Abstract:The enthusiasm for research on lanthanide-doped upconversion nanoparticles is driven by both a fundamental interest in the optical properties of lanthanides embedded in different host lattices and their promise for broad applications ranging from biological imaging to photodynamic therapy. Despite the considerable progress made in the past decade, the field of upconversion nanoparticles has been hindered by significant experimental challenges associated with low upconversion conversion efficiencies. Recent experimental and theoretical studies on upconversion nanoparticles have, however, led to the development of several effective approaches to enhancing upconversion luminescence, which could have profound implications for a range of applications. Herein we present the underlying principles of controlling energy transfer through lanthanide doping, overview the major advances and key challenging issues in improving upconversion luminescence, and consider the likely directions of future research in the field.

Abstract:Lanthanide-doped upconversion nanoparticles (UCNPs) have attracted considerable interest due to their superior physicochemical features, such as large anti-Stokes shifts, low autofluorescence background, low toxicity and high penetration depth, which make them extremely suitable for use as alternatives to conventional downshifting luminescence bioprobes like organic dyes and quantum dots for various biological applications. A fundamental understanding of the photophysics of lanthanide-doped UCNPs is of vital importance for discovering novel optical properties and exploring their new applications. In this review, we focus on the most recent advances in the development of lanthanide-doped UCNPs as potential luminescent nano-bioprobes by means of our customized lanthanide photophysics measurement platforms specially designed for upconversion luminescence, which covers from their fundamental photophysics to bioapplications, including electronic structures (energy levels and local site symmetry of emitters), excited-state dynamics, optical property designing, and their promising applications for in vitro biodetection of tumor markers. Some future prospects and efforts towards this rapidly growing field are also envisioned.

Abstract:Along with the development of science and technology, lanthanide‐doped upconversion nanostructures as a new type of materials have taken their place in the field of nanomaterials. Upconversion luminescence is a nonlinear optical phenomenon, which absorbs two or more photons and emits one photon. Compared with traditional luminescence materials, upconversion nanostructures have many advantages, such as weak background interference, long lifetime, low excitation energy, and strong tissue penetration. These interesting nanostructures can be applied in anticounterfeit, solar cell, detection, bioimaging, therapy, and so on. This review is focused on the current advances in lanthanide‐doped upconversion nanostructures, covering not only basic luminescence mechanism, synthesis, and modification methods but also the design and fabrication of upconversion nanostructures, like core–shell nanoparticles or nanocomposites. At last, this review emphasizes the application of upconversion nanostructure in detection and bioimaging and therapy. Learning more about the advances of upconversion nanostructures can help us better exploit their excellent performance and use them in practice.

Abstract:Through the process of photon upconversion, trivalent lanthanide doped nanocrystals convert long-wavelength excitation radiation in the infrared or near infrared region to higher energy emission radiation from ultraviolet to infrared. Such materials offer potential for numerous advantages in analytical applications in comparison to molecular fluorophores and quantum dots. The use of IR radiation as an excitation source reduces autofluorescence and scattering of excitation radiation, which leads to a reduction of background in optical experiments. The upconverting nanocrystals offer excellent photostability and are composed of materials that are not particularly toxic to biological organisms. Excitation at long wavelengths also minimizes damage to biological materials. In this review, the different mechanisms responsible for the upconversion process, and methods that are used to synthesize and decorate upconverting nanoparticles are presented to indicate how absorption and emission can be tuned. Examples of recent applications of upconverting nanoparticles in bioassays for the detection of proteins, nucleic acids, metabolites and metal ions offer indications of analytical advantages in the development of methods of analysis. Examples include multi-color and multi-modal imaging, and the use of upconverting nanoparticles in theranostics.

