional gene analysis, protein subcellular localization, and biochemical manipulations. This unit describes transient gene expression by electroporation of DNA into protoplasts of Arabidopsis or tobacco suspension-cultured cells and by polyethylene glycol (PEG)-mediated DNA transformation into protoplasts derived from rice leaf sheaths. PEG-mediated DNA transformation for transient gene expression in rice protoplasts in suspension culture is also described as an alternative technique. Methods for collecting intracellular and secreted proteins are also provided.

Abstract:The pollen-mediated transformation approach could not only circumvent the tedious tissue culture procedures like in Agrobacterium infection, but also circumvent the incertitude of the pollen tube path-way. So,its transformants combined from the transformed pollen and eggs would not be chimera.At last this approach is simple, easy to operate, and could be widely used in practice. For the transformation happened in the pollen culture medium, the solution with a suitable solute has to be selected. This study showed the medium consisting of (0.1%) H_3BO_3 、(0.3%) Ca(NO_3)_2、(0.2%) MgSO_4·7H_2O、(0.1%) KNO_3 and 45% sucrose could maintain normal turgidity of the pollens and facilitate them to be germinated. There are more than 95% pollens don't dispose in this medium and the germinating rate was up to 58.5%. A transient GUS expression assay and the exploded pollen rate proved that the transformation method with 1‰ Tween-20 in the medium using vacuum-infiltration was a better method. Finally, four transformed seedlings were confirmed by Kanamycin selection, GUS expression and Northern blot. Therefore this transformation approach is available.

Abstract:In genetics and molecular biology Gene transformation is a gene alteration technique that involves the introduction and expression of a foreign gene into the host organism. There are many gene transformation methods like particle bombardment electroporation micro-injection, (PEG), for different biotechnological experiments But Plant gene transformation is a widely used procedure for obtaining transgenic plants and plant models to understand gene functions. Agrobacterium tumefaciens is a natural genetic engineer which is rod-shaped gram-negative soil-born barteri. Initially Agrobacterium was utilized to transform only dicot plants but over the year’s modification in plant transformation protocol. It was now utilized in monocot plants as well as in fruits plants too. Agrobacterium tumefaciens inserts its (DNA), (Transfer DNA-T-DNA), into the host plant. The transmitted (DNA), is randomly integrated into the host cell's genetic material inside the infected plant cell nucleus. Alternatively bacterial DNA, can transiently remain in the nucleus without integrating into the genome but it still replicates alongside the plant genome, using its machinery and expressing its genes to make separate gene products. Besides the traditional method new research has also been done to transform the plants through agrobacterium. Various methods have been developed to transform monocotyledonous plants such as wheat maize rice and fruity plants. Generally dicotyledonous plants can be transformed by the traditional method of agrobacterium but various methods have also been developed for dicots for various applications. Here, we have taken an example of a tobacco plant (Nicotiana tabacum), transformed with different methods.

Abstract:Genetic transformation is an important strategy for enhancing plant biomass or resistance in response to adverse environments and population growth by imparting desirable genetic characteristics. Research on plant genetic transformation technology can promote the functional analysis of plant genes, the utilization of excellent traits, and precise breeding. Various technologies of genetic transformation have been continuously discovered and developed for convenient manipulation and high efficiency, mainly involving the delivery of exogenous genes and regeneration of transformed plants. Here, currently developed genetic transformation technologies were expounded and compared. Agrobacterium-mediated gene delivery methods are commonly used as direct genetic transformation, as well as external force-mediated ways such as particle bombardment, electroporation, silicon carbide whiskers, and pollen tubes as indirect ones. The regeneration of transformed plants usually involves the de novo organogenesis or somatic embryogenesis pathway of the explants. Ectopic expression of morphogenetic transcription factors (Bbm, Wus2, and GRF-GIF) can significantly improve plant regeneration efficiency and enable the transformation of some hard-to-transform plant genotypes. Meanwhile, some limitations in these gene transfer methods were compared including genotype dependence, low transformation efficiency, and plant tissue damage, and recently developed flexible approaches for plant genotype transformation are discussed regarding how gene delivery and regeneration strategies can be optimized to overcome species and genotype dependence. This review summarizes the principles of various techniques for plant genetic transformation and discusses their application scope and limiting factors, which can provide a reference for plant transgenic breeding.

