Abstract:Abstract Despite the remarkable efficacy of immune checkpoint inhibitor (ICI) immunotherapy in various types of human cancers, colon cancer, except for the approximately 4% microsatellite-instable (MSI) colon cancer, does not respond to ICI immunotherapy. ICI acts through activating CTLs that use the Fas–FasL pathway as one of the two effector mechanisms to suppress tumor. Cancer stem cells are often associated with resistance to therapy including immunotherapy, but the functions of Fas in colon cancer apoptosis and colon cancer stem cells are currently conflicting and highly debated. We report here that decreased Fas expression is coupled with a subset of CD133+CD24lo colon cancer cells in vitro and in vivo. Consistent of the lower Fas expression level, this subset of CD133+CD24loFaslo colon cancer cells exhibits decreased sensitivity to FasL-induced apoptosis. Furthermore, FasL selectively enriches CD133+CD24loFaslo colon cancer cells. CD133+CD24loFaslo colon cancer cells exhibit increased lung colonization potential in experimental metastatic mouse models and decreased sensitivity to tumor-specific CTL adoptive transfer and ICI immunotherapies. Interestingly, FasL challenge selectively enriched this subset of colon cancer cells in microsatellite-stable (MSS) but not in the MSI human colon cancer cell lines. Consistent with the downregulation of Fas expression in CD133+CD24lo cells, lower Fas expression level is significantly correlated with decreased survival in patients with human colon cancer. Implications: Our data determine that CD133+CD24loFaslo colon cancer cells are capable to evade Fas-FasL cytotoxicity of tumor-reactive CTLs and targeting this subset of colon cancer cells is potentially an effective approach to suppress colon cancer immune evasion.

Abstract:Immunotherapy is currently approved for CRC whose tumors have high MSI-H. To find additional biomarkers for immunotherapy in CRC, targeted sequencing was performed on tumor tissues from a discovery cohort of 161 CRC patients. Validation cohorts from the cBioPortal were also used for survival and tumor cell infiltration analyses. The FAT1-mutated CRC group often co-occurred with MSI events and displayed a higher tumor mutational burden compared to the FAT1 wild-type CRC. Overall survival was higher in patients with FAT1 mutations than in patients with wild type FAT1. The altered PI3K-AKT pathway and immune pathways were enriched in the FAT1-mutated CRC. A higher infiltration rate of immune cells including CD4+ T cells, CD8+ T cells, macrophages M1 and regulatory T cells were also observed in the colorectal tumors with FAT1 mutation compared to tumors with wild type FAT1. The results showed that CRC patients with FAT1 mutations exhibited an immunotherapy-favorable profile.

Abstract:AIM:To detect the expression of Fas ligand (FasL) in colon cancer tissues and cell lines and analyze the function of FasL-expressing colon cancer cells in inducing Fas-sensitive T lymphocyte apoptosis.METHODS:Ninety surgically resected colon cancer tissues and 15 hepatic metastasis specimens were investigated by immunohistochemical method with normal colon mucosa and colon adenoma as control. The relationship between FasL expression and pathologic features was also analyzed. FasL expression of 4 colon cancer cell lines, SW620, Lovo, LS-174T and SW1116, were detected by Western blotting assay. The function of FasL expressed on colon cancer cells was determined by coculture assay with Jurkat T lymphocytes, the apoptotic rate of which was detected by flow cytometry assay.RESULTS:Fifty-six (62.22%) cases of all the 90 colon cancer tissues and all (100%) the liver metastasis specimens expressed FasL, significantly higher than normal colon mucosa and colonic adenoma. Higher expression of FasL was found in more advanced stage of colon cancer and in cancer tissues with lymphatic or hepatic metastasis. All the colon cancer cell lines were found to express FasL. After coculture with the SW1116 cells for 24 h with an effector: target ratio 10:1, the rate of apoptosis of Jurkat cells rose from 1.9% to 21.0%.CONCLUSION:The expression of FasL is upregulated in colon cancer and the functionally expressed FasL can induce apoptosis of Fas-expressing T lymphocytes.

