Abstract:Copper-based nanoparticles (NPs) display a strong potential to replace copper salts (e.g., CuSO4) for application in agricultures as antimicrobial agents or nutritional amendments. Yet, their effects on crop quality are still not comprehensively understood. In this study, the Cu contents in soybeans grown in soils amended with Cu NPs and CuSO4 at 100-500 mg Cu/kg and the subsequent effects on the plant physiological markers were determined. The Cu NPs induced 29-89% at the flowering stage (on Day 40) and 100-165% at maturation stage (on Day 100) more Cu accumulation in soybeans than CuSO4. The presence of particle aggregates in the root cells with deformation upon the Cu NP exposure was observed by transmission electron microscopy. The Cu NPs at 100 and 200 mg/kg significantly improved the plant height and biomass, yet significantly inhibited at 500 mg/kg, compared to the control. In leaves chlorophyll-b was more sensitive than chlorophyll-a and carotenoids to the Cu NP effect. The Cu NPs significantly decreased the root nitrogen and phosphorus contents, while they significantly increased the leaf potassium content in comparison with control. Our results imply that cautious use of Cu NPs in agriculture is warranted due to relatively high uptake of Cu and altered nutrient quality in soybeans.

Abstract:Abstract Background Selenium (Se) in soil mainly consists of selenite, selenate, and elemental Se. However, little is known about the mechanism involved in the uptake and biotransformation of elemental Se by plants. Results In this study, the uptake, translocation, subcellular distribution and biotransformation of selenium nanoparticles (SeNPs) in rice ( Oryza sativa L.), and a comparison with selenite and selenate, were investigated through hydroponic experiments. The study revealed that SeNPs could be absorbed by rice plants; and aquaporin inhibitor was responsible for a 60.4% inhibition of SeNP influx, while metabolic inhibitor was ineffective. However, the SeNPs uptake rate of rice roots was approximately 1.7 times slower than that of selenite or selenate. Under the SeNPs or selenite treatment, Se was primarily accumulated in roots rather than in shoots, whereas an opposite trend was observed with selenate treatment. Additionally, most of the absorbed Se was distributed in cell wall of the SeNPs or selenite treated-rice plants, while its proportion was the highest in soluble cytosol of the selenate treated-rice plants. The absorbed SeNPs or selenite was rapidly assimilated to organic forms, with SeMet being the most predominant species in both shoots and roots of the rice plants. However, following selenate treatment, Se(VI) remained as the most predominant species, and only a small amount of it was converted to organic forms. Conclusion Therefore, this study provides a deeper understanding of the mechanisms associated SeNPs uptake and biotransformation within plants.

Abstract:Nanotechnology has been increasingly used in plant sciences, with engineered nanoparticles showing promising results as fertilizers or pesticides. The present study compared the effects in the foliar application of Se nanoparticles (SeNPs) or sodium selenite-Se(IV) on rice seedlings. The degree of plant growth, photosynthetic pigment content, and concentrations of Se, Na, Mg, K, Ca, Mn, Co, Cu, Zn, As, Cd, and Pb were evaluated. The results showed that the application of SeNPs at high concentrations (5 mg L-1), as well as the application of Se(IV), inhibited plant growth and increased the root concentrations of As and Pb. The application of SeNPs at 0.5 mg L-1 significantly increased Se accumulation in the aerial part from 0.161 ± 0.028 mg kg-1 to 0.836 ± 0.097 mg kg-1 without influencing physiological, chemical, or biochemical parameters. When applied to leaves, SeNPs tended to remain in the aerial part, while the application of Se(IV) caused a higher Se translocation from the shoots to the roots. This study provides useful information concerning the uptake, accumulation, and translocation of different Se formulations in rice seedlings and their effect on plant ionomic profiles, thus showing that the foliar application of SeNPs at low concentrations can be an effective and safe alternative for rice biofortification.