Abstract:For years, isolation of foreign genes from one plant and transferring them to another and then observing effects of new genes in transferred plants has only been a dream for a plant biologist. Today, many commercially important species are routinely transformed by different biotechnological methods. Methods available for plant transformation are arranged in three main groups: using biological vectors (virus- or bacteria-mediated transformation), direct DNA transfer techniques (chemical-, electrical-, or laser-induced permeability of protoplasts or cells) and non-biological vector systems (microprojectiles, microinjection or liposome fusion). Today in many countries a number of transgenic important crops such as soybean, maize, cotton, canola, sugarbeet, sugarcane and alfalfa are available and the mostly preferred method is Agrobacterium-mediated transformation. In this chapter, some information about this important bacterium and mechanisms of Agrobacterium-mediated gene transfer are presented.

Abstract:Gene transformation can be done in direct and indirect (Agrobacterium-mediated) ways. The most efficient method of gene transformation to date is Agrobacterium-mediated method. The main problem of Agrobacterium-method is that some plant species and mutant lines are recalcitrant to regeneration. Requirements for sterile conditions for plant regeneration are another problem of Agrobacterium-mediated transformation. Development of genotype-independent gene transformation method is of great interest in many plants. Some tissue culture-independent Agrobacterium-mediated gene transformation methods are reported in individual plants and crops. Generally, these methods are called in planta gene transformation. In planta transformation methods are free from somaclonal variation and easier, quicker, and simpler than tissue culture-based transformation methods. Vacuum infiltration, injection of Agrobacterium culture to plant tissues, pollen-tube pathway, floral dip and floral spray are the main methods of in planta transformation. Each of these methods has its own advantages and disadvantages. Simplicity and reliability are the primary reasons for the popularity of the in planta methods. These methods are much quicker than regular tissue culture-based Agrobacterium-mediated gene transformation and success can be achieved by non-experts. In the present review, we highlight all methods of in planta transformation comparing them with regular tissue culture-based Agrobacterium-mediated transformation methods and then recently successful transformations using these methods are presented.

Abstract:Gene transfer technology in crop plants has tremendous potential to introduce newer and better traits through development of transgenics and broaden the genetic base of crop plants by transferring genes from novel sources overcoming the species and genus barriers. Nevertheless, development of efficient transformation systems remains a prerequisite and might involve many years of exhaustive research. This chapter overviews the different methods of alien gene transfer through genetic transformation and factors affecting efficient transformation across different crop species. A comparative study on Agrobacterium and biolistics-mediated transformation including methods for production of marker-free transgenics are described in detail. Addressing the growing concerns over the biosafety issue constraining wider application of GM products in agriculture this chapter also focuses on improved methods of choice with respect to a crop family and also deals with future strategies which can help in further exploiting the existing technologies to develop improved crop varieties which can help to combat poverty, hunger and global agro-climatic changes.

