Abstract:Background: Secreted protein acidic and rich in cysteines-like 1 (SPARCL1) is an extracellular matrix glycoprotein with malignancy suppressing potential. The hypothesis that SPARCL1 reduces cancer invasiveness and predicts better survival in colorectal cancers (CRC) was investigated. Material and Methods: Stable SPARCL1 transfectants, RKO-SPRCL1, and corresponding vector control were constructed and implanted into nude mice to generate a mouse xenograft model of liver metastasis. Also, a retrospective outcome study was conducted on the COH set (222 CRCs) and ZJU set (412 CRCs). The protein expression level of SPARCL1 was determined by immunohistochemistry. The Kaplan-Meier and COX analyses were employed for survival analysis. The association of SPARCL1 with MET was sought by RT-PCR and Western blot analysis. Results: The ectopic expression of SPARCL1 significantly reduced the potential for anchorage-independent growth, migration and invasion, and it induced cell differentiation in RKO and SW620 cells. In mouse xenograft model, the expression of SPARCL1 significantly reduced the liver metastasis (P = 0.001). The patient-based studies revealed that the expression of SPARCL1 was related to better differentiation (P < 0.01), less lymph node involvement (Odds Ratio, OR=0.67, 95% CI 0.45-1.00) and less distant metastasis (OR=0.38, 95% CI 0.18-0.79). The Kaplan-Meier and COX analysis demonstrated that the expression of SPARCL1 was associated with better overall survival (Log–rank P < 0.01; hazard ratio, HR=0.57, 95% CI 0.39-0.84). Transfection of SPARCL1 induced MET of colon cancer cells. Conclusion: SPARCL1 functions as a tumor suppressor promoting differentiation possibly via MET, which inhibits the aggressiveness of CRC. Abstract:Proceedings: AACR 102nd Annual Meeting 2011‐‐ Apr 2‐6, 2011; Orlando, FL Secreted protein that's acidic and rich in cysteine, like 1(SPARCL1), an extracellular de-adhesion glycoprotein, was down-regulated in several solid cancers such as non-small cell lung cancer and pancreatic cancer. Our previous study revealed that expression of SPARCL1 was negatively related to liver metastasis of colorectal cancer (CRC). To clarify the function of SPARCL1 and its mechanism in CRC, the overexpression cell clone, RKO-SPARCL1, was created for illustrating biological behavior of SPARCL1 in vitro and in vivo. It was shown that SPARCL1 significantly reduced colony formation (p=0.027), invasion ability (p=0.001) and adhesion (p=0.04) of CRC cells in vitro. In comparison with vector control (RKO-pLXSN), RKO-SPARCL1 displayed significantly reduction in liver metastatic nodules in intrasplenic injection nude mice model (p=0.009). Meanwhile, tissue samples from 412 CRC patients were reassembled as tissue microarray for an outcome study. The antibody against SPARCL1 was developed for Immunohistochemistry (IHC) staining to assess the expression level of SPARCL1. Uni- and multivariate Logistic analysis demonstrated that SPARCL1 was negatively related with poorly tumor differentiation (p=0.001), lymph node metastasis (Odds ratio, OR=0.67, 95% 0.45-1.00) and distant metastasis (OR=0.39, 95% 0.18-0.82). The Kaplan-Meier analysis revealed that overexpression of SPARCL1 was significantly related to better overall survival in CRCs (p=0.004). Furthermore, the multivariate COX proportional hazard analysis demonstrated that SPARCL1 significantly reduced the risk of death from CRC (Hazard ratio, HR=0.57, 95% 0.39-0.84). Based on bioinformatics gene ontology analysis for gene expression microarray, overexpression of SPARCL1 significantly inhibited focal adhesion pathway and other tumor-associated oncogenes, such as S100A4 and ERBB3, in RKO cell line. Therefore, it is concluded that SPARCL1 may function as a tumor suppressor inhibiting the aggressiveness of cancer cells and reducing the risk of death from CRCs. Abstract:Abstract Introduction: Despite the numerous drugs available for patients with metastatic colorectal cancer (mCRC), median overall survival for this group of patients remains at ~20-24 months, with no significant advances in the last 7 years. In the US ~50,000 patients die each year from mCRC refractory to systemic therapy. Inhibiting angiogenesis as a therapy has led to a great deal of enthusiasm. However, the overall benefit of classical-antiangiogenic therapy remains modest and has not lived up to their expectations in treating CRC. Our recent studies suggest that VEGF intracrine signaling, rather than autocrine/paracrine signaling, regulates cell survival in CRC cells. Further studies to understand the significance and mechanisms of this novel function have led us to identify factors that intracellularly interact with VEGF. Our studies further indicate that one such interacting protein, SPECC1L, may