Abstract:The exploration of original computed tomography (CT) imaging contrast agents with enhanced sensitivity and specificity is currently one of the major challenging tasks for precision medicine. Herein, we develop an innovative nanoprobe of dendrimer-stabilized gold nanoparticles (Au DSNs) linked with folic acid (FA) as a targeting ligand and diatrizoic acid (DTA) for specific enhanced tumor CT imaging. In current work, poly-(amidoamine) (PAMAM) dendrimers of generation 5 (G5) with amine termini were adopted to entrap Au NPs through a stepwise complexation/reduction method to achieve a higher Au loading than the conventional one-step complexation/reduction method. The prepared [(Au-0)(120)-G5.NH2] NPs were sequentially functionalized with diatrizoic acid (DTA), a typical CT contrast agent based on iodine(I), FA through a poly(ethylene glycol) (PEG) spacer, and carboxylated PEG monomethyl ether (mPEG-COOH), ended with complete acetylation of the leftover dendrimer amine termini. The generated Au DSNs-DTA-FA (Au core diameter = 5.9 nm) were thoroughly characterized. Our data reveal that the Au DSNs-DTA-FA containing Au and I dual radiodense elements are stable, display enhanced CT imaging performance, much higher than the single-radiodense elemental material solely based on Au or I, and possess a quite good cytocompatibility. With the demonstrated FA-rendered specific targeting, the developed Au DSNs-DTA-FA can be employed as a highly efficient nanoprobe for targeted enhanced CT imaging of cancer cells and a subcutaneous tumor model. Overall, the created Au DSNs-DTA-FA may be a powerful nanoprobe for specific enhanced CT imaging of various kinds of FA receptor-expressing tumors or biosystems.
Recent advances in multifunctional dendrimer-based complexes for cancer treatment

Abstract:The application of nanotechnology in biological and medical fields have resulted in the creation of new devices, supramolecular systems, structures, complexes, and composites. Dendrimers are relatively new nanotechnological polymers with unique features; they are globular in shape, with a topological structure formed by monomeric subunit branches diverging to the sides from the central nucleus. This review analyzes the main features of dendrimers and their applications in biology and medicine regarding cancer treatment. Dendrimers have applications that include drug and gene carriers, antioxidant agents, imaging agents, and adjuvants, but importantly, dendrimers can create complex nanosized constructions that combine features such as drug/gene carriers and imaging agents. Dendrimer-based nanosystems include different metals that enhance oxidative stress, polyethylene glycol to provide biosafety, an imaging agent (a fluorescent, radioactive, magnetic resonance imaging probe), a drug or/and nucleic acid that provides a single or dual action on cells or tissues. One of major benefit of dendrimers is their easy release from the body (in contrast to metal nanoparticles, fullerenes, and carbon nanotubes), allowing the creation of biosafe constructions. Some dendrimers are already clinically approved and are being used as drugs, but many nanocomplexes are currently being studied for clinical practice. In summary, dendrimers are very useful tool in the creation of complex nanoconstructions for personalized nanomedicine. Development of Magnetic Resonance Imaging Contrast Agents with PAMAM Dendrimer Cores

Abstract:Magnetic resonance imaging (MRI) contrast agents with dendrimer cores have larger proton relaxation enhancements and higher molar relaxivities, better imaging visualization and longer blood retention compared to those agents with small molecular weight. It suggests that this new class of contrast agent has the potential for diverse and extensive applications.The properties and application of polyamidoamine (PAMAM) dendrimer-based gadolinium complex contrast agents are reviewed briefly in the present paper. This entire series of dendrimer-based macromolecular MRI contrast agents have a spherical shape, and changes in molecular size alter the route of excretion. Smaller sized contrast agents(PAMAM-G2,G3,G4)are excreted through the kidney resulting in these agents being suitable as functional renal contrast agents. Larger sized contrast agents(PAMAM-G7,G8)are found better suited for use as blood pool contrast agents, and larger contrast agents(PAMAM-G6)are also useful for lymphatic imaging. Finally, macromolecular MRI contrast agents conjugated with either folate or with avidin are able to function as tumor-specific contrast agents and might also be employed as therapeutic drugs for gadolinium neutron capture therapy(Gd-NCT).
Design of biocompatible dendrimers for cancer diagnosis and therapy: current status and future perspectives.

