Abstract:Particulate platforms capable of delivering multiple actives as well as providing diagnostic features have gained considerable interest over the last few years. In this study, magnetic polymer yolk-shell particles (YSPs) were engineered using a tri-needle coaxial electrospraying technique enabling dual-mode (ultrasonic and magnetic resonance) imaging capability with specific multidrug compartments via an advanced single-step encapsulation process. YSPs comprised magnetic Fe3O4 nanoparticles (MNPs) embedded in the polymeric shell, an interfacing oil layer, and a polymeric core (i.e., composite shell-oil interface-polymeric core). The frequency of the ultrasound backscatter signal was modulated through YSP loading dosage, and both T-1- and T-2-weighted magnetic resonance imaging signal intensities were shown to decrease with increasing MNP content (YSP outer shell). Three fluorescent dyes (selected as model probes with varying hydrophobicities) were coencapsulated separately to confirm the YSP structure. Probe release profiles were tuned by varying power or frequency of an external auxiliary magnetic field (AMF, 0.7 mT (LAMF) or 1.4 mT (HAMF)). In addition, an inversion phenomenon for the AMF-enhanced drug release process was studied and is reported. A low YSP cytotoxicity (5 mg/mL) and biocompatibility (murine, L929) was confirmed. In summary, magnetic YSPs demonstrate timely potential as multifunctional theranostic agents for dual-imaging modality and magnetically controlled coactive delivery.

Abstract:Composite worm-like micelles (CWMs) that encapsulate a large number of superparamagnetic magnetite nanoparticles (SMNPs) within the core while still having a small width should have a high magnetic resonance sensitivity and simultaneously a desirable circulation behaviour in biological systems. However, the preparation of such CWMs is still a challenge. Herein we report the fabrication of CWMsviatwo-stage self-assembly of SMNPs guided by a block copolymer and DNA, respectively. First, poly(ethylene glycol)-b-poly(4-vinylpyridine) co-assembled with SMNPs to form spherical composite micelles, guiding the clustering of multiple SMNPs within the core of the micelles. Then, at the second stage, DNA co-assembled with and guided the linear aggregation and fusion of the micelles, resulting in the CWM encapsulating a large number of SMNPs in the linear core. As a proof of concept, CWMs with aT(2)relaxivity as high as 704 mM(-1)s(-1)and a low MRI detection limit of 2.2 mu g mL(-1)(similar to 0.02 nM CWMs) were prepared; it is significant that theT(2)relaxivity of the SMNPs assembled in the CWMs is around 20 times that of the same SMNPs in the same amount but individually dispersed in the suspension. Additionally, although heavily loaded with SMNPs, the CWMs have a width as small as 20.5 nm and a core-shell structure with PEG as the shell. These features should make the CWMs very promising in their use as ultra-sensitive magnetic resonance imaging probes.
Fabrication of Uniform, Biocompatible and Multifunctional PCL-b-PAA Copolymer-Based Hybrid Micelles for Magnetic Resonance Imaging

Abstract:In this paper, a simple strategy was developed to fabricate a novel kind of uniform, biocompatible and PEGylated multifunctional hybrid micelles with multiple magnetite nanocrystal loaded cores and dye-doped silica cross-linked shells based on the self-assembly between poly(epsilon-caprolactone)-b-poly(acrylic acid) copolymer and inorganic nanoparticles. The loading of multiple magnetite nanoparticles in the core part of the hybrid micelles endows them with T2-weighted MR imaging functionalities. Small dye molecules (Rhodamine B) were directly incorporated into the silica layer framework during the cross-linking process, imparting the hybrid micelles with fluorescent imaging modality. Poly(ethylene glycol) (PEG) was grafted to reduce the phagocytic capture of nanoparticles by cellular components of the immune system. Importantly, the potential application of magnetite incorporated PEGylated hybrid micelles as T2 contrast agents for MRI was demonstrated both in vitro and in vivo, with the passive targeting behavior via EPR effect due to the leaky vasculature and poor lymphatic drainage in tumors.
Multifold Enhanced T-2 Relaxation Of Znfe2o4 Nanoparticles By Jamming Them Inside Chitosan Nanospheres

