Abstract:Recent studies have shown that YKL-40, a secreted glycoprotein, is highly expressed in patients with advanced cancers including breast cancer, colorectal cancer, ovarian cancer, lung cancer, and brain tumors. We have identified YKL-40 as an angiogenic molecule capable of stimulating endothelial cells in different types of cancers. Nevertheless, the therapeutic value of targeting YKL-40 in human cancers remains unknown. Therefore, it was the goal of this study to develop a monoclonal antibody against YKL-40 that could bind specifically and inhibit YKL-40 function with regard to cancer progression. A mouse monoclonal anti-YKL-40 antibody (mAY) was developed using hybridoma technology. mAY could specifically recognize recombinant YKL-40, as well as secreted YKL-40 from the media of two cancerous cell lines, MG63 and U87. Additionally, in functional testing, we demonstrated that mAY inhibited in vitro tube formation on Matrigel of endothelial cells induced by the conditioned media of MG63 and U87 cells. Recombinant YKL-40 treated endothelial cells displayed increased tube formation. mAY significantly abrogated tube formation by blocking recombinant YKL-40. Moreover, mAY reduced YKL-40 activated VEGF receptor 2 (Flk-1/KDR) and intracellular signaling MAP (mitogen-activated protein) kinase extracellular signal-regulated kinase (Erk) 1 and Erk 2. In relation to radiation therapy, treatment of gamma-irradiated U87 cells with mAY induced cell death through a down-regulation of p-AKT and AKT protein expression. Lastly, mAY treatment of U87 xenograft tumors diminished tumor volume, angiogenesis, and progression. These results demonstrate for the first time the therapeutic value of targeting YKL-40 in human cancers which highlights the potential benefits of anti-YKL-40 therapy in patients with a wide spectrum of cancers that over-produce YKL-40. Abstract:Drugs specifically targeting YKL-40, an over-expressed gene ( CHI3L1 ) in various diseases remain developed. The current study is to create a humanized anti-YKL-40 neutralizing antibody and characterize its potentially therapeutic signature. We utilized in silico CDR-grafting bioinformatics to replace the complementarity determining regions (CDRs) of human IgG1 with mouse CDRs of our previously established anti-YKL-40 antibody (mAY). In fifteen candidates (VL1-3/VH1-5) of heavy and light chain variable region combination, one antibody L3H4 named Rosazumab demonstrated strong binding affinity with YKL-40 (K D = 4.645 × 10 −8 M) and high homology with human IgG (80 %). In addition, we established different overlapping amino acid peptides of YKL-40 and found that Rosazumab specifically bound to residues K337, K342, and R344, the KR-rich functional domain of YKL-40. Rosazumab inhibited migration and tube formation of YKL-40-expressing tumor cells and induced tumor cell apoptosis. Mechanistically, Rosazumab induced interaction of N-cadherin with β-catenin and activation of downstream MST1/RASSF1/Histone H2B axis, leading to chromosomal DNA breakage and cell apoptosis. Treatment of xenografted tumor mice with Rosazumab twice a week for 4 weeks inhibited tumor growth and angiogenesis, but induced tumor apoptosis. Rosazumab injected in mice distributed to blood, tumor, and other multiple organs, but did not impact in function or structure of liver and kidney, indicating non-detectable toxicity in vivo . Collectively, the study is the first one to demonstrate that a humanized YKL-40 neutralizing antibody offers a valuable means to block tumor development.

