Abstract:miRNAs are single-stranded small RNAs that do not encode proteins. They can combine complementarily with the 3′-UTRs of target gene mRNA molecules to promote targeted mRNA degradation or inhibit mRNA translation, thereby regulating gene expression at the post-transcriptional level. MiRNAs participate in regulation of cell cycling, growth, apoptosis, differentiation, and stress responses. MiRNA-145 (miR-145) is a tumor suppressor that targets various tumor-specific genes and proteins, thereby influencing related signaling pathways. MiR-145 not only regulates tumor growth, invasion, and metastasis, but is also important for tumor angiogenesis and tumor stem cell proliferation. Here, we review the roles and mechanisms of miR-145 in the diagnosis and treatment of malignant tumors. Published data confirm that miR-145 expression in various tumors is significantly lower than that in normal tissues and that overexpression of miR-145 inhibits the growth of different tumor cells, significantly reduces the ability of tumors to spread, and improves sensitivity to chemotherapeutic drugs. We conclude that miR-145 is a potential marker for use in the early diagnosis and prognostic evaluation of patients with cancer, has a role as a tumor suppressor, and is a promising cancer treatment target candidate.

Abstract:MicroRNAs (miRNAs) are a recently discovered family of endogenous, non-coding RNA molecules approximately 22 nt in length [1]. They negatively modulate gene expression post-transcriptionally by binding to the complementary sequence in the 3′ untranslated region of target messenger RNAs (mRNAs) [1]. miRNAs are transcribed from genomic DNA by RNA polymerase II but not further translated into protein (non-coding RNA). Eventually, they are processed from primary transcripts known as pri-miRNAs to short stem-loop structures called pre-miRNA and finally to become functionally mature miRNA. Mature miRNA molecules are partially complimentary to target mRNA where they either repress translation or direct destructive cleavage [2]. The first miRNA was described in 1993 by Lee and colleagues, who found miRNA-lin-4 is essential for the normal temporal control of diverse post-embryonic development in Caenorhabditis elegans by negatively regulating the level of LIN-14 protein via antisense RNA–RNA interaction [3]. miRNAs have a large-scale effect as a new layer of gene regulation mechanism. It has been estimated that the vertebrate genome encodes up to 1000 unique miRNAs, which can regulate expression of at least 30% of genes [4,5].

Abstract:MicroRNAs (miRNAs) are a type of small non-coding RNA molecule that performs an important role in post-transcriptional gene regulation. Since miRNAs were first identified in 1993, a number of studies have demonstrated that they act as tumor suppressors or oncogenes in human cancer, including colorectal, lung, brain, breast and liver cancer, and leukemia. Large high-throughput studies have previously revealed that miRNA profiling is critical for the diagnosis and prognosis of patients with cancer, while certain miRNAs possess the potential to be used as diagnostic and prognostic biomarkers or therapeutic targets in cancer. The present study reviews the studies and examines the roles of miRNAs in cancer diagnosis, prognosis and treatment, and discusses the potential therapeutic modality of exploiting miRNAs.

Abstract:MicroRNAs (miRNAs) are approximately 22 nt non-coding RNAs, which regulate gene expression in a sequence-specific manner via translational inhibition or messenger RNA (mRNA) degradation. Since the discovery of their fundamental mechanisms of action, the field of miRNAs has opened a new era in the understanding of small noncoding RNAs. By molecular cloning and bioinformatic approaches, miRNAs have been identified in viruses, plants and animals. miRNAs are predicted to negatively target up to one-third of human mRNAs. Cancer is a complex genetic disease caused by abnormalities in gene structure and expression. Previous studies have heavily focused on protein-coding genes; however, accumulating evidence is revealing an important role of miRNAs in cancer. Epigenetics is defined as mitotically and/or meiotically heritable changes in gene expression that are not accompanied by changes in DNA sequence. Given the critical roles of miRNAs and epigenetics in cancer, characterizing the epigenetic regulation of miRNAs will provide novel opportunities for the development of cancer biomarkers and/or the identification of new therapeutic targets in the foreseeable future.

Abstract:microRNAs (miRNAs) are a class of non-coding RNAs that function as endogenous triggers of the RNA interference pathway. Studies have shown that thousands of human protein-coding genes are regulated by miRNAs, indicating that miRNAs are master regulators of many important biological processes, such as cancer development. miRNAs frequently have deregulated expression in many types of human cancers, and play critical roles in tumorigenesis, which functions either as tumor suppressors or as oncogenes. Recent studies have shown that miRNAs are highly related with cancer progression, including initiating, growth, apoptosis, invasion, and metastasis. Furthermore, miRNAs are shown to be responsible for the cancer-related inflammation, anti-cancer drug resistance, and regulation of cancer stem cells. Therefore, miRNAs have generated great interest as a novel strategy in cancer diagnosis and therapy. Here we review the versatile roles of miRNAs in cancers and their potential applications for diagnosis, prognosis, and treatment as biomarkers.

