Morin Protects Chicks with T-2 Toxin Posioning by Decreasing Heterophile Extracellular Traps, Oxidative Stress and Inflammation

Abstract:T-2 toxin is the most toxic type A trichothecenes which cause serious damage to poultry industry. Morin belongs to the class of flavone compounds, and possess anticancer, antioxidant and anti-inflammatory effects. Our previous research confirm that morin alleviates Aflatoxin B1-induced liver and kidney injury in chicks, but whether morin also protect chicks from T-2 toxin poisoning remains unknown. In this study, the effects of morin on chicks with T-2 toxin posioning were observed, and the protective mechanism were further investigated. Biochemical dectection found that T-2 toxin elevated AST, ALT, BUN and Cre in serum; Pathological observations revealed haemorrhagic spots on the surface of the liver, disturbed arrangement of the hepatic cords, enlarged kidneys and infiltration of inflammatory cells; Oxidative stress assays revealed that T-2 toxin activated the oxidase MDA and inhibited the antioxidant enzymes SOD and CAT; Quantitative real-time PCR revealed that T-2 toxin increased the expression of TNF-α, COX-2, IL-1β, IL-6, caspase 1, caspase 3, and caspase 11, and all these were changed by morin treatment. Moreover, morin also significantly decreased T-2 toxin-induced heterophile extracellular traps (HETs) release in vivo and in vitro. In summary, the chick model of T-2 toxin poisoning was successfully established, and morin protected chicks with T-2 toxin poisoning by decreasing HETs, oxidative stress and inflammation, indicating that morin might used as potential feed additives in poultry industry.
