Antibacterial Effect and Osteoblast Differentiation Promotion of Rose Bengal-Mediated Photodynamic Therapy

Abstract:Background and Objectives: Antibacterial and osteogenesis promotion are both required in peri-implantitis treatment. Photodynamic therapy (PDT) has been proven as an effective tool to inactivate pathogenic bacteria. Fusobacteriun nucleatum is the most frequently detected pathogenic periodontal bacteria species and the bridge of early and late bacteria adhesion. The aim of the present study was to investigate the effect of RB mediated PDT on F. nucleatum disruption as well as its effect on bone marrow mesenchymal stem cells proliferation and osteoblastic differentiation.Study design/materials and methods: Effect of RB in combination with 100mW 532nm diode laser PDT treatment on Fusobacteriun nucleatum (ATCC 25586)’s growth curve, viability, morphology, gene expression were detected by spectrometer, MTT, live/dead assay, SEM, RT-qPCR analysis. Intracellular reactive oxygen species (ROS) was detected by flow cytometry. Amount of singlet oxygen was demonstrated in the 399-nm OD decay curve. Phototoxicity of RB mediated PDT towards Bone marrow mesenchymal stem cells was investigated by CCK-8, alkaline phosphate (ALP) activity was stained and measured on day 7 and deposited calcium was detected by Alizarin-red staining test on day 21.Results: The MIC value for RB-mediated PDT was 50μg/mL+30s for Fusobacterium nucleatum inactivation, and the MBC value was 152μg/mL+30s. The dose-and time-dependent disruption of biofilm was confirmed by MTT, SEM and live/dead assay. 100μg/mLRB+30s produce more ROS and singlet oxygen than 100μg/mLRB+60s. 50μg/mLRB+30s group demonstrated the least phototoxicity towards Bone marrow mesenchymal stem cells. 50μg/mLRB+30s group can induce better alkaline phosphate (ALP) activity on day 7 after irradiation only inferior to 100μg/mLRB+30s and 50μg/mL+60s. And there was a significant calcium nodule formation on day 21 in 50μg/mLRB+30s group compared with the control one.Conclusion: These results demonstrate RB in combination with 532nm diode laser irradiation mediated PDT is effective in F. nucleatum inactivation with MIC of 50μg/mL+30s and MBC of 175μg/mL+30s. Meanwhile, 50μg/mL RB in combination with 30s 1.42mW/cm2 532nm diode laser irradiation mediated PDT demonstrated best in BMSCs osteoblast differentiation promotion with regard to less phototoxicity.