Abstract:Tumor angiogenesis is of paramount importance in solid tumor development. Elevated serum levels of YKL-40, which is a secreted heparin-binding glycoprotein, have been associated with a worse prognosis from various advanced human cancers. Yet the role of YKL-40 activity in these cancers is still missing. In this study, we showed that ectopic expression of YKL-40 in MDA-MB-231 breast cancer cells and in HCT-116 colon cancer cells led to larger tumor formation with an extensive angiogenic phenotype than did control cancer cells in mice. Affinity-purified recombinant YKL-40 protein promoted vascular endothelial cell angiogenesis in vitro , the effects of which are similar to the activities observed using MDA-MB-231 and HCT-116 cell-conditioned medium after transfection with YKL-40. Furthermore, YKL-40 was found to induce coordination of membrane-bound receptor syndecan-1 and integrin α v β 3 and to activate an intracellular signaling cascade, including focal adhesion kinase and mitogen-activated protein kinase extracellular signal-related kinase1/2 in endothelial cells. Moreover, blockade of YKL-40 using small-interfering RNA gene knockdown suppressed tumor angiogenesis in vitro and in vivo . Immunohistochemical analysis of human breast cancer showed a correlation between YKL-40 expression and blood vessel density. These findings provide novel insights into angiogenic activities and molecular mechanisms of YKL-40 in cancer development.
Anti-YKL-40 Antibody and Ionizing Irradiation Synergistically Inhibit Tumor Vascularization and Malignancy in Glioblastoma

Abstract:Chemo/radiotherapies are the most common adjuvant modality treated for patients with glioblastoma (GBM) following surgery. However, the overall therapeutic benefits are still uncertain, as the mortality remains high. Elevated expression of YKL-40 in GBM was correlated with increases in mural cell-associated vessel coverage, stability and density, and decreases in vessel permeability and disease survival. To explore the potential role of YKL-40 in mural cell-mediated tumor vascularization, we employed an anti-YKL-40 neutralizing antibody (mAY) and ionizing irradiation (IR) in xenografted brain tumor models. Although single treatment with mAY or IR partially increased mouse survival, their combination led to dramatic inhibition in tumor growth and increases in mouse survival. mAY blocked mural cell-mediated vascular stability, integrity and angiogenesis; whereas IR merely promoted tumor cell and vascular cell apoptosis. Vascular radioresistance is at least partially attributed to expression of YKL-40 in mural cells. These divergent effects were also recapitulated in cultured systems using endothelial cells and mural cells differentiated from glioblastoma stem-like cells (GSCs). Dysfunction of intercellular contact N-cadherin was found to mediate mAY-inhibited vascularization. Collectively, the data suggest that the conjunction therapy with mAY and IR synergistically inhibit tumor vascularization and progression. The evidence may shed light on a new adjuvant therapy in clinic.

Abstract:Glioblastoma is one of the most fatal cancers, characterized by a strong vascularized phenotype. YKL-40, a secreted glycoprotein, is overexpressed in patients with glioblastomas and has potential as a novel tumor biomarker. The molecular mechanisms of YKL-40 in glioblastoma development, however, are poorly understood. Here, we aimed to elucidate the role YKL-40 plays in the regulation of VEGF expression, tumor angiogenesis, and radioresistance. YKL-40 up-regulated VEGF expression in glioblastoma cell line U87, and both YKL-40 and VEGF synergistically promote endothelial cell angiogenesis. Interestingly, long term inhibition of VEGF up-regulated YKL-40. YKL-40 induced coordination of membrane receptor syndecan-1 and integrin αvβ5, and triggered a signaling cascade through FAK(397) to ERK-1 and ERK-2, leading to elevated VEGF and enhanced angiogenesis. In addition, γ-irradiation of U87 cells increased YKL-40 expression that protects cell death through AKT activation and also enhances endothelial cell angiogenesis. Blockade of YKL-40 activity or expression decreased tumor growth, angiogenesis, and metastasis in xenografted animals. Immunohistochemical analysis of human glioblastomas revealed a correlation between YKL-40, VEGF, and patient survival. These findings have shed light on the mechanisms by which YKL-40 promotes tumor angiogenesis and malignancy, and thus provide a therapeutic target for tumor treatment.

