Abstract:Herein, a general strategy for chemo- and regioselective 1,2-reduction of chromium-bound arenes was developed, thus providing rapid access to 1,3-cyclohexadienes. Selective arene activation via π-complexation along with the use of mild hydride Ph3SiH can overcome the inherently low reactivity of arene π-bonds while tolerating various reduction-sensitive functional groups. Its versatility further enables a regiodivergent deuteration. Using different sequences of (non)deuterated hydride and acid reagents, the deuterated positions as well as the degrees of deuterium incorporation can be controlled precisely, which leads to a large and previously inaccessible chemical space for 1,3-cyclohexadiene isotopologues. A reasonable mechanism was proposed based on intermediate capture and control experiments. The synthetic value of this selective 1,2-reduction was demonstrated in the formal total synthesis of (±)-galanthamine and (±)-lycoramine.

Abstract:Under the catalysis of PdBr2 and a chiral phosphoramidite ligand, the upper-rim mono (2-bromoaroyl)-substituted calix[4]arene derivatives underwent a facile enantioselective desymmetrization reaction to afford 9H-fluorene-embedded inherently chiral calixarenes in good yields with excellent enantioselectivities. The transannular dehydrogenative arene-arene coupling reaction proceeded most probably through an oxidative addition of the C-aryl-Br bond to a ligated palladium catalyst followed by a sequence of an enantioselective 1,5-palladium migration and an intramolecular C-H arylation sequence. This new family of inherently chiral calixarenes possesses unique chiroptical properties thanks to their highly rigid structure induced by the 9H-fluorene segment.

Abstract:Inherently chiral calixarenes have garnered significant attention due to their distinctive properties, yet the development of efficient catalytic asymmetric synthesis methods remains a critical challenge. Herein, we report the asymmetric synthesis of calix[4]arenes featuring inherent or both inherent and axial chirality via a cobalt-catalyzed C–H activation/annulation strategy in high yield with excellent enantio- and diastereoselectivity (up to > 99% ee and > 20:1 dr). Electrooxidation was also suitable for this transformation to obviate the sacrificial metal oxidants, underscoring the environmentally friendly potential of this approach. A key octahedral cobaltacycle intermediate was synthesized and characterized, providing valuable insights into the mode of enantio- and diastereocontrol of this protocol. Noteworthy photoluminescence quantum yields of up to 0.94 were measured, underscoring the potential of these compounds in the domain of organic fluorescent materials.

Abstract:We describe herein an intriguing method for the synthesis of biaryl aldehydes bearing both axial and central chirality through a desymmetric [3 + 2] cycloaddition reaction of activated isocyanides with prochiral biaryl dialdehydes under silver catalysis. This protocol features excellent enantioselectivity, 100% atom economy, good functional group compatibility, and operational simplicity.

Abstract:We report herein an efficient approach for the enantioselective synthesis of inherently chiral calix[4]arenes via palladium-catalyzed asymmetric intramolecular C-H arylations. Using a chiral bifunctional phosphine-carboxylate ligand, the inherent chirality on macrocyclic scaffolds was induced successfully, from which a wide range of calix[4]arenes with fluorenone motifs were obtained with good yields and excellent enantioselectivities (up to >99% ee). The synthetic utility of this method was demonstrated by diverse transformations of the products, thus substantially expanding the chemical space of chiral calix[4]arenes. Further investigations of photophysical and chiroptical properties revealed that calix[4]arenes bearing two fluorenone moieties displayed remarkable glum values (up to 0.019), highlighting the great potential of inherent chirality in the development of organic optoelectronic materials.

Abstract:Inherently chiral calix[4]arenes are an excellent structural scaffold for asymmetric synthesis, chiral recognition, sensing, and circularly polarized luminescence. However, their catalytic asymmetric synthesis remains challenging. Herein, we report an efficient synthesis of inherently chiral calix[4]arene derivatives via cascade asymmetric cyclization and oxidation reactions. The three-component reaction features a broad substrate scope (33 examples), high efficiency (up to 90% yield), and excellent enantioselectivity (>95% ee on average). The potential applications of calix[4]arene derivatives are highlighted by their synthetic transformation and a detailed investigation of their photophysical and chiroptical properties.