Abstract:Cancer is a disease involving multistep dynamic changes in the genome. However, studies to date on the cancer genome have focused most heavily on protein-coding genes, and our knowledge on alterations of the functional noncoding sequences in cancer is largely absent. MicroRNAs (miRNAs) are ~22 nucleotide (nt) noncoding RNAs, which regulate gene expression in a sequence-specific manner via translational inhibition or mRNA degradation. Mounting evidence shows that miRNAs may play an important role in tumor development, and a better understanding of their alteration in cancer genome and oncogenic property should contribute to the diagnosis and treatment of cancer.

Abstract:MicroRNAs (miRNAs) are ~22 nucleotide long, noncoding, endogenous RNA molecules which exert their functions by base pairing with messenger RNAs (mRNAs), thereby regulate protein-coding gene expression. In eukaryotic cells, miRNAs play important roles in regulating biological processes such as proliferation, differentiation, apoptosis, and stem cell self-renewal. The human genome may contain as many as 1,000 miRNAs, and more than 700 of them have been identified. miRNAs are predicted to target up to one third of mRNAs. Each miRNA can target hundreds of transcripts directly or indirectly, while more than one miRNA can converge on a single transcript target. Therefore, the potential regulatory circuitry afforded by miRNA is enormous. Recently, mounting evidence implicates miRNAs as a new class of modulator for human tumor initiation and progression. Therefore, it has been proposed that manipulating miRNA activity and miRNA biogenesis may be a novel avenue for developing efficient therapies against cancer.

Abstract:Since the discovery of the first microRNA (miRNA), the exploration of miRNA biology has come to a new era in recent decades. Monumental studies have proven that miRNAs can be dysregulated in different types of cancers and the roles of miRNAs turn out to function to either tumor promoters or tumor suppressors. The interplay between miRNAs and the development of cancers has grabbed attention of miRNAs as novel tools and targets for therapeutic attempts. Moreover, the development of miRNA delivery system accelerates miRNA preclinical implications. In this review, we depict recent advances of miRNAs in cancer and discuss the potential diagnostic or therapeutic approaches of miRNAs.

Abstract:MicroRNAs (miRNAs), small non-coding RNAs (ncRNAs) of about 22 nucleotides in size, play important roles in gene regulation, and their dysregulation is implicated in human diseases including cancer. A variety of miRNAs could take roles in the cancer progression, participate in the process of tumor immune, and function with miRNA sponges. During the last two decades, the connection between miRNAs and various cancers has been widely researched. Based on evidence about miRNA, numerous potential cancer biomarkers for the diagnosis and prognosis have been put forward, providing a new perspective on cancer screening. Besides, there are several miRNA-based therapies among different cancers being conducted, advanced treatments such as the combination of synergistic strategies and the use of complementary miRNAs provide significant clinical benefits to cancer patients potentially. Furthermore, it is demonstrated that many miRNAs are engaged in the resistance of cancer therapies with their complex underlying regulatory mechanisms, whose comprehensive cognition can help clinicians and improve patient prognosis. With the belief that studies about miRNAs in human cancer would have great clinical implications, we attempt to summarize the current situation and potential development prospects in this review.

Abstract:MicroRNAs (miRNAs) are a group of endogenous non-coding RNAs that regulate gene expression. Alteration in miRNA expression results in changes in the profile of genes involving a range of biological processes, contributing to numerous human disorders. With high stability in human fluids, miRNAs in the circulation are considered as promising biomarkers for diagnosis, as well as prognosis of disease. In addition, the translation of miRNA-based therapy from a research setting to clinical application has huge potential. The aim of the current review is to: (i) discuss how miRNAs traffic intracellularly and extracellularly; (ii) emphasize the role of circulating miRNAs as attractive potential biomarkers for diagnosis and prognosis; (iii) describe how circulating microRNA can be measured, emphasizing technical problems that may influence their relative levels; (iv) highlight some of the circulating miRNA panels available for clinical use; (v) discuss how miRNAs could be utilized as novel therapeutics, and finally (v) update those miRNA-based therapeutics clinical trials that could potentially lead to a breakthrough in the treatment of different human pathologies.

Abstract:MicroRNAs (miRNAs) are a class of short noncoding RNAs that participate in mastering the balance of gene-regulating networks. By targeting and controlling expression of messenger RNA, miRNAs can control highly complex signal transduction pathways and other biological pathways. Unique aberrant expression of miRNA at each stage of cancer development suggests that miRNA could play a novel role in cancer diagnosis and therapeutic strategies. Accumulated information on epigenetic modification of miRNA suggests a promising platform for miRNA in cancer therapy. Clinical applications exploiting the understanding of miRNA's function will be the next great challenge in cancer research.

