Abstract:The first Lewis acid catalyzed asymmetric Friedel-Crafts alkylation reaction of ortho-hydroxybenzyl alcohols with C3-substituted indoles is described. A chiral N,N'-dioxide Sc(OTf)3 complex served not only to promote formation of ortho-quinone methides (o-QMs) in situ but also induced the asymmetry of the reaction. This methodology enables a novel activation of ortho-hydroxybenzyl alcohols, thus affording the desired chiral diarylindol-2-ylmethanes in up to 99 % yield and 99 % ee. A range of functional groups were also tolerated under the mild reaction conditions. Moreover, this strategy gives concise access to enantioenriched indole-fused benzoxocines.

Abstract:An inexpensive and environmentally friendly FeCl3 catalyzed Friedel-Crafts alkylation of alpha-hydroxy ketene dithioacetals with indoles including free-NH indoles and N or C(2)-substituted indoles has been developed. The reaction was efficiently performed in the presence of FeCl3 (2.5 mol%) at room temperature (25 degrees C) in CH2Cl2, affording alpha-indolyl ketene dithioacetals in excellent yields. The procedure was characterized by lower amount of inexpensive catalyst, mild reaction condition, environmentally benign and similicity.

Abstract:In the past two decades, substantial advances have been made on the asymmetric alkyne functionalization by the activation of inert alkynes. However, these asymmetric transformations have so far been mostly limited to transition metal catalysis, and chiral Bronsted acid-catalyzed examples are rarely explored. Here, we report a chiral Bronsted acid-catalyzed dearomatization reaction of phenol- and indole-tethered homopropargyl amines, allowing the practical and atom-economical synthesis of a diverse array of valuable fused polycyclic enones and indolines bearing a chiral quaternary carbon stereocenter and two contiguous stereogenic centers in moderate to good yields with excellent diastereoselectivities and generally excellent enantioselectivities (up to >99% enantiomeric excess). This protocol demonstrates Bronsted acid-catalyzed asymmetric dearomatizations via vinylidene-quinone methides.

Abstract:Over the course of the present studies, a series of optically pure phosphines functionalized by chiral aziridines was synthesized in reasonable/good chemical yields. Their catalytic activity was checked in the enantioselective Friedel–Crafts alkylation of indoles by β-nitrostyrene in the presence of a copper(I) trifluoromethanesulfonate benzene complex. The corresponding Friedel–Crafts products were achieved efficiently in terms of chemical yield and enantioselectivity (up to 85% in some cases).

Abstract:A cooperative catalytic system established by the combination of an iron salt and a chiral Bronsted acid has proven to be effective in the asymmetric Friedel-Crafts alkylation of indoles with beta-aryl alpha'-hydroxy enones. Good to excellent yields and enatioselectivities were observed for a variety of alpha'-hydroxy enones and indoles, particularly for the beta-aryl alpha'-hydroxy enones bearing an electron-with-drawing group at the para position of the phenyl ring (up to 90% yield and 91% ee). The proton of the chiral Bronsted acid, the Lewis acid activation site, as well as the inherent basic site for the hydrogen-bonding interaction of the Bronsted acid are responsible for the high catalytic activities and enantioselectivities of the title reaction. A possible reaction mechanism was proposed. The key catalytic species in the catalytic system, the phosphate salt of Fe-III, which was thought to be responsible for the high activity and good enantioselectivity, was then confirmed by ESIMS studies.

Abstract:The synthesis of diverse products from the same starting materials is always attractive in organic chemistry. Here, a palladium-catalyzed substrate-controlled regioselective functionalization of unactivated alkenes with trifluoroacetimidoyl chlorides has been developed, which provides a direct but controllable access to a variety of structurally diverse trifluoromethyl-containing indoles and indolines. In more detail, with respect to γ,δ-alkenes, 1,1-geminal difunctionalization of unactivated alkenes with trifluoroacetimidoyl chloride enables the [4 + 1] annulation to produce indoles; as for β,γ-alkenes, a [3 + 2] heteroannulation with the hydrolysis product of trifluoroacetimidoyl chloride through 1,2-vicinal difunctionalization of alkenes occurs to deliver indoline products. The structure of alkene substrates differentiates the regioselectivity of the reaction.

Abstract:Diarylethenes (DAEs) with a bromine atom at the ring-closing position catalyze C–C bonding reactions induced by UV or sunlight. Upon photo-irradiation, bromodiarylethenes undergo 6π-electrocyclization (6π-EC), followed by the release of an acid species that catalyzes the double Friedel–Crafts addition of indoles to aldehydes and isatins to form the corresponding triarylmethanes and 3,3′-diarylindolin-2-ones. This protocol is applicable to a wide spectrum of aldehydes and isatins, as well as chalcones as electrophiles. Acid or oxidant-sensitive functional groups, such as ferrocene, 4-methoxyphenyl, thiophene, pyrrole are tolerated. Mechanistic studies show that light is needed to initiate the reaction. Abstract:Asymmetric synthesis of enantioenriched C3-arylated oxindoles bearing tetrasubstituted stereocenter has been achieved using chiral N,N′-dioxide/transition metal complex which promote Friedel–Crafts alkylation of 3-bromo-3-substituted oxindoles with arenes and heteroarenes i.e., anisole derivatives, furans, pyrroles, 2-methoxythiophene, anilines, and phenols. A series of 3-aryl-indolinones with quaternary stereocenters have been obtained up to 99% yield, 99% ee. In addition, the rationale of the reactivity is based on the nucleophilicity parameters of arenes and heteroarenes were predicted from Mayr's Database. Also, a gram-scale synthesis has been achieved and this method has been applied to synthesis of bioactive compounds. Additionally, a plausible catalytic cycle has been proposed.