Abstract:Under the catalysis of PdBr2 and a chiral phosphoramidite ligand derived from R-3,3'-bis(trifluoromethyl)-[1,1'-binaphthalene]-2,2'-diol, the upper-rim mono (2-bromoaroyl)-substituted calix[4]arene derivatives underwent facile enantioselective desymmetrization reaction to afford 9H-fluorene-embedded inherently chiral cyclophanes in good yields with ee up to 94%. The transannular arene-arene cross coupling reaction proceeded most probably through an oxidation addition of Caryl-Br bond to ligated palladium catalyst, an unusual long range 1,5-palladium migration fol-lowed by the intramolecular arylation sequence. Because of the macrocyclic strain, the newly formed 9H-fuorene segment adopts curved structure, rendering highly rigid inherently chiral macrocycles unique chiroptical property.

Abstract:Inherently chiral calix[4]arenes exhibit broad applications in asymmetric catalysis, chiral recognition, and materials science. However, their synthesis remains a significant challenge. Herein, we report a highly efficient method for the construction of calix[4]arenes with both axial and inherent chiralities through palladium/chiral norbornene (Pd/NBE*) cooperative catalysis. In this cascade reaction, the initial introduced axial chirality is dictated by Pd/NBE* cooperative catalysis, while the latter inherent chirality is controlled by the preestablished axial chirality via an unprecedented axial-to-inherent diastereoinduction process. This method employs ortho-calix[4]arene-tethered aryl iodides and β-substituted naphthyl bromides as starting materials, enabling the synthesis of a wide range of five- and six-membered benzo-fused calix[4]arenes with axial and inherent chiralities (29 examples) in one step with excellent enantioselectivities and diastereoselectivities. Preliminary studies of photophysical and chiroptical properties reveal that these axially and inherently chiral calix[4]arenes possess promising glum values, demonstrating their potential in developing new organic optoelectronic materials.

Abstract:This paper describes our attempts to synthesize inherently chiral heteroatom-bridged calixaromatics. Based on a stepwise fragment-coupling approach using a chiral 3,5-dihydroxybenzamide or benzoate, 2,4-dihydroxyacetophenone, and cyanuric chloride as reactants, chiral tetraoxacalix[2]arene[2]triazine derivatives 16 and 17 were synthesized in good yields. Subsequent macrocyclic condensation with a diamine 6 furnished efficiently the pairs of diastereomers of inherently chiral tetraoxacalix[2]arene[2]triazine azacrowns 18 and 19.

Abstract:The asymmetric cross-coupling reaction is developed as a straightforward strategy toward 1,1-diaryl alkanes, which are a key skeleton in a series of natural products and bioactive molecules in recent years. Here we report an enantioselective benzylic C(sp(3))-H bond arylation via photoredox/nickel dual catalysis. Sterically hindered chiral biimidazoline ligands are designed for this asymmetric cross-coupling reaction. Readily available alkyl benzenes and aryl bromides with various functional groups tolerance can be easily and directly transferred to useful chiral 1,1-diaryl alkanes including pharmaceutical intermediates and bioactive molecules. This reaction proceeds smoothly under mild conditions without the use of external redox reagents.

Abstract:The simultaneous construction of multiple stereogenic elements in a single step is highly appealing and desirable in the field of asymmetric synthesis. Furthermore, the catalytic enantioselective synthesis of inherently chiral calix[n]arenes with high enantiopurity has long been a challenging endeavor. Herein, we report an enantioselective cobalt-catalyzed C-H activation/annulation for the efficient construction of inherently chiral calix[4]arenes bearing multiple C-N axially chiral element. By employing the benzamide tethered calix[4]arene as the substrate, the C-H annulation with alkynes can be successfully accomplished, leading to the generation of multiple stereogenic elements. A wide range of calix[4]arenes and alkynes are found to be well compatible, and exhibit good yields, high enantioselectivity and excellent diastereoselectivity. Notably, the gram-scale reaction, catalytic application, synthetic transformations, and chiral recognition further showcase the potential applications of this protocol.