Abstract:Objective To study the FasL expression in colorectal carcinoma and its significance in immune escape of cancer.Methods The expression of FasL protein and the number of TIL in 80 specimens of colorectal carcinoma were detected by immunohistochemitry.The mRNA of FasL was measured by in situ hybridization in the consecutive tissue slices of 80 colorectal carcinomas respectively.Using terminal deoxynucleotidyl transferase-mediaed dUTP nick end labeling (TUNEL),apoptotic cells were detected in 80 specimens of colorectal carcinoma.Results The expression of FasL were detected in all 80 specimens,but it was not even in the same or among different tissues.In the consecutive tissue slices, the location of expression of FasL protein corresponded with that of FasL mRNA.In those FasL extensive expression, the number of TIL was lower than that of FasL weak expression (P<0.05),and the apoptotic index (AI) of TIL was higher and the AI of tumor cells was lower than that of FasL weak expression respectively (P<0.01).The AI of TIL was correlated with the AI of tumor cells (r=-0.631,P<0.01).Conclusion Colorectal carcinoma cells can induce the apoptosis of TIL through the expression of FasL,which can counterattack the immune system.This maybe a mechanism of immune evasion in colorectal carcinoma.

Abstract:OBJECTIVE:To investigate the role of Fas/Fas ligand (FasL) in tum orgenesis, immune escape, counterattack in colonic cancer. METHODS:Fas and FasL were measured by immunohitochemistry in 52 colon cancer, 27 adenomas, 28 ulcerative colitis (UC), 30 normal colon tissues and 23 lymph node metastases of colon cancer. In different FasL expression regions, the apoptosis of tumor cells was detected by TUNEL. RESULTS:(1) The expression rate of Fas was lowest in colon cancer tissue. It was lower than that in adenomas and UC, while it was highest in normal tissue. Compare of Fas, FasL was highest in colon cancer tissue, lowest in the normal tissue. The FasL in adenomas and UC were higher than that in the normal tissue. (2)Expression of Fas/FasL was not correlated with the typ e of tumor histology, but it was correlated with Dukes'stage, tumor differentiation and lymph node metastasis.(3) In the same tumor tissue, FasL expression was not even. Apoptotic rate of tumor cells was higher in FasL positive region(n = 25)(81.2%) than that in FasL negative region(n = 25)(47.4%)(P < 0.01). (4) FasL was strong positive in all 23 lymph node metastases of large intestine cancer, and more than 75% tumor cells were FasL positive in each tissue. CONCLUSION:Fas/FasL play an important role in tumorgenesis, immune escape, counterattack in colon cancer.

Abstract:Immunotherapy based on checkpoint inhibitors is providing substantial clinical benefit, but only to a minority of cancer patients. The current priority is to understand why the majority of patients fail to respond. Besides T-cell dysfunction, T-cell apoptosis was reported in several recent studies as a relevant mechanism of tumoral immune resistance. Several death receptors (Fas, DR3, DR4, DR5, TNFR1) can trigger apoptosis when activated by their respective ligands. In this review, we discuss the immunomodulatory role of the main death receptors and how these are shaping the tumor microenvironment, with a focus on Fas and its ligand. Fas-mediated apoptosis of T cells has long been known as a mechanism allowing the contraction of T-cell responses to prevent immunopathology, a phenomenon known as activation-induced cell death, which is triggered by induction of Fas ligand (FasL) expression on T cells themselves and qualifies as an immune checkpoint mechanism. Recent evidence indicates that other cells in the tumor microenvironment can express FasL and trigger apoptosis of tumor-infiltrating lymphocytes (TIL), including endothelial cells and myeloid-derived suppressor cells. The resulting disappearance of TIL prevents anti-tumor immunity and may in fact contribute to the absence of TIL that is typical of "cold" tumors that fail to respond to immunotherapy. Interfering with the Fas-FasL pathway in the tumor microenvironment has the potential to increase the efficacy of cancer immunotherapy.
Loss of Fas Expression and High Expression of HLA-E Promoting the Immune Escape of Early Colorectal Cancer Cells.