Abstract:In the past decade, nanomedicine with its promise of improved therapy and diagnostics has revolutionized conventional health care and medical technology. Dendrimers and dendrimer-based therapeutics are outstanding candidates in this exciting field as more and more biological systems have benefited from these starburst molecules. Anticancer agents can be either encapsulated in or conjugated to dendrimer and be delivered to the tumour via enhanced permeability and retention (EPR) effect of the nanoparticle and/or with the help of a targeting moiety such as antibody, peptides, vitamins, and hormones. Imaging agents including MRI contrast agents, radionuclide probes, computed tomography contrast agents, and fluorescent dyes are combined with the multifunctional nanomedicine for targeted therapy with simultaneous cancer diagnosis. However, an important question reported with dendrimer-based therapeutics as well as other nanomedicines to date is the long-term viability and biocompatibility of the nanotherapeutics. This critical review focuses on the design of biocompatible dendrimers for cancer diagnosis and therapy. The biocompatibility aspects of dendrimers such as nanotoxicity, long-term circulation, and degradation are discussed. The construction of novel dendrimers with biocompatible components, and the surface modification of commercially available dendrimers by PEGylation, acetylation, glycosylation, and amino acid functionalization have been proposed as available strategies to solve the safety problem of dendrimer-based nanotherapeutics. Also, exciting opportunities and challenges on the development of dendrimer-based nanoplatforms for targeted cancer diagnosis and therapy are reviewed (404 references).

Abstract:<p>Cancer threatens the life and health of human beings, while the effective treatment of cancer is a major challenge. The design of fluorescent probe provides the strategy for developing effective therapeutic nanomaterials that can detect and identify cancer cells as well as achieve targeted and image-guided cancer therapy. Among the fluorescent nanomaterials studied, carbon dots (CDs) have attracted increasing attention owing to their most significant advantages in terms of diverse imaging features, high drug loading as well as low toxicity, and thus they have emerged as one of the most appealing candidates in the field of imaging-guided theranostics. In this review, we firstly overview the imaging analysis of cancer cells, including single-modal imaging and multi-modal imaging. After the introduction of diverse imaging features, we focus on the design of imaging-assisted drug delivery system. Finally, the current development of CDs in imaging-guided photothermal therapy (PTT) and photodynamic therapy (PDT) are further described.</p>

Abstract:Cancer phototherapy with non-invasiveness and high therapeutical efficiency has emerged as a hot spot research in cancer management. Various nanomaterials have been involved in the development of novel photoactive agents to overcome the current limitations in cancer phototherapy. Dendrimers, as an excellent nanocarrier with unique physicochemical properties, have received extensive attention and much effort has been made in the development of dendrimer-based hybrid platforms for photomedicine applications. Dendrimers can be entrapped with photosensitive agents within their internal cavities and be surface modified with reactive molecules, constructing multifunctional nanoplatforms for cancer treatment. In this review, we concisely survey the design of several different kinds of dendrimer-based nanohybrids for cancer photomedicine applications, and provide an overview of their recent applications in molecular imaging, single-modality photothermal therapy or photodynamic therapy, combination therapy, and theranostics of cancer. In addition, we also briefly discuss the future perspectives in the area of dendrimer-based nanohybrids for cancer photomedicine.

Abstract:Poly (amidoamine) (PAMAM) dendrimers are well-defined, highly branched macromolecules with numerous active amine groups on the surface. Because of their unique properties, PAMAM dendrimers have steadily grown in popularity in drug delivery, gene therapy, medical imaging and diagnostic application. This review focuses on the recent developments on the application in PAMAM dendrimers as effective carriers for drug and gene (pDNA, siRNA) delivery in cancer therapy, including: a) PAMAM for anticancer drug delivery; b) PAMAM and gene therapy; c) PAMAM used in overcoming tumor multidrug resistance; d) PAMAM used for hybrid nanoparticles; and e) PAMAM linked or loaded in other nanoparticles.