Abstract:Proceedings: AACR 101st Annual Meeting 2010‐‐ Apr 17‐21, 2010; Washington, DC Growing evidence from clinical investigations has shown that increased serum levels of YKL-40 are correlated with short survival and metastasis in a broad type of human cancers. These data suggest that YKL-40 in the blood may present a powerful tool for cancer diagnosis and prognosis. However, pathological activity of YKL-40 in cancer development is poorly understood. Here, we explored a functional role for YKL-40 in vasculature development of human cancer. In a survey of twenty cases of breast ductal carcinoma in situ (DCIS) and thirty-eight cases of infiltrating ductal carcinoma (IDC), we found that expression levels of YKL-40 in specimens from DCIS were barely detectable, but different levels of YKL-40 were present in IDC. Nine out of thirty-eight IDC cases showed high levels of YKL-40 and nine cases displayed medium levels of YKL-40, whereas the rest twenty cases were low or negative. Interestingly, in an analysis of vasculature formation in these IDC samples, we found that different blood vessel densities significantly correlated with different degrees of YKL-40 expression. To test this possible angiogenic activity of YKL-40 in tumor development, we utilized angiogenic xenograft models by subcutaneous injection of SCID/Beige mice with a brain tumor line U87 cells which express strong endogenous YKL-40 and U87 cells with siRNA knockdown of YKL-40. siRNA knockdown of YKL-40 notably suppressed tumor angiogenesis and tumor development relative to high levels of YKL-40 expressed in control U87 cells. Further, to dissect cellular and molecular mechanisms underlying YKL-40-induced tumor angiogenesis, we treated human microvascular endothelial cells with YKL-40 and found that YKL-40 was capable of binding to heparan sulfate on ectodomain of membrane receptor syndecan-1 (S1). The activated S1 prompted its interaction with adjacent receptor integrin αvβ3, leading to activation of intracellular cascades including phosphorylation of focal adhesion kinase and MAP kinase. A series of individual signaling inhibitors blocked YKL-40-induced angiogenesis in vitro. Collectively, these data strongly suggest that YKL-40 expressed in cancer activates vascular endothelial cell angiogenesis to promote tumor development. These findings may hold promise for developing novel therapeutic agents targeting YKL-40, a cancer biomarker that is overexpressed in a wide range of human cancers. Abstract:A secreted glycoprotein YKL-40 also named chitinase-3-like-1 is normally expressed by multiple cell types such as macrophages, chondrocytes, and vascular smooth muscle cells. However, a prominently high level of YKL-40 was found in a wide spectrum of human diseases including cancers and chronic inflammatory diseases where it was strongly expressed by cancerous cells and infiltrating macrophages. Here, we summarized recent important findings of YKL-40 derived from cancerous cells and smooth muscle cells during tumor angiogenesis and development. YKL-40 is a potent angiogenic factor capable of stimulating tumor vascularization mediated by endothelial cells and maintaining vascular integrity supported by smooth muscle cells. In addition, YKL-40 induces FAK-MAPK signaling and up-regulates VEGF receptor 2 in endothelial cells; but a neutralizing antibody (mAY) against YKL-40 inhibits its angiogenic activity. While YKL-40 is essential for angiogenesis, little is known about its functional role in tumor-associated macrophage (TAM)-mediated tumor development. Therefore, significant efforts are urgently needed to identify pathophysiological function of YKL-40 in the dynamic interaction between tumor cells and TAMs in the tumor microenvironment, which may offer substantial mechanistic insights into tumor angiogenesis and metastasis, and also point to a therapeutic target for treatment of cancers and other diseases.