Abstract:Previous studies have investigated the mechanisms of immune evasion of tumor cells in numerous types of advanced solid malignant tumor, and several types of immune preparations have been administered as antitumor adjuvant therapies. However, in the majority of studies, the efficacy of therapies has been revealed to be limited. The present study aimed to investigate the immune evasion mechanisms employed by early colorectal cancer cells and the expression of the molecules associated with immune evasion during the malignant transformation process of normal colorectal epithelial cells to measure the effects of immune intervention for early colorectal cancer, and to improve the efficacy of immunotherapy. A total of 60 colorectal tissues, including 15 normal mucosa, 15 adenoma, 15 early cancer and 15 advanced cancer tissues, from patients undergoing endoscopic procedures in Huadong Hospital Affiliated to Fudan University (Shanghai, China) were collected. A comparison of baseline characteristics among these four groups was performed. The expression levels of human leukocyte antigen-A (HLA-A), apoptosis antigen 1 (Fas), c-c chemokine receptor type 5 (CCR5), Fas ligand (FasL) and HLA-E in each group were detected by immunohistochemical analysis. Furthermore, 15 patients with advanced colorectal cancer were enrolled into the present study. Advanced cancer and paracancer tissues (normal mucosal tissues 3 cm away from the margin of cancer tissues) were collected from each patient by colonoscopic biopsy. The expression levels of HLA-A, Fas, CCR5, FasL and HLA-E in each group were detected by western blot analysis. During the malignant transformation process of normal colorectal epithelial cells, the expression levels of CCR5, FasL and HLA-E increased significantly (P<0.001), whilst the expression levels of Fas reduced significantly (P=0.0271). In the early cancer group, the expression levels of Fas reduced significantly (P=0.0239), whilst the expression levels of HLA-E increased significantly (P<0.001) compared with adenoma group. In conclusion, a loss of Fas expression and high expression levels of HLA-E may promote the immune evasion of early colorectal cancer cells.
Blockade of Fas Signaling in Breast Cancer Cells Suppresses Tumor Growth and Metastasis Via Disruption of Fas Signaling-initiated Cancer-related Inflammation*

Abstract:Mechanisms for cancer-related inflammation remain to be fully elucidated. Non-apoptotic functions of Fas signaling have been proposed to play an important role in promoting tumor progression. It has yet to be determined if targeting Fas signaling can control tumor progression through suppression of cancer-related inflammation. In the current study we found that breast cancer cells with constitutive Fas expression were resistant to apoptosis induction by agonistic anti-Fas antibody (Jo2) ligation or Fas ligand cross-linking. Higher expression of Fas in human breast cancer tissue has been significantly correlated with poorer prognosis in breast cancer patients. To determine whether blockade of Fas signaling in breast cancer could suppress tumor progression, we prepared an orthotopic xenograft mouse model with mammary cancer cells 4T1 and found that blockade of Fas signaling in 4T1 cancer cells markedly reduced tumor growth, inhibited tumor metastasis in vivo, and prolonged survival of tumor-bearing mice. Mechanistically, blockade of Fas signaling in cancer cells significantly decreased systemic or local recruitment of myeloid derived suppressor cells (MDSCs) in vivo. Furthermore, blockade of Fas signaling markedly reduced IL-6, prostaglandin E2 production from breast cancer cells by impairing p-p38, and activity of the NFκB pathway. In addition, administration of a COX-2 inhibitor and anti-IL-6 antibody significantly reduced MDSC accumulation in vivo. Therefore, blockade of Fas signaling can suppress breast cancer progression by inhibiting proinflammatory cytokine production and MDSC accumulation, indicating that Fas signaling-initiated cancer-related inflammation in breast cancer cells may be a potential target for treatment of breast cancer.
[Apoptosis of Lymphocytes Induced by Fas/FasL of Colon Cancer Cells].