Abstract:Near infrared (NIR) excited lanthanide-doped upconversion nanoparticles (UCNPs) are emerging as a new type of fluorescent tag for biological applications, which can emit multi-photon ultraviolet, visible or NIR luminescence for imaging or activation of photosensitive molecules. Here, we present a comprehensive review on recent advances of UCNPs for a manifold of biological applications, including upconversion mechanisms, building bright multicolor upconversion nanocrystals, single nanoparticle and super resolution imaging, in vivo optical and multimodal imaging, photodynamic therapy, light-controlled drug release, biosensing, and toxicities. Our perspectives on the future development of UCNPs are also described.

Abstract:Lanthanide-doped upconversion nanomaterials that convert low energy excitation wavelength to high energy emission have attracted considerable attention due to the advantages of high penetration depth and high resistance to autofluorescence interference capability. This review highlights the literature of the past several years covering recent advances of upconversion nanoprobes for biodetection. We begin by discussions of the key points in fabrication of lanthanide-activated nanoprobes. We further conduct a careful evaluation of various detection applications of upconversion nanoprobes, including temperature, ions, small molecules, DNA, enzyme, virus, and etc. We finish the discussion by assessing current challenges and future prospects of upconversion nanoprobes for fundamental studies and clinical applications.

Abstract:The lanthanide-doped up-conversion (UC) nanocrystals exhibit unique luminescence properties, such as the conversion of near-infrared long wavelength excitation into short visible wavelength emission. These UC nanoparticles symbolize a new class of phosphor materials. Recently, lanthanide-doped UC nanocrystals have been developed as a biological class for various applications including bio-imaging, drug delivery, and sensing which require low auto-fluorescence background, large anti-Stokes shifts, sharp emission bandwidths, high resistance to photo-bleaching, high penetration depth tissue, and high temporal resolution. Furthermore, these UC nanocrystals show promising poten-tial for improving the selectivity and sensitivity compared with those in common traditional methods. This review focuses on the fundamental mechanism of UC optical processes, synthetic methods with focus on the different techniques used, energy transfer mecha-nisms of lanthanide-doped UC nanocrystals, and in particular recent developments in biological applications, such as bio-imaging, drug delivery, fluorescence resonance energy transfer sensing, and heavy metal ion detections.

Abstract:Owing to their unique photo-physical properties, rare-earth ions-doped upconversion nanoparticles (UCNPs) have attracted extensive attention in recent years. UCNPs have many special merits, such as a long luminescence lifetime, narrow emission band widths, high quantum yields and low toxicity, which allows their potential applications in bio-medical field, biological luminescent labels and drug delivery carriers. Compared with traditional fluorescence labels exited by UV (ultraviolet), such as organic dyes and quantum dots, UCNPs can transfer near-infrared (NIR) light into visible light, which is commonly called upconversion luminescence (UCL). This paper reviews the recent advances of several typical synthesis methods of UCNPs in detail as well as the fabrication and optimization of the particle morphology, and the latest advances of UCNPs for multimode imaging, surface passivation and functionalization are also described.