Abstract:ZnFe2O4 nanoparticles were tightly packed and encapsulated in chitosan nanospheres by a nonsolvent-aided counterion complexation method. The obtained hybrid nanospheres exhibited not only the superparamagnetic properties provided by pure ZnFe2O4 nanoparticles but also a much higher r(2) relaxivity value than separate ZnFe2O4 nanoparticles, representing an approximately 3 to 16 fold increase. This was attributed to the high ZnFe2O4 payload and ZnFe2O4 nanoparticle clustering effect in the core of the nanospheres. The MR images generated in vivo demonstrated that the hybrid nanospheres showed an excellent contrast in T-2 weighted MRI and a high MRI sensitivity in the tumor sites for both passive and active targeting samples. In addition, the biodistribution analysis revealed that the obtained hybrid nanospheres could accumulate at tumor sites via passive and active targeting strategies.

A paramagnetic polyoxometalate cluster, K13 [Gd(β2 -SiW11 O39 )2 ], is first encapsulated with a cationic 3-generation dendron (D-3) bearing triethylene glycol monomethyl ether terminal groups through ionic substitution. The formed electrostatic complex, (D-3)13 [Gd(β2 -SiW11 O39 )2 ] (D-GdSiW), which displays an ambivalent micellar structure, is then demonstrated to act as a suitable carrier for the uptake of cationic fluorescent dyes, forming a loaded complex. The dye-loaded complex D-GdSiW-D is thus proven to display high relaxivity as a contrast agent, maintaining its structural stability under physiological conditions with low cell cytotoxicity. Confocal laser scanning microscopic observation also demonstrates the fluorescence imaging property of the prepared complex upon consumption by MCF-7 cells. Both in vivo fluorescence imaging and MRI further confirm that the complex micelle with dual micellar structure may be a suitable liver-specific contrast agent for clinical diagnosis.

Abstract:Magnetite (Fe3O4) – loaded polymer micelles (denoted as “magnetomicelles”) are produced by self-assembly of fluorine-containing amphiphilic poly(HFMA-g-PEGMA) copolymers with oleic acid modified Fe3O4 nanoparticles in an aqueous medium. The oleic acid modified Fe3O4 nanoparticles form small clusters in the poly(HFMA-g-PEGMA) micelles with a mean diameter of 100 nm and the magnetomicelles show high stability in an aqueous medium due to the high hydrophobic fluorine segments in graft copolymers enhance the stability of the micelles. The magnetomicelles also show good cytocompatibility based on the MTT cytotoxicity assay and possess paramagnetic properties with saturation magnetization of 17.14 emu/g.Their good stability, cytocompatibility, and paramagnetic properties render the materials attractive in drug delivery and in vivo magnetic resonance imaging (MRI) applications. Controlled release of hydrophobic drug-5-fluorouracil is achieved from the magnetomicelles with a loading efficiency of 20.94 wt%. The magnetomicelles have transverse relaxivity rates (r2) of 134.27 mM−1 s−1 and exhibit high efficacy as a negative MRI agent in T2-weighted imaging. In vivo MRI studies demonstrate that the contrast between liver and spleen is enhanced by the magnetomicelles. These favorable properties suggest clinical use as nanocarriers in drug delivery applications and contrast agents in MRI.
Ultra-high relaxivity iron oxide nanoparticles confined in polymer nanospheres for tumor MR imaging

Abstract:Superparamagnetic iron oxide nanoparticles encapsulated in hydrophilic chitosan nanospheres were prepared by nonsolvent-aided counterion complexation completely in an aqueous solution. The T-2 relaxation of these hybrid nanospheres in vitro and in vivo was investigated. It was found that the molar transverse relaxivity rate r(2) of hybrid nanospheres highly depends upon the payload of iron oxide nanoparticles within hybrid nanospheres. Compared to free iron oxide nanoparticles, the molar transverse relaxivity rate, r(2) of hybrid nanospheres shows an approximately 8-fold increase and reaches the maximum of 533 Fe mM(-1) s(-1). Such a high r(2) value is probably associated with the clustering effect of iron oxide nanoparticles, which are confined in the chitosan nanospheres. The in vivo magnetic resonance imaging (MRI) demonstrates that the hybrid nanospheres shorten transverse relaxation time, T-2 and significantly decrease the signal intensity of the tumor area, giving rise to high contrast tumor MR imaging at a relatively low dose.

