Abstract:Elemental tellurium (Te) nanoparticles are increasingly important in a variety of applications such as thermoelectricity, photoconductivity, and piezoelectricity. However, they have been explored with limited success in their biomedical use, and thus a tremendous challenge still exists in the exploration of Te nanoparticles that can treat tumors as an effective anticancer agent. Here, we introduce bifunctional Te nanodots with well-defined nanostructure as an effective anticancer agent for photo-induced synergistic cancer therapy with tumor ablation, which is accomplished using hollow albumin nanocages as a nanoreactor. Under near-infrared light irradiation, Te nanodots can produce effective photothermal conversion, as well as highly reactive oxygen species such as center dot O-2(-) and dismutated center dot OH via a type-I mechanism through direct electron transfer, thereby triggering the potent in vivo hyperthermia and simultaneous intracellular reactive oxygen species at tumors. Moreover, Te nanodots possess perfect resistance to photobleaching, effective cytoplasmic translocation, preferable tumor accumulation, as well as in vivo renal elimination, promoting severe photo-induced cell damage and subsequent synergy between photothermal and photodynamic treatments for tumor ablation. These findings provide the insight of elemental Te nanodots for biomedical research.

Abstract:Nanoparticles containing elemental tellurium (Te) have been aroused extensive attention in biomedical applications. However, the ideal synthesis methods and effective anti-cancer strategies of Te nanoparticles remain great challenges. In this study, genetically engineered polypeptide (PC10ARGD)-capped Te nanorods (P-TeNRs) was in situ synthesized. The excellent dispersion of P-TeNRs with average lengths of 25.2 ± 4.65 nm and diameters of 6.5 ± 0.83 nm were confirmed by TEM. PC10ARGD was integrated onto the surface of Te nanorods to enhance the stability and biocompatibility of P-TeNRs. P-TeNRs exhibited the excellent photothermal conversion performance under irradiation of NIR laser and the reactive oxygen species (ROS) generating capacity under ultrasound (US) irradiation, which can achieve photothermal therapy and sonodynamic therapy of tumors. Compared with single photothermal therapy and sonodynamic therapy, the results of in vitro and in vivo experiments showed that the combination therapy has a synergistic effect on breast 4T1 tumors. In addition, the results of in vivo toxicity assay suggested that synthesized P-TeNRs presented negligible toxicity. These results demonstrate that this P-TeNRs can be engineered as a novel theranostic nanoplatform for synergistic photothermal-sonodynamic therapy of tumors.

Abstract:As an important inorganic material,titanium dioxide gets more and more attention and is used in various fields,especially in the biomedical field.This paper summarizes the main synthesis methods of titanium dioxide nanomaterial and its applications in cancer therapy area.The titanium dioxide nanomaterial can be prepared by gas,liquid and solid phase methods.Among them,we mainly introduce liquid phase methods including sol-gel method,hydrothermal method,solvothermal,and microemulsion method.We also further describe the principles of photodynamic therapy and photoacoustic treatment in cancer treatments and applications.Finally,we get the conclusions and have an outlook at the application of titanium dioxide nanomaterials in biomedicine.

Abstract:Selenium/tellurium containing polymers have been attracting growing interest due to their unique properties and potential applications as bio-nanomaterials. The introduction of selenium or tellurium chemistry into block copolymers endows self-assembled nanomaterials with novel stimuli responsiveness and enhanced sensitivity. This review article provides an overview on recent advances of selenium/tellurium containing polymers. We summarize the redox response and reversible self-assembly behaviors, gamma-ray responsive systems for combined chemo- and radio-therapies, and coordination-responsive systems for controlled delivery are highlighted. The review then reveals how selenium-containing polymers can modulate reactive oxygen species and work as gene delivery vehicles or self-delivery therapeutics. (C) 2015 Elsevier Ltd. Ultrathin tellurium nanosheets for simultaneous cancer thermo-chemotherapy