Abstract:BACKGROUND & OBJECTIVE:It has been reported that apoptotic rate of tumor infiltrating lymphocytes(TIL) in Fas ligand(FasL) positive region was higher than in FasL negative region, which indicated that tumor cells could kill tumor infiltrating lymphocytes through high-expressed FasL. This study was designed to determine whether colon cancer cells SW480 could induce apoptosis of lymphocytes by Fas system in vitro.METHODS:Colon cancer cells SW480 were co-cultured with Jurkat cells with different effector-target(E-T) ratios of 20:1, 10:1, 5:1. Growth curve method, flow cytometry(FCM) analysis, fluorescence microscope were used to detect apoptosis of Jurkat cells.RESULTS:Flow cytometry revealed that the apoptotic rate of Jurkat cells were 1.8 +/- 0.21, 5.49 +/- 0.17, 11.18 +/- 0.14, and 18.22 +/- 0.11 with the SW480 planting concentrations at 0(control), 0.5 x 10(5)/ml, 1 x 10(5)/ml, 2 x 10(5)/ml, respectively. Fluorescence microscopy showed that the apoptotic rate of Jurkat cells were 1.58 +/- 0.12, 5.22 +/- 0.13, 9.74 +/- 0.21, and 19.33 +/- 0.18 with the SW480 planting concentrations at 0(control), 0.5 x 10(5)/ml, 1 x 10(5)/ml, and 2 x 10(5)/ml, respectively. Based on the three different E-T ratios, SW480 cell could all induce apoptosis of Jurkat. Furthermore, Jurkat cell apoptosis rate increased with the increase of planting concentration of SW480 and co-culture time. There were significant difference between each test group and control(All values P < 0.01).CONCLUSION:Colon cancer cells SW480 could induce apoptosis of lymphocytes throught Fas system, which providing theoretical foundation for metastasis and counterattack of colon cancer.
FasL Expression in Human Colon Carcinomas

Abstract:Objective: The Fas and Fas ligand (FasL) play an important role in maintaining immune privilege on malignant tumors. In present study, we investigated the expression of FasL in SW480 and LS174 human colon carcinoma cell lines and twenty primary colon carcinoma specimens. Methods: The expression of FasL in human colon carcinoma cell lines and primary colon carcinomas specimens was detected by immunohistochemistry and Reverse Transcription-PCR (RT-PCR). Results: We found that all of detected human colon carcinoma cell lines and primary colon carcinoma specimens constitutively expressed FasL at the mRNA and protein level. However, the expression of FasL was not found in normal colon epithelial cells. Conclusion: The expression of FasL may occur during malignant transformation from normal colon epithelial cells to colon carcinoma cells. Our results suggest that tumor cells kill cytotoxic T lymphocytes (CTLS) and natural killer (NK) cells by expression of FasL. It may be a new mechanism for tumor cells to escape the host’s immune surveillance. The expression of FasL may contribute to the formation of colon carcinomas.
Fas-mediated Pathway and Apoptosis in Normal Cervix, Cervical Intraepithelial Neoplasia and Cervical Squamous Cancer.