Abstract:Agricultural soil is gradually becoming a primary sink for metal -based nanoparticles (MNPs). The uptake and accumulation of MNPs by crops may contaminate food chain and pose unexpected risks for human health. Here, we investigated the fate and transformation of CuO nano particles (NPs) in the soil -rice system during the rice lifecycle. The results show that at the maturation stage, 1000 mg/kg CuO NPs significantly decreased redox potential by 202.75 triV but enhanced electrical conductivity by 497.07 mS/cm compared to controls. Moreover, the bioavailability of highest CuO NPs in the soil was reduced by 69.84% along with the plant growth but then was significantly increased' y 165% after drying -wetting cycles. Meanwhile, CuO and Cu coMbined with humic acid were transformed to Cu2S and Cu associated with goethite by X-ray absorption near edge structure analysis. AdditiOnally, CuO NPs had an acute negative effect on the plant growth than bulk particles, which dramatically reduced the fresh weight of grains to 6.51% of controls. Notably, CuO NPs were found to be translocated from soil to plant especially to the chaff and promoted the Cu accumulation in the aleurone layer of rice using micro X-ray fluorescence technique, but could not reach the polished rice.
Bioavailability of cerium oxide nanoparticles to Raphanus sativus L. in two soils

Abstract:Cerium oxide nanoparticles (CeO2 NP) are a common component of many commercial products. Due to the general concerns over the potential toxicity of engineered nanoparticles (ENPs), the phytotoxicity and in planta accumulation of CeO2 NPs have been broadly investigated. However, most previous studies were conducted in hydroponic systems and with grain crops. For a few studies performed with soil grown plants, the impact of soil properties on the fate and transport of CeO2 NPs was generally ignored even though numerous previous studies indicate that soil properties play a critical role in the fate and transport of environmental pollutants. The objectives of this study were to evaluate the soil fractionation and bioavailability of CeO2 NPs to Raphanus sativus L (radish) in two soil types. Our results showed that the silty loam contained slightly higher exchangeable fraction (F1) of cerium element than did loamy sand soil, but significantly lower reducible (F2) and oxidizable (F3) fractions as CeO2 NPs concentration increased. CeO2 NPs associated with silicate minerals or the residue fraction (F4) dominated in both soils. The cerium concentration in radish storage root showed linear correlation with the sum of the first three fractions (r2 = 0.98 and 0.78 for loamy sand and silty loam respectively). However, the cerium content in radish shoots only exhibited strong correlations with F1 (r2 = 0.97 and 0.89 for loamy sand and silty loam respectively). Overall, the results demonstrated that soil properties are important factors governing the distribution of CeO2 NPs in soil and subsequent bioavailability to plants.

Abstract:The application of selenium nanoparticles (Se NPs) in agriculture has garnered significant attention, yet there remains a knowledge gap regarding the correlation between the size of Se NPs and their availability in soil. To address this gap, adsorption experiments, soil column experiments, and pot experiments were conducted to investigate the relationship between the availability in soil of Se NPs and their particle size. The results showed that the adsorption capacities of Se NPs with various sizes (30, 50, 80, 110 nm) to fulvic acid (FA) was decreased with increasing particle sizes and pH. Small sized Se NPs (30 nm) are more stable because they adsorb more organic matter, which significantly decreases aggregation. Moreover, the mobility of Se NPs was decreased by increasing clay content in soil and particle size. These results demonstrated that small-sized Se NPs have higher bioavailability than large-sized Se NPs. The Se content in rhizosphere soil and Brassica chinensis shoot was significantly increased by 319.4% and 220.2% upon exposure to Se NPs (30 nm), higher than that for others (50, 80, 110 nm and 2 μm). The reason is that small-sized Se NPs are more stable in the soil, more easily migrate from the non-rhizosphere to the rhizosphere and interact with crop roots, leading to increased root exudations. These interactions recruit beneficial microorganisms in the rhizosphere. In turn, the improved root exudations and the presence of these microorganisms enhance the bioavailability of Se NPs. Linear regression analysis indicated that the size of soil-applied Se NPs should fall within the range of 11-631 nm, as those outside of this range may not meet the standard for Se-enriched food. Besides, the nutrient accumulation in leaves promoted photosynthesis (81.1%) and increased the yield (113.1%) of Brassica chinensis by small-sized Se NPs (30 nm). Therefore, these information highlights the critical role of Se NPs size in crop Se biofortification for agriculture production.

