Abstract:Asymmetric expansion: A catalytic asymmetric ring-expansion reaction of the title compounds occurs in the presence of a Sc(OTf)(3) catalyst bearing an N,N'-dioxide-based ligand. Highly functionalized 2-quinolone derivatives containing a chiral C4-quaternary stereocenter were obtained in high yields and high levels of selectivity under mild reaction conditions (see scheme; Tf=trifluoromethanesulfonyl).

Abstract:Herein, we successfully inhibited the preferential homodimerization and C−Si/Si−H bond cross-exchange of benzosilacyclobutenes and monohydro-silacyclobutanes and achieved the first highly selective C−Si/C−Si bond cross-exchange reaction by deliberately tuning the Ni-catalytic system, which constitutes a powerful and atom-economical ring expansion method for preparing medium-sized cyclic compounds bearing two silicon atoms at the ring junction, which are otherwise inaccessible. The DFT calculation explicitly elucidated the pivotal role of Si−H bond at silacyclobutanes and the high ring strain of two substrates in realizing the two C−Si bonds cleavage and reformation in the catalytic cycle.

Abstract:A highly regioselective base-mediated ring expansion of 2,4-diiminoazetidines via cleavage of C-N and C(sp(3))-H bonds is achieved for the first time to afford efficiently 2,3-dihydropyrimidinesulfonamides. The mechanism of the ring expansion via tandem 4π electrocyclic ring-opening/1,5-H shift/6π electrocyclic ring-closing is well confirmed by the trapping experiments of two key intermediates and deuterium labeling studies.

Abstract:A ring distortion approach for the synthesis of an advanced intermediate en route to rhodomolleins XIV and XLII was described, which led to successful construction of the 5/8/5/5 tetracyclic core framework of the kalmane diterpenoids. Key steps of the strategy include an oxidative dearomatization-induced (ODI)-Diels-Alder cycloaddition, a Dowd-Beckwith rearrangement, and a bioinspired Wagner-Meerwein rearrangement.

Abstract:Unusual insertion of carbodiimide into the Y-Cp (Cp = C(5)H(5)) bond and isomerization of the resulting cyclopentadienyl (Cp)-substituted amidinate complex to the amidino-substituted Cp complex have been established, representing an efficient and simple method for ring modification of sensitive metallocenes. All products, including the rare four-center interaction precursor of the insertion, have been characterized by X-ray structural analyses.

Herein, nitrene equivalent mediated metal-free ring expansions of alkylidenecyclopropanes (ACPs) and an alkylidenecyclobutane (ACB) were described. Abstract:The dearomative expansion of aromatic rings has long been pursued by chemists due to its potential to provide tractable approaches for synthesizing valuable non-aromatic molecules. To circumvent the conventional use of hazardous and unstable diazo compounds, photochemical synthesis has recently emerged as a promising platform. However, protocols that can effentively handle both arenes and azaarenes remain scarce. Herein, we introduce a generic strategy that efficiently converts of β-(aza)aryl-β-substituted enones into biologically significant cycloheptatriene derivatives, including their aza-variants. This method allows for the easy modulation of diverse functional groups on the product and demonstrates a wide substrate scope, evidenced by its excellent tolerance for various drug motifs and good compatibility with five-membered azaarenes undergoing ring expansion. Moreover, DFT calculations of plausible mechanisms have motivated the implementation of an important cascade diradical recombination strategy for 1,3-dienones, thus facilitating the synthesis of valuable 2-oxabicyclo[3.1.0]hex-3-ene derivatives.

Abstract:Silacycle is one of the most essential core frameworks in silicon-containing functional molecules. However, the structural diversity of silacycles has been largely limited due to the lack of general synthetic methods. Here, we report an efficient synthesis of exo-cyclic enoate-substituted silacyclohexenes by the ring expansion of silacyclobutanes with allenoates. The reaction proceeds with two regioselectivities during Si–C bond insertion. In the presence of the Pd/PR3 catalyst, unsubstituted allenoates undergo β, γ-insertion to form a Si–Cβ bond, giving 2-(E)-enoate-substituted silacyclohexenes. In this pathway, a chiral phosphoramidite ligand is used to construct the stereogenic silicon center enantioselectively. In the second pathway, in the presence of the PtCl2 catalyst, α-substituted allenoates undergo γ, β-insertion to form a Si–Cγ bond, leading to 3-(E)-enoate-substituted silacyclohexenes. The control experiments and density functional theory calculations have been performed to understand the regio- and stereochemical outcome of both Pd- and Pt-catalyzed ring expansion reactions.

Abstract:The development of innovative strategies for the synthesis of N-heterocyclic compounds is an important topic in organic synthesis. Ring expansion methods to form large N-heterocycles often involve the cycloaddition of strained aza rings with π bonds. However, in some cases such strategies suffer from some limitations owing to the difficulties in controlling the regioselectivity and the accessibility of specific π-bond synthons. Here, we report the development of a general ring expansion strategy that involves a formal cross-dimerization between three-membered aza heterocycles and three- and four-membered-ring ketones through synergistic bimetallic catalysis. These formal cross-dimerizations of two different strained rings are efficient and scalable, and provide a straightforward and broadly applicable means of assembling diverse N-heterocycles, such as 3-benzazepinones, dihydropyridinones and uracils, which are versatile units in numerous drugs and biologically active compounds. Preliminary mechanistic studies revealed that the C–C bond of strained ring ketones is first cleaved by the Pd<sup>0</sup> species during the reaction.

Abstract:Saturated N-heterocycles are ubiquitous structures among natural products and biologically active compounds. Therefore, the development of synthetic methods for the construction of N-heterocycles is of great importance in the synthetic community. Altering the ring system of these motifs to analogues with different ring sizes by employing molecular editing techniques would be highly appealing in medicinal chemistry. We present herein the direct insertion of glycine derivatives as two-carbon synthons into unstrained five- or six-membered saturated cyclic amines at predictable sites, enabling the construction of synthetically challenging medium-sized azacycles through sequential Ru-catalysed C‒C bond formation, retro-aza-Michael addition and a lactamization process. Upon further derivation, we leverage this homologation platform to realize modular insertion of one- or two-carbon units into the aliphatic rings. The conversion of a single azacycle into up to five others provides a promising toolbox for diversifying existing drug candidates and increasing the prospects for clinical success.
