Abstract:Hydrogen sulfide (H 2 S), as the third gasotransmitter, plays important roles in promoting the growth, proliferation, and metastasis of cancer cells. Recently, emerging nanotherapeutics have been developed for cancer therapy by regulating the H 2 S concentration at tumor sites. The action of H 2 S in cancer therapy can be demonstrated by a bell-shaped dose-response curve. In addition to the H 2 S donation from gas therapy, reducing the level of intratumoral H 2 S can also exert an antitumor effect by disturbing the redox balance of tumor cells. On top of that, the integration of nanotechnology further enhances the diversity of H 2 S-involved cancer treatments. As a result, phototherapy, chemotherapy, immunotherapy, and ultrasonic therapy can all be realized by designing nanotherapeutics based on the physicochemical properties of H 2 S. Furthermore, the active/passive targeting effects and onsite H 2 S-regulating ability of nanotherapeutics greatly improve the safety of treatment. To track the development and promote the clinical transformation of H 2 S-involved nanotherapeutics, this review introduced the antitumor-related properties of H 2 S gas and the evolution of H 2 S-involved cancer therapy. Then, we discussed the design concepts and mechanisms of H 2 S-involved nanotherapeutics in depth. Finally, the future expectations and challenges were spotlighted in the concept of translational medicine.

Abstract:The last two decades have seen one of the greatest excitements and discoveries in science, gasotransmitters in biology and medicine. Leading the trend by nitric oxide and extending the trudge by carbon monoxide, here comes hydrogen sulfide (H2S) who builds up the momentum as the third gasotransmitter. Being produced by different cells and tissues in our body, H2S, alone or together with the other two gasotransmitters, regulates an array of physiological processes and plays important roles in the pathogenesis of various diseases from neurodegenerative diseases to diabetes or heart failure, to name a few. As a journal dedicated to serve the emergent and challenging field of H2S biology and medicine, Antioxidant and Redox Signaling assembles the most recent discoveries and most provoking ideas from leading scientists in H2S fields, which were communicated in the First International Conference of H2S in Biology and Medicine, and brings them to our readers in two Forum Issues. Through intellectual exchange and intelligent challenge with an open-mind approach, we can reasonably expect that sooner rather than later the exploration of metabolism and function of H2S will provide solutions for many of the biological mysteries of life and pave way for the arrival of many more gasotransmitters. Abstract:Key PointsHydrogen sulfide (H2S) is an important signalling molecule; it influences physiological and pathophysiological processes throughout the body.Accurate measurement of hydrogen sulfide remains a challenge, largely because it is highly reactive. H2S synthesis from the microbiome contributes substantially to 'tissue levels' of H2S.In circumstances of anoxia or hypoxia, H2S can 'rescue' mitochondria. H2S also exerts important anti-inflammatory and cytoprotective effects while driving the resolution of tissue injury.Animal studies of several H2S-releasing drugs show considerable promise for the safe treatment of a wide range of disorders, and several such drugs are now in clinical trials.Coupling of an H2S-releasing moiety to other drugs, such as anti-inflammatory drugs, can improve efficacy and/or substantially reduce toxicity.Based on studies in animal models, H2S-based drugs may be effective in chemoprevention of cancer, and have also shown promise in the treatment of several neurological disorders, such as Alzheimer disease.

Abstract:SIGNIFICANCE: An abundance of experimental evidence suggests that hydrogen sulfide (H(2)S) plays a prominent role in physiology and pathophysiology. Many targets exist for H(2)S therapy. The molecular targets of H(2)S include proteins, enzymes, transcription factors, and membrane ion channels.RECENT ADVANCES: Novel H(2)S precursors are being synthesized and discovered that are capable of releasing H(2)S in a slow and sustained manner. This presents a novel and advantageous approach to H(2)S therapy for treatment of chronic conditions associated with a decline in endogenous H(2)S, such as diabetes and cardiovascular disease.CRITICAL ISSUES: While H(2)S is cytoprotective at physiological concentrations, it is not universally cytoprotective, as it appears to have pro-apoptotic actions in cancer cells and is well known to be toxic at supraphysiological concentrations. Many of the pleiotropic effects of H(2)S on health are associated with the inhibition of inflammation and upregulation of prosurvival pathways. The powerful anti-inflammatory, cytoprotective, immunomodulating, and trophic effects of H(2)S on the vast majority of normal cells seem to be mediated mainly by its actions as an extremely versatile direct and indirect antioxidant and free radical scavenger. While the overall effects of H(2)S on transformed (i.e., malignant) cells can be characterized as pro-oxidant and pro-apoptotic, they contrast sharply with the cytoprotective effects on most normal cells.FUTURE DIRECTIONS: H(2)S has become a molecule of great interest, and several slow-releasing H(2)S prodrugs are currently under development. We believe that additional agents regulating H(2)S bioavailability will be developed during the next 10 years.

