Abstract:AbstractTumor cell survival in the hostile distant organ is a rate-limiting step in cancer metastasis. Bone marrow–derived myeloid cells can form a premetastatic niche and provide a tumor-promoting microenvironment. However, it is unclear whether these myeloid cells in the premetastatic site have any direct effect on tumor cell survival. Here, we report that chemokine CCL9 was highly induced in Gr-1+CD11b+ immature myeloid cells and in premetastatic lung in tumor-bearing mice. Knockdown of CCL9 in myeloid cells decreased tumor cell survival and metastasis. Importantly, CCL9 overexpression in myeloid cells lacking TGFβ signaling rescued the tumor metastasis defect observed in mice with myeloid-specific Tgfbr2 deletion. The expression level of CCL23, the human orthologue for CCL9, in peripheral blood mononuclear cells correlated with progression and survival of cancer patients. Our study demonstrates that CCL9 could serve as a good candidate for anti-metastasis treatment by targeting the rate-limiting step of cancer cell survival. Abstract:Abstract Tumor cell survival in the hostile distant organ is a rate-limiting step in cancer metastasis. However, it is unclear whether these myeloid cells in the premetastatic site have any direct effect on tumor cell survival. Here we report that chemokine CCL9 was highly induced in Gr-1+CD11b+ immature myeloid cells and in premetastatic lung of mice bearing 4T1 mammary tumor and B16 melanoma. Knockdown of CCL9 in myeloid cells decreased tumor cell survival and metastasis. The expression level of CCL23, the human orthologue for CCL9, in peripheral blood mononuclear cells correlated with progression and survival of cancer patients. Our studies identify CCL9 as a key pro-survival factor for cancer cells in the premetastatic organ. Interestingly, the expression of CCL9 in myeloid cells is regulated by TGF-β signaling as overexpression of CCL9 in myeloid cells lacking TGF-β signaling rescued the tumor metastasis defect observed in mice with myeloid-specific deletion of TGF-β signaling. These data together with our previous report of a fundamental role of myeloid TGF-β signaling in host compromised immune surveillance (Cancer Discovery 2013); demonstrate that myeloid-specific TGF-β signaling constitutes an essential component of the metastasis-promoting puzzle of TGF-β. This finding is different from TGF-β signaling in fibroblasts and a variety of epithelial cells, as well as T cells in which the TGF-β signaling functions as a tumor suppressor. This new understanding of TGF-β at the interface of tumor and host may provide insight into how TGF-β switches from a tumor suppressor to a tumor promoter, a long-standing challenge in cancer biology field. Our study demonstrates that CCL9 could serve as a good candidate for anti-metastasis treatment by targeting the rate-limiting step of cancer cell survival. Abstract:Objectives Spleen is involved in multiple diseases, the role of the spleen and spleen-derived factors in hepatocellular carcinoma (HCC) is still not clarified.Methods In the current study, a murine H22 orthotopic hepatoma model was established. Three groups were divided: normal mice, tumor-bearing mice with spleen-preserving, and tumor-bearing mice with splenectomy. Spleen and tumor weights were recorded by weeks 1 and 2. The proportion of myeloid-derived suppressor cell (MDSC) in peripheral blood and tumor tissue was detected using flow cytometry. Protein chip assay was used to compare the differential cytokines between normal liver supernatant and tumor supernatant. The common upregulated cytokines both in spleen and tumor were focused and analyzed using gene expression profiling interactive analysis (GEPIA) database. Enzyme-linked immunosorbent assay was performed to verify the chip result, and to examine CCL9 expression before and after splenectomy. Spleen MDSC was sorted using flow cytometry, and chemotaxis assay was performed to demonstrate whether CCL9 attracted spleen MDSC.Results The spleen enlarged during tumor progression, and compared with splenectomy group, there were faster tumor growth, shorter survival time, and higher proportions of MDSC in spleen-preserving group. Protein chip assay and GEPIA database revealed CCL9 was the most promising chemokine involved in HCC upregulated both in spleen and tumor tissue. CCL9 attracted MDSC in vitro, the level of CCL9 in tumor tissue was downregulated, and the percentage of MDSC was decreased after splenectomy.Conclusion The results demonstrate that CCL9 may be derived from spleen; it facilitated HCC growth via the chemotaxis of MDSC, targeting CCL9 may be a promising strategy in HCC treatment.

Abstract:Background & Aims: Different immune cells in tumor microenvironment shape tumor progression. CCL20 over-expression was reported as one of the "stemness" trait in TP53 mutated hepatocellular carcinoma (HCC). We aimed to understand the effect of CCL20 on HCC progression. Methods: In two HCC cohort patients (n=95, n=85 respectively), serum CCL20 concentration was quantified by using ELISA. Expressions of CCL20 and CCR6 in 41 paired HCC tumor and adjacent non-tumor tissues were determined by quantitative Real-Time PCR, confirmed by immunohistochemistry (CCL20) or by flow cytometry analysis (CCR6). Chemotaxis of splenocytes or purified CD19(+) B cells to tumor cell-derived CCL20, and angiogenesis of different CD19(+) B subtypes responding to tumor cell-derived CCL20 were measured in vitro. H22 murine hepatoma cells were inoculated into immunocompetent or immuno-deficient SCID mice, tumor growth and metastasis were monitored after the mice were treated with anti-CCL20 neutralizing antibody or depleted B cells by anti-CD20. Results: Elevation of pretherapy serum CCL20 in HCC patients and increase of CCR6 expression in HCC tissues were closely associated with tumor metastasis and disease poor prognosis. In HCC tissues, CCL20 expression was positively correlated with CCR6 (R-2=0.3134, P=0.0002), and CCR6 was exclusively identified in tumor infiltrated immune cells. CD19(+)CD5(+) B lymphocytes expressed higher CCR6, responded to tumor cell-derived CCL20 and enhanced angiogenesis in vitro. Neutralizing CCL20 activity in immunocompetent mice, not in SCID mice, attenuated tumor incidence, restrained tumor growth and distal metastasis. Tumor angiogenesis was significantly inhibited after CCL20 activity was blockade. In addition, inhibiting B lymphocyte infiltration into tumor mileum also attenuated tumor growth. Conclusions: Tumor cell-derived CCL20 interacts with CCR6 highly expressed CD19(+)CD5(+) B cells, to promote HCC progression, which might be via enhancing angiogenesis.