Abstract:Introduction: Approved angiogenesis inhibitors generally target certain growth factors or their receptors, which exist in both normal and cancerous cells; this results in suppression of cell-signaling pathways throughout the body and causes adverse effects. For this reason, there is a need for a new angiogenesis inhibitor that can specifically target the tumor vasculature. In our previous study, we revealed that doppel, a prion-like protein, was overexpressed specifically in the tumor vasculatures, but not in normal endothelium, and its expression enhanced blood vessel formation. To develop our new anti-angiogenic agent, we produced monoclonal antibodies which target doppel as the antigen. Methods: We evaluated whether the doppel antibody inhibits angiogenesis by a spheroid assay. We cultured human colonic tumor-associated endothelial cells (HCTEC) and collected the cells by a hanging drop method. After producing compact aggregates, the spheroids were collected and embedded into collagen-based 3D cultures. The culture media/growth factor/doppel antibody mixture was added to each well that contained 3D spheroid cultures. After 24 hours, the number of sprouts was counted and analyzed. Further, HCTECs were incubated with culture media, growth factor, and doppel antibodies for investigating the underlying mechanisms. Each well was lysed, and we conducted western blot and microarray experiments using the lysates. In addition, we injected fluorescent dye-conjugated doppel antibody into tumor xenograft mouse models and visualized the tumor-targeting efficacy using in vivo imaging system (IVIS). Results: In spheroid assays, we found that doppel antibody SNU-H01 and SNU-H02 inhibit angiogenesis (by 32% and 29%, respectively) compared to control. In anti-phosphorylation assays using western blot, the amount of phosphorylated VEGFR2 and phosphorylated FGFR1 was decreased in doppel antibody SNU-H01 and SNU-H02-treated groups. Also, when SNU-H02-treated lysate was added to the microarray kit, STAT5A and beta-catenin were inhibited. Inhibition of STAT5A may promote apoptosis and increase sensitivity to anticancer drugs, and inhibition of beta-catenin may further increase suppression of angiogenesis. In addition, doppel antibody SNU-H04 accumulates in the tumor significantly higher compared to IgG injected mouse group. Conclusion: Doppel antibodies in development, SNU-H01, -H02, -H03, -H04, will be screened and selected for their anti-angiogenic efficacy and favorable pharmacodynamic features. We hope that doppel antibody is expected to be a new and superior tumor vasculature-specific angiogenesis inhibitor that can overcome the limitations of existing anti-angiogenic agents. Abstract:Over the past two decades, emerging data have found that YKL-40, a secreted glycoprotein, is elevated in a broad spectrum of human diseases including cancers, liver injury, asthma, diabetes, inflammatory diseases, and cardiac disorders. In breast cancer, increased serum levels of YKL-40 are correlated with cancer metastasis and short survival, suggesting that serum levels of YKL-40 serve as a cancer biomarker. YKL-40 has the ability to stimulate vascular endothelial cell activation and suppress mammary epithelial cell differentiation, the pathophysiological events associated with tumor angiogenesis and poor differentiation. Neutralization of YKL-40 via an anti-YKL40 monoclonal antibody in animal trials demonstrates the ability of YKL-40 blockade to impede tumor angiogenesis and tumor growth, thus holding therapeutic promise for cancer therapy. Apart from these findings, substantial efforts are urgently required to decipher the key molecular mechanisms that mediate cancer metastasis and malignancy, which is expected to significantly offer translational value for breast cancer diagnosis, prognosis and therapy. This review discusses the current status of research on YKL-40’s expression, biophysiological and pathological activities and functional inhibition, which is instrumental for future clinical practice.

Abstract:Angiogenesis is an important process in cell development, especially in cancer. Vascular endothelial growth factor (VEGF) signaling is an important regulator of angiogenesis. Several therapies that act against VEGF signal transduction have been developed, including YN968D1, which is a potent inhibitor of the VEGF signaling pathway. This study investigated the antitumor activity of YN968D1 (apatinib mesylate) in vitro and in vivo. YN968D1 potently suppressed the kinase activities of VEGFR-2, c-kit and c-src, and inhibited cellular phosphorylation of VEGFR-2, c-kit and PDGFR beta. YN968D1 effectively inhibited proliferation, migration and tube formation of human umbilical vein endothelial cells induced by FBS, and blocked the budding of rat aortic ring. In vivo, YN968D1 alone and in combination with chemotherapeutic agents effectively inhibited the growth of several established human tumor xenograft models with little toxicity. A phase I study of YN968D1 has shown encouraging antitumor activity and a manageable toxicity profile. These findings suggest that YN968D1 has promise as an antitumor drug and might have clinical benefits. (Cancer Sci 2011; 102: 1374-1380)

