Abstract:The Cover Feature shows the Samuel Beckett bridge and the Convention Centre Dublin beside the river Liffey. The Convention Centre is the venue for the 9th EuChemS Congress being held in July 2024. The chemistry scheme shows a racemic β‐keto allyl ester and the two diastereomeric transition states, the one on top leading to the preferred enantiomer (98 %) and the one on the bottom leading to the disfavoured enantiomer (2 %). Stereospecific Csp3 Suzuki–Miyaura Cross-Coupling That Evades Β-Oxygen Elimination

Abstract:Stereospecific Csp(3) Suzuki-Miyaura cross-coupling could transform stereochemically complex small molecule synthesis into a simple and broadly accessible process. However, most current methods are not compatible with complex building blocks that represent densely packed, multistereogenic center-containing motifs commonly found in natural products and other complex targets. Here, we report a method that enables the alpha-methyl-beta-hydroxyl motif, which is found in > 18 000 natural products as well as other Csp(3)-rich organic fragments, to be embedded within stereochemically defined secondary alkyl boronic ester building blocks that are readily cross-coupled in a stereospecific manner. The steric effect-mediated decrease in reactivity toward transmetalation and deleterious side reactions associated with the cross-coupling of beta-oxygen-containing Csp(3) boronic esters are concomitantly addressed using beta-aryloxysilyl groups as dual-purpose transmetalation-promoting groups and stable beta-oxygen surrogates. Mechanistic studies including real-time HPLC analysis show that a five-membered pinacol siloxaborolate generated in situ is then hydrolyzed to a dihydroxysiloxaborolate that acts as an activated transmetalation partner in a stereospecific process that proceeds with retention of configuration.

Abstract:Stereochemical editing strategies have recently enabled the transformation of readily accessible substrates into rare and valuable products. Typically, site selectivity is achieved by minimizing kinetic complexity by using protecting groups to suppress reactivity at undesired sites (substrate control) or by using catalysts with tailored shapes to drive reactivity at the desired site (catalyst control). We propose "network control," a contrasting paradigm that exploits hidden interactions between rate constants to greatly amplify modest intrinsic biases and enable precise multisite editing. When network control is applied to the photochemical isomerization of hexoses, six of the eight possible diastereomers can be selectively obtained. The amplification effect can be viewed as a mesoscale phenomenon between the limiting regimes of kinetic control in simple chemical systems and metabolic regulation in complex biological systems.

Similar to the click reaction, the click polymerization enjoys the advantages of high efficiency, mild reaction conditions, and high regio- and stereoselectivity etc. In this highlight, we summarize the recent progress on click polymerizations, with focus on the alkyne-based ones. Abstract:The year 1874 is considered to be the birth of stereochemistry when the third dimension in describing and discussing structures in chemistry was first introduced. Stereochemistry and structural chemistry are closely related though they are not each other's synonyms. Structural chemistry implies greater emphasis on metrical aspects and stereochemistry on shape, symmetry, and chirality. We review some of the nodal points in the early development of stereochemistry.

Abstract:Tetracoordinate organoborons are a highly important class of molecules, that have found wide applications in areas ranging from synthetic chemistry, medicinal chemistry, to materials science. In this review, we summarized and discussed the significant progress and recent advances in the synthesis and application of enantioenriched tetracoordinate boron‐stereogenic compounds, according to the synthetic method: (1) resolution of racemates, (2) chiral substrate induced asymmetric synthesis, and (3) asymmetric catalysis. Comprehensive Summary The 21st century has witnessed a continuous evolution in the development of boron‐stereogenic chemistry. Since the 1990s, various innovations for the synthesis of tetracoordinate boron‐stereogenic compounds, which exhibited great potential applications, have been demonstrated by synthetic chemists. This paper reviews the significant progress and recent advances towards the assembly of enantioenriched boron‐stereogenic compounds, and hopes to shed light on new perspectives and inspire further research in this emerging field.

Strategies and techniques for identification of novel bioactive compounds--CHI's second annual conference. 7-9 October 1998, Zurich, Switzerland

Abstract:This conference on recent developments in the discovery of novel therapeutic candidates was organized by Amy Dasch (Cambridge Healthtech Institute, Newton Upper Falls, MA, USA; http://www.xensei.com/conferences). The conference provided an overview of all relevant aspects of the rapidly changing paradigms in drug research. Gene technology creates a vast number of new biological targets. The progress in combinatorial chemistry and high-throughput screening (HTS) is accompanied by the development of virtual libraries, large screening programs, and the generation of enormous sets of data. Correspondingly, the lectures covered such different topics as target identification and assay development, HTS technology, combinatorial library design and synthesis, chemoinformatics, and the integration of these components into the discovery of novel pharmaceutical compounds, the development of agricultural chemicals, and other applications. A most valuable addition was reports on case histories in drug development from pharmaceutical companies utilizing these technologies. About 100 scientists, many of them from European countries, attended the meeting. In total, 25 lectures were presented in four sessions: molecular diversity and library design; combinatorial synthesis; HTS; computational methodologies and chemoinformatics. Like other commercially organized conferences, this meeting was well-planned. The balance of speakers from small venture capital companies, large pharmaceutical and agricultural firms gave a broad overview of recent progress in the rational design, combinatorial synthesis, and HTS of new bioactive compounds, as well as on different approaches to handling large data sets and deriving structure-activity relationships from such data.
Revealing the effect of stereocontrol on intermolecular interactions between abiotic, sequence-defined polyurethanes and a ligand

