Abstract:Transglutaminases (TGases) have been widely used in food, pharmaceutical, biotechnology, and other industries because of their ability to catalyze deamidation, acyl transfer, and crosslinking reactions between Ƴ-carboxamide groups of peptides or protein-bound glutamine and the Ɛ-amino group of lysine. In this study, we demonstrated an efficient systematic engineering strategy to enhance the synthesis of TGase in a recombinant Streptomyces mobaraensis smL2020 strain in a 1000-L fermentor. Briefly, the enzymatic properties of the TGase TGL2020 from S. mobaraensis smL2020 and TGase TGLD from S. mobaraensis smLD were compared to obtain the TGase TGLD with perfected characteristics for heterologous expression in a recombinant S. mobaraensis smL2020ΔTG without the gene tgL 2020. Through multiple engineering strategies, including promoter engineering, optimizing the signal peptides and recombination sites, and increasing copies of the expression cassettes, the final TGLD activity in the recombinant S. mobaraensis smL2020ΔTG: (PL2020-spL2020-protgLD-tgLD)2 (tgL2020and BT1) reached 56.43 U/mL and 63.18 U/mL in shake flask and 1000-L fermentor, respectively, which was the highest reported to date. With the improvement of expression level, the application scope of TGLD in the food industry will continue to expand. Moreover, the genetic stability of the recombinant strain maintained at more than 20 generations. These findings proved the feasibility of multiple systematic engineering strategies in synthetic biology and provided an emerging solution to improve biosynthesis of industrial enzymes.

Abstract:Microbial transglutaminase (TGase) is widely used in food processing because of its ability to catalyze protein cross-linking. In this study, ultraviolet (UV) mutagenesis was used to create a mutant of Streptomyces mobaraensis TX1 with a high TGase yield, with a maximum yield of 37.51 ± 0.46 U/mL after 32 h of flask fermentation, and a 2.15-fold increase compared to the wild-type (WT) strain. The transcriptional level of the transglutaminase gene increased 50-fold in the mutant strain compared to the WT strain, and a point mutation was found in the promoter of transglutaminase gene. The evaluation results of the promoter function revealed that the high yield of microbial transglutaminase was primarily caused by the point mutation. Our results provided a significant information on the high-yield mechanism of microbial transglutaminase biosynthesis.
Engineering the C-Terminal Region to Enhance the Thermal Stability of Streptomyces hygroscopicus Transglutaminase

Abstract:To enhance the applied value of transglutaminase (TGase), various methods have been employed to improve its catalytic properties. However, most modifications have targeted the N-terminus, while the role of the C-terminus in determining TGase properties has been overlooked. In this study, we focused on enhancing the thermal stability of Streptomyces hygroscopicus TGase by engineering its C-terminal region. Modeling revealed that the C-terminal loop interacts with the N-terminal loop through hydrogen bonds between Trp331 and N-terminal residues (Asp19, Ala20, Tyr21). Removing the last C-terminal residue (Ser322) had no significant effect on TGase stability, but deleting additional residues (Trp331, Gly330, Gln299) led to inactivation. Substituting Trp331 with Ala reduced TGase's half-life at 50 °C and specific activity by 50% and 70%, respectively, highlighting the importance of C-terminal interactions in TGase stability. We also attempted to fuse three self-assembling amphipathic peptides (SAPs) (EAK16, KL15, ELK16) and a C-terminal sequence (IGCIILT) from Sulfolobus tokodaii RNase HI to TGase. The fusion of IGCIILT increased TGase's half-life by 1.5-fold without affecting specific activity, while the three SAPs had little effect on stability. Structural analysis showed that the fusion of IGCIILT raised TGase's melting temperature by 5.2 °C and altered its tertiary structure. Our results indicate that the C-terminus is important for modulating TGase properties, and fusing "stabilization tags" like IGCIILT at the C-terminus is a promising strategy to enhance thermal stability.