Abstract:Inherently chiral tetraoxacalix[2]arene[2]pyridines containing C(2) symmetry were synthesized efficiently from a macrocyclic condensation reaction of resorcinol derivatives with 2,6-dichloro-3-nitropyridine in a one-pot reaction manner, while tetraoxacalix[2]arene[2]pyridine with an ABCD-substitution pattern was prepared in a good yield by means of a stepwise fragment coupling approach. Postmacrocyclization chemical manipulations led to functionalized tetraoxacalix[2]arene[2]pyridines. A racemic sample was resolved into enantiopure (+)- and (-)-inherently chiral compounds.

Abstract:Inherent chirality was generated by regioselective monoalkylation of 1,3-substituted calix[5]crown-6 incorporating an axially chiral BINOL moiety, with ethyl bromoacetate. The resultant diastereomers 4a and 4b, which possess both axial chirality and inherent chirality, were readily separated by column chromatography. Respective hydrolysis afforded diastereomeric acids 5a and 5b. 1H NMR spectra and X-ray crystallography established that esters 4a and 4b feature a cone-in conformation, which disappears in the corresponding acids 5a and 5b due to intramolecular hydrogen-bonding between the carboxyl group and a glycolic oxygen atom. Amino alcohol-induced fluorescence quenching was observed for 5a and 5b. The highest enantioselectivity, based on the ratio of Stern–Volmer constants KSV(S)/KSV(R), reached 1.90 in the case of 5a with 2-amino-1-phenylethanol (G1), and 5b with phenylalaninol (G2). The highest diastereoselectivity of 1.83, measured by KSV(5a)/KSV(5b), occurs in the case of 5a/5b with (R)-G2.

Abstract:The use of calixarenes in asymmetric catalysis was described. The current progress of asymmetric catalysts based on calixarene scaffold for the enantioselective reactions including allylation, alkylation, addition of diethylzinc to benzaldehyde, Micheal-type addition, aldol reaction, hydrogenation, Friedal Crafts reaction, epoxidation, hydroformylation, and Biginelli reaction was illustrated. In these cases, calixarenes were used as organocatalysts or ligands in metal-catalyzed asymmetric reactions.

Abstract:The catalytic enantioselective desymmetrizing bromoaminocyclization of prochiral cyclohexa-1,4-dienes has been achieved by using chiral anion phase-transfer catalysis, providing a range of enantioenriched cis-3a-arylhydroindoles bearing an all-carbon quaternary stereocenter in good yields (up to 78%) and excellent enantioselectivities (up to 97% ee). Abstract:As a novel type of macrocycles with attractive planar chirality, pillar[5]arenes have gained increasing research interest over the past decades, enabling their widespread applications in diverse fields such as porous materials, molecular machines, and chiral luminescence materials. However, the catalytic methodology towards the enantioselective synthesis of planar chiral pillar[5]arenes remains elusive. Here we report a novel method for the enantioselective synthesis of planar chiral pillar[5]arenes via asymmetric Sonogashira coupling, giving access to a wide range of highly functionalized planar chiral pillar[5]arenes, including both homo- and hetero-rimmed ones, with excellent enantioselectivities. Attractively, the resultant planar chiral pillar[5]arenes show great potential for widespread use in many areas such as chiral luminescent materials. This work not only enables the successful synthesis of planar chiral pillar[5]arenes with abundant structural and functional diversity as key building blocks for practical applications but also enriches the asymmetric cross-coupling methodologies in organic synthetic chemistry.

Abstract:We report herein the synthesis of highly enantiopure inherently chiral N3,O-calix[2]arene[2]triazines from enantioselective macrocyclization enabled by chiral phosphoric acid-catalyzed intramolecular nucleophilic aromatic substitution reaction. In contrast to documented examples, the inherent chirality of the acquired compounds arises from one heteroatom difference in the linking positions of heteracalix[4](het)arenes.