Abstract:The important life-supporting role of hydrogen sulfide (H 2 S) has evolved from bacteria to plants, invertebrates, vertebrates, and finally to mammals. Over the centuries, however, H 2 S had only been known for its toxicity and environmental hazard. Physiological importance of H 2 S has been appreciated for about a decade. It started by the discovery of endogenous H 2 S production in mammalian cells and gained momentum by typifying this gasotransmitter with a variety of physiological functions. The H 2 S-catalyzing enzymes are differentially expressed in cardiovascular, neuronal, immune, renal, respiratory, gastrointestinal, reproductive, liver, and endocrine systems and affect the functions of these systems through the production of H 2 S. The physiological functions of H 2 S are mediated by different molecular targets, such as different ion channels and signaling proteins. Alternations of H 2 S metabolism lead to an array of pathological disturbances in the form of hypertension, atherosclerosis, heart failure, diabetes, cirrhosis, inflammation, sepsis, neurodegenerative disease, erectile dysfunction, and asthma, to name a few. Many new technologies have been developed to detect endogenous H 2 S production, and novel H 2 S-delivery compounds have been invented to aid therapeutic intervention of diseases related to abnormal H 2 S metabolism. While acknowledging the challenges ahead, research on H 2 S physiology and medicine is entering an exponential exploration era.

Abstract:Hydrogen sulfide (H2S) along with carbon monoxide and nitric oxide is an important signaling molecule that has undergone large numbers of fundamental investigations. H2S is involved in various physiological activities associated with the regulation of homeostasis, vascular contractility, pro- and anti-inflammatory activities, as well as pro- and anti-apoptotic activities etc. However, the actions of H2S are influenced by its concentration, reaction time, and cell/disease types. Therefore, H2S is a signaling molecule without definite effect. The use of existing H2S donors is limited because of the instant release and short lifetime of H2S. Thus, translational medicine involving the sustained and controlled release of H2S is of great value for both scientific and clinical uses. H2S donation can be manipulated by different ways, including where H2S is given, how H2S is donated, or the specific structures of H2S-releasing drugs and H2S donor molecules. This review briefly summarizes recent progress in research on the physiological and pathological functions of H2S and H2S-releasing drugs, and suggests hope for future investigations.

Abstract:Sulfur is not only one of the most abundant elements on the Earth, but it is also essential to all living organisms. As life likely began and evolved in a hydrogen sulfide (H2S)-rich environment, sulfur metabolism represents an early form of energy generation via various reactions in prokaryotes and has driven the sulfur biogeochemical cycle since. It has long been known that H2S is toxic to cells at high concentrations, but now this gaseous molecule, at the physiological level, is recognized as a signaling molecule and a regulator of critical biological processes. Recently, many metabolites of H2S, collectively called reactive sulfur species (RSS), have been gradually appreciated as having similar or divergent regulatory roles compared with H2S in living organisms, especially mammals. In prokaryotes, even in bacteria, investigations into generation and physiology of RSS remain preliminary and an understanding of the relevant biological processes is still in its infancy. Despite this, recent and exciting advances in the fields are many. Here, we discuss abiotic and biotic generation of H2S/RSS, sulfur-transforming enzymes and their functioning mechanisms, and their physiological roles as well as the sensing and regulation of H2S/RSS.