Abstract:MicroRNA (miRNA) dysregulation is causal in most, if not all, cancers. They silence the gene’s expression by modulating messenger RNA (mRNA) through employing translational repression or degradation. MiRNA has the ability to target various genes, which involve various significant signaling pathway in cancer cells, to control cancer development and progression. As a result, manipulating miRNA expression levels for cancer therapy seems an attractive and novel therapeutic approach. However, certain miRNAs, which possess the bi-directional roles, could promote cancer cells growth, but also enhance T-cell immunity. Therefore, miRNA-based therapy could also kill T cells and other lymphocytes, ultimately resulting in promoting cancer growth, which makes miRNA-based cancer therapy process with caution. In this view, we will focus on these miRNAs with the bi-directional roles, and discuss their potential effects to cancer therapy. Ultimately, identified and dissected the miRNAs crossing both T-cell-mediated anti-tumor immunity and tumor cell growth will prove vital for the design of more effective and safer strategies for cancer therapy.

Abstract:Background: MicroRNAs (miRNAs), small, non-coding single-stranded RNAs with the potential to regulate more than 30 percent of the human protein coding genes, are associated with a wide array of biological processes and human disease pathologies, including cancer. To date, thymidylate synthase ( TS) and Dihydrofolate reductase (DHFR) are the major targets of cancer chemotherapy, making the TS and DHFR inhibitors, TDX and MTX, the most widely used therapeutic agents by clinicians. However, increased DHFR levels in certain types of cancer reportedly are associated with drug resistance, requiring vital new therapeutic compounds that target DHFR inhibitors.

Abstract:MicroRNAs (miRNAs) are small noncoding RNA molecules which regulate the target gene expression posttranscriptionally. Increasing studies have shown that microRNAs play important roles in multiple biological pathways. For instance, aberrant expression of microRNA-224 (miR-224) plays a vital role in tumor biology in various types of human cancer. Here, we aim to summarize the molecular mechanisms that lead to the overexpression of miR-224 in cancers, analyze the effect of miR-224 on tumor biology, and reveal the clinical significance of miR-224. MiR-224 regulates its targets by modulating messenger RNA (mRNA) stability and/or protein translation, and it would provide new insight into molecular targeting cancer treatment.

Abstract:MicroRNAs (miRNAs) comprise an important class of small single-stranded non-coding RNAs that regulate several important biological processes, including cell differentiation, growth, and apoptosis. The dysregulations of miRNAs are usually correlated with several diseases because of their regulatory functions in organisms. Over the past few years, miRNAs are often observed to be aberrantly expressed in cancers, indicating their potential correlations with cancer pathogenesis. Recently, evidence has increased to prove this assumption. Further investigations show that miRNAs are involved extensively in various cancer cell processes on the molecular level, including promotions of cancer cell development. Considering the important functions of miRNAs in cancer development, investigating the relationship between miRNAs and cancers can lead to a better understanding of the latter. Therefore, this review summarizes recent developments from related studies, particularly focusing on the functions of miRNAs and their potential applications as therapeutic strategies for cancers.

Abstract:MicroRNAs (miRNAs) are small RNA molecules that regulate more than 30% of genes in humans. Recent studies have revealed that miRNAs play a crucial role in tumorigenesis. Large sets of miRNAs in human tumors are under-expressed compared to normal tissues. Furthermore, experiments have shown that interference with miRNA processing enhances tumorigenesis. Multiple studies have documented the causal role of miRNAs in cancer, and miRNA-based anticancer therapies are currently being developed. This review primarily focuses on two key points: (1) miRNAs and their role in human cancer and (2) the regulation of tumor suppressors by miRNAs. The review discusses (a) the regulation of the tumor suppressor p53 by miRNA, (b) the critical role of the miR-144/451 cluster in regulating the Itch-p63-Ago2 pathway, and (c) the regulation of PTEN by miRNAs. Future research and the perspectives of miRNA in cancer are also discussed. Understanding these pathways will open avenues for therapeutic interventions targeting miRNA regulation.

Abstract:Background & Objective: The incidence of non-small cell lung cancer (NSCLC) has been rising over the past several decades. Despite various therapeutic regimens and modern diagnostic techniques are developed, NSCLC still have an extremely poor prognosis due to drug resistance. Therefore, it is critical to find a novel precise diagnosis and effective treatment approach for NSCLC patients. MicroRNAs (MiRNAs) are a class of 18-25nt non-coding small RNAs, which have been shown to be involved profoundly in the pathogenesis such as cellular proliferation, differentiation, development, apoptosis and tumorigenesis in many human tumors including of NSCLC. We reviewed existing research literature regarding correlations between miRNAs and their target's response to anticancer treatment, and summarized the recent findings between miRNAs and therapy availability in NSCLC.