Abstract:Copper oxide nanoparticles (CuO NPs) are widely used as fungicides in agriculture. Arsenic (As) is a ubiquitous contaminant in paddy soil. The present study was focused on the adsorption behavior of CuO NPs with regard to As as well as the characteristics of the microbial community changes in As-contaminated soil-rice systems in response to CuO NPs. The study found that CuO NPs could be a temporary sink of As in soil; a high dose of CuO NPs promoted the release of As from crystalline iron oxide, which increased the As content in the liquid phase. The study also found that the As bioavailability changed significantly when the dose of CuO NPs was higher than 50 mg kg-1 in the soil-rice system. The addition of 100 mg kg-1 CuO NPs increased the microbial diversity and the abundance of genes involved in As cycling, decreased the abundance of Fe(III)-reducing bacteria and sulfate-reducing genes, and decreased As accumulation in grains. Treatment with 500 mg kg-1 CuO NPs increased the abundance of Fe(III)-reducing bacteria and sulfate-reducing genes, decreased Fe plaques, and increased As accumulation in rice. The adverse effects of CuO NPs on crops and associated risks need to be considered carefully.

Abstract:To determine the bioavailability and translocation of metal oxide nanoparticles (MONPs) in the soil-rice plant system, we examined the accumulation and micro-distribution of ZnO nanoparticles (NPs), CuO NPs and CeO2 NPs (50, 100 and 500 mg/kg) in the paddy soil and rice plants under flooded condition for 30 days using single-step chemical extraction and diffusive gradients in thin films (DGT) technique combined with micro Xray fluorescence spectroscopy (mu-XRF). The results show that various MONPs changed the soil properties, especially the redox potential was enhanced to -165.33 to -75.33 mV compared to the control. The extraction efficiency of Zn, Cu and Ce in the paddy soil from high to low was EDTA, DTPA CaCl2 and DGT. Moreover, exposure to 500 mg/kg CuO NPs and CeO2 NPs induced the primary accumulation of Cu and Ce elements in rice roots as high as 235.48 mg Cu/kg and 164.84 mg Ce/kg, respectively, while the Zn concentration in shoots was up to 313.18 mg/kg under highest ZnO NPs with a 1.5 of translocation factor. The effect of MONPs on the plant growth was mainly related to the chemical species and solubility of MONPs. Micro-XRF analysis shows that Zn was mostly located in the root cortex while Cu was primarily accumulated in the root exodermis and few Ce distributed in the root. Pearson correlation coefficients indicate that only DTPA-extracted metals in soil were significantly and well correlated to the Zn, Cu and Ce accumulation in rice seedlings exposed to MONPs. Abstract:Enzyme release was studied in various tissues in intact animals or by means of tissue slices, organ perfusion or isolated and cultured cells. The factors that determine the extent and the time course of the leakage were studied extensively with several liver enzymes. It was found that the molecular properties of the enzymes, their topology within the organ, their intracellular localisation, their inducibility on the one hand, the nutritional and energetic state of the organ, and the respective protein turnover on the other, and finally, the type of the organ lesion all contribute to the different release patterns, which are known from experimental studies, and which are utilized for the diagnosis of diseases. While these preconditions of enzyme release are fairly well understood, the nature of the membrane alterations and the mechanism of detachment and transport of the enzyme proteins are less clear. Different models have been proposed, which may not exclude each other. Alterations of the cytoskeleton and the lipid composition of the plasma membranes are closely linked to ion imbalance, which is, in turn, dependent on the energy state of the individual cellular compartments.