Abstract:Hydrogen sulfide (H2S) is a gaseous signaling molecule involved in a plethora of physiological and pathological processes. It is primarily synthesized by cystathionine-β-synthase, cystathionine-γ-lyase, and 3-mercaptopyruvate sulfurtransferase as a metabolite of the transsulfuration pathway. H2S has been shown to exert beneficial roles in lung disease acting as an anti-inflammatory and antiviral and to ameliorate cell metabolism and protect from oxidative stress. H2S interacts with transcription factors, ion channels, and a multitude of proteins via post-translational modifications through S-persulfidation ("sulfhydration"). Perturbation of endogenous H2S synthesis and/or levels have been implicated in the development of accelerated lung aging and diseases, including asthma, chronic obstructive pulmonary disease, and fibrosis. Furthermore, evidence indicates that persulfidation is decreased with aging. Here, we review the use of H2S as a biomarker of lung pathologies and discuss the potential of using H2S-generating molecules and synthesis inhibitors to treat respiratory diseases. Furthermore, we provide a critical appraisal of methods of detection used to quantify H2S concentration in biological samples and discuss the challenges of characterizing physiological and pathological levels. Abstract:Hydrogen sulfide (H2S) was recognized as a gaseous signaling molecule, similar to nitric oxide (-NO) and carbon monoxide (CO). The aim of this review is to provide an overview of the formation of hydrogen sulfide (H2S) in the human body. H2S is synthesized by enzymatic processes involving cysteine and several enzymes, including cystathionine-β-synthase (CBS), cystathionine-γ-lyase (CSE), cysteine aminotransferase (CAT), 3-mercaptopyruvate sulfurtransferase (3MST) and D-amino acid oxidase (DAO). The physiological and pathological effects of hydrogen sulfide (H2S) on various systems in the human body have led to extensive research efforts to develop appropriate methods to deliver H2S under conditions that mimic physiological settings and respond to various stimuli. These functions span a wide spectrum, ranging from effects on the endocrine system and cellular lifespan to protection of liver and kidney function. The exact physiological and hazardous thresholds of hydrogen sulfide (H2S) in the human body are currently not well understood and need to be researched in depth. This article provides an overview of the physiological significance of H2S in the human body. It highlights the various sources of H2S production in different situations and examines existing techniques for detecting this gas.

Abstract:Hydrogen sulfide (H2S) is an important gasotransmitter that is produced by mammalian cells and performs profound physiological and pathophysiological functions. Biomedical research on H2S metabolism and function in China began 20 years ago, which pioneered the examination of the correlation of abnormal H2S metabolism and cardiovascular diseases. Over the last two decades, research teams in China have made numerous breakthrough discoveries on the effects of H2S metabolism on hypertension, atherosclerosis, pulmonary hypertension, shock, angiogenesis, chronic obstructive pulmonary disease, pain, iron homeostasis, and testicle function, to name a few. These research developments, carried by numerous research teams all over China, build nationwide research network and advance both laboratory study and clinical applications. An integrated and collaborative research strategy would further promote and sustain H2S biomedical research in China and in the world.

Abstract:The discovery of endogenous gasotransmitters puts forwards a new concept, "waste gas is not waste". Hydrogen sulfide (H(2)S) is considered as a new member of gasotransmitter family, following nitric oxide (NO) and carbon monoxide (CO). Recently, the understanding of H(2)S biological effect and its mechanisms has been deepened, especially the pathophysiological significance of H(2)S in the various diseases such as cardiovascular diseases, neurological diseases, respiratory diseases, endocrine diseases, etc. This article reviews recent progress of basic, clinical and pharmacological researches related to endogenous H(2)S, including the regulatory effects of H(2)S on the cell proliferation, apoptosis, inflammation, angiogenesis and ion channels, the role of endogenous H(2)S pathway in the pathogenesis of various diseases, as well as the study of the H(2)S donor and H(2)S-related drugs.

Abstract:In recent years, research has unveiled the significant role of hydrogen sulfide (H2S) in many physiological and pathological processes. The role of endogenous H2S, H2S donors, and inhibitors has been the subject of studies that have aimed to investigate this intriguing molecule. The mechanisms by which H2S contributes to different diseases, including inflammatory conditions, cardiovascular disease, viral infections, and neurological disorders, are complex. Despite noteworthy progress, several questions remain unanswered. H2S donors and inhibitors have shown significant therapeutic potential for various diseases. This review summarizes our current understanding of H2S-based therapeutics in inflammatory conditions, cardiovascular diseases, viral infections, and neurological disorders.