have a significant role in CRC cell proliferation and may be a potential target in mCRC therapy. Methods: Lysates from CRC cells expressing Myc-tagged VEGF protein were immunoprecipitated and analyzed by mass spectrometry to identify VEGF-interacting protiens. SPECC1L was identified as a co-precipitated protein with high level of confidence. SPECC1L was depleted using siRNA and effects of such depletion on CRC cell growth and morphology were measured by cell growth assays (MTT), microscopy, FACS and western blot analyses. Localization of the protein and its interaction with microtubules and actin were visualized by immunostaining of FLAG-tagged recombinant SPECC1L protein. Results: SPECC1L was identified as a protein that co-immunoprecipitated with Myc-tagged VEGF in CRC cells using mass spectroscopy. Previous literature suggests a role for SPECC1L in cell division. As a fraction of VEGF overexpressing CRC cells have a large multinucleated phenotype, likely arising due to defects in cell division, it was hypothesized that a VEGF mediated regulation of SPECC1L may lead to such phenotype. Depletion of SPECC1L by siRNAs in multiple CRC cell lines led to strong defects in cell division. The effects of SPECC1L depletion were manifested as accumulation of doublet-cells failing to complete cytokinesis following mitosis and resulted in reduced cell proliferation. Failure to complete cell division also led to the formation of multinucleated cells and enhanced cell death. Conclusions: Inhibition of SPECC1L strongly inhibits CRC cell proliferation and enhances cell death. Abstract:Background Secreted protein acidic and rich in cysteine-like 1 (SPARCL1) plays an important role in tumor pathogenesis. We aim to evaluate the clinical significance and potential biological roles of SPARCL1 in colorectal cancer (CRC). Methods Datasets from the Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) databases were downloaded to evaluate the expression levels of SPARCL1 in CRC. Receiver operating characteristic (ROC) curve was constructed to evaluate the diagnostic value of SPARCL1. Then, comprehensive database search was conducted for published clinical studies to explore clinical significance of SPARCL1. In addition, coexpression genes of SPARCL1 were identified through the cBioPortal database and enrichment analysis of SPARCL1 and its coexpression genes were performed by the “clusterProfiler” R package. Finally, the correlations between SPARCL1 and tumor microenvironment scores, tumor-infiltrating immune cells in CRC were determined by “ESTIMATE” and “GSVA” R packages. Results SPARCL1 was significantly downregulated in CRC tissues, and SPARCL1 showed high accuracy for diagnosis of primary CRC in both GEO and TCGA datasets. Pooled results from published clinical studies showed SPARCL1 expression was associated with differentiation (OR = 1.89, 95% CI: 1.38-2.59), tumor stage (OR = 0.47, 95% CI: 0.29-0.77), distant metastasis (OR = 0.53, 95% CI: 0.33-0.84), and overall survival (HR = 0.56, 95% CI: 0.43-0.74). SPARCL1 and its top 300 coexpression genes were involved in several KEGG pathways, such as focal adhesion, cell adhesion molecules, PI3K-Akt signaling pathway, cGMP-PKG signaling pathway, and ECM-receptor interaction. Besides, the SPARCL1 expression was significantly correlated with stromal score, immune score, ESTIMATE score, and diverse immune cells. Conclusion SPARCL1 significantly correlated with clinicopathological features and tumor microenvironment in CRC.

Abstract:Secreted protein acidic and rich in cysteines-like protein 1 (SPARCL1) has been implicated in tumor initiation, formation, and progression of various cancers, yet its role in hilar cholangiocarcinoma remains largely uncharacterized. In the present study, tissue microarrays containing resected hilar cholangiocarcinoma specimens from 92 patients were used to evaluate the expression of SPARCL1 protein by immunohistochemistry (IHC). In vitro assays were used to determine the effect of SPARCL1 overexpression on cell growth and migration. Loss of SPARCL1 expression was observed in 46 (50.0 %) of the 92 primary tumors. SPARCL1 expression is inversely associated with poorly or undifferentiation specimens ( P = 0.030) in addition to lymph node metastasis ( P = 0.047). Survival analysis demonstrated that SPARCL1 is an independent factor in predicting the outcome of patients with hilar cholangiocarcinoma. SPARCL1 overexpression suppressed tumor cell migration in vitro by inhibiting MMP-9, MMP-2, Vimentin, and Fibronectin expression, whereas did not inhibit cell proliferation in vitro. Our results suggest that loss of SPARCL1 is involved in the tumorigenesis of hilar cholangiocarcinoma and may serve as a novel molecular biomarker for patients’ outcome.