Abstract:Agricultural soil pollution with potentially toxic trace elements (PTEs) has emerged as a significant environmental concern, jeopardizing food safety and human health. Although, conventional remediation approaches have been used for PTEs-contaminated soils treatment; however, these techniques are toxic, expensive, harmful to human health, and can lead to environmental contamination. Nano-enabled agriculture has gained significant attention as a sustainable approach to improve crop production and food security. Silicon nanomaterials (SiNMs) have emerged as a promising alternative for PTEs-contaminated soils remediation. SiNMs have unique characteristics, such as higher chemical reactivity, higher stability, greater surface area to volume ratio and smaller size that make them effective in removing PTEs from the environment. The review discusses the recent advancements and developments in SiNMs for the sustainable remediation of PTEs in agricultural soils. The article covers various synthesis methods, characterization techniques, and the potential mechanisms of SiNMs to alleviate PTEs toxicity in plant-soil systems. Additionally, we highlight the potential benefits and limitations of SiNMs and discusses future directions for research and development. Overall, the use of SiNMs for PTEs remediation offers a sustainable platform for the protection of agricultural soils and the environment.

Abstract:Expression of the cytokine, receptor activator of NF‐κB ligand (RANKL), is stimulated by both parathyroid hormone (PTH) and calcitriol in osteoblasts. Most studies have examined the effects on RANKL mRNA, and less information is available on the protein products. We have determined the effects of PTH, the adenylate cyclase stimulator forskolin, and calcitriol, alone and in combination, on endogenous RANKL protein expression in UMR‐106 rat osteoblastic osteosarcoma cells by Western blotting and enzyme immunoassay (EIA). PTH and forskolin dose dependently increased a ∼52 kDa band in whole cell lysates that was detected by both C‐ and N‐terminal directed RANKL antibodies. Calcitriol treatment produced little or no expression of this ∼52 kDa band, but markedly increased the expression of a ∼32 kDa band that was only detected with an antibody directed to the N‐terminus of RANKL. An EIA based on RANKL binding to OPG detected a large increase in RANKL expression following calcitriol treatment, and much smaller increases with PTH or forskolin. The combination of PTH and calcitriol or forskolin and calcitriol elicited effects similar to those of PTH and forskolin alone, as detected by both Western blotting and EIA. In contrast to the effects on protein, all agents increased RANKL mRNA expression, with the greatest effects seen with the co‐treatments. The results indicate that PTH, likely through effects on cyclic AMP, has a different effect on RANKL processing than calcitriol. The ∼52 and ∼32 kDa RANKL products appear to interact differently with OPG, which could affect responses to the agents in target cells. © 2005 Wiley‐Liss, Inc.

Biosynthesized Selenium Nanoparticles as an Effective Tool to Combat Soil Metal Stresses in Rice (Oryza sativa L.)

Abstract:Nanotechnology has demonstrated significant potential to improve agricultural production and increase crop tolerance to abiotic stress including exposure to heavy metals. The present study investigated the mechanisms by which aloe vera extract gel-biosynthesized (AVGE) selenium nanoparticles (Se NPs) alleviated cadmium (Cd)-induced toxicity to rice (Oryza sativa L.). AVGE Se NPs, chemically synthesized bare Se NPs, and NaSeO3 as an ionic control were applied to Cd-stressed rice seedlings via root exposure in both hydroponic and soil systems. Upon exposure to AVGE Se NPs at 15 mg Se/L, the fresh root biomass was significantly increased by 100.7% and 19.5% as compared to Cd control and conventional bare Se NPs. Transcriptional analyses highlighted that AVGE Se NPs activated stress signaling and defense related pathways, including glutathione metabolism, phenylpropanoid biosynthesis and plant hormone signal transduction. Specifically, exposure to AVGE Se NPs upregulated the expression of genes associated with the gibberellic acid (GA) biosynthesis by and 4.79- and 3.29-fold as compared to the Cd-alone treatment and the untreated control, respectively. Importantly, AVGE Se NPs restored the composition of the endophyte community and recruit of beneficial species under Cd exposure; the relative abundance of Azospirillum was significantly increased in roots, shoots, and the rhizosphere soil by 0.73-, 4.58- and 0.37-fold, respectively, relative to the Cd-alone treatment. Collectively, these findings highlight the significant potential of AVGE Se NPs to enhance plant growth and to minimize the Cd-induced toxicity in rice and provide a promising nanoenabled strategy to enhance food safety upon crop cultivation in contaminated agricultural soils.