Abstract:Highly sensitive and safe contrast agents (CAs) are essential for magnetic resonance imaging (MRI) to achieve accurate tumor detection and imaging. Dendrimer-based macromolecular MRI contrast agents are advantageous owing to their tumor-targeting ability, enhanced imaging contrast and enlarged imaging window. However, most of them have drawbacks of non-degradability and thereby long-term retention in body and toxicity. Herein, a tumor-targeting biodegradable dendritic CA (DCA) (FA-PEG-G2-DTPA-Gd) was prepared from a polyester dendrimer conjugated with gadolinium (Gd) chelates and PEG chains with distal folic acid. The DCA had a high longitudinal relaxivity up to 17.1mM−1s−1, 4 times higher than the clinically used CA Magnevist. The MRI contrasted by FA-PEG-G2-DTPA-Gd outlined the inoculated tumor more clearly, and had much higher contrast enhancement for a much longer time than Magnevist. More importantly, the biodegradable FA-PEG-G2-DTPA-Gd gave much less Gd retentions in all the organs or tissues than non-degradable DCAs. Thus, the high efficiency in MRI contrast enhancement and low Gd retention merit it a promising CA for contrast enhanced tumor MRI.
Core–Shell Tecto Dendrimers Enable Enhanced Tumor MR Imaging Through an Amplified EPR Effect

Abstract:Development of nanoplatforms that can amplify the passive tumor targeting effect based on enhanced permeability and retention (EPR) effect is crucial for precision cancer nanomedicine applications. Herein, we present the development of core-shell tecto dendrimers (CSTDs) as a platform for enhanced tumor magnetic resonance (MR) imaging through an amplified EPR effect. In this work, poly(amidoamine) (PAMAM) dendrimers of generation 5 (G5) were decorated with beta-cydodextrin (CD) and then assembled with G3 PAMAM dendrimers premodified with adamantane (Ad) via supramolecular recognition of CD and Ad. The formed G5-CD/Ad-G3 CSTDs were conjugated with tetraazacyclododecane tetraacetic acid (DOTA)-Gd(III) chelators and further acetylated to neutralize the remaining CSTD periphery amines. We reveal that the formed CSTD.NHAc-DOTA(Gd) (CSTD-D-Gd) complexes have a narrow size distribution and satisfactory colloidal stability, and are cytocompatible within the concentration range studied. Compared to the single dendrimer counterpart of GS.NHAc-DOTA(Gd) (GS-D-Gd) complexes, the CSTD-D-Gd complexes with a higher molecular weight and volume possess a longer rotation correlation time, hence having a longitudinal relaxivity (r(1)) of 7.34 mM(-1) s(-1), which is 1.5 times larger than that of GS-D-Gd complexes (4.92 mM(-1) s(-1)). More importantly, the CSTD-D-Gd complexes display better permeability in the three-dimensional (3D) cell spheroids in vitro through fluorescence imaging and a more significant EPR effect for improved tumor MR imaging in vivo than the G5-DOTA-Gd complexes. The generated CSTD-D-Gd complexes may be adopted for enhanced tumor MR imaging through an amplified passive EPR effect and also be further extended for different cancer theranostic applications.

Abstract:Dendrimers have become effective drug carriers and gene transfection vectors for medical biomaterials. In this experiment, UCNPs (NaGdF4:Tm, Yb@NaGdF4) and three-generation PAMAM dendrimers were prepared firstly; then NaGdF4:Tm, Yb@NaGdF4 was modified by Polyacrylic acid(PAA); PAMAM were grafted to the surface of the modified UCNPs by the reaction between amino groups and carboxyl groups; finally, O-nitrobenzaldehyde was grafted to the surface by Schiff base reaction.The synthesized upconversion materials were coated with PAA and PAMAM, then loaded with O-nitrobenzaldehyde to form spherical nanoparticles. The particle size of prepared nanoparticles is 75 similar to 80 nm. The dendrimer complex can emit ultraviolet light to regulate the pH gradient of aqueous solution under the excitation of near-infrared light (980 nm), and can be used to kill cancer cells by changing the acidity of tissue cells; secondly, the cavity between dendrimers and dendrimers on the surface of the complex can be used in many fields such as drug delivery and gene transfection; the nanoparticles provide a new method for cancer treatment.

