Modulation of the Ventrolateral Part of Ventromedial Hypothalamus Affects Post-Mi Cardiac Remodeling Via the Sympathetic Pathway

Abstract:Backgrounds: The ventrolateral part of the ventromedial hypothalamus (VMHVL) reflects anger and is associated with sympathetic activity. We aim to investigate whether there is a sympathetic pathway projecting from VMHVL to the heart and mediates cardiac remodeling.Methods: Left artery descending occlusion (LADO) was performed to establish 14-day myocardial infarction (MI) and assess the activities of VMHVL for preliminary VMHVL-heart link evidence. Then in other rats, activating (hM3dGq) and inhibiting (hM4dGi) receptors exclusively activated by clozapine N-oxide (CNO) were delivered into VMHVL to modulate it and LADO was operated a month later, accompanying with two-week intraperitoneally CNO injection (3 mg/ml/kg). Trace VMHVL-heart projections retrogradely via pseudorabies virus (PRV). Immunofluorescence of neurons, echocardiography, electrocardiogram, infarct area, serum norepinephrine, and RNA-seq was used to verify the effects of VMHVL on cardiac remodeling and its connections with the heart.Findings: VMHVL was enhanced post-MI and its inhibition ameliorated cardiac remodeling. RNA-seq analysis revealed the alternation of the adrenergic pathway in the myocardium. The paraventricular nucleus (PVN), rostral ventrolateral medulla (RVLM), superior cervical ganglion (SCG), and VMHVL were labeled by PRV, and VMHVL inhibition decreased their activities and downregulated the heart rate variability and serum norepinephrine.