Abstract:Selenium (Se) is an essential element for human and animal health and has antioxidant, anticancer, and antiviral effects. However, more than 100 million people in China do not have enough Se in their diets, resulting in a state of low Se in the human body. Since the absorption of Se by crop seeds depends not only on the Se content in soil, there are many omissions and misjudgments in the division of Se-rich producing areas. Soil pH, total iron oxide content (TFe 2 O 3 ), soil organic matter (SOM), and P and S contents were the main factors affecting Se migration and transformation in the soil–rice system. In this study, we compared the performance of the back propagation neural network (BP network) and multiple linear regression (MLR) using 177 pairs of soil–rice samples. Our results showed that the BP network had higher accuracy than MLR. The accuracy and precision of the prediction data met the requirements, and the prediction data were reliable. Based on the Se data of surface paddy fields, 26,900 ha of Se-rich rice planting area was planned using this model, accounting for 77% of the paddy field area. In the planned Se-rich area for rice, the proportion of soil Se content greater than 0.4 mg·kg −1 was only 5.29%. Our research is of great significance for the development of Se-rich lands.

Abstract:Cadmium (Cd) pollution restricts the rice growth and poses a threat to human health. Nanosized selenium (NanoSe) is a new nano material. However, the effects of NanoSe application on aromatic rice performances under Cd pollution have not been reported. In this study, a pot experiment was conducted with two aromatic rice varieties and a soil Cd concentration of 30 mg/kg. Five NanoSe treatments were applied at distinct growth stages: (T1) at the initial panicle stage, (T2) at the heading stage, (T3) at the grain-filling stage, (T1+2) at both the panicle initial and heading stages, and (T1+3) at both the panicle initial and grain-filling stages. A control group (CK) was maintained without any application of Se. The results showed that, compared with CK, the T1+2 and T1+3 treatments significantly reduced the grain Cd content. All NanoSe treatments increased the grain Se content. The grain number per panicle, 1000-grain weight, and grain yield significantly increased due to NanoSe application under Cd pollution. The highest yield was recorded in T3 and T1+3 treatments. Compared with CK, all NanoSe treatments increased the grain 2-acetyl-1-pyrroline (2-AP) content and impacted the content of pyrroline-5-carboxylic acid and 1-pyrroline which are the precursors in 2-AP biosynthesis. In conclusion, the foliar application of NanoSe significantly reduced the Cd content, increased the Se content, and improved the grain yield and 2-AP content of aromatic rice. The best amendment was applying NanoSe at both the panicle initial and grain-filling stages.