Abstract:A tumor-targeting carrier, peptide HAIYPRH (T7)-conjugated polyethylene glycol-modified polyamidoamine dendrimer (PAMAM-PEG-T7) was explored to deliver magnetic resonance imaging (MRI) contrast agents targeting to the tumor cells specifically. Two different types of tumors, liver cancer and early brain glioma model (involved with the blood–brain barrier), were chosen to evaluate the imaging capacity of this contrast agent. PAMAM-PEG-T7 was synthesized, conjugated with diethylene triamine pentaacetic acid (DTPA) and further chelated gadolinium (Gd), yielding GdDTPA-PAMAM-PEG-T7. The result of ICP-AES showed that about 92 Gd ions could be loaded per PAMAM molecule. The calculated longitudinal relaxivity R1 of the GdDTPA-PAMAM-PEG-T7 was 10.7 mm−1 S−1 per Gd (984.4 mm−1 S−1 per PAMAM), while that of GdDTPA was only 4.8 mm−1 S−1. PAMAM-PEG-T7 had better targeting capacity to the liver cancer cells in vitro and in vivo, compared with PAMAM-PEG. The accumulation of PAMAM-PEG-T7 was 162.5% times that of PAMAM-PEG. But for glioma cells, PAMAM-PEG-T7 did not show its specificity. Furthermore, GdDTPA-PAMAM-PEG-T7 could improve the diagnostic efficiency of liver cancer with the enhanced signal (187%), compared to 130% for PAMAM-PEG and 121% for GdDTPA. GdDTPA-PAMAM-PEG-T7 could selectively identify liver cancer but not early glioma. This nanoscaled MRI contrast agent GdDTPA-PAMAM-PEG-T7 might allow for selective and efficient diagnosis of tumors without the natural barrier including liver cancer.
Multifunctional Low-Generation Dendrimer Nanogels As an Emerging Probe for Tumor-Specific CT/MR Dual-Modal Imaging

Abstract:The development of nanoprobes that have amplified enhanced permeability and retention (EPR) effect is crucial for their precise cancer diagnosis performance. Here, we present the development of functional dendrimer-based nanogels (DNGs) with the generation three primary amine-terminated poly(amidoamine) (PAMAM) dendrimers (G3·NH2) cross-linked by N,N'-bis(acryloyl) cystamine (BAC). The DNGs were prepared through a Michael addition reaction between G3·NH2 dendrimers and BAC via an inverse microemulsion method and entrapped with gold nanoparticles (Au NPs) to form Au-DNGs. The Au-DNGs were sequentially modified with diethylenetriamine penta-acetic acid (DTPA)-gadolinium (Gd) complex, poly(ethylene glycol) (PEG)-linked arginine-glycine-aspartic (RGD) peptide, and 1,3-propanesultone (1,3-PS). The formed multifunctional RGD-Gd@Au-DNGs-PS (R-G@ADP) possessing an average diameter of 122 nm are colloidally stable and display a high X-ray attenuation coefficient, excellent r1 relaxivity (9.13 mM-1 s-1), desired protein resistance rendered by the zwitterionic modification, and cytocompatibility. With the targeting specificity mediated by RGD and the much better tumor penetration capability than the counterpart material of single dendrimer-entrapped Au NPs, the developed multifunctional R-G@ADP enable targeted and enhanced computed tomography (CT)/magnetic resonance (MR) dual-modal imaging of a pancreatic tumor model in vivo. The current work demonstrates a unique design of targeted and zwitterionic DNGs with prolonged blood circulation time as an emerging nanoprobe for specific tumor CT/MR imaging through amplified passive EPR effect.
PAMAM dendrimer and carbon quantum dots complexes as theranostic nanocarrier: Synthesis, optimization and photophysical characterization

Abstract:The current research work proposes the optimization of highly fluorescent carbon quantum dots (CQDs) and photophysical characterization of its theranostic nanocomplexes with PAMAM dendrimers. In recent years, theranostic nanocarriers have been emerged as a promising nanocarrier for personalized medicine. The half generation (G3.5 and G4.5) and full generation (G4.0 and 5.0) PAMAM dendrimer nanocomplexes are comparatively less explored as theranostic nanocarrier, thus to study the photophysical behavior of these complexes in biological environment we have planned various studies at different conditions of pH, salt, temperature and radiation. The amphiphilic CQDs were synthesized using a facile microwave assisted pyrolysis method and optimized for various parameters to get highly fluorescent CQDs. Further, we have prepared CQDs-PAMAM dendrimer nanocomplexes via electrostatic interaction, which was confirmed by change in the emission maxima of CQDs and FTIR spectroscopy. The nanocomplexes showed 37.32 ± 4.57 % fluorescent quenching upon increasing the temperature 20 °C to 60 °C. The photostability study showed enhanced photobleaching upon increasing the generation (G4.0 to G5.0) due to structural changes, although it was significantly lower than organic dyes. The stability study of nanocomplexes showed suitable fluorescence intensity after 3 months at −20 ± 0.5 °C. Overall, these nanocomplexes holds great potential to serve as theranostic nanocarrier, thus offering an indispensable platform for personalized medicine in various aspects of cancer treatment.

