NKD2 As a Mediator of IFIX Anti-Oncogene-Induced Wnt Signaling and Epithelial-Mesenchymal Transition In Human Oral Squamous Cell Carcinoma

Abstract:Background: Activation of the human interferon-inducible protein X (IFIX) isoform is linked to a stable cytoskeleton and the inhibition of epithelial-mesenchymal transition (EMT). However, the precise mechanisms and pathways underlying IFIX-mediated oncogenesis remain poorly understood.Methods and Results: IFIX overexpression and knockdown in CAL-27 and SCC-25 oral squamous cell carcinoma cells induced active variations in the expression of E-cadherin, N-cadherin, vimentin, and Snail, as well as changes in Wingless/Integrated (Wnt) signaling. These findings were substantiated by data from oral squamous cell carcinoma (OSCC) cells with IFIX overexpression and knockdown, indicating a link between IFIX and EMT. Quantitative reverse transcription-PCR and western blotting revealed that Wnt3a and Wnt4 pathway components were regulated in IFIX overexpressing or knockdown CAL-27 and SCC-25 cells, with naked cuticle 2 (NKD2) showing the strongest positive correlation. Both IFIX overexpression and knockdown modulated NKD2 expression. The silencing NKD2 mimicked the phenotypic effects of IFIX knockdown by inhibiting E-cadherin expression and increasing N-cadherin, Snail, and vimentin expression. Silencing of NKD2 restored the anti-carcinogenic phenotype associated with IFIX overexpression in terms of cell proliferation, invasion, and migration.Conclusions: Our results provide mechanistic insights into the anti-oncogenesis effect of IFIX in OSCC.
