Xiaoyankangjun Tablet Alleviates Dextran Sulfate Sodium-Induced Colitis by Regulating Nrf2/HO-1 and JAK2/STAT3 Pathway and Gut Microbiota

Abstract:Ethnopharmacological relevance: Xiaoyankangjun Tablet (XYKJP) is a Traditional Chinese Medicine compound preparation for gastroenteritis, which shown good therapeutic effects on bacillary dysentery and gastroenteritis in clinical practice in Renmin Hospital of Wuhan University for more than 20 years. Given that some of the clinical symptoms of gastroenteritis are similar to those of colitis, and our study found that XYKJP had a therapeutic effect on experimental colitis, but the corresponding mechanism need to be clarified.Aim of the study: To investigate the pharmacodynamics and related mechanisms for the treatment of colitis about XYKJP, and to clarify the antioxidative, anti-inflammatory and regulating intestinal flora effects of XYKJP and its main components.Materials and methods: Dextran sulfate sodium (DSS)-induced colitis C57BL/6 mice, lipopolysaccharide (LPS)-induced RAW264.7 cells and NCM460 cells were used as an inflammatory model to evaluate the effect of XYKJP in vitro and vivo.Results: XYKJP and its ingredients alleviated pathological symptoms in DSS-induced colitis mice. In addition, XYKJP regulated the activation of the Nrf2/HO-1 and JAK2/STAT3 pathways and significantly decreased the levels of ROS, proinflammatory factors and anti-inflammatory factors. Moreover, XYKJP upregulated the protein expression of claudin-1, ZO-1 and occludin-1. The same results were confirmed in LPS-stimulated RAW264.7 cells and NCM460 cells in vitro. Furthermore, 16s rRNA gene sequencing results showed that XYKJP increased the relative abundance of Akkermansia, Ruminococcaceae_UCG-013 and Bifidobacterium and reduced the relative abundance of Coriobacteriaceae_UCG-002. Meanwhile, XYKJP increased the content of SCFAs, including acetic acid and butyric acid.Conclusion: XYKJP and its components could improve colitis by antioxidant and anti-inflammatory and regulating the imbalance of the gut microbiota and SCFAs. The Nrf2/HO-1 and JAK2/STAT3 pathways might be possible signaling pathways.
