Abstract:Natural Polysaccharides have been considered to be the most promising materials in recent years because of their numerous advantages including their accessibility, non-toxic, biocompatible, biodegradable, and ease of modification.Natural substanceswith pharmacotherapy potential are currently viewed as a promising future alternative to conventional therapy.With the rapid development of analytical techniquesandbiotechnologies,a large number of methods for identifying and quantifying polysaccharides without causing adverse effects have been created, resulting in the use of natural substances with established safety. Some bioactive polysaccharides obtained from natural sources have received a lot of attention in the field of biochemistry and pharmacology in recent years.Natural polysaccharides have been found in studies to boost innate immunity by activating upstream immune cells, hence regulating adaptive immunological pathways such as T cells and improving the efficacy of immunotherapy, implying that polysaccharides have a promising future in immunotherapy. As there is no targeted therapy available but these natural polysaccharides have been shown to possess complex, important and multifaceted biological activities including antitumor, anticoagulant, antioxidant, antiviral, immunomodulatory and antihyperlipidemic activities. Their properties are mainly due to their structural characteristics and are useful for combating various diseases as well improving immunity. Thus, polysaccharides can be used as an activating agent of immune system, minimising the damage caused by infectious agents such as SARS-CoV-2. Finally, these biopolymers can still be exploited to produce new treatments and vaccines.The present paper summarizes an overview of natural polysaccharides with immune boosting properties.

Abstract:Edible fungus polysaccharides are safety and natural polymers from edible fungus,they have many biologic activities.The studies on medical science,pharmacology and food science of the polysaccharides were made by domestic and abroad scholars.The best biologic activity of the polysaccharides was immunological regulation effect.The immunological regulation mechanisms of it were promoting immune allelotaxy,enhancing macrophage phagocytosis function,promoting lymphocyte proliferation,increasing humoral immune function,promoting immune cytokine and its mRNA expression,influencing transduction or activation of immunocyte signal as Ca2+,NO,cAMP and cGMP.Some edible fungus polysaccharides,such as lentinan and grifolan were used as adjunctive therapy on low immune function,tumor and infectious diseases.This article reviewed the immunological regulation effects and mechanisms of edible fungus polysaccharides in order to provide references on domestic fungus resource utilization.

Abstract:The human gut is colonized with a vast and diverse microbial ecosystem, and these bacteria play fundamental roles in the well being of our bodies. Gut-associated lymphoid tissues, the largest mucosal immune system, should never be overlooked for their profound effect in maintaining the host immunity. Therefore, we discussed the relationship between gut immunity and host health, primarily from two aspects: the homeostasis of gut microbiota, and the function of gut-associated lymphoid tissues. Polysaccharides, widely concerned as bioactive macromolecules in recent centuries, have been proved to benefit the intestinal health. Dietary polysaccharides can improve the ratio of probiotics, regulate the intestinal microenvironment like decreasing the gut pH, and stimulate the macrophages or lymphocytes in gut tissues to fight against diseases like cancer. Based on various experimental and clinical evidence, the impacts of dietary polysaccharides on intestinal health are summarized, in order to reveal the possible immunomodulatory mechanisms of polysaccharides.

Abstract:Polysaccharides,as a kind of important biological information macromolecules,have been indicated to exist widely in animals,plants and microbials.Currently,the role of polysaccharides in regulating immune system has been generally recognized by the academic community.The immunoregulatory mechanism of polysaccharides on the immune system was mainly via binding to pattern recognition receptors on the surface of immune cells such as Toll like receptors,mannose receptor,Dectin-1,scavenger receptors and complement receptors.Thus stimulating immune cells involves protomoting the phagocytosis of macrophages,regulating the proliferation and transformation of lymphocyte,enhancing the activation of complement system and inducing the secretion of cytokines so as to achieve the immunoregnlatory effects.In addition,the regulation of neuroendocrineimmunoregulatory network is regarded as a vital way to exert immunoregulatory effects of polysaccharides as well.The biological activity of polysaccharide and its immunomodulatory effects have always been one of the hot projects.The aim of this article was to review the effects and mechanisms of polysaccharides on immunoregulatory effects related to immune system.

