Abstract:Starch is the predominant reserve in rice (Oryza sativa L.) endosperm, which is synthesized by the coordinated efforts of a series of starch biosynthetic-related enzymes in the form of a multiple enzyme complex. Whether the enzyme complex changes during seed development is not fully understood. Here, we investigated the dynamic change in multi-protein complexes in an indica rice variety IR36 (wild type, WT) and its BEIIb-deficient mutant (be2b) at different developmental stages. Gel permeation chromatography (GPC) and Western blotting analysis of soluble protein fractions revealed most of the enzymes except for SSIVb were eluted in smaller molecular weight fractions at the early developing stage and were transferred to higher molecular weight fractions at the later stage in both WT and be2b. Accordingly, protein interactions were enhanced during seed development as demonstrated by co-immunoprecipitation analysis, suggesting that the enzymes were recruited to form larger protein complexes during starch biosynthesis. The converse elution pattern from GPC of SSIVb may be attributed to its vital role in the initiation step of starch synthesis. The number of protein complexes was markedly decreased in be2b at all development stages. Although SSIVb could partially compensate for the role of BEIIb in protein complex formation, it was hard to form a larger protein complex containing over five proteins in be2b. In addition, other proteins such as PPDKA and PPDKB were possibly present in the multi-enzyme complexes by proteomic analyses of high molecular weight fractions separated from GPC. Two putative protein kinases were found to be potentially associated with starch biosynthetic enzymes. Collectively, our findings unraveled a dynamic change in the protein complex during seed development, and potential roles of BEIIb in starch biosynthesis via various protein complex formations, which enables a deeper understanding of the complex mechanism of starch biosynthesis in rice.

Abstract:Loss of starch branching enzyme IIb (BEIIb) leads to altered starch structure and increased amylose content. The changes in starch fine structure and function during seed development were investigated as a result from differentially expressed genes between wildtype (WT) and be2b rice. The expression patterns of all starch synthesis related genes except the AGPS1 were altered in be2b. From five to 15 days after flowering (DAF), the amylose content and proportion of A chains of amylopectin increased, while those of B2, B3 chains and average chain length declined in both WT and be2b. The mutant had a C-type crystalline pattern and a higher relative crystallinity (RC) at five and 10 DAF, which was transferred to a B-type and a lower RC at 15 DAF in be2b, while the WT had A-type starch at all developmental stages. A possible model for amylose and amylopectin structure in WT and be2b was proposed.

Abstract:Physicochemical properties of endosperm starches in milled rice determine its cooking and eating quality. Amylose is synthesized by granule-bound starch synthase I (GBSSI), whilst amylopectin is synthesized by the synergistic activities of starch synthases (SSs), branching enzymes (BEs) and debranching enzymes (DBEs). However, the complexes formed by starch biosynthetic enzymes are not well characterized. Gene expression profiles and protein complexes were determined in white-core (GM645) and waxy (GM077) mutants derived from a high amylose indica rice Guangluai 4 (GLA4). In GM645, genes including AGPS1, GBSSI, SSIIa, BEI, BEIIa, BEIIb, PUL, ISA1 and SP were significantly downregulated during seed development. In GM077, the expression levels of AGPL2, AGPS1, AGPS2b, SSIIIa, BEI, PUL and ISA1 were significantly upregulated. Co-immunoprecipitation assays revealed interactions of SSs-BEs, SSs-PUL and BEs-PUL in developing seeds. However, weak SSI-SSIIa interaction was detected in GM077, whilst SSI-PUL interaction was absent. Weak interaction signals for SSI-SSIIa, SSIIa-BEI, SSIIa-BEIIb, BEI-BEIIb and SSI-BEI were also observed in GM645. These results suggest that the protein-protein interactions for starch biosynthesis are modified in mutants, which provides insight into the mechanisms of starch biosynthesis, particularly in indica rice.