Abstract:The development of precision polymer synthesis has facilitated access to a diverse library of abiotic structures, wherein chiral monomers are positioned at specific locations within macromolecule chains. These structures are anticipated to exhibit folding characteristics similar to biotic macromolecules and possess comparable functionalities. However, the extensive sequence space and numerous variables make selecting a sequence with the desired function challenging. Therefore, revealing sequence-function dependencies and developing practical tools are necessary to analyze their conformation and molecular interactions. In this study, we investigate the effect of stereochemistry, which dictates the spatial location of backbone and pendant groups, on the interaction between sequence-defined oligourethanes and bisphenol A ligands. Various methods are explored to analyze the receptor-like properties of model oligomers and the ligand. The accuracy of molecular dynamics simulations and experimental techniques is assessed to uncover the impact of discrete changes in stereochemical arrangements on the structure of the resulting complexes and their binding strength. Detailed computational investigations providing atomistic details show that the formed complexes demonstrate significant structural diversity depending on the sequence of stereocenters, thus affecting the oligomer-ligand binding strength. Among the tested techniques, the fluorescence spectroscopy data, fitted to the Stern-Volmer equation, are consistently aligned with the calculations, thus validating the developed simulation methodology. The developed methodology opens a way to engineer the structure of sequence-defined oligomers with receptor-like functionality to explore their practical applications, e.g., as sensory materials.

Abstract:This review explores the fascinating world of molecules featuring multiple stereogenic elements, unraveling the different strategies designed over the years for their enantioselective synthesis. Specifically, (dynamic) kinetic resolutions, desymmetrisations and simultaneous installation of stereogenic elements exploiting either metal- or organo-catalysis are the principal approaches to efficiently create and control the three-dimensional shapes of these attractive molecules. Although most molecules presented in this review possess a stereogenic carbon atom in combination with a stereogenic axis, other combinations with helices or planes of chirality have started to emerge, as well as molecules displaying more than two different stereogenic elements.

Abstract:The control of chemo-, regio-, diastereo-, and enantioselectivity is a central theme in organic synthesis. The capability to obtain the full set of stereoisomers of a molecule would significantly enhance the efficiency for the synthesis of natural product analogues and creation of chiral compound libraries for drug discovery. Despite the tremendous progress achieved in the field of asymmetric synthesis in the past decades, the precise control of both relative and absolute configurations in catalyst-controlled reactions that create multiple stereocenters remains a significant synthetic challenge. We report here the development of a catalyst-controlled hydroboration with hitherto unattainable selectivity. The Rh-catalyzed hydroboration of a, beta-unsaturated carbonyl compounds with pinacolborane proceeds with high levels of regio-, diastereo-, and enantioselectivities to provide a hydroboration product with two vicinal stereocenters. Through the appropriate choice of substrate geometry (E or Z) and ligand enantiomer (S or R), all the possible diastereoisomers are readily accessible. The boron-containing products underwent many stereospecific transformations, thus providing a strategy for collective stereodivergent synthesis of diverse valuable chiral building blocks.

Abstract:β,γ-Unsaturated α-ketoesters prove to be versatile organic synthons participating in diverse catalytic asymmetric transformations with the breathtaking development of organo-catalysis, new catalytic systems including ingenious chiral ligands as well as Lewis acid cations. The highly efficient creation of stereogenic centers with excellent enantioselectivity is not a surprise, but owes to the bidentate coordination of its unique 1,2-dicarbonyl motif to artful chiral messenger, establishing a rigid system for the precise chiral-identification of the attack. In the past five years, various reaction modes of β,γ-unsaturated α-ketoesters have been developed, involving their multiple reaction sites, such as the carbon-carbon double bond (C=C), the carbonyl group (C=O), the entire C=C-C=O fragment, and the ester group. In this review, we summarize the state-of-the-art catalytic asymmetric reactions of β,γ-unsaturated α-ketoesters, to provide an updated overview to chemists working in this and related fields, facilitating their discoveries in asymmetric catalysis, natural products synthesis, and drug development.

Abstract:##IMG## [http://ej.iop.org/images/0036-021X/89/12/1507/rc4980c.gif] Representative examples of the application of 3D printing in organic synthesis, biochemistry, biotechnology, analytical chemistry, pharmaceutics and chemical education are considered. It is shown that additive technologies open up new prospects for the development of these fields of science. The characteristics of widely used 3D printing methods (fused deposition modelling and stereolithography) are discussed in the context of chemical applications. It is noted that the key feature of these methods is the wide accessibility of technologies and materials. The bibliography includes 498 references.