Abstract:Microbial transglutaminase (MTG) from Streptomyces mobaraensis is widely used in the food and pharmaceutical industries for cross-linking and post-translational modification of proteins. It is believed that its industrial applications could be further broadened by improving its thermostability. In our previous study, we showed that the introduction of structure-based disulfide bonds improved the thermostability of MTG, and we succeeded in obtaining a thermostable mutant, D3C/G283C, with a T50 (incubation temperature at which 50% of the initial activity remains) 9 °C higher than that of wild-type MTG. In this study, we performed random mutations using D3C/G283C as a template and found several amino acid substitutions that contributed to the improvement of thermostability, and investigated a thermostable mutant (D3C/S101P/G157S/G250R/G283C) with three amino acid mutations in addition to the disulfide bond. The T50 of this mutant was 10 °C higher than that of the wild type, the optimal temperature for enzymatic reaction was increased to 65 °C compared to 50 °C for the wild type, and the catalytic efficiency (kcat/Km) at 37.0 °C was increased from 3.3 × 102 M-1 s-1 for the wild type to 5.9 × 102 M-1 s-1. X-ray crystallography of the D3C/G283C MTG showed no major structural differences against wild-type MTG. Structural differences were found that may contribute to thermostabilization and improve catalytic efficiency. Abstract:Objectives: To improve the fermentation production of transglutaminase (TGase) from Streptomyces mobaraensis for applications in the food industry, the atmospheric and room-temperature plasma (ARTP) mutagenesis was applied to breed S. mobaraensis mutants with increased TGase production.Results: After eight rounds of iterative ARTP mutagenesis, four genetically stable mutants, Sm5-V1, Sm6-V13, Sm2V10, and Sm7-V12, were identified, which showed increased TGase production by 27, 24, 24, and 19%, respectively. The best mutant Sm5-V1 exhibited a maximum TGase activity of 5.85 U/mL during flask fermentation. Compared to the wild-type strain, the transcription levels of the zymogen TGase genes in the mutants increased significantly as indicated by quantitative real-time PCR, while the gene nucleotide sequences of the mutants did not change at all. It was shown that the overexpression of TGase zymogen gene in the mutants contributes to the increase in TGase production.Conclusions: ARTP is a potentially efficient tool for microbial mutation breeding to bring some significant changes required for the industrial applications.

Abstract:Stress-mediated bioprocess is a strategy designed to enhance biological target productivity. In this study, an attempt was made to enhance transglutaminase (TGase) production by Streptomyces mobaraensis by using different stress conditions including heat shock, alcohols and salt stress. Results showed that the effects of stress on TGase production depended on the type applied. For heat shock, TGase production (1.32 U/ml) was recorded maximum in the culture treated at 48 h post inoculation in water bath at 60 °C for 1 min. For alcohols treatment, the maximum activity of TGase (1.77 and 1.75 U/ml) was obtained when 3% methanol was added into the medium at 0 or 24 h of fermentation. However, a 3.5-fold increased production of TGase (3.8 U/ml) was observed in the medium supplemented with 0.2 mol/l MgCl 2 compared with the basic medium at the beginning of fermentation. In conclusion, TGase production from S. mobaraensis was improved by heat shock, methanol and salt stress treatments, MgCl 2 stress was the most effective.
Purification and Characterization of A High-Salt-Resistant Microbial Transglutaminase from Streptomyces Mobaraensis

Abstract:A novel microbial transglutaminase (MTG-TX) was obtained from Streptomyces mobaraensis by fermentation and purification. The enzyme was purified by ethanol precipitation via a two-step purification method, with a 44.0% yield and a specific activity of 39.2 U mg(-1). The purified enzyme exhibited stable performance over a range of pH 5.0-pH 10.0 and displayed maximal activity at pH 6 and 48 degrees C. We verified through biochemical analyses that the enzyme is a novel MTG variant possessing the same zymogen characteristics as that of another reported MTG from Streptoverticillium ladakanum B1. Furthermore, the loss of enzyme activity by MTG-TX in the presence of high salt was only 79.8% that observed in a control MTG from S. mobaraensis DSM40847. On the basis of salt resistance, the novel MTG-TX presented here also performed well in food-related applications by successfully crosslinking proteins in high-salt environments, thereby enhancing the cohesiveness of bacon. Integrated strategies for efficient production of Streptomyces mobaraensis transglutaminase in Komagataella phaffii