Abstract:Seaweeds polysaccharides possess diverse structures and functions, extending beyond culinary uses to potentially offer health benefits. Notable among seaweed polysaccharides are fucoidans, carrageenans, galactans, agars, alginates, and laminarin etc. These bioactive compounds exhibit a range of biological activities, including antioxidant, anti-inflammatory, antiviral, and immunomodulatory effects. Seaweed polysaccharides, characterized by their unique structural complexity, present themselves as promising candidates for immunomodulation. This review provides an overview of the intricate connections between gut microbiota, immune system dynamics, and the prebiotic potential of seaweed polysaccharides. The review highlights the prebiotic effects of seaweed polysaccharides, examining their fermentation in the colon, promotion of beneficial bacteria, and inhibition of pathogens. This section presents the significant discoveries from the review, categorizing them based on the types of seaweeds studied which are brown, red, and green. It highlights the distinct prebiotic effects associated with the polysaccharide. The conclusion summarizes the key insights derived from the dynamic interplay of gut microbiota, seaweed polysaccharides, and their prebiotic effects.

Abstract:Bioactive substances derived from natural products are valued for effective health-related activities. As extremely important component of plants, animal cell membrane and microbes cytoderm, polysaccharides have been applied as medications, foods and cosmetics stemming from their prominent biological functions and minor side-effects. Recent studies indicate that polysaccharides exert biological effects also through epigenetic mechanism. Through the intervention of DNA methylation, histone modification, and non-coding RNA, polysaccharides participatate in regulation of immunity/inflammation, glucose and lipid metabolism, antioxidant damage and anti-tumor, which presents novel mechanism of polysaccharide exerting various functions. In this review, the latest advances in the biological functions of dietary polysaccharides via epigenetic regulations were comprehensively summarized and discussed. From the view point of epigenetic regulation, investigating the relationship between polysaccharides and biological effects will enhance our understandings of polysaccharides and also means huge breakthrough of molecular mechanism in the polysaccharide research fields. The paper will provide important reference to these investigators of polysaccharide research and expand the applications of dietary polysaccharides in the functional food developments.

Abstract:Pharmacological and clinical studies have consistently demonstrated that polysaccharides exhibit great potential on immune regulation. Polysaccharides can interact directly or indirectly with the immune system, triggering cell-cell communication and molecular recognition, leading to immunostimulatory responses. Gut microbiota is adept at foraging polysaccharides as energy sources and confers benefits in the context of immunity and chronic autoimmune disease, such as multiple sclerosis. A compelling set of interconnectedness between the gut microbiota, natural polysaccharides, and immune regulation has emerged. In this review, we highlighted the available avenues supporting the existence of these interactions, with a focus on cytokines-mediated and SCFAs-mediated pathways. Additionally, the neuroimmune mechanisms for gut microbiota communication with the brain in multiple sclerosis are also discussed, which will lay the ground for ameliorate multiple sclerosis via polysaccharide intervention.

Abstract:Central nervous system (CNS) disorders present a growing and costly global health challenge, accounting for over 11% of the diseases burden in high-income countries. Despite current treatments, patients often experience persistent symptoms that significantly affect their quality of life. Dietary polysaccharides have garnered attention for their potential as interventions for CNS disorders due to their diverse mechanisms of action, including antioxidant, anti-inflammatory, and neuroprotective effects. Through an analysis of research articles published between January 5, 2013 and August 30, 2023, encompassing the intervention effects of dietary polysaccharides on Alzheimer's disease, Parkinson's disease, depression, anxiety disorders, autism spectrum disorder, epilepsy, and stroke, we have conducted a comprehensive review with the aim of elucidating the role and mechanisms of dietary polysaccharides in various CNS diseases, spanning neurodegenerative, psychiatric, neurodevelopmental disorders, and neurological dysfunctions. At least four categories of mechanistic bases are included in the dietary polysaccharides' intervention against CNS disease, which involves oxidative stress reduction, neuronal production, metabolic regulation, and gut barrier integrity. Notably, the ability of dietary polysaccharides to resist oxidation and modulate gut microbiota not only helps to curb the development of these diseases at an early stage, but also holds promise for the development of novel therapeutic agents for CNS diseases. In conclusion, this comprehensive review strives to advance therapeutic strategies for CNS disorders by elucidating the potential of dietary polysaccharides and advocating interdisciplinary collaboration to propel further research in this realm.
Immune-enhancing Activity of Extracellular Polysaccharides Isolated from Rhizopus Nigricans