Abstract:This study examined the effects of the combinations of Waxy (Wx) and starch branching enzyme IIb (BEIIb) alleles on starch fine structure and functional properties in indica rice cultivars. The results showed that be2b mutant starches with BEIIb deficiency had higher amylose content, shorter amylose long chains, a higher proportion of amylopectin long chains and molecular order, but a lower proportion of amylopectin short chains and relative crystallinity, resulting in higher gelatinization temperature but lower enthalpy and paste viscosity. Compared with the rice lines carrying Wxb allele, Wxa allele contributed to relatively higher amylose content, longer amylopectin chains, less short-range ordered structure and lower relative crystallinity, leading to a little lower gelatinization enthalpy. This study provides new insight into structure-function relations among rice lines with different allele combinations of starch synthesis related genes, which is a useful strategy for rice quality breeding.

Abstract:Starch biosynthesis is controlled by multiple enzymes, including granule-bound starch synthase I (GBSSI), soluble starch synthases (SSs), branching enzymes (BEs), and debranching enzymes (DBEs). Although the role of individual isoforms has been primarily elucidated, the precise information about how they work together in the synthesis of specific amylose and amylopectin chains is still unclear. In this study, starch molecular chain-length distributions (CLDs) of five rice varieties with different amylose contents were measured by fluorophore-assisted carbohydrate electrophoresis and size-exclusion chromatography and fitted with two mathematical models, and the protein abundance of 11 starch synthesis-related enzymes was measured by western blotting. The correlation between model fitting parameters of amylose and amylopectin CLDs demonstrated that amylose and amylopectin syntheses are closely dependent. GBSSI could interact with BEI, BEIIb, SSIIa, SSIVb, ISA1, PUL, and PHO1 to synthesize the amylopectin intermediate and long chains as well as amylose chains. In addition, the interaction among SSIVb and SSI, SSIIa, BEI, BEIIb, ISA1, and PUL possibly suggests that SSIVb assists them to synthesize the amylopectin chains. The results can help understand the mechanisms about the functional interaction of different enzyme isoforms in starch biosynthesis.

Abstract:Starch branching enzyme IIb (BEIIb) and soluble starch synthase IIa (SSIIa) play important roles in starch biosynthesis in cereals. Deficiency in the BEIIb gene produces the amylose extender (ae) mutant rice strain with increased amylose content (AC) and changes in the amylopectin structure. The SSIIa gene is responsible for the genetic control of gelatinization temperature (GT). The combined effects of BEIIb and SSIIa alleles on the AC, fine structures, and physicochemical properties of starches from 12 rice accessions including 10 recombinant inbred lines (RIL) and their two parents were examined in this study. Under the active BEIIb background, starches with the SSIIa-GC allele showed a higher GT than those with the SSIIa-TT allele, resulting from a lower proportion of A chain and a larger proportion of B1 chains in the amylopectin of SSIIa-GC. However, starch with the BEIIb mutant allele (be2b) in combination with any SSIIa genotype displayed more amylose long chains, higher amylose content, B2 and B3 chains, and molecular order, but smaller relative crystallinity and proportion of amylopectin A and B1 chains than those with BEIIb, leading to a higher GT and lower paste viscosities. These results suggest that BEIIb is more important in determining the structural and physicochemical properties than SSIIa. These results provide additional insights into the structure-function relationship in indica rice rather than that in japonica rice and are useful for breeding rice with high amylose content and high resistant starch.