Abstract:The emerging two-dimensional monoelemental materials (2D Xenes) have been commonly supposed as promising drug delivery carriers, photothermal and photodynamic therapeutic agents, biosensors, theranostics, and some other candidates for biomedical applications. Here, high-performance and bioactive ultrathin 2D Tellurium nanosheets (Te NSs) are prepared by a simple but efficient liquid-phase exfoliation approach. The as-obtained Te NSs possess a mean size of ∼90 nm and a mean thickness of ∼5.43 nm. The pegylation Te NSs (Te-PEG NSs) possess excellent biocompatibility and stability. The Te-PEG NSs could generate local hyperthermia with a remarkable photothermal conversion efficiency of about 55% under 808 nm laser irradiation. Additionally, Te-PEG NSs exhibit an extremely high loading capacity of chemo drug (∼162%) owing to their ultra-high surface area and tumor microenvironment-triggered drug release superiority. The results of <i>in vivo</i> experiments show that the Te-PEG NSs have higher tumor elimination efficiency via the combination of photothermal and chemotherapy, comparing to any other single therapeutic modalities. Therefore, our work not only highlights the promising potentials of tellurene as an ideal anti-cancer platform but also expands the application of 2D Te for cancer nanomedicine.

Double-camouflaged tellurium nanoparticles for enhanced photothermal immunotherapy of tumor

Abstract:The photothermal conversion properties of tellurium (Te) nanoparticles have been extensively investigated, rendering them a promising candidate for tumor photothermal therapy. However, there is still room for improvement in the development of efficient Te-based drug delivery systems. Here, Te nanoparticles are mineralized with bioactive molecules within attenuated Salmonella (S-Te), which are subsequently taken up by macrophages (RAW264.7) to construct a double-camouflaged delivery platform (RS-Te). Remarkably, RS-Te retains superior photothermal properties under near-infrared irradiation. The mineralization process eliminates bacterial proliferation potential, thereby mitigating the risk of excessive bacterial growth in vivo. Furthermore, the uptake of bacteria by macrophages not only polarizes them into M1 macrophages to induce an anti-tumor immune response but also circumvents any adverse effects caused by complex antigens on the bacterial surface. The results show that RS-Te can effectively accumulate and retain in tumors. RS-Te-mediated photothermal immunotherapy largely promotes the maturation of dendritic cells and priming of cytotoxic T cells induced by near-infrared laser irradiation. Moreover, RS-Te can switch the activation of macrophages from an immunosuppressive M2 phenotype to a more inflammatory M1 state. The double-camouflaged delivery system may offer highly efficient and safe cancer treatment.

Abstract:Nanomaterials play crucial roles in the diagnosis and treatment of diseases. Photothermal and photodynamic therapy, as two minimally invasive therapeutic methods, have promising potential in the diagnosis and prevention of cancer. Recently, many photothermal materials (such as noble metal material, transition metal sulfur oxides, carbon material and upconversion nanomaterial) and photodynamic materials (such as phthalein cyanogen, porphyrins and other dye molecules) have been applied in photothermal therapy (PTT) and photodynamic therapy (PDT). Moreover, as nanomaterials have suitable biocompatibility, these materials have been applied in cancer therapy. In the present review, we summarized the effects of different material types, synthesis methods, material morphologies and surface modifications on the outcomes of cancer therapy. The application of nanomaterials in PTT and PDT was introduced and the advantages and disadvantages of PTT and PDT in the prevention of cancer were discussed. Finally, we discussed the application of nanomaterials in the combination of PTT and PDT in cancer treatment.

Abstract:<p>The research of titanium-based nanomaterials (Ti-based NMs) has gradually expanded from the fields of photocatalysis, electronics, energy, and engineering to biomedicine in recent years. As a non-toxic transitional metal, Ti has been used to construct various nanomaterials with unparalleled structural, compositional, and physicochemical features combined with multitudinous alluring properties. These excellent and unique properties endow Ti-based NMs that have broad biomedical application potential. Ti-based NMs mainly include titanium oxides, titanium carbides, titanium nitrides, titanium sulfides, titanium hydrides, titanium-organic coordination compounds, and their nanocomposites. In this review, we mainly summarize the preparation, surface modification, and their recent progress of Ti-based NMs in cancer theranostics. The application of Ti-based NMs has gradually expanded from the simple drug delivery systems to versatile platforms for multiple therapies, including photothermal therapy (PTT), photodynamic therapy (PDT), and sonodynamic therapy (SDT), etc. To significantly improve the therapeutic efficiency of tumors and reduce the side effects compared with the single therapy, Ti-based NMs have been widely used in combination therapy. Moreover, the single/multi-modal imaging based on Ti-based NMs are also briefly introduced, as well as the strategies to enhance the diagnostic effectiveness. This review highlights the theranostic applications of Ti-based NMs with various composition, structure, and size. To promote the pre-clinical transformation of Ti-based NMs, the toxicity evaluation is also systematically discussed. In addition, the future challenge and perspective are briefly summarized.</p>