Abstract:Lanthanide (Ln)-doped upconversion nanoparticles (UCNPs) with appropriate surface modification can be used for a wide range of biomedical applications such as bio-detection, cancer therapy, bio-labeling, fluorescence imaging, magnetic resonance imaging and drug delivery The upconversion phenomenon exhibited by Ln-doped UCNPs renders them tremendous advantages in biological applications over other types of fluorescent materials (e.g.. organic dyes, fluorescent proteins, gold nanoparticles, quantum dots, and luminescent transition metal complexes) for: (i) enhanced tissue penetration depths achieved by near-infrared (NIR) excitation; (ii) improved stability against photobleaching, photoblinking and photochemical degradation; (iii) non-photodamaging to DNA/RNA due to lower excitation light energy; (iv) lower cytotoxicity; and (v) higher detection sensitivity. Ln-doped UCNPs are therefore attracting increasing attentions in recent years. In this review, we present recent advances in the synthesis of Ln-doped UCNPs and their surface modification, as well as their emerging applications in biomedicine. Abstract:Upconversion nanoparticles (UCNPs) are a class of nanomaterials composed of lanthanide ions with great potential for paraclinical applications, especially in laboratory and imaging sciences. UCNPs have tunable optical properties and the ability to convert long-wavelength (low energy) excitation light into short-wavelength (high energy) emission in the ultraviolet (UV)-visible and near-infrared (NIR) spectral regions. The core-shell structure of UCNPs can be customized through chemical synthesis to meet the needs of different applications. The surface of UCNPs can also be tailored by conjugating small molecules and/or targeting ligands to achieve high specificity and selectivity, which are indispensable elements in biomedical applications. Specifically, coatings can enhance the water dispersion, biocompatibility, and efficiency of UCNPs, thereby optimizing their functionality and boosting their performance. In this context, multimodal imaging can provide more accurate in vivo information when combined with nuclear imaging. This article intends to provide a comprehensive review of the core structure, structure optimization, surface modification, and various recent applications of UCNPs in biomolecular detection, cell imaging, tumor diagnosis, and deep tissue imaging. We also present and discuss some of their critical challenges, limitations, and potential future directions.

Abstract:This review focuses on the enhancement of upconversion luminescence from lanthanide-doped nanoparticles. The unique luminescence capability of long-wavelength absorption and then efficient short-wavelength radiation makes these nanoparticles have attractive potential in numerous photonic and biophotonic fields. However, the dark luminescence limited by the low upconversion quantum yield hinders their applications in in vivo and in photovoltaics. In this work, we first present the fundamental concepts of upconversion luminescence based on energy transfer upconversion, which is the most widespread and the efficient upconversion process. Subsequently, we present the varying strategies to enhance upconversion luminescence intensity from the excitation, energy transfer, and radiation perspectives.

Abstract:Various fluctuations of intracellular ions, biomolecules, and other conditions in the physiological environment play crucial roles in fundamental biological processes. These factors are of great importance for analysis in biomedical detection. Nevertheless, developments of the simple, rapid, and accurate proof for specific detection still encounter major challenges. Upconversion nanoparticles (UCNPs), which could absorb multiple low-energy near-infrared light (NIR) photon excitation and emits high-energy photons caused by anti-Stokes shift, show unique upconversion luminescence (UCL) properties, for example, sharp emission band, high physicochemical stability like near-zero photobleaching, photo blinking in biological tissues, and long luminescence lifetime. Furthermore, the NIR used for the light source to excite UCNPs enable lower photo-damage effect and deeper penetration of tissue, and in the meantime, it can avoid the auto-fluorescence and light scattering from biological tissue interference. Thus, the lanthanide-doped UCNP-based functional platform with controlled structure, crystalline phase, size, and multicolor emission has become an appropriate nanomaterial for bioapplications such as biosensing, bioimaging, drug release, and therapies. In this review, the recent progress about synthesis and biomedical applications of UCNPs related to sensing and bioimaging is summarized. Firstly, the different luminescence mechanisms of the upconversion process are presented. Secondly, four of the most common methods for synthesizing UCNPs are compared as well as the advantages and disadvantages of these synthetic routes. Meanwhile, the surface modification of lanthanide-doped UCNPs was introduced to pave the way for their biochemistry applications. Next, this review detailed the biological applications of lanthanide-doped UCNPs, particularly in bioimaging, including UCL and multi-modal imaging and biosensing (monitoring intracellular ions and biomolecules). Finally, the challenges and future perspectives in materials science and biomedical fields of UCNPs are concluded: the low quantum yield of the upconversion process should be considered when they are executed as imaging contrast agents. And the biosafety of lanthanide-doped UCNPs needs to be evaluated.