Abstract:Computed tomography (CT) is one of the most widely used noninvasive clinical imaging modalities because of its wide availability, low cost, high efficiency and high resolution. Although the resolution of CT is much higher than that of conventional radiography and many other imaging techniques, it is still difficult to distinguish subtle changes of soft tissues because of physical limitations. To increase the contrast of soft tissues and improve the accuracy of disease diagnosis, a variety of CT contrast agents have been developed. Small iodinated compounds have been widely used as clinical CT contrast agents, but their toxicity and short circulation time restrict their further application. To resolve these problems, nanosized CT contrast agents have been developed. In addition to iodine, nanoparticles based on heavy atoms such as gold, tantalum, lanthanides, and bismuth have been investigated as more efficient CT contrast agents than small iodinated compounds. These novel nanosized CT contrast agents have high biocompatibility and long circulation times, which are advantageous for visualization of blood vessels and tumors. Moreover, the combination of CT with other imaging techniques, such as magnetic resonance imaging and ultrasound imaging, could realize higher sensitivity and more personalized diagnostic information. In particular, theranostic CT contrast agents that integrate diagnosis with therapy in a single nanoplatform may serve as a nanomedicine capable of both noninvasive imaging and remote-controlled therapy. Herein, we introduce the principles of CT imaging and commonly used nanoscale CT contrast elements, including iodine, gold, bismuth, tantalum, and lanthanides. The recent advances, future development and existing problems of multifunctional nanosized CT contrast agents are reviewed.

Abstract:Numerous macromolecular MRI contrast agents prepared employing relatively simple chemistry may be readily available that can provide sufficient enhancement for multiple applications. These agents operate using a ~100-fold lower concentration of gadolinium ions in comparison to the necessary concentration of iodine employed in CT imaging. Herein, we describe some of the general potential directions of macromolecular MRI contrast agents using our recently reported families of dendrimer-based agents as examples. Changes in molecular size altered the route of excretion. Smaller-sized contrast agents less than 60 kDa molecular weight were excreted through the kidney resulting in these agents being potentially suitable as functional renal contrast agents. Hydrophilic and larger-sized contrast agents were found better suited for use as blood pool contrast agents. Hydrophobic variants formed with polypropylenimine diaminobutane dendrimer cores created liver contrast agents. Larger hydrophilic agents are useful for lymphatic imaging. Finally, contrast agents conjugated with either monoclonal antibodies or with avidin are able to function as tumor-specific contrast agents, which also might be employed as therapeutic drugs for either gadolinium neutron capture therapy or in conjunction with radioimmunotherapy.

Targeted Tumor CT Imaging Using Folic Acid-Modified PEGylated Dendrimer-Entrapped Gold Nanoparticles

Abstract:The development of multifunctional nanoprobes with a targeting capability for efficient molecular imaging of tumors still remains a great challenge. Herein, we report the synthesis and characterization of folic acid (FA)-modified dendrimer-entrapped gold nanoparticles (Au DENPs) via a facile polyethylene glycol (PEG) linking strategy for in vivo targeted tumor computed tomography (CT) imaging applications. In this study, amine-terminated poly(amidoamine) dendrimers of generation 5 (G5.NH2) sequentially modified by two types of PEG moieties (PEG monomethyl ether with one end of carboxyl group (mPEG-COOH), and FA-modified PEG with one end of carboxyl group (FA-PEG-COOH)) were used as templates to synthesize AuNPs within the dendrimer interiors, followed by acetylation of the remaining dendrimer terminal amines. The formed multifunctional Au DENPs were characterized via different techniques. Cell viability assay, flow cytometric analysis of the cell cycles, and hemolysis assay were used to assess the cytotoxicity and hemocompatibility of the particles. We show that the formed multifunctional Au DENPs are stable at different pH and temperature conditions and in different aqueous media, cytocompatible and hemocompatible in the given Au concentration range, and display much higher X-ray attenuation intensity than Omnipaque (an iodine-based CT contrast agent) under similar concentrations of the active element (Au or iodine). Moreover, the developed Au DENPs enable targeted CT imaging of the model cancer cells with high FA receptor expression in vitro and the corresponding xenografted tumor model in vivo. These findings suggest that the designed Au DENPs may be used as promising contrast agents for targeted CT imaging of tumors.

