Abstract:Many monoclonal antibodies (mAbs) have been extensively used in the clinic, such as rituximab to treat lymphoma. However, resistance and non-responsiveness to mAb treatment have been challenging for this line of therapy. Complement is one of the main mediators of antibody-based cancer therapy via the complement-dependent cytolysis (CDC) effect. CD59 plays a critical role in resistance to mAbs through the CDC effect. In this paper, we attempted to investigate whether the novel CD59 inhibitor, recombinant ILYd4, was effective in enhancing the rituximab-mediated CDC effect on rituximab-sensitive RL-7 lymphoma cells and rituximab-induced resistant RR51.2 cells. Meanwhile, the CDC effects, which were mediated by rituximab and anti-CD24 mAb, on the refractory multiple myeloma (MM) cell line ARH-77 and the solid tumor osteosarcoma cell line Saos-2, were respectively investigated. We found that rILYd4 rendered the refractory cells sensitive to the mAb-mediated CDC effect and that rILYd4 exhibited a synergistic effect with the mAb that resulted in tumor cells lysis. This effect on tumor cell lysis was apparent on both hematological tumors and solid tumors. Therefore, rILYd4 may serve as an adjuvant for mAb mediated-tumor immunotherapy.

Abstract:Antibody-dependent cellular cytotoxicity (ADCC) is a mechanism by which immune effector cells engage the Fc portion of antibodies bound to antigen on the surface of another cell, thereby targeting that cell for destruction. In mice, the Fcgr4 receptor is primarily responsible for triggering ADCC, while the FCGR3A receptor (CD16A) mediates this function in human NK cells, which are an attractive target cell type for cancer immunotherapy. Given these differences, we sought to generate humanized CD16A preclinical models to evaluate the efficacy of antibodies in triggering human CD16A-mediated ADCC in vivo. Previously, we established humanized CD16A mice on immunocompetent backgrounds for evaluating efficacy of novel antibodies in preventing growth of syngeneic tumor models. Here, we introduce a novel humanized CD16A mouse on the CB-17-SCID background, in order to inoculate human tumor cells for therapeutic efficacy testing. We confirmed expression of CD16A in human NK cells and monocytes/macrophages, but not granulocytes in the model; expression of murine Fcgr4 was no longer detectable, indicating successful generation of the model. Assessment of leukocyte populations in the humanized model indicated no significant alterations in the percentages of NK cells, dendritic cells, granulocytes, monocytes and macrophages in the spleen or blood of the humanized mice compared with wild-type CB-17 SCID mice. Subsequently, the ability of these mice to grow human tumor cells was evaluated: B-hHep3B-Luc, HC116, K-562, and NCI-N87 tumors were all successfully established. Finally, efficacy of an anti-human CLDN18.2 antibody was evaluated against B-hCLDN18.2 BxPC-3 tumors in the humanized mice, which showed modest anti-tumor efficacy at 30 mg/kg when compared to isotype control-treated mice. In summary, humanized B-hCD16A mice (CB-17-SCID) provide a suitable model for evaluating the ability of antibody candidates to induce ADCC. Abstract:We evaluated the capacity of freshly isolated blood monocytes to mediate antibody-dependent cellular-mediated cytotoxicity (ADCC) in cooperation with murine anti-tumor monoclonal antibodies (MAbs). Blood monocytes isolated from most donors by adherence selection to fibronectin-coated plastic surfaces and subsequently depleted of natural killer/killer (NK/K) cells exhibited significant ADCC activity against tumor cell lines in combination with an IgG3 antitumor MAb (BR55-2). However, significant variation in ADCC competence was observed among donors. Culture parameters influencing monocyte ADCC activity were evaluated and optimized. The influence of MAb isotype on ADCC capacity of anti-tumor MAbs was also evaluated using anti-tumor class-switch variant hybridoma proteins and a panel of anti-tumor MAbs. MAbs of the IgG2a and IgG3 subclasses exhibited high ADCC potential, whereas MAbs of the IgG2b subclass exhibited no ADCC activity. One of two IgG1 MAbs tested exhibited high ADCC activity with monocyte effectors. The role of monocytes or macrophages in tumor remission of cancer patients undergoing MAb immunotherapy is not known. However, correlative studies of monocyte ADCC capacity and responsiveness of cancer patients to MAb immunotherapy may help to establish the role of these effectors in MAb-mediated tumor remissions.
