Structural Basis of Ion Transport and Inhibition in Ferroportin

Abstract:Ferroportin (SLC40A1, Fpn) is an iron transporter essential for releasing cellular ferrous ions (Fe2+) into circulation. Hepcidin is a peptide hormone that inhibits Fpn and regulates iron homeostasis. in humans, mutations in Fpn are associated with hemochromatosis, which is a hereditary disease characterized by a paradoxical accumulation of iron in macrophages and symptoms of iron-deficient anemia. We expressed and purified Fpn from human (hFpn) and tarsier monkey (TsFpn) and reconstituted the Fpns in liposomes to measure metal ion transport using a fluorescence-based transport assay. We found that Fpn is an electroneutral H+/Fe2+ antiporter and thus transport of each Fe2+ is coupled to the movement of two H+ in the opposite direction. We confirmed that hepcidin, which is known to bind Fpn from the extracellular side, inhibits metal ion transport through the purified Fpns. We used purified TsFpn to produce mouse monoclonal antibodies and found that the antigen-binding fragment (Fab) binds to both TsFpn and hFpn, and that the Fab inhibits metal ion transport from the intracellular side of Fpn. We solved the structures of TsFpn and hFpn in complex with the Fab by cryo-electron microscopy to nominal resolutions of ∼3.1 Å. The structures identify two novel metal ion binding sites that are coordinated differently from previously known transition metal ion binding sites. Mutations to the metal ion binding sites reduce the rate of transport and render the transport process electrogenic. We also solved the structure of Fpn in complex with hepcidin to define the hepcidin binding pocket and found that hepcidin reaches one of the metal ion binding sites. These results established the structural basis of metal ion binding and transport in Fpn and provided a blueprint for targeting Fpn for pharmacological intervention.