Abstract:Development of dual-mode or multi-mode imaging contrast agents is important for accurate and self-confirmatory diagnosis of cancer. We report a new multifunctional, dendrimer-based gold nanoparticle (AuNP) as a dual-modality contrast agent for magnetic resonance (MR)/computed tomography (CT) imaging of breast cancer cells in vitro and in vivo. In this study, amine-terminated generation 5 poly(amidoamine) dendrimers modified with gadolinium chelate (DOTA-NHS) and polyethylene glycol monomethyl ether were used as templates to synthesize AuNPs, followed by Gd(III) chelation and acetylation of the remaining dendrimer terminal amine groups; multifunctional dendrimer-entrapped AuNPs (Gd-Au DENPs) were formed. The formed Gd-Au DENPs were used for both in vitro and in vivo MR/CT imaging of human MCF-7 cancer cells. Both MR and CT images demonstrate that MCF-7 cells and the xenograft tumor model can be effectively imaged. The Gd-Au DENPs uptake, mainly in the cell cytoplasm, was confirmed by transmission electron microscopy. The cell cytotoxicity assay, cell morphology observation, and flow cytometry show that the developed Gd-Au DENPs have good biocompatibility in the given concentration range. Our results clearly suggest that the synthetic Gd-Au DENPs are amenable for dual-modality MR/CT imaging of breast cancer cells.
Selective Cellular Uptake and Druggability Efficacy through Functionalized Chitosan-Conjugated Polyamidoamine (PAMAM) Dendrimers

Abstract:Nanotechnology has ushered in significant advancements in drug design, revolutionizing the prevention, diagnosis, and treatment of various diseases. The strategic utilization of nanotechnology to enhance drug loading, delivery, and release has garnered increasing attention, leveraging the enhanced physical and chemical properties offered by these systems. Polyamidoamine (PAMAM) dendrimers have been pivotal in drug delivery, yet there is room for further enhancement. In this study, we conjugated PAMAM dendrimers with chitosan (CS) to augment cellular internalization in tumor cells. Specifically, doxorubicin (DOX) was initially loaded into PAMAM dendrimers to form DOX-loaded PAMAM (DOX@PAMAM) complexes via intermolecular forces. Subsequently, CS was linked onto the DOX-loaded PAMAM dendrimers to yield CS-conjugated PAMAM loaded with DOX (DOX@CS@PAMAM) through glutaraldehyde crosslinking via the Schiff base reaction. The resultant DOX@CS@PAMAM complexes were comprehensively characterized using Fourier-transform infrared (FTIR) spectroscopy, transmission electron microscopy (TEM), and dynamic light scattering (DLS). Notably, while the drug release profile of DOX@CS@PAMAM in acidic environments was inferior to that of DOX@PAMAM, DOX@CS@PAMAM demonstrated effective acid-responsive drug release, with a cumulative release of 70% within 25 h attributed to the imine linkage. Most importantly, DOX@CS@PAMAM exhibited significant selective cellular internalization rates and antitumor efficacy compared to DOX@PAMAM, as validated through cell viability assays, fluorescence imaging, and flow cytometry analysis. In summary, DOX@CS@PAMAM demonstrated superior antitumor effects compared to unconjugated PAMAM dendrimers, thereby broadening the scope of dendrimer-based nanomedicines with enhanced therapeutic efficacy and promising applications in cancer therapy.

Abstract:Star-burst dendrimers represent a superior carrier platform for targeted drug delivery. Partially acetylated generation 5 (G5) polyamidoamine (PAMAM) dendrimer was conjugated with the targeting moiety (biotin) and the imaging moiety (fluoresceinisothiocyanate, FITC), and the resulting dendrimer–biotin conjugate was characterized by 1H NMR, UV–vis spectrum. As revealed by flow cytometry and confocal microscopy, the bifunctional conjugate (dendrimer–biotin–FITC) exhibited much higher cellular uptake into HeLa cells than the conjugate without biotin. The uptake was energy-dependent, dose-dependent, and could be effectively blocked by dendrimer-conjugated biotin. Our results indicated that the biocompatible biotin–dendrimer conjugate might be a promising nano-platform for cancer therapy and cancer diagnosis.