Abstract:Background: Secreted protein acidic and rich in cysteine-like 1 (SPARCL1) was a kind of extracellular matrix glycoprotein. SPARCL1 was strongly inhibited in most cancers. However, the potential functions of SPARCL1 in the pan-cancer cohort have not been widely studied. Methods: We evaluated the transcriptional level and the prognostic value of SPARCL1 in 33 types of cancer and revealed the relationship between genetic alterations of SPARCL1 and the tumor mutation burden. Meanwhile, we assessed the correlations between SPARCL1 and tumor-infiltrating lymphocytes across cancers. Results: The transcriptional level of SPARCL1 was inhibited in most cancers. Although SPARCL1 was down-regulated in most cancers, SPARCL1 might play a protective or detrimental role in different cancers. We demonstrated that mutation count was elevated in the altered SPARCL1 group in several cancers. Additionally, we found a significant positive correlation between SPARCL1 and macrophage infiltration levels in most cancers. Especially, marker sets of M2 macrophages were strongly related to SPARCL1 in cholangiocarcinoma, colon adenocarcinoma, rectum adenocarcinoma, and pancreatic adenocarcinoma. Conclusion: Our study found that SPARCL1 might work as a biomarker for prognosis and immune infiltration in pan-cancer analysis.
Secreted Protein Acidic and Rich in Cysteine Expression in Human Colorectal Cancer Predicts Postoperative Prognosis

Abstract:OBJECTIVE: Secreted protein acidic and rich in cysteine (SPARC) is an extracellular matrix glycoprotein involved in cell proliferation, migration and angiogenesis. The aim of this study was to assess its expression in colorectal cancer, see whether and how it correlates with clinicopathological features, and evaluate its potential prognostic significance.PATIENTS AND METHODS: SPARC expression was detected by microarrays containing 847 immunohistochemically stained specimens, and further correlated with the clinicopathological and prognostic data. The prognostic significance of its expression was assessed using Kaplan-Meier survival with log-rank tests. Multivariate regression utilizing Cox's proportional hazard model was used to evaluate prognostic factors.RESULTS: SPARC expression in the normal colorectal mucosa and colorectal cancer tissue was significantly different (p < 0.001). Low SPARC expression was found to be associated with poor prognosis, and it was unfavorably correlated with overall survival and disease-free survival in colorectal cancer patients. In addition, SPARC expression in surrounding mesenchymal and stromal cells, bowel wall invasion, lymph node metastasis, and distant metastasis were independent prognostic factors for overall survival and disease-free survival.CONCLUSIONS: Reduced expression of SPARC in colorectal cancer tissue is associated with poor prognosis and aggressive clinicopathological features. Therefore, SPARC expression could potentially be used as a prognostic predictor for colorectal cancer patients.

Abstract:Abstract Secreted protein, acidic and rich in cysteine‐like 1 (SPARCL1) belongs to the SPARC family of matricellular proteins. However, underlying functions of SPARCL1 in bladder cancer (BCa) remain understudied. We performed an integrated search for the expression patterns of SPARCL1 in relation to various clinicopathological features of BCa. We then carried out Gene Ontology (GO) enrichment analysis, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis, and gene set enrichment analysis (GSEA). Furthermore, we investigated the correlations between SPARCL1 and immunological features, such as tumour mutation burden (TMB), immune activation processes, immune checkpoint expression, tumour immune dysfunction and exclusion (TIDE) scores, and chemotherapeutic sensitivity in BCa. Our analysis revealed that SPARCL1 was downregulated across multiple cancers. In BCa, elevated SPARCL1 was linked with advanced histopathologic stage, higher T and N stage, and poorer prognosis in the clinical cohort. In vitro experiments demonstrated that increased SPARCL1 expression inhibited cell proliferation, migration, and invasion. Additionally, highly expressed SPARCL1 was linked to elevated immune, stromal and ESTIMATE scores, as well as an increase in naive B cells, M2 macrophages, and resting mast cells. We observed a moderate correlation between SPARCL1 expression and CD163, VSIG4 and MS4A4A, which are markers of M2 macrophages. Furthermore, SPARCL1 expression was positively related to TMB, immune activation processes, TIDE scores, immune checkpoint expression, and chemotherapeutic sensitivity in BCa. Our study highlights the potential involvement of SPARCL1 in macrophage recruitment and polarization and suggests its utility as a biomarker for prognosis in BCa.