Abstract:Stable and cytocompatible hybrid PEGylated micelles with multimodal imaging capabilities are described. The F3O4-encapsulated polymeric micelles composed of cores containing magnetic nanoparticles and polyethylene glycol (PEG) shells are synthesized by self-assembly of amphiphilic poly(HFMA-co-VBK)-g-PEG copolymers and oleic acid stabilized Fe3O4 nanoparticles. The Fe3O4 magnetic nanoparticles in the core produce T2-weighted MR imaging functionalities, whereas the small fluorescent monomer carbazole in the polymer shell introduces good fluorescent properties. The multifunctional micelles exhibit excellent paramagnetic properties with the maximum saturation magnetization of 9.61 emu/g and transverse relaxivity rate of 157.44 mm−1 S−1. In vivo magnetic resonance imaging (MRI) studies reveal enhanced contrast between the liver and spleen. Fluorescence spectra show characteristic emission peaks from carbazole at 350 nm and 365 nm and vivid blue fluorescence can be observed by 2-photon confocal scanning laser microscopy (CLSM). In vivo optical imaging demonstrates the unique fluorescent characteristics of the Fe3O4-encapsulated polymeric micelles in the liver and spleen and the excellent multifunctional properties suggest potential clinical use as nanocarriers in magnetic resonance imaging and optical imaging.

Abstract:Uniform magnetic nanoparticle-loaded polymer nanospheres with different loading contents of manganese ferrite nanoparticles were successfully synthesized using a flexible emulsion process. The MnFe2O4-loaded polymer nanospheres displayed an excellent dispersibility in both water and phosphate buffer saline. The effect of loading ratio and size of MnFe2O4 nanoparticles within the nanospheres on the specific absorption rate (SAR) under an alternating magnetic field was investigated. Our results indicate that a large size (here 18 nm) and a low loading ratio are preferable for a high SAR. For a smaller particle size (6 nm), the low loading ratio did not result in an enhancement of the SAR value, while a very low SAR value is expected for 6 nm. In addition, the SAR of low-content MnFe2O4 (18 nm)-loaded polymer nanospheres in the agarose gel which is simulated for in vivo environment is the highest among the samples and does not change substantially in physiological environments. This differs largely from the behaviour of singly dispersed nanoparticles. Our results have paved the way for the design of MnFe2O4-loaded polymer nanospheres as magnetic hyperthermia agents for in vivo bio-applications.

Abstract:Control of the morphology, mesostructure, and surface chemistry of multifunctional materials is important for their applications. We report here the fabrication of multifunctional spheres which possess a silica-coated magnetite core and a highly aminated mesoporous silica shell with radially aligned ordered pore channels. The well-designed core-shell structured spheres have superparamagnetism, high magnetization (32 emu g-1), large surface area (133 m2 g-1), uniform accessible mesopores (3.4 nm), and abundant amino groups on the mesoporous shell. Owing to the presence of the amino groups, the composite spheres exhibit high capacity for convenient connection of fluorescent dyes, which make them promising candidates for applications in various biomedical fields, such as cell imaging and cell sorting. Furthermore, a fibrin-specific magnetic resonance imaging (MRI) contrast agent was successfully prepared by connecting the composite spheres with pentapeptide Gly-Pro-Arg-Pro-Pro, which can significantly increase the MRI signal of thrombus.
Multi-Functional Thermosensitive Composite Microspheres with High Magnetic Susceptibility Based on Magnetite Colloidal Nanoparticle Clusters