Abstract:Most studies have focused on regulation in a metabolic pathway in response to exogenous selenium under cadmium stress, rather than the change of key factors in soil and pepper plants. In this study, the correlations in environmental variables, microorganisms, metabolic pathways, Se and Cd morphology under nano-Se intervention were examined using metabolomics and microbial diversity in rhizosphere soil and pepper plants. The principal forms of Se in the soils were Se (VI) and SeCys, while SeMet and MeSeCys were the main components in the root, stem, leaves, and fruits in the treatment of nano-Se (5 and 20 mg/L) relative to the control. Soil enzymes,metabolites (fluorescein diacetate, urease, brassinolide, and p-hydroxybenzonic acid), and plant metabolites (rutin, luteolin, brassinolide, and abscisic acid) were remarkably enhanced by nano-Se fortification. The bio-enhancement of nano-Se can boost the beneficial microorganisms of Gammaproteobacteria, Alphaproteobacteria, Bacteroidia, Gemmatimonadetes, Deltaproteobacteria, and Anaerolineae in rhizosphere soil. Changes in microbial community were found to be strongly linked to the environment index, enzymes, soil metabolites, Se forms, which reduced Cd bioavailability and Cd accumulation in pepper plants. In conclusion, the nano-Se application integrates soil-plant balance by improving soil qualities and assigning signaling molecule levels in rhizosphere soil and pepper plants.
Absorption and Bio-Transformation of Selenium Nanoparticles by Wheat Seedlings (Triticum aestivum L.)

Abstract:Elemental selenium is one of the dominant selenium species in soil, but the mechanism of its uptake by plants is still unclear. In this study, nanoparticles of elemental selenium (SeNPs) with different sizes were prepared, and their uptake and transformation in wheat (Triticum aestivum L.) were analyzed in hydroponic experiments by HPLC-ICP-MS. We found that the SeNPs can be absorbed by wheat seedlings, and the process is energy independent. The addition of aquaporins inhibitor caused 92.5 and 93.4% inhibition of chemosynthesized SeNPs (CheSeNPs) and biosynthesized SeNPs (BioSeNPs) absorption by wheat roots, respectively. The 40 nm SeNPs uptake by wheat roots was 1.8-fold and 2.2-fold higher than that of 140 and 240 nm, respectively. The rate of SeNPs uptake in wheat was much slower than that of selenite [Se (IV)], and CheSeNPs were more efficiently absorbed than BioSeNPs. The SeNPs were rapidly oxidized to Se (IV) and converted to organic forms [selenocystine (SeCys2), se-methyl-selenocysteine (MeSeCys), and selenomethionine (SeMet)] after they were absorbed by wheat roots. Additionally, we demonstrated that the aquaporin function in some way is related to the absorption of SeNPs. The particle size and synthesis method of the SeNPs affected their uptake rates by plants. Taken together, our results provide a deep understanding of the SeNPs uptake mechanism in plants.

Abstract:The contamination of paddy fields by cadmium and lead is a major issue in China. The consumption of rice grown in heavy metals contaminated areas poses severe health risks to humans, where bioavailability and bioaccessibility remains the critical factor for risk determination. Selenium nanoparticles (Se-NPs) can mitigate the toxicity of heavy metals in plants. However, there exists limited information regarding the role of Se-NPs in dictating cadmium (Cd) toxicity in rice for human consumption. Moreover, the impact of Se-NPs under simultaneous field and laboratory controlled conditions is rarely documented. To address this knowledge gap, a field experiment was conducted followed by laboratory scale bioavailability assays. Foliar application of Se-NPs and selenite (at 5, 10 mg L-1) was performed to assess their efficiency in lowering Cd accumulation, promoting Se biofortification in rice grains, and evaluating Cd exposure risk from contaminated rice. Obtained results indicate that foliar treatments significantly reduced the heavy metal accumulation in rice grains. Specifically, Se-NP 10 mg L-1 demonstrated higher efficiency, reducing Cd and Pb by 56 and 32 % respectively. However, inconsistent trends for bioavailable Cd (0.03 mg kg-1) and bioaccessible (0.04 mg kg-1) were observed while simulated human rice intake. Furthermore, the foliage application of Se-NPs and selenite improved rice quality by elevating Se, Zn, Fe, and protein levels, while lowering phytic acid content in rice grains. In summary, this study suggests the promising potential of foliage spraying of Se-NPs in lowering the health risks associated with consuming Cd-contaminated rice.