Abstract:Trivalent lanthanide (Ln(3+))-doped luminescent inorganic nanoparticles (NPs), characterized by long-lived luminescence, large Stokes and/or anti-Stokes shifts, narrow emission bands and high photochemical stability, are considered to be promising candidates as luminescent bioprobes in biomedicine and biotechnology. In this feature article, we provide a brief overview of the most recent advances in Ln(3+)-doped luminescent inorganic NPs as sensors, which covers from their chemical and physical fundamentals to biodetection, such as controlled synthesis methodology, surface modification chemistry, optical physics, and their promising applications in diverse bioassays, with an emphasis on heterogeneous and homogeneous in vitro biodetection. Finally, some of the most important emerging trends and future efforts toward this active research field are also proposed.

Abstract:Lanthanide-based upconversion nanophosphors are able to convert lower-energy near-infrared photons to higher-energy ones as emission. This anti-Stokes photoluminescence process will result in low background noise, large tissue penetration depth, and low photo-damage in bioimaging applications. Due to their special photoluminescent properties, upconversion nanophosphors have been considered as the next generation of photoluminescent probes for sensing and bioimaging, operating in the near-infrared range. This review summarizes recent advances in the synthesis of water-soluble lanthanide upconversion nanophosphors and their application in functional bioimaging (including lymphatic imaging, multimodality bioimaging, ion monitoring in vivo, targeted tumor imaging) that have been developed by our group in the past five years. In addition, the set-up for laser scanning upconversion luminescence confocal microscope and the in vivo imaging system developed by our group are also included in this review. Finally, we discuss the challenges and opportunities in the development of lanthanide upconversion nanophosphors for bioimaging.

Abstract:Upconversion fluorescence is the process of converting optical energy from low absorption energies to high emission energies. The upconversion (UC) materials exhibit significant advantages such as high penetration depth, low unwanted autofluorescence and light scattering from biological species at near-infrared excitation. Nowadays, UC materials are widely applied in optical applications, including solid-state lasers, temperature sensor and chemical sensors. The upconversion materials consist of low phonon host material and sensitizer-activator pair. The sensitizer and activator are often rare earth elements, where energy transfers from the sensitizer to the activator. This review describes the general picture and recent progress of lanthanide upconversion fluorescent materials along with applications in biochemical sensing and imaging. We also provide novel strategies to enhance upconversion intensity and tunable emissions with doping metal ions and core-shell structures, and to integrate magnetic properties to upconversion materials.

Abstract:Organic lanthanide complexes have garnered significant attention in various fields due to their intriguing energy transfer mechanism, enabling the upconversion (UC) of two or more low-energy photons into high-energy photons. In comparison to lanthanide-doped inorganic nanoparticles, organic UC complexes hold great promise for biological delivery applications due to their advantageous properties of controllable size and composition. This review aims to provide a summary of the fundamental concept and recent developments of organic lanthanide-based UC materials based on different mechanisms. Furthermore, we also detail recent applications in the fields of bioimaging and solar cells. The developments and forthcoming challenges in organic lanthanide-based UC offer readers valuable insights and opportunities to engage in further research endeavors.

Abstract:With abundant energy levels of 4f electron configurations, trivalent lanthanide ions (Ln(3+)) are endowed with unique and fascinating luminescent properties. Inheriting the native transition behaviour of the lanthanide ions, Ln(3+) based nanomaterials have aroused great interest for a wide range of applications, including lighting and displays, optical fibers and amplifiers, responsive luminescent stains for biomedical analysis, in vivo and in vitro imaging, and enhancement for silicon solar cell devices. It should be noted that the application depends completely on the corresponding luminescent behaviour. To deepen the understanding of the luminescent mechanism is important for the developing of the field and the design of new Ln(3+) based luminescent materials toward applications. In this review, we focused mainly on the recent developments on upconversion (UC) emission studies. Firstly, the emphasis was put on the introduction of basic luminescent properties of Ln(3+) with f-f transitions, and then the corresponding mechanisms and properties of UC emission were discussed in detail, the potential researches with respect to UC mechanisms and properties were finally outlined.