Abstract:Monodisperse organic/inorganic composite microspheres with well-defined structure were prepared through the encapsulation of silica coated superparamagnetic magnetite colloidal nanoparticle clusters (CNCs) with cross-linked poly(N-isopropylacrylamide) (PNIPAM) shell. At first, the sub-micrometer-sized CNCs were fabricated by the solvothermal process, and then a silica layer was coated on the surface of CNCs through a sol-gel process, and finally, a thermoresponsive shell of PNIPAM was deposited onto the surface of the core/shell magnetic microspheres by a precipitation polymerization. The experimental results showed that the size of Fe(3)O(4) core, the thickness of SiO(2) shell, as well as volume phase transition temperature (VPTT) of PNIPAM shell could be well controlled, and this structured modulation could satisfy different requirements. The superparamagnetic behavior, high magnetization (the saturation magnetization of Fe(3)O(4)/SiO(2)/PNIPAM microspheres with a 10% cross-linking density is 41.6 emu/g), and good thermosensitivity make these composite microspheres an ideal candidate for various important applications such as in controlled drug delivery, bioseparation, and catalysis.
Magnetic colloidal superparticles of Co, Mn and Ni ferrite featured with comb-type and/or linear amphiphilic polyelectrolytes; NMR and MRI relaxometry

Abstract:The ability to encapsulate hydrophobic ferrites in colloidal superparticle structures of an a-telechelic hexadecyl-functionalized poly(methacrylic acid) (C16H33-PMAA) polymer with a linear architecture was investigated and compared with that of two amphiphilic comb-type water-soluble copolymers, namely, P(ANa-co-DAAm) and P(MANa-co-DMA), which are comprised of a poly(sodium acrylate) or poly(sodium methacrylate) backbone and pendent dodecyl acrylamide or dodecyl methacrylate chains, respectively. In the case of C16H33-PMAA, the pH-sensitive self-assembly behavior, which was studied through Nile Red probing and TEM, was related to its encapsulation properties. Hydrophobic MFe2O4 nanoparticles coated with oleylamine (MFe2O4@OAm MNPs, where M = Co, Mn, Ni) with a similar shape and size (∼9 nm) and magnetization values of 87.4, 63.1 and 55.0 emu g(-1) for CoFe2O4@OAm, MnFe2O4@OAm and NiFe2O4@OAm, respectively, were successfully encapsulated into the hydrophobic cores of spherical micellar structures formed by the copolymers in an aqueous solution through a solvent mixing procedure. The synthesized magnetic colloidal superparticles fell in the static dephasing regime (SDR). NMR relaxivity measurements of MFe2O4@P(ANa-co-DAAm), MFe2O4@P(MANa-co-DMA) and MFe2O4@C16H33-PMAA at pH = 4.5 and pH = 7 (where M = Co, Mn, Ni) at 11.7 T were recorded and the transverse relaxivity (r2) (mM(-1) s(-1)) was determined. Among all, the CoFe2O4@polymers demonstrated the highest r2 relaxivity values, ranging from 61.6 for CoFe2O4@C16H33-PMAA (pH = 7) to 316.0 mM(-1) s(-1) for CoFe2O4@P(ANa-co-DAAm). The relaxation efficiency (r1 and r2) of CoFe2O4@P(ANa-co-DAAm) was investigated further by magnetic resonance imaging (MRI) at 1.5 T and 3 T and the r2/r1 ratios were found to be 16.5 and 18.2, respectively, indicating its potential use as a T2 contrast agent.

Abstract:Starlike polymers have been widely used in various fields, such as tissue engineering, imaging, gene and drug delivery because of their unique structures and properties. Dextran has long been used as a temporary plasma substitute because of its excellent biocompatibility. In this study, starlike polysaccharide with multiple dextran arms was designed and developed by attaching dextran to a β-cyclodextrin core through click chemistry. Next, starlike dextran was modified with aliphatic chains and these amphiphilic polymers can self-assemble into nanoscale micelles in water, and their critical micelle concentration values (3.7 × 10(-8) M) are much lower comparing to its linear analogs (1.7 × 10(-7) M), resulting in more stable nanostructures in aqueous environment. These micelles can encapsulate multiple superparamagnetic iron oxide nanoparticles and forming clustering particle nanostructures in water, and the resulting nanocomposites have a high T(2) relaxivity of 436.8 Fe mm(-1) s(-1) under a 1.5T clinical magnetic resonance imaging (MRI) scanner. Further, dual functional probes were developed by loading both superparamagnetic iron oxide nanoparticles and small molecule anticancer drug doxorubicin into polymeric micelles. Multidrug-resistant breast cancer cells MCF-7/Adr treated with these probes can be characterized under MRI.