Abstract:Transglutaminase (TGase) from Streptomyces mobaraensis commonly used to improve protein-based foods due to its unique enzymatic reactions, which imply considerable attention in its production. Recently, TGase exhibit broad market potential in non-food industries. However, achieving efficient synthesis of TGase remains a significant challenge. Herein, we achieved a substantial amount of a fully functional and kinetically stable TGase produced by Komagataella phaffii (Pichia pastoris) using multiple strategies including Geneticin (G418) screening, combinatorial mutations, promoter optimization, and co-expression. The active TGase expression reached a maximum of 10.1 U mL-1 in shake flask upon 96 h of induction, which was 3.8-fold of the wild type. Also, the engineered strain exhibited a 6.4-fold increase in half-life and a 2-fold increase in specific activity, reaching 172.67 min at 60 °C (t1/2(60 °C)) and 65.3 U mg-1, respectively. Moreover, the high-cell density cultivation in 5-L fermenter was also applied to test the productivity at large scale. Following optimization at a fermenter, the secretory yield of TGase reached 47.96 U mL-1 in the culture supernatant. Given the complexity inherent in protein expression and secretion, our research is of great significance and offers a comprehensive guide for improving the production of a wide range of heterologous proteins.

[Methods] The gene of pro and TG from Streptoverticillium ladakanum were cloned into pET-22b to generate co-expression and fusion expression, respectively. Based on these two expression patterns, the first four amino acids of the mature TGase N-terminal were modified by site-directed mutagenesis to detect the effect of different expression patterns and mutation on enzyme activity. [Results] When co-expression of the pro-peptide with TGase, the active form of TGase can be obtained directly, and the specific activity reaches 37.71 U/mg. Based on the fusion expression, the first three amino acid DSD of the N-terminal of TGase were mutated to AAA, and the specific enzyme activity reached 14.04 U/mg, which was 14.05% higher than the original expression pattern. [Conclusion] The co-expression of pro-peptide with TGase can directly produce the active form of TGase. For the fusion expression pattern, the mutation of the appropriate site is beneficial to increase the TGase activity.
Genome Shuffling Enhances Transglutaminase Production of Streptomyces Mobaraensis

Abstract:Transglutaminase can catalyze Trans-phosphatidylation reaction,and is widely used in food industry.But the production of transglutaminase cannot meet the requirement of industrial production.Nowadays,random mutagenesis is the most frequent but inefficiency method to improve transglutaminase production.Transglutaminase production of Streptomyces mobaraensis is enhanced by genome shuffling.With 96-well plates high-throughput screening,the time and work are reduced.Firstly,protoplasts presentation conditions are optimized.The optimization conditions are treating mycelium by 6mg/ml lysozyme solution for 2 hours.Protoplasts are inactivated by UV illumination for 2 minutes and water-bath in 60℃ for 40min.Then fusants are picked up regenerating in regeneration medium and then screened by 96-well plates high-throughput screening and verified the high-production strains by fermentation in shaking flask.After two rounds of genome shuffling,a strain can produce Tgase 65.5％ higher than incipient strain is obtained and the production reaches 7.12U/ml.
Heterologous Expression of Recombinant Transglutaminase in Bacillus subtilis SCK6 with Optimized Signal Peptide and Codon, and Its Impact on Gelatin Properties

Abstract:Microbial transglutaminases (MTGs) are widely used in the food industry. In this study, the MTG gene of <i>Streptomyces</i> sp. TYQ1024 was cloned and expressed in a food-grade bacterial strain, <i>Bacillus subtilis</i> SCK6. Extracellular activity of the MTG after codon and signal peptide (SP Ync M) optimization was 20 times that of the pre-optimized enzyme. After purification, the molecular weight of the MTG was 38 kDa and the specific activity was 63.75 U/mg. The optimal temperature and pH for the recombinant MTG activity were 50°C and 8.0, respectively. MTG activity increased 1.42- fold in the presence of β-ME and 1.6-fold in the presence of DTT. Moreover, 18% sodium chloride still resulted in 83% enzyme activity, which showed good salt tolerance. Cross-linking gelatin with the MTG increased the strength of gelatin 1.67 times and increased the thermal denaturation temperature from 61.8 to 75.8°C. The MTG also significantly increased the strength and thermal stability of gelatin. These characteristics demonstrated the huge commercial potential of MTG, such as for applications in salted protein foods.

