Abstract:OBJECTIVE: Recent studies indicate that as immunoactive substance, cytokine plays an important ajustive role in the inflammatory reaction of Alzheimer disease. This study summarized the role of interleukin-1 in the pathophysiological processes of Alzheimer disease. DATA SOURCES: A computer-based online search of Medline database was undertaken to identify for the related articles on the relationship between interleukin-1 and Alzheimer disease published in English between August 1995 and August 2005 with the of "interleukin-1, Alzheimer disease". At the same time, Chinese related articles were researched in China National Knowledge Infrastructure (CNKI) published between August 1995 and August 2005 with the same keywords in Chinese. STUDY SELECTION: Inclusive criteria: All the investigated subjects, both animals and patients, were involved, control group should be established, those with similar contents and published in the latest 3 years in authoritative journals were in superiority. The repetitive clinical or animal experimental studies were excluded. DATA EXTRACTION: Totally 207 articles were screened, and 21 articles, which were mainly concerned with the role of interleukin-1 in the pathophysiological processes of Alzheimer disease, were selected, and then sorted according to the participation of interleukin-1 in the pathophysiological processes of Alzheimer disease, including 5 about the relationship between the polymorphism of interleukin-1 and Alzheimer disease, 15 relevant to different pathophysiological processes, and 1 about the prospects of the effects of interleukin-1 on Alzheimer disease. The other 186 repetitive studies or other without control group were excluded. DATA SYNTHESIS: ① The interleukin-1 β was increased in the serum of patients with Alzheimer disease, indicating that interleukin-1 may have certain correlation with the attack of Alzheimer disease. ② It has been proved that interleukin-1 A(-889) polymorphism was associated with the increased level of interleukin-1α in vivo. Interleukin-1RN can adjust the function of interleukin-1A and interleukin-1B, and their interactions may be involved in the attack of Alzheimer disease. ③ Interleukin-1 may adjust the amyloid precursor protein mRNA translation through 5' nontranslated region. It can also produce more amyloid β-protein through accelerating the degradation of amyloid precursor protein. ④ In the brain of Alzheimer disease, many interleukin-1 immune positive microglias surrounded the brain regions with pathological changes, and their distribution in cortex was correlated with that in amyloid β-protein. ⑤ Interleukin-1 can also lead to the formation of neurofibrillary tangles and facilitating neurite outgrowth, etc, and then participate in the whole pathophysiological processes of Alzheimer disease. CONCLUSION: Interleukin-1 plays an important role in the pathophysiological processes of Alzheimer disease. Aim at that it seeks therapeutic targets in the process of its action on Alzheimer disease, such as interleukin-1Ra, further study on the effects of interleukin-1 would provide new clues for the clinical treatment of Alzheimer disease.