Abstract:Multifunctional magnet–fluorescent nanocomposites are widely applied in biomedical applications. Incorporating biocompatible quantum dots with highly ferrimagnetic magnetic nanoparticles into one nanoplatform for achieving efficient magnetic hyperthermia therapy (MHT) is very important. Herein, we reported an amphiphilic block copolymer with a flowable hydrophobic chain to encapsulate highly ferrimagnetic magnetic nanoparticles and ZnS/InP quantum dots via a facile self-assembly method. The obtained ferrimagnetic fluorescent micelle (FMFM) exhibited a uniform diameter of about 180 nm. In stark contrast, larger aggregation (400 nm in diameter) inevitably occurred using common poly( D , L -lactide) (PLA)-based amphiphilic block copolymer with a rigid hydrophobic chain, which was readily cleared by the reticuloendothelial system (RES). The flowable FMFM exhibited long-term colloidal stability within one month and desired fluorescent stability within 84 h. Benefiting from the high ferrimagnetism, the FMFM revealed excellent magnetic heating effect and magnetic resonance imaging capability. With accurate manipulation under an external magnetic field, FMFM realized in vitro enhanced fluorescence imaging sensitivity and accumulation efficiency at the tumor region, achieving in vitro and vivo improved MHT efficacy.

Abstract:This article describes the fabrication of nanosized magneto-vesides (MVs) comprising tunable layers of densely packed superparamagnetic iron oxide nanopartides (SPIONs) in membranes via cooperative assembly of polymer tethered SPIONs and free poly(styrene)-b-poly(acrylic acid) (PS-b-PAA). The membrane thickness of MVs could be well controlled from 9.8 to 93.2 nm by varying the weight ratio of PS-b-PAA to SPIONs. The increase in membrane thickness was accompanied by the transition from monolayer MVs, to double layered MVs and to multilayered MVs (MuMVs). This can be attributed to the variation in the hydrophobic/hydrophilic balance of polymer-grafted SPIONs upon the insertion and binding of PS-b-PAA onto the surface of nanopartides. Therapeutic agents can be efficiently encapsulated in the hollow cavity of MVs and the release of payload can be tuned by varying the membrane thickness of nanovesicles. Due to the high packing density of SPIONs, the MuMVs showed the highest magnetization and transverse relaxivity rate (r2) in magnetic resonance imaging (MRI) among these MVs and individual SPIONs. Upon intravenous injection, doxorubicin-loaded MuMVs conjugated with RGD peptides could be effectively enriched at tumor sites due to synergetic effect of magnetic and active targeting. As a result, they exhibited drastically enhanced signal in MRI, improved tumor delivery efficiency of drugs as well as enhanced antitumor efficacy, compared with groups with only magnetic or active targeting strategy. The unique nanoplatform may find applications in effective disease control by delivering imaging and therapy to organs/tissues that are not readily accessible by conventional delivery vehicles.
Electrostatic Polyion Micelles with Fluorescence and Mri Dual Functions

Abstract:We report in this work the formation of fluorescence and MRI bimodal imaging nanoparticles achieved by electrostatic self-assembly. The nanoparticles are micelles formed with Gd(3+) ion, a bisligand that contains aggregation induced emission (AIE) group, and a block copolymer. The coordination between the Gd(3+) ion and the bisligand produces a negatively charged coordination complex, which interacted with the positive-neutral block copolymer to form polyion micelles. The micelles exhibit considerable fluorescence owing to the rotation restriction of the AIE group; meanwhile, the longitudinal relaxation of water was significantly slowed down which provide T1 contrast for magnetic resonance imaging. In vitro fluorescence imaging and in vivo MRI measurements verified this micelle indeed exhibit dual imaging ability. We expect that this orthogonal imaging may provide more accurate diagnosis in practical applications and will pave the way for the development of an advanced technique for diagnosis.