Abstract:Finding new crosslinking enzymes and elucidating internal cross-linking mode are beneficial to meeting the requirement of various protein substrates and processing conditions in food industry. Transglutaminase (TG) can lead to the covalent crosslink of ε-(γ-glutamine)-lysine isopeptidic bonds in proteins through acyl-transfer reaction. This work reported the characterization of transglutaminases (TGs) from four Bacillus strains with obvious differences in amino acid sequences and enzymatic properties. Among them, TG from B. clausii (BCLTG) displayed the highest optimal temperature (70 °C) and specific activity (282 U/mg). The molecular dynamics simulation indicated that enlargement of substrate accessible tunnel in BCLTG with the increased temperature might result in the easy approach of substrate molecule to the active center. Moreover, BCLTG could cause the crosslinking of bovine serum albumin and almost all protein constituents in whey protein and soy protein isolate, and the crosslinking path of BCLTG towards three proteins was revealed. This study provides an ideal candidate for proteins crosslinking and theoretical foundation and preliminary information for applying TG in food processing.

Abstract:Transglutaminase can generate covalent cross-links between protein molecules or intramolecular covalent cross-links to change functional properties of proteins, and is applied in industries of food processing, medicine, cosmetic and textile. Microbial transglutaminase has aroused much more attention because of its calcium-independent and broader substrate specificity. This article summarized the gene expression of microbial transglutaminase, reviewed the gene characteristics and enzymatic characteristics, analyzed the relationship between crystal structures and function, and introduced the advantages and problems of microbial transglutaminase production using genetic engineering technique. In order to improve the yield of the enzyme, the activity and thermo-stability, the suggestions about expression strategies and molecular modification of microbial transglutaminase were also provided.

Abstract:Mannooligosaccharides are released by mannan-degrading endo-β-1,4-mannanase and are known as functional additives in human and animal diets. To satisfy demands for biocatalysis and bioprocessing in crowed environments, in this study, we employed a recently developed enzyme-engineering system, isopeptide bond-mediated molecular cyclization, to modify a mesophilic mannanase from Bacillus subtilis. The results revealed that the cyclized enzymes showed enhanced thermostability and ion stability and resilience to aggregation and freeze-thaw treatment by maintaining their conformational structures. Additionally, by using the SpyTag/SpyCatcher system, we generated a mannanase-xylanase bifunctional enzyme that exhibited a synergistic activity in substrate deconstruction without compromising substrate affinity. Interestingly, the dual-enzyme ring conformation was observed to be more robust than the linear enzyme but inferior to the single-enzyme ring conformation. Taken together, these findings provided new insights into the mechanisms of molecular cyclization on stability improvement and will be useful in the production of new functional oligosaccharides and feed additives.
Enhancing the Functional Characteristics of Soy Protein Isolate Via Cross-Linking Catalyzed by Bacillus Subtilis Transglutaminase.