Abstract:AIM: To investigate the expression of markers that are correlated with the prognosis of colorectal cancer (CRC) patients.METHODS: One hundred and fifty-six CRC patients were followed up for more than 3 years after radical surgery. Immunohistochemical (IHC) analysis was performed to detect the expression of 14 pathway-related markers (p53, APC, p21ras, E-cadherin, endothelin-B receptor, Shp2, ADCY-2, SPARCL1, neuroligin1, hsp27, mmp-9, MAPK, MSH2 and rho) in specimens from these patients. Bioinformatics analysis involving a Support Vector Machine (SVM) was used to determine the best prognostic model from combinations of these markers.RESULTS: Seven markers (SPARCL1, Shp2, MSH2, E-cadherin, p53, ADCY-2 and MAPK) were significantly related to the prognosis and clinical pathological features of the CRC patients (P < 0.05). Prognostic models were established through SVM from combinations of these 7 markers and proved able to differentiate patients with dissimilar survival, especially in stage II/III patients. According to the best prognostic model, the p53/SPARCL1 model, patients having high p53 and low SPARCL1 expression had about 50% lower 3-year survival than others (P < 0.001).CONCLUSION: SPARCL1, Shp2, MSH2, E-cadherin, p53, ADCY-2 and MAPK are potential prognostic markers in CRC. Abstract:Aim: The authors previously found that SPARCL1 functions to suppress colorectal cancer metastasis. Here, the epigenetic mechanism of SPARCL1 regulation and its relationship with clinicopathological features in colon cancer were investigated. Materials & methods: SPARCL1 expression was evaluated by immunohistochemistry staining in a tissue array containing 271 left-sided colon cancer samples and 257 right-sided colon cancer samples. In vivo and in vitro DNA methylation states were measured by biochemical sulfide potential assay. The transcription and DNA methylation states in cells were altered by siRNA or decitabine treatment, respectively. Cellular motility properties were compared through transwell assay. Results & conclusion: SPARCL1, mediated by its DNA methylation, may arrest colorectal carcinoma motility. Furthermore, SPARCL1 expression is higher and may have a specific prognostic value in left-sided colon cancer.

Abstract:To analyze the heterogeneous functions of secreted protein acidic and rich in cysteine (SPARC) from different origins and in different tumor microenvironments with the purpose of determining its clinical significance. Methods: The PubMed, CINAHL, Cochrane, Web of Science and Embase databases were utilized. Studies that focused on the effects of SPARC expression on solid tumor progression and clinical implications were used. The different outcomes including overall survival and disease-free survival were analyzed to evaluate their relations with tumor- and stroma-derived SPARC expression. Results: A total of 26 studies including 5,939 patients were enrolled in the present meta-analysis. Tumor-derived SPARC overexpression was significantly related with poor overall survival (hazard ratio: 1.478; 95% CI: 1.143-1.910; p=0.003), and a similar tendency was also observed in disease-free survival (hazard ratio: 1.476; 95% CI: 0.993-2.195; p=0.054). However, the hazard ratios for overall survival and disease-free survival did not present a statistical trend in stromal SPARC overexpression. Tumor type subgroup analysis revealed marked heterogeneity among outcomes. In pancreatic cancer, SPARC overexpression in the stroma was significantly associated with poorer overall survival and disease-free survival. In colorectal cancer, SPARC overexpression in the stroma was associated with better disease-free survival. Conclusion: For the majority of solid tumors, SPARC in cancer cells may be an unfavorable indicator for long-term survival for patients. As for stromal expression, SPARC indicates a poorer prognosis in pancreatic cancer, but a better disease-free survival in colorectal cancer. Secreted protein acidic and rich in cysteine might be a potential biomarker for solid tumor prognosis.