Abstract:Great progress has been made in recent years in studies on the mechanism of Agrobacterium-mediated transformation and its application. Many details of the key molecular events within the bacterial cells involved in T-DNA transfer have been elucidated, and it is notable that some plant factors which were elusive before are purified and characterized. Vast kinds of species, which were either recalcitrant to or not included in the host range of Agrobacterium, can' now be transformed by this bacterium, and they include the very important cereal species, gymnosperms, yeast and many filamentous fungi. The simple in vivo transformation of tissue in intact plants and the 'agrolistic' methods to transform recalcitrant plants are the two novel technical achievements. Combined with other powerful techniques such as bacterial artificial chromosome, very large DNA fragment can be transformed into the plant genome by Agrobacterium. Abstract:Agrobacterium tumefaciens -mediated plant transformation is the most dominant technique for the transformation of plants. It is used to transform monocotyledonous and dicotyledonous plants. A. tumefaciens apply for stable and transient transformation, random and targeted integration of foreign genes, as well as genome editing of plants. The Advantages of this method include cheapness, uncomplicated operation, high reproducibility, a low copy number of integrated transgenes, and the possibility of transferring larger DNA fragments. Engineered endonucleases such as CRISPR/Cas9 systems, TALENs, and ZFNs can be delivered with this method. Nowadays, Agrobacterium-mediated transformation is used for the Knock in, Knock down, and Knock out of genes. The transformation effectiveness of this method is not always desirable. Researchers applied various strategies to improve the effectiveness of this method. Here, a general overview of the characteristics and mechanism of gene transfer with Agrobacterium is presented. Advantages, updated data on the factors involved in optimizing this method, and other useful materials that lead to maximum exploitation as well as overcoming obstacles of this method are discussed. Moreover, the application of this method in the generation of genetically edited plants is stated. This review can help researchers to establish a rapid and highly effective Agrobacterium-mediated transformation protocol for any plant species.

Abstract:The most widely used method for the introduction of new genes into plants is based on the natural DNA transfer capacity of Agrobacterium tumefaciens. Over the past decades, intensive studies on the mechanism of T-DNA transfer have resulted in a reasonably detailed insight into this process. This research revealed that the genetic transformation of plant cells by Agrobacterium evolved by adaptation of processes that existed long before the evolution of the plant–Agrobacterium association. This conclusion holds true for the two-component signal transduction system leading to induction of the virulence genes and for the mechanism responsible for T-DNA transfer to plant cells, which is very similar to that of bacterial conjugation. Integration of the T-DNA in the plant chromosomes is accomplished by the plant’s illegitimate recombination system, but with the help of agrobacterial proteins, which enhance the precision and the efficiency of the integration process. This chapter starts with an overview of the current knowledge on the molecular basis of Agrobacterium-mediated plant transformation. The main emphasis is on the role of the virulence proteins and on recent insights into the later steps of the transformation process. Then, a summary is given on the characteristics of the T-DNA inserted into the plant nuclear DNA. The chapter concludes with a critical assessment of the different approaches that have been used to exploit Agrobacterium tumefaciens, a natural “engineer”, for the production of transgenic plants.

Abstract:Plant genetic transformation heavily relies on the bacterial pathogen Agrobacterium tumefaciens as a powerful tool to deliver genes of interest into a host plant. Inside the plant nucleus, the transferred DNA is capable of integrating into the plant genome for inheritance to the next generation (i.e. stable transformation). Alternatively, the foreign DNA can transiently remain in the nucleus without integrating into the genome but still be transcribed to produce desirable gene products (i.e. transient transformation). From the discovery of A. tumefaciens to its wide application in plant biotechnology, numerous aspects of the interaction between A. tumefaciens and plants have been elucidated. This article aims to provide a comprehensive review of the biology and the applications of Agrobacterium-mediated plant transformation, which may be useful for both microbiologists and plant biologists who desire a better understanding of plant transformation, protein expression in plants, and plant-microbe interaction.