Abstract:BACKGROUND: Although Streptomyces mobaraense transglutaminase (MTG) has been extensively applied to enhance the functional characteristics of soy protein isolate (SPI) through cross-linking, various transglutaminases (TGs) in nature may provide more choice in the food industry. Previous research reported that TG derived from Bacillus subtilis (BTG) exhibited better pH stability and thermostability than MTG. RESULTS: An attempt was made to study the influence of BTG induced cross-linking on the properties of SPI. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) results indicated that almost all protein constituents (alpha', alpha, beta, AS, and BS) in SPI could be cross-linked with BTG treatment. The BTG treatment also resulted in a significant increase (*P < 0.05) in SPI mean particle size. Emulsifying activity and stability were improved from 0.11535 m(2) g(-1) and 48.3% for native SPI to 0.13252 m(2) g(-1) and 83.9% for SPI treated with BTG at 6 h. Similarly, the modified SPI showed better foam activity (1.32 mL) and stability (87.6%) than the original SPI (0.93 mL and 56.8%). The water-holding capacity of SPI gel was found to increase with time, with a value of 95.43% at 6 h. Furthermore, SPI gel's texture profiles were greatly improved by adding BTG (*P < 0.05). CONCLUSION: The results of the present study indicated that BTG could be a promising cross-linking agent for improving the functional characteristics of SPI. As a substitute for MTG, BTG could thus potentially be used for food structure engineering to enhance the functional characteristics of multiple proteins to advance the development of food chemistry. Abstract:Transglutaminase (TGase) can catalyze the crosslinking within or between protein molecules, improving the structural and functional properties of proteins. It has been widely used in artificial meat, yogurt products, flour products and other foods. In this study, a high yield TGase strain was screened from fermented soybean, and it was identified as the Bacillus amyloliquefaciens XCT-09. Then, the transglutaminase (X9Tgl) from XCT-09 was expressed in Escherichia coli BL21(DE3), and the enzyme activity of purified X9Tgl reached 5.73 U/mg. Subsequently, the catalytic mechanism of X9Tgl was elucidated, and the X9Tgl contained the active center of E115, C116 and E187, as well as the substrate binding sites W149 and F69. Furthermore, this study characterized the enzymatic properties of X9Tgl, and the optimal catalytic temperature and pH were 60°C and 6, respectively. Moreover, X9Tgl exhibited stronger thermal stability and pH tolerance than STG, and it was not inhibited by the majority of metal ions. This study successfully identified a X9Tgl with high thermal stability, and pH tolerance, which provided a potential TGase resource for application in the food and medicine fields.
Enhancement of transglutaminase activity by NMR identification of its flexible residues affecting the active site

Abstract:Incorporation of inter- or intramolecular covalent cross-links into food proteins with microbial transglutaminase (MTG) improves the physical and textural properties of many food proteins, such as tofu, boiled fish paste, and sausage. By using nuclear magnetic resonance, we have shown that the residues exhibiting relatively high flexibility in MTG are localized in the N-terminal region; however, the N-terminal region influences the microenvironment of the active site. These results suggest that the N-terminal region is not of primary importance for the global fold, but influences the substrate binding. Therefore, in order to increase the transglutaminase activity, the N-terminal residues were chosen as candidates for site-directed replacement and deletion. We obtained several mutants with higher activity, del1-2, del1-3, and S2R. We propose a strategy for enzyme engineering targeted toward flexible regions involved in the enzymatic activity. In addition, we also briefly describe how the number of glutamine residues in a substrate protein can be increased by mixing more than two kinds of TGases with different substrate specificities.

Abstract:Microbial transglutaminases(MTG)are a family of enzymes that catalyze post-translational modification of proteins by inter or intra-molecular crosslinking through acyl-transfer reaction.Because of the modification of protein functional properties,MTG showed a broad range of applications in the food industry,biomedicine and textile industry.In this paper,correlation techniques,research progress of constructing engineered strains which can produce microbial transglutaminase were reviewed and direction of future research was discussed detailed.

Abstract:The microbial transglutaminase(TGase) especially which come from Streptoverticillium is an important enzyme. It is widely used in food industry. The recent progresses in the study of the enzyme, especially on the molecular biology and genetic engineering of it were summarized here. In addition, the future development is discussed in this paper. We consider that with the tool of genetic engineering, we'll soon gain a recombine bacterium, which can highly and activated expression TGase.

Abstract:Microbial transglutaminase (MTG) from Streptomyces mobaraensis is an important enzyme widely applied in food processing for the improvement of protein properties by catalyzing the cross-linking of proteins. In this work we aimed at improving the production and enabling an easy and efficient purification process from culture supernatants. Thus, recombinant vectors, with either a constitutive promoter (Pp5) or an inducible promoter (PnisA), controlling the expression of the MTG gene fused to the signal peptide of Usp45 (SPusp45) were constructed and then expressed in Lactococcus lactis. After purification, 43.5 ± 0.4 mg/L mature MTG-6His was obtained. It displayed 27.6 ± 0.5 U/mg enzymatic activity cross-linking soy protein isolate effectively. The purified mature MTG was immobilized with magnetic porous Fe3O4 nanoparticles, which improved its activity up to 29.1 ± 0.4 U/mg. The immobilized MTG maintained 67.2% of the initial activity after being recycled for 10 times. The high production and secretion of functional S. mobaraensis MTG from L. lactis and the magnetic immobilized MTG-6His onto Fe3O4 nanoparticles reported in this study would have potential industrial applications.