Abstract:The aim of this study was to examine the expression patterns of apoptosis-related antigens, such as Fas, FasL, and cFLIP, in cervical squamous cells, and investigate the role of Fas-mediated apoptosis in the pathogenesis of cervical neoplasia. Using specific antibodies for Fas, FasL, and cFLIP, we examined protein expression in 19 specimens of normal cervix, 15 mild dysplasia (CIN I), 22 moderate dysplasia (CIN II), 23 severe dysplasia (CIN III), and 34 invasive squamous cell carcinoma (SCC) by immunohistochemistry. We detected the apoptotic indices by TUNEL in the same specimens. Fas expression levels were quite similar in CIN I, CIN II and the normal cervix. Though Fas expression tended to increase in grade 2 cancer compared to grade 1 cancer and CIN III, and a slight decline was present in grade 3 compared with grade 2 cancer, these differences did not reach statistical significance. Almost all CINs did not express FasL, while FasL expression increased with the grade of the tumor. Statistically significant differences could be observed between grade 1 and grade 2 (p<0.01) and between grade 2 plus grade 3 and grade 1 (p<0.001). All cases of normal cervix and CIN, except two cases of CIN III, did not express cFLIP. cFLIP expression tended to increase with the grade of the tumor. Apoptosis was determined in all samples by TUNEL. There was a decreasing tendency of apoptotic cells from normal cervix to cancers. A negative correlation between cFLIP and apoptosis (r=-0.499 and p=0.000) was observed. Deregulated Fas/FasL system and constitutive expression of cFLIP in SCC may be an important mechanism by which SCCs escape apoptosis during the malignant transformation and progression of these tumors.

Abstract:The cellular Fas-associated death domain-like interleukin-1β-converting enzyme inhibitory protein-long form (c-FLIPL) is a key regulator of Fas signaling, although owing a dominant-negative homologue of caspase-8, the role of c-FLIPL remains controversial. In the present study, two pairs of small interfering RNA (siRNA) directed against c-FLIPL were used to assess the effect of c-FLIPL on Fas-mediated apoptosis of colon carcinoma in vitro. HT-29 cell line was selected for overexpression of c-FLIPL and Fas with RT–PCR and flow cytometry analyses. After electroporation, the mRNA level of c-FLIPL was significantly decreased (control siRNA versus c-FLIPL siRNA, 77.97±5.61% versus 26.22±3.79%) and the maximum interfering efficiency was around 66.49% using semi-quantitative RT–PCR analysis. Knockdown of c-FLIPL with the specific siRNA sensitized colon carcinoma cells to Fas-mediated apoptosis (control siRNA versus c-FLIPL siRNA, 5.68±2.11% versus 29.50±2.27%) using DNA content analysis and Annexin V–FITC analysis. In conclusion, our study indicated that c-FLIPL might be a suppressor of Fas-mediated apoptosis in Fas antigen expressing colon carcinoma and therefore a potential target for novel anticancer therapies.

We performed genome-wide chromatin immunoprecipitation sequencing analysis and identified that the FAS promoter is enriched with H3K9me3 in metastatic human colon carcinoma cells. The H3K9me3 level in the FAS promoter region is significantly higher in metastatic than in primary cancer cells, and it is inversely correlated with Fas expression level. We discovered that verticillin A is a selective inhibitor of histone methyltransferases SUV39H1, SUV39H2, and G9a/GLP that exhibit redundant functions in H3K9 trimethylation and FAS transcriptional silencing. Genome-wide gene expression analysis identified FAS as one of the verticillin A target genes. Verticillin A treatment decreased H3K9me3 levels in the FAS promoter and restored Fas expression. Furthermore, verticillin A exhibited greater efficacy than decitabine and vorinostat in overcoming colon carcinoma resistance to FasL-induced apoptosis. Verticillin A also increased DR5 expression and overcame colon carcinoma resistance to DR5 agonist drozitumab-induced apoptosis. Interestingly, verticillin A overcame metastatic colon carcinoma resistance to 5-fluorouracil in vitro and in vivo. Using an orthotopic colon cancer mouse model, we demonstrated that tumor-infiltrating cytotoxic T lymphocytes are FasL(+) and that FasL-mediated cancer immune surveillance is essential for colon carcinoma growth control in vivo. Our findings determine that H3K9me3 of the FAS promoter is a dominant mechanism underlying FAS silencing and resultant colon carcinoma immune evasion and progression.
Suppression of Tumor Cell Immune Counterattack by Blocking FasL Expression in Pancreatic Cancer Cells

