Abstract:Enantioselective trifluoromethylation of aromatic ketones promoted by the cinchona alkaloid-derived ammonium bromide and sodium hydride was described. A series of trifluoromethyl-substituted aryl alcohols could be obtained in up to 82% ee with 98% yield under mild conditions. A possible catalytic cycle was also presented.

Abstract:Cinchona alkaloid is one of the most effective organocatalysts that facilitate a wide range of transformations. To elucidate the mechanistic details and especially the origins of the observed high enantio- and diastereoselectivities, computational chemists have contributed dramatic efforts in this important area. This review covers the recent computational studies on the cinchona alkaloid-catalyzed asymmetric Michael additions, including both CC and CS bond formations, to present a general viewpoint of these fruitful advances. Based on the types of bond formation and the cinchona alkaloid catalysts, key information regarding the activation model and the origins of enantioselectivities is discussed.

Abstract:A novel enantioselective [4 + 2] annulation of the allenoates having a unique positive ortho-effect with in situ generated ortho-quinone methides has been developed under the catalysis of Cinchona alkaloid. This chiral amine-catalyzed reaction provides an alternative route to asymmetric catalytic construction of synthetically interesting, highly functionalized chiral chromans in good to excellent enantioselectivities (up to 97% ee).

Abstract:2-Perfluoroalkylated quinolines were efficiently synthesized via the Michael addition of perfluoroalk-2-ynoates with isatins, followed by the reaction with alcohols through an intramolecular rearrangement cyclization in the presence of Na2CO3. Under the mild and metal-free reaction conditions, a broad scope of quinolines was synthesized and the mechanism was proposed.

Abstract:We developed an amide-based Cinchona alkaloids catalyzed enantioselective fluorination of 4-substituted pyrazolones, the products with a quaternary chiral fluorine-attached carbon centre were afforded in high yields (up to 99%) with excellent enantioselectivities (up to 98% ee). Our method is highlighted by low catalyst loading (0.05-0.5 mol %), short reaction times (30 min) and broad substrate scopes (35 examples). Furthermore, we carried out this asymmetric fluorination under sustainable conditions in 10 cylces, the optically pure product (>99.5% ee) could be easily obtained by simple extraction and recrystallization on a multigram scale. Controlled experiments and DFT studies showed that the multiple hydrogen bonding interactions and the steric hindrance at ortho-position in benzene ring of catalyst were the main reasons for high enantioselectivity and reactivity. With a simple operational protocol and low catalyst loading, this transformation provides a practical method for preparation of chiral fluorine-containing compounds.

Abstract:A natural quinidine-catalyzed asymmetric chlorination of 4-substituted pyrazolones is developed, affording products with a quaternary chiral chlorine-attached carbon centre in high yield with excellent enantio-selectivity. Abstract:Asymmetric conjugate addition of ethyl 4,4,4-trifluoroacetoacetate and other trifluoromethyl substituted nucleophiles to β,γ-unsaturated α-keto esters has been developed. The reaction efficiently provided dihydropyrans via cascade Michael–hemiketalization pathways. Quinine-derived thiourea was identified to be the best catalyst. A number of trifluoromethyl substituted dihydropyrans with three consecutive chiral centers were obtained in excellent yields, diastereoselectivities, and enantioselectivities. The product was readily transformed to the corresponding tetrahydropyridine without the loss of the optical purity.

Abstract:The development of efficient syntheses of complex natural products has long been a major challenge in synthetic chemistry. Designing cascade reactions and employing bioinspired transformations are an important and reliable means of achieving this goal. Presented here is a combination of these two strategies, which allow efficient asymmetric synthesis of the cinchona alkaloid (+)-cinchonidine. The key steps of this synthesis are a controllable, visible-light-induced photoredox radical cascade reaction to efficiently access the tetracyclic monoterpenoid indole alkaloid core, as well as a practical biomimetic cascade rearrangement for the indole to quinoline transformation. The use of stereoselective chemical transformations in this work makes it an efficient synthesis of (+)-cinchonidine.

Abstract:Cinchona alkaloid squaramide can effectively catalyze the asymmetric Michael addition of α-aryl isocyanoacetates to β-trifluoromethylated enones, affording the corresponding adducts with an adjacent chiral tertiary carbon center bearing a CF3 group and a quaternary carbon center in moderate to good yields along with excellent stereoselectivities. The adduct can be easily transformed into biologically attractive chiral β-trifluoromethylated pyrroline carboxylate in high yield via an isocyano group hydrolysis/cyclization/dehydration cascade reaction by treating with acid. The one-pot enantioselective Michael addition/isocyano group hydrolysis/cyclization/dehydration sequential protocol has also been investigated.

Abstract:An efficient bifunctional cinchona alkaloids‐catalyzed asymmetric tandem intramolecular oxa‐Michael addition/electrophilic thiocyanation of alkylidene β‐ketoesters with N‐thiocyanatosuccinimide (NTS) was developed. A series of chiral α‐thiocyanato flavanones containing two vicinal stereocenters including an all‐carbon quaternary center were prepared in good yields with excellent diastereo‐ and enantioselectivities (up to 97% ee) under mild conditions, and a successful scale up preparation of our protocol was also demonstrated.

Abstract:A novel asymmetric halogenation/semipinacol rearrangement reaction catalyzed by cinchona alkaloid derivatives was developed. Two types of beta-haloketones (X = Br, Cl) were obtained with up to 95% yield and 99% enantiomeric excess. The desired (+) and (-) enantiomers of the beta-haloketones were readily obtained.

Abstract:We have demonstrated that bidentate Lewis base catalysts can be constructed based on the Cinchona alkaloid structure that promote highly stereoselective reactions of allyl- and crotyltrichlorosilane with aromatic as well as aliphatic aldehydes (90–99% ee, >98% diastereoselectivity). The catalysts are available in a one-pot procedure in >70% yield from cheap starting materials and promote the allylation reactions at ambient temperature. Gram scale reactions with catalyst recovery and reuse showcased the practicality of the catalytic system.
