Abstract:Recent years have seen novel modalities emerge for the treatment of human diseases resulting in an increase in beyond rule of 5 (bRo5) chemical matter. As a result, synthetic innovations aiming at enabling rapid access of complex bRo5 molecular entities become increasingly valuable for medicinal chemists’ tool-kits. Herein, we report the general synthesis of a new class of noncanonical amino acids (ncAA) with a cyclopropyl back-bone to achieve conformational constraint and bearing C(sp3)-rich benzene bioisosteres. We also demonstrate preliminary studies towards utilities of these ncAA as building blocks for medicinal chemistry research.

What problem does this paper attempt to address?
The paper aims to address the following issues: 1. **Design of Novel Non-Canonical Amino Acids (ncAAs)**: The paper introduces a method for designing novel non-canonical amino acids that feature a cyclopropane scaffold to achieve conformational constraint and are rich in sp3-hybridized benzene bioisosteres. These designs aim to minimize the number of freely rotating bonds, reduce protease metabolism, and fine-tune physicochemical properties by increasing the proportion of sp3 hybridization. 2. **Synthetic Methodology**: The paper proposes a general synthetic method that can prepare these non-canonical amino acids in a one-step reaction from 2-aminoacrylate and the corresponding N-tosylhydrazone. This method allows for the rapid acquisition of various stereoisomers, facilitating subsequent medicinal chemistry research. 3. **Functional Validation**: Preliminary studies indicate that these non-canonical amino acids can be used as building blocks in medicinal chemistry research and demonstrate their potential applications in peptide chemistry. For example, complex tripeptides can be synthesized through selective deprotection and coupling reactions. 4. **Optimization of Pharmacological Properties**: By using non-canonical amino acids in small molecule drug discovery, the physicochemical properties of drugs, such as polarity, hydrophobicity, and solubility, can be optimized. For instance, one example in the paper shows that replacing a phenyl group with a cyclopropyl group can improve the solubility and permeability of a compound. In summary, the main objective of this paper is to develop a new synthetic method for non-canonical amino acids and demonstrate their potential application value in medicinal chemistry research.