Abstract:Cancer remains a major global health problem, causing significant economic burden worldwide. Current clinical cancer treatments are unsatisfactory in terms of both antitumor efficacy and biocompatibility. Meanwhile, nanomaterials attract much attention as potential treatment strategies with improved antitumor efficacy and reduced side effects. Selenium-containing nanomaterials are considered to be a promising biomaterial candidate among many different approaches during the last decade. Selenium-containing nanomaterials exhibit good biocompatibility since selenium is an essential trace element in human body. With rational design, selenium-containing nanomaterials show high sensitivity to redox stimuli and anticancer activity. Here, we discuss the development of selenium-containing nanomaterials as drug delivery carriers and anticancer drugs for cancer treatment. The nanomaterials with promising performance both in vitro and in vivo could advance to clinical therapies.

Abstract:Recent advances in tumor immunotherapy have highlighted the pivotal role of carbon nanomaterials, such as carbon dots, graphene quantum dots, and carbon nanotubes. This review examines the unique benefits of these materials in cancer treatment, focusing on their mechanisms of action within immunotherapy. These include applications in immunoregulation, recognition, and enhancement. We explore how these nanomaterials when combined with specific biomolecules, can form immunosensors. These sensors are engineered for highly sensitive and specific detection of tumor markers, offering crucial support for early diagnosis and timely therapeutic interventions. This review also addresses significant challenges facing carbon nanomaterials in clinical settings, such as issues related to long-term biocompatibility and the hurdles of clinical translation. These challenges require extensive ongoing research and discussion. This review is of both theoretical and practical importance, aiming to promote using carbon nanomaterials in tumor immunotherapy, potentially transforming clinical outcomes and enhancing patient care.

Abstract:Photo-therapeutics have attracted increasing attention for the treatment of tumors because of their controllable and tunable nature and low potential for side effects. Photocatalytic materials triggered by targeted wavelengths of light facilitate the generation of reactive oxygen species for tumor DNA damage, allowing for high efficacy at low radiation doses with limited damage of normal tissues. This short review emphasizes the mechanisms and recent breakthroughs achieved in photocatalytic materials for tumor treatment, mainly focusing on light wavelengths in the respective near-infrared (NIR), UV-vis, and X-ray regions. Lastly, current limitations and future perspectives are also highlighted.

Abstract:Probiotic Escherichia coli Nissle 1917 (EcN) are employed as a bioreactor for intracellularly synthesizing tellurium nanorods (TeNRs) providing a biohybrid therapeutic platform (Te@EcN) for the elimination of advanced malignant tumor by photothermal immunotherapy. Te@EcN is found to possess superior photothermal property upon near-infrared irradiation, and can efficiently accumulate and retain in tumors, although EcN loses proliferation ability after the synthesis of TeNRs, thus inducing considerable immunogenic tumor cell death. Under co-stimulation by EcN acting as immunoadjuvants, maturation of dendritic cells and priming of cytotoxic T cells are largely promoted. In addition, Te@EcN can reprogram tumor-associated macrophages to ameliorate the immunosuppressive tumor microenvironment. Thus, tumor metastasis and recurrence can be efficiently suppressed. Most importantly, owing to the non-pathogenicity of probiotic EcN and their non-proliferative characteristics after TeNRs synthesis, Te@EcN is found to be rapidly metabolized and cleared from the normal tissues, showing very slight acute side effects in healthy mice even at a relatively high administration dose. Therefore, the proposed combined therapeutic strategy based on bacteria-synthesized TeNRs may find great potential in improving bacteria-mediated tumor therapy with increased antitumor efficacy and reduced toxicity.

Abstract:Conventional chemotherapy for cancer treatment is usually compromised by shortcomings such as insufficient therapeutic outcome and undesired side effects. The past decade has witnessed the rapid development of combination therapy by integrating chemotherapy with hyperthermia for enhanced therapeutic efficacy. Near-infrared (NIR) light-mediated photothermal therapy, which has advantages such as great capacity of heat ablation and minimally invasive manner, has emerged as a powerful approach for cancer treatment. A variety of nanomaterials absorbing NIR light to generate heat have been developed to simultaneously act as carriers for chemotherapeutic drugs, contributing as heat trigger for drug release and/or inducing hyperthermia for synergistic effects. This review aims to summarize the recent development of advanced nanomaterials in chemo-photothermal combination therapy, including metal-, carbon-based nanomaterials and particularly organic nanomaterials. The potential challenges and perspectives for the future development of nanomaterials-based chemo-photothermal therapy were also discussed.