Abstract:Abstract Introduction: Intrinsic or acquired resistance to cancer therapy is a major problem in the treatment of many cancers. Therapy-resistant cancer cells are able to evade the effect of cancer drugs, resulting in highly aggressive clones that can cause poor long-term prognosis. Neuropilin-2 (NRP2) is a cell surface receptor that acts as a co-receptor for vascular endothelial growth factor (VEGF) and has been shown to be associated with therapy resistance. The VEGF-C signaling pathway has also been linked to chemoresistance in several cancer types including breast, bladder, and gastric cancer. Previously we have shown that ATYR2810—a highly specific humanized monoclonal antibody that blocks NRP2/VEGF-C signaling—sensitizes triple negative breast cancer (TNBC), a highly aggressive and chemoresistant subtype of breast cancer, to chemotherapy and targeted anti-VEGF therapy both in vitro and in vivo. In this study we examined the role of enhanced VEGF-C signaling in tumor growth and therapeutic resistance, and the ability of ATYR2810 to neutralize these effects using in vivo models for lung and kidney cancers. Results: We generated a stable VEGF-C overexpressing non-small cell lung cancer (NSCLC) cell line and found that tumor growth was enhanced by increased VEGF-C signaling. Importantly, ATYR2810 monotherapy counteracted this VEGF-C-driven enhanced tumor growth as tumor growth inhibition was increased in the VEGF-C overexpressing tumors compared to control tumors. Further, VEGF-C overexpression was also found to enhance resistance to platinum-based chemotherapy in this model. However, sensitivity to chemotherapy was restored when ATYR2810 was used in combination with cisplatin. In clear cell renal cell carcinoma (ccRCC), the use of sunitinib—a small molecule inhibitor that inhibits the signaling of several receptor tyrosine kinases (RTKs), including vascular endothelial growth factor receptors (VEGFRs)—has reportedly been associated with the upregulation of VEGF-C and NRP2 expression as well as increased metastasis. In line with this, we found that the use of sunitinib leads to increased levels of circulating VEGF-C in the serum of ccRCC xenograft mice. The use of ATYR2810 in combination with sunitinib was found to act synergistically to inhibit tumor growth in ccRCC xenograft tumors leading to complete tumor regression in some animals. Conclusions: Here we demonstrate that high levels of VEGF-C expression increase tumor dependency on the VEGF-C/NRP2 signaling axis thereby promoting tumor growth and chemoresistance. ATYR2810 counteracts the aggressive traits of tumors expressing high levels of VEGF-C resulting in enhanced tumor growth inhibition and sensitivity to both chemotherapy and targeted therapy. Abstract:AB598 is a CD39 inhibitory antibody being pursued for the treatment of solid tumors in combination with chemotherapy and immunotherapy. CD39 metabolizes extracellular ATP (eATP), an alarmin capable of promoting anti-tumor immune responses, into adenosine, an immuno-inhibitory metabolite. By inhibiting CD39, the consumption of eATP is reduced, resulting in a pro-inflammatory milieu in which eATP can activate myeloid cells to promote anti-tumor immunity. The preclinical characterization of AB598 provides a mechanistic rationale for combining AB598 with chemotherapy in the clinic. Chemotherapy can induce ATP release from tumor cells and, when preserved by AB598, both chemotherapy-induced eATP and exogenously added ATP promote the function of monocyte-derived dendritic cells via P2Y11 signaling. Inhibition of CD39 in the presence of ATP can promote inflammasome activation in in vitro-derived macrophages, an effect mediated by P2X7. In a MOLP8 murine xenograft model, AB598 results in full inhibition of intratumoral enzymatic activity, an increase in intratumoral ATP, a decrease of extracellular CD39 on tumor cells, and ultimately, control of tumor growth. In cynomolgus monkeys, systemically dosed AB598 results in effective enzymatic inhibition in tissues, full peripheral and tissue target engagement, and a reduction in cell surface CD39 both in tissues and in the periphery. Taken together, these data support a promising therapeutic strategy of harnessing the eATP generated by standard-of-care chemotherapies to prime the tumor microenvironment for a productive anti-tumor immune response.