Abstract:Hydrogen sulfide (H2S) has been recognized and studied for nearly 300 years, but past researches mainly focus on its toxicity effect. During the past two decades, the majority of researches have reported that H2S is a novel endogenous gaseous signal molecule in organisms, and play an important role in various systems and diseases. H2S is mainly produced by three enzymes, including cystathionine β-synthase, cystathionine γ-lyase and 3-mercaptopyruvate sulfurtransferase along with cysteine aminotransferase. H2S had been firstly reported as a neuromodulator in the brain, because of its essential role in the facilitating hippocampal long-term potentiation at physiological concentration. It is subsequently reported that H2S may have relevance to neurologic disorders through antioxidative, anti-inflammatory, anti-apoptotic and additional effects. Recent basic medical studies and preclinical studies on neurologic diseases have demonstrated that the administration of H2S at physiological or pharmacological levels attenuates brain injury. However, the neuroprotective effect of H2S is concentration-dependent, only a comparatively low dose of H2S can provide beneficial effect. Herein, we review the neuroprotevtive role of H2S therapy in brain diseases from its mechanism to clinical application in animal and human subjects, and therefore provide the potential strategies for further clinical treatment.

Abstract:Significance: Inflammation and the immune response are associated with many pathological disorders including rheumatoid arthritis (RA), lupus, cancer, heart failure and cancer(s). In recent times, important roles of hydrogen sulfide (H2S) have been evidenced by researchers in inflammatory responses, as well as immunomodulatory effects in several disease models. Recent Advances: Numerous biological targets including cytochrome c oxidase, various kinases, enzymes involved in epigenetic changes, transcription factors viz. nuclear factor kappa B (NF-κB) and nuclear factor erythroid 2-related factor 2 (Nrf-2), and several membrane ion channels, are shown to be sensitive to H2S and have been widely investigated in various pre-clinical models. Critical Issues: A complete understanding of the effects of H2S in inflammatory and immune response is vital in the development of novel H2S generating therapeutics. In this review, the biological effects, and pharmacological properties of H2S in inflammation and immune response will be addressed. The review will also cover some of the novel H2S releasing pro-drugs developed in recent years as tools to study this fascinating molecule. Future Directions: H2S plays important roles in inflammation and immunity related processes. Future researches are needed to further assess the immunomodulatory effects of H2S and to assist in the design of more efficient H2S carrier systems, or drug formulations for the management of immune related conditions in humans.

Abstract:Hydrogen sulfide (H2S), together with nitric oxide (NO) and carbon monoxide (CO), belongs to the gasotransmitter family and plays important roles in mammals as a signaling molecule. Many studies have also shown the various therapeutic effects of H2S, which include protection against myocardial ischemia injury, cytoprotection against oxidative stress, mediation of neurotransmission, inhibition of insulin signaling, regulation of inflammation, inhibition of the hypoxia-inducible pathway, and dilation of blood vessels. One major challenge in the development of H2S-based therapeutics is its delivery. In this manuscript, we assess the various drug delivery strategies in the context of being used research tools and eventual developability as therapeutic agents.

Abstract:Significance: Hydrogen sulfide (H2S) is a recently recognized gasotransmitter involved in physiological and pathological conditions in mammals. It protects organs from oxidative stress, inflammation, hypertension, and cell death. With abundant expression of H2S-production enzymes, the liver is closely linked to H2S signaling. Recent Advances: Hepatic H2S comes from various sources, including gut microbiota, exogenous sulfur salts, and endogenous production. Recent studies highlight the importance of hepatic H2S in liver diseases such as nonalcoholic fatty liver disease (NAFLD), liver injury, and cancer, particularly at advanced stages. Endogenous H2S production deficiency is associated with severe liver disease, while exogenous H2S donors protect against liver dysfunction. Critical Issues: However, the roles of H2S in NAFLD, liver injury, and liver cancer are still debated, and its effects depend on donor type, dosage, treatment duration, and cell type, suggesting a multifaceted role. This review aimed to critically evaluate H2S production, metabolism, mode of action, and roles in liver function and disease. Future Direction: Understanding H2S's precise roles and mechanisms in liver health will advance potential therapeutic applications in preclinical and clinical research.
