QSAR Tools for Toxicity Prediction in Risk Assessment—comparative Analysis

Abstract:Computational toxicology techniques, represented by quantitative structure–activity relationship models, enable high-throughput prediction of chemical toxicity parameters, which are necessary for chemical risk assessment. In this chapter, a total of eight commonly used toxicity prediction software packages were collected and compared in terms of basic information and modeling methods, including development background, prediction endpoints, user experience, types of prediction endpoints, raw datasets, structural characterization methods, modeling algorithms, and application domain characterization methods. The results show that the current software packages mainly cover 10 types of toxicological data in two categories: health data and ecotoxicology data. The modeling methods of different toxicity prediction software packages have different characteristics in terms of assessment endpoint types, raw datasets, modeling algorithms, structural characterization methods, and application domain characterization. Current toxicity prediction software packages still have much room for improvement in terms of information completeness, raw dataset quality, and toxicity mechanisms.