Abstract:The utilization of light for therapeutic interventions, also known as phototherapy, has been extensively employed in the treatment of a wide range of illnesses, including cancer. Despite the benefits of its non-invasive nature, phototherapy still faces challenges pertaining to the delivery of phototherapeutic agents, phototoxicity , and light delivery. The incorporation of nanomaterials and bacteria in phototherapy has emerged as a promising approach that leverages the unique properties of each component. The resulting nano-bacteria biohybrids exhibit enhanced therapeutic efficacy when compared to either component individually. In this review, we summarize and discuss the various strategies for assembling nano-bacteria biohybrids and their applications in phototherapy. We provide a comprehensive overview of the properties and functionalities of nanomaterials and cells in the biohybrids. Notably, we highlight the roles of bacteria beyond their function as drug vehicles, particularly their capacity to produce bioactive molecules. Despite being in its early stage, the integration of photoelectric nanomaterials and genetically engineered bacteria holds promise as an effective biosystem for antitumor phototherapy. The utilization of nano-bacteria biohybrids in phototherapy is a promising avenue for future investigation, with the potential to enhance treatment outcomes for cancer patients.

Abstract:Abstract Cancer is a disease with complex pathological process. Current chemotherapy faces problems such as lack of specificity, cytotoxicity, induction of multi-drug resistance and stem-like cells growth. Nanomaterials are materials in the nanorange 1–100 nm which possess unique optical, magnetic, and electrical properties. Nanomaterials used in cancer therapy can be classified into several main categories. Targeting cancer cells, tumor microenvironment, and immune system, these nanomaterials have been modified for a wide range of cancer therapies to overcome toxicity and lack of specificity, enhance drug capacity as well as bioavailability. Although the number of studies has been increasing, the number of approved nano-drugs has not increased much over the years. To better improve clinical translation, further research is needed for targeted drug delivery by nano-carriers to reduce toxicity, enhance permeability and retention effects, and minimize the shielding effect of protein corona. This review summarizes novel nanomaterials fabricated in research and clinical use, discusses current limitations and obstacles that hinder the translation from research to clinical use, and provides suggestions for more efficient adoption of nanomaterials in cancer therapy.

Abstract:In recent years, titanium dioxide (TiO2), one of the most vital and classical metal oxides, has been thoroughly investigated and extensively used in many fields of science and technology owing to its outstanding photo-activity. TiO2-based nanosystems become the potential candidate which has been widely utilized in cancer therapy and antimicrobial field because of the high photo-therapeutic activity. In this review, the recent advances in TiO2-based nanosystems for cancer therapy, including photodynamic therapy (PDT), photothermal therapy (PTT) and combination therapy with other strategies, are summarized. The photo-therapies of TiO2-based nanosystems could be promising alternative strategy for traditional cancer therapies due to the deeper penetration depth and the absence of phototoxicity in clinic. Meanwhile, the frontiers in TiO2-based nanosystems for bacteria-induced infectious diseases management are introduced and summarized here. This potential therapy could efficiently and effectively eliminate the bacteria, manage the infection, and avoid the microbial drug-resistance. Finally, the challenges for the future development of TiO2-based nanosystems in biomedical applications are concluded and discussed. Collectively, we think that TiO2-based nanosystems could be potentially applied in clinical fields to promote the improvement of cancer and infections treatments.

Abstract:Anticancer treatment has become a research highlight in recent years. Despite several techniques have been developed and applied in the clinic, this area still meets great challenges in the construction of smart anticancer devices with accurate targeting, controlled release and microenvironment response properties. Most of the carbon-based materials are biocompatible, possessing abundant and tunable pore structures and particularly large surface areas. These properties make them suitable materials as drug carriers. In addition, some carbon-based materials are capable of absorbing near-infrared radiation (NIR) and have highly efficient photothermal effects. The generated heat in situ can be used to kill cancer cells in short time on the position. This review describes the recent and significant application of four kinds of carbon materials including carbon nanotubes, graphene, carbon dots and mesoporous carbon for drug delivery and photothermal therapy. After a short introduction of the structures and properties of these materials, the construction and application of these nanoplatforms in drug delivery, photothermal therapy or their combination will be summarized and discussed in depth. In addition, other carbon allotropes as drug carriers will be introduced briefly. Finally, the risk assessments and the perspectives and challenges of these materials used in cancer therapies are enclosed.