Abstract:Abstract Molecular systems known as single-molecule magnets (SMMs) exhibit magnet-like behaviour of slow relaxation of the magnetisation and magnetic hysteresis and have potential application in high-density memory storage or quantum computing. Often, their intrinsic magnetic properties are plagued by low-energy molecular vibrations that lead to phonon-induced relaxation processes, however, there is no straightforward synthetic approach for molecular systems that would lead to a small amount of low-energy vibrations and low phonon density of states at the spin-resonance energies. In this work, we apply knowledge accumulated over the last decade in molecular magnetism to nanoparticles, incorporating Er 3+ ions in an ultrasmall sub-3 nm diamagnetic NaYF 4 nanoparticle (NP) and probing the slow relaxation dynamics intrinsic to the Er 3+ ion. Furthermore, by increasing the doping concentration, we also investigate the role of intraparticle interactions within the NP. The knowledge gained from this study is anticipated to enable better design of magnetically high-performance molecular and bulk magnets for a wide variety of applications, such as molecular electronics.

Abstract:A facile and effective approach to preparation of dual-responsive magnetic core/shell composite microspheres is reported. The magnetite(Fe3O4)/poly(methacrylic acid) (PMAA) composite microspheres were synthesized through encapsulating c-methacryloxypropyltrimethoxysilane (MPS)-modified magnetite colloid nanocrystal clusters (MCNCs) with crosslinked PMAA shell. First, the 200-nm-sized MCNCs were fabricated through solvothermal reaction, and then the MCNCs were modified with MPS to form active vinyl groups on the surface of MCNCs, and finally, a pH-responsive shell of PMAA was coated onto the surface of MCNCs by distillation-precipitation polymerization. The transmission electron microscopy (TEM) and vibrating sample magnetometer characterization showed that the obtained composite microspheres had welldefined core/shell structure and high saturation magnetization value (35 emu/g). The experimental results indicated that the thickness and degree of crosslinking of PMAA shell could be well-controlled. The pH-induced change in size exhibited by the core/shell microspheres reflected the PMAA shell contained large amount of carboxyl groups. The carboxyl groups and high saturation magnetization make these microspheres have a great potential in biomolecule separation and drug carriers. Moreover, we also demonstrated that other magnetic polymeric microspheres, such as Fe3O4/PAA, Fe3O4/PAM, and Fe3O4/PNIPAM, could be synthesized by this approach. (C) 2011 Wiley Periodicals, Inc. Fabrication and Functionalization of Dendritic Poly(Amidoamine)-Immobilized Magnetic Polymer Composite Microspheres

Abstract:The synthesis of functionalized magnetic polymer micro spheres was described by a process involving (1) preparation of the monodisperse magnetic seeds according to a two-step procedure including the preparation of bilayer-oleic acid-coated Fe3O4 nanoparticles followed by soap-free emulsion polymerization with methyl methacrylate (MMA) and divinyl benzene (a cross-linking agent, DVB); (2) seeded emulsion polymerization proceeding under the continuous addition of glycidyl methacrylate (GMA) monomers in the presence of the magnetic PMMA seeds; and (3) chemical modification of the PGMA shells with ethylenediamine (EDA) to yield amino groups. As such, the magnetic poly(MMA-DVB-GMA) microspheres were prepared possessing monodispersity, uniform magnetic properties, and abundant surface amino groups. Then, the dendritic poly(amidoamine) (PAMAM) shells were coated on the magnetic particles on the basis of the Michael addition of methyl acrylate and the amidation of the resulting ester with a large excess of EDA, which could achieve generational growth under such uniform stepwise reactions. For improving the luminescence properties of the composite particles, fluorescein isothiocyanate, which is a popular organic dye, was reacted with the terminal -NH2 groups from the dendritic PAMAM shells, resulting in the formation of multifunctional microspheres with excellent photoluminescence, superparamagnetic, and pH-sensitive properties. In this case, it can be expected that an extension of the functionalization of these microspheres is to immobilize other target molecules onto the PAMAM shells to introduce other desired functions for potential chemical and biological applications.