Alternative methods of plant transformation--a short review

Abstract:Several methods of transformation are currently available for delivering exogenous DNA to plant cells. Agrobacterium-mediated transformation, microprojectile bombardment and direct protoplast transformation are routinely used today. However, each of them has certain disadvantages, which led to research into the development of novel alternative systems such as infiltration, electroporation of cells and tissues, electrophoresis of embryos, microinjection, pollen-tube pathway, silicon carbide- and liposome-mediated transformation. The low efficiency of transformation is considered to be the main reason for the limited popularity of the alternative transformation methods, other than infiltration and silicon carbide-mediated transformation, which seem to be the most promising ones for practice.
Combination of Particle Bombardment-mediated and Agrobacterium-mediated Transformation Methods in Tea Plant

Abstract:A microprojection protocol using particle delivery system and GUS report gene was developed for tea plant (Camellia sinensis) callus. Comparison of several pre-bombardment culture media showed the highest transient gene expression in the media with PVP. Instead of making contribution to improving GUS expression, osmotic pre-treatment would cause the target tissue having difficulty in regeneration. High transformation rate was obtained using 125 μg tungsten coated with 0.25 μg DNA per shot. Bombarding tea callus at 7 MPa with a target distance of 5 cm and one time was suitable for both GUS activity and regeneration. Several transformation methods, including Agrobacterium-mediated transformation(AGR), particle bombardment (BOM), and combination of the two methods (BTA, BPA and BOA), have been tested and compared with each other. Based on the results of GUS expression and survival rates of the resistant calli, the method combining Agrobacterium and particle bombardment systems is suitable to apply for tea plant transfortion.

Abstract:The transformation effects of BADH gene (encod ing betaine aldehyde dehydrogenase) into rice by particle bombarment and Agr obacterium tumefaciens mediated were compared in transgenic plant number, t ransformation efficiency, rate of PCR positive plantlets, foreign gene copies and their expression activity, and seed setting rate of T 0 generation. Resu lts showed that transformation efficiency and rate of PCR positive plantlets by Agrobacterium tumefaciens mediated were higher than those by particle bombardment method. Moreover, the performance in seed setting rate of T 0 ge neration and some other agronomic traits of transgenic rice obtained from A grobacterium tumefaciens mediated gene transfer system were better compared w ith particle bombarment methodology.

Despite the popularity of the method, low efficiency of transformation is a major challenge for scientists. Abstract:Plant genetic engineering is essential for improving crop yield, quality, and resistance to abiotic/biotic stresses for sustainable agriculture. Agrobacterium-, biolistic bombardment-, electroporation-, and poly(ethylene glycol) (PEG)-mediated genetic-transformation systems are extensively used in plant genetic engineering. However, these systems have limitations, including species dependency, destruction of plant tissues, low transformation efficiency, and high cost. Recently, nanotechnology-based gene-delivery methods have been developed for plant genetic transformation. This nanostrategy shows excellent transformation efficiency, good biocompatibility, adequate protection of exogenous nucleic acids, and the potential for plant regeneration. However, the nanomaterial-mediated gene-delivery system in plants is still in its infancy, and there are many challenges for its broad applications. Herein, the conventional genetic transformation techniques used in plants are briefly discussed. After that, the progress in the development of nanomaterial-based gene-delivery systems is considered. CRISPR-Cas-mediated genome editing and its combined applications with plant nanotechnology are also discussed. The conceptual innovations, methods, and practical applications of nanomaterial-mediated genetic transformation summarized herein will be beneficial for promoting plant genetic engineering in modern agriculture.

Abstract:The ability to efficiently genetically modify plant species is crucial, driving the need for innovative technologies in plant biotechnology. Existing plant genetic transformation systems include Agrobacterium -mediated transformation, biolistics, protoplast-based methods, and nanoparticle techniques. Despite these diverse methods, many species exhibit resistance to transformation, limiting the applicability of most published methods to specific species or genotypes. Tissue culture remains a significant barrier for most species, although other barriers exist. These include the infection and regeneration stages in Agrobacterium, cell death and genomic instability in biolistics, the creation and regeneration of protoplasts for protoplast-based methods, and the difficulty of achieving stable transformation with nanoparticles. To develop species-independent transformation methods, it is essential to address these transformation bottlenecks. This review examines recent advancements in plant biotechnology, highlighting both new and existing techniques that have improved the success rates of plant transformations. Additionally, several newly emerged plant model systems that have benefited from these technological advancements are also discussed.