Abstract:OBJECTIVE: To investigate if the supression of FasL expression in pancreatic cancer cells can inhibite tumor cell immune in vivo.METHODS: A cDNA fragment complementary,when transcribed,from nucleotides 35 to 371 of the mouse FasL mRNA sequence was amplified by PCR.The fragment was cloned into the StuI site of pBlast expression vector.Panc-1 cells were transfected with plasmid DNA.The suppression of FasL expression in stably transfected clones was determined by Western blot for FasL protein.Soluble FasL was detected in the culture supernatant of Panc-1 cells by ELISA.C57BL/6 mice were injected with tumor cells suspended.Tumor growth was monitored by measuring the width and length of the tumors.The level of lymphocyte infiltration in the excised tumors was assessed.RESULTS: FasL expression in hepatic cancer cells was down-regulated following stable transfection with a plasmid encoding antisense FasL cDNA,and with 80% decrease of FasL expression in plasmidtrans-trans pected cells.Down-regulation of FasL expression on pancreatic cancer cells had no effect on tumor growth in vitro,but significantly reduced tumor development in syngeneic immunocompetent mice in vivo.Mean tumor diameters was 1.53 mm in the group transfected with plasmid DNA,versus 2.98 mm in the group not transfected with plasmid DNA,and the difference was significant between the two groups,P0.01.Compared with the group not transfected with plasmid DNA,the absolute number of CD45+ TIL was increased by 3.23 times in the group transfected with plasmid DNA,P0.01.CONCLUSION: Down-regulation of FasL expression by pancreatic cancer cells results in an improved anti-tumor immune challenge in vivo,providing experimental evidence for counterattack as a mechanism of tumor immune evasion.

Abstract:Expression of membrane-bound Fas ligand (FasL) by colorectal cancer cells may allow the development of an immune-privileged site by eliminating incoming tumour-infiltrating lymphocytes (TILs) in a Fas-mediated counter-attack. Sporadic colorectal cancer can be subdivided into three groups based on the level of DNA microsatellite instability (MSI). High-level MSI (MSI-High) is characterized by the presence of TILs and a favourable prognosis, while microsatellite-stable (MSS) cancers are TIL-deficient and low-level MSI (MSI-Low) is associated with an intermediate TIL density. The purpose of this study was to establish the relationship between MSI status and FasL expression in primary colorectal adenocarcinoma. Using immunohistochemistry and a selected series of 101 cancers previously classified as 31 MSI-High, 30 MSI-Low, and 40 MSS, the present study sought to confirm the hypothesis that increased TIL density in MSI-High cancers is associated with low or absent membrane-bound FasL expression, while increased FasL in MSS cancers allows the killing of host TILs. TUNEL/CD3 double staining was also used to determine whether MSS cancers contain higher numbers of apoptotic TILs in vivo than MSI-High or MSI-Low cancers. Contrary to the initial hypothesis, it was found that MSI-High cancers were associated with higher FasL expression (p = 0.04) and a stronger intensity of FasL staining (p = 0.007). In addition, mucinous carcinomas were independently characterized by increased FasL expression (p = 0.03) and staining intensity (p = 0.0005). Higher FasL expression and staining intensity did not correlate with reduced TIL density or increased numbers of apoptotic TILs. However, consistent with the hypothesis that curtailment of the host anti-tumour immune response contributes to the poor prognosis in MSS cancers, it was found that apoptotic TILs were most abundant in MSS carcinomas and metastatic Dukes' stage C or D tumours (p = 0.004; p = 0.046 respectively). This study therefore suggests that MSS colorectal cancers are killing incoming TILs in an effective tumour counter-attack, but apparently not via membrane-bound FasL.