Abstract:Amylopectin is synthesized by the coordinated actions of many (iso)enzymes, including ADP-glucose pyrophosphorylase (AGPase), starch synthases (SSs), branching enzymes (BEs), and debranching enzymes (DBEs). Here, two polymorphic forms of starch synthase I (SSI) and pullulanase (PUL) in rice-developing seeds, designated as SSI-1/SSI-2 and PUL-1/PUL-2, were discovered for the first time by zymographic analysis. The SSI and PUL polymorphisms were strongly associated with the SSI microsatellite marker (p = 3.6 × 10(-37)) and PUL insertion/deletion (InDel) markers (p < 3.6 × 10(-51)). Western blotting and mass spectrometric analysis confirmed that the polymorphic bands were truly the SSI and PUL enzymes. Only one non-synonymous variation in SSI DNA sequence (the SNP A/G) causing the change of the amino acid K438 to E438 was observed, which coincided well with the polymorphic forms of SSI. Nine non-synonymous variations were found between PUL-1 and PUL-2. Two non-synonymous variations of PUL (F316L and D770E) were identified by mass spectrometric analysis, but all of the variations did not change the structure of PUL. The co-immunoprecipitation results revealed the differences in protein-protein interaction patterns, i.e., strong or weaker signals of SSI-BEI and SSI-BEIIb, between the two forms of SSI. The results will enhance our understanding of SSI and PUL properties and provide helpful information to understand their functions in starch biosynthesis in rice endosperm.

Abstract:Rice grain-filling is an important agronomic trait that contributes greatly to grain weight.A grain mutant from the japonica cultivar Nipponbare by mutagenesis with ethyl methane sulfonate (EMS), which had no accumulation of starch granules in endosperm with a transparent liquid during grain filling stage, was used to analyze the caryopsis development and starch biosynthesis metabolism in present study.Measurement of soluble substances in the liquid of developing endosperm showed that there was remarkably higher soluble sugar content in this no starch mutant.Semi-quantitative reverse transcription-PCR (RT-PCR) analysis of the starchsynthesizing genes revealed that soluble starch synthase1 (SSS1) gene could be normally expressed in the mutant.Substantially lower expressions of starch branching enzyme1 (SBE1), isoamylase1 (ISA1) and pullulanase (PUL) were detected in the no starch mutant compared with the wild type, whereas the expression of ADP-glucose pyrophosphorylase large subunit 1 (AGPL1) and ADP-glucose pyrophosphorylase small subunit 1 (AGPS1) were visibly increased.The measurements of the key enzymatic activities involving in starch biosynthesis suggested that there was a significant reduction in the activities of starch debranching enzyme (DBE) and starch branching enzyme (SBE) in the transparent liquid endosperm of the no starch mutant, which coincided with their down-regulation expression at transcriptional levels.These results suggest that the transparent liquid with no starch accumulation at the filling stage in the glume of the mutant was likely due to abnormal transcriptional and enzymatic activities related to the DBE and/or SBE genes loci mutant.

Abstract:With the rice sugary endosperm mutant (Sug-1 1 )and its wild-type (Zhonghua 1 1 )as materials,the total soluble sugar,sucrose contents,starch accumulation in filling grains and the starch physicochemical properties were compared to further reveal the temporal patterns of sugar content,several key enzymes activities and their isoform genes transcriptional expression level during the grain filling so as to analyze the physiological mechanism for Sug-1 1 whose sugars contents change and 1000-grain weight decline from the point of starch biosynthesis pathway in rice grains.The result showed that the differences in soluble sugar and sucrose contents between Sug-1 1 and its wild-type were not obvious at the initial stage of grain filling,but with the filling process it gradually became apparent.Secondly, the amylose(Am)content and BV(Am)of Sug-1 1 were significantly decreased while the starch solubility and BV (amylopectin,Ap)were increased compared with Zhonghua 1 1 ,indicating that the physicochemical properties of grain starch were significantly affected in the sugary endosperm mutant.Finally,among the key enzymes in starch synthetic metabolism,debranching enzyme(DBE),existed in Sug-1 1 seeds,were quite different from its wild type in activity and dynamic in the process of grain filling,and PUL transcriptional expression level and DBE activity strikingly decreased at the middle-late stage,resulting in the declined starch accumulation and increased sugar content in Sug-1 1 . However the poor filling and 1000-grain weight decline were,to some extent,associated with dropped ADPGase activity at the early grain filling stage.