Abstract:Extracellular polysaccharides (EPS1-1) was extracted from fermentation liquor of Rhizopus nigricans and evaluated its immune-enhancing activities in vitro and in vivo. Results suggested that the proliferation of lymphocyte was stimulated after treated with EPS1-1. Moreover, the activities of macrophages were enhanced by increasing the activities of phagocytosis and acid phosphatase, the production of NO and the mRNA levels of IL-2, TNF-α and iNOS. Furthermore, EPS1-1 could significantly boost the immunity of normal and immunosuppressed mice, which included the increase of loaded swimming time, footpad swelling, organ index and the secretion of IL-2 and TNF-α in serum, thus suggesting that EPS1-1 could improve the body immunity through cellular immunity and humoral immunity. These findings provided further insights into the potential use of EPS1-1 as immunopotentiator or new function food.
Bioactive Polysaccharides Promote Gut Immunity Via Different Ways.

Abstract:Numerous kinds of bioactive polysaccharides are identified as having intestinal immunomodulatory activity; however, the ways in which the different polysaccharides work differ. Therefore, we selected nine representative bioactive polysaccharides, including xanthan gum, inulin, guar gum, arabinogalactan, carrageenan, glucomannan, araboxylan, xylan, and fucoidan, and compared their intestinal immunomodulatory mechanisms. A cyclophosphamide (CTX)-induced immunosuppressed model was used in this experiment, and the effects of these polysaccharides on the number of T cells in the intestinal mucosa, expression of transcription factors and inflammatory factors, intestinal metabolome and gut microbiota were compared and discussed. The results revealed that the nine polysaccharides promote intestinal immunity in different ways. In detail, guar gum, inulin and glucomannan better alleviated immune suppression in intestinal mucosal T cells. Inulin improved the intestinal microenvironment by significantly upregulating the abundance of Lactobacillus and Monoglobus and promoted short chain fatty acid (SCFA) production. Fucoidan and carrageenan promoted the colonization of the beneficial bacteria Rikenella and Roseburia. In addition, fucoidan, inulin and carrageenan inhibited the colonization of harmful bacteria Helicobacter, upregulated the abundance of Clostridia_UCG-014 and alleviated the accumulation of amino acids, bile acids and indoles in the large intestine. In conclusion, our study uncovered the different intestinal immunomodulatory mechanisms of the different polysaccharides and provided a guideline for the development of superior intestinal immunomodulatory polysaccharides.

Abstract:The COVID-19 pandemic has caused significant global impact, resulting in long-term health effects for many individuals. As more patients recover, there is a growing need to identify effective management strategies for ongoing health concerns, such as post-COVID-19 syndrome, characterized by persistent symptoms or complications beyond several weeks or months from the onset of symptoms. In this review, we explore the potential of dietary polysaccharides as a promising approach to managing post-COVID-19 syndrome. We summarize the immunomodulatory, antioxidant, antiviral, and prebiotic activities of dietary polysaccharides for the management of post-COVID-19 syndrome. Furthermore, the review investigates the role of polysaccharides in enhancing immune response, regulating immune function, improving oxidative stress, inhibiting virus binding to ACE2, balancing gut microbiota, and increasing functional metabolites. These properties of dietary polysaccharides may help alleviate COVID-19 symptoms, providing a promising avenue for effective treatment strategies.

Abstract:Immunoregulatory activity is one of the most important biological activities of polysaccharides.Polysaccharides can enhance immune activity while offer no side-effects on cells. Appropriate modification on polysaccharide could be good to exert immune function. The common methods for structure modification to improve its immune activity are the chemical method,biological method and physical method. This paper had expounded different methods of polysaccharide modification on the immune activity in recent years and discussed the mechanism of action. It provided reference and basis for further study on the immunologic mechanism and structure-function relationship of polysaccharides.

Abstract:Natural polysaccharide is a kind of natural macromolecular which can be extracted from plants, fungi, algae, animals, and bacteria. The monosaccharide compositions and glucosidic bonds of polysaccharides from different origins vary substantially. Natural polysaccharides have been shown to possess complex, important and multifaceted biological activities including antitumor, anticoagulant, antioxidative, antiviral, immunomodulatory, antihyperlipidemic and antihepatotoxic activities. Their properties are mainly due to their structural characteristics. It is necessary to develop polysaccharide immunomodulators with potential for preventive or therapeutic action. The present paper summarizes the structural features, immunostimulatory activity and the immunomodulatory mechanisms of natural polysaccharides. In particular, it also provides an overview of representative natural polysaccharide immunomodulators.
