Abstract:This study demonstrated the combined use of sonolysis with the H2O2/ascorbic acid (Vc) redox reaction to degrade chitosan (CS). The influence of operating parameters, such as the initial CS concentration (1-15 mg mL(-1)), the input power level (15-45% of the total input power), the H2O2 and Vc concentration (10-70 mM) and the pH (3.0-5.0) of the reaction solution, on the molecular weight and degradation kinetics of CS were investigated and optimized. Based on the degradation kinetics, a synergetic effect of sonolysis with H2O2 and Vc on the degradation of CS was observed. Structural analysis by FT-IR and H-1 and C-13 NMR indicated no significant difference between the chemical structure of chitosan before and after degradation. Moreover, intermolecular hydrogen bonds were broken, and the crystallinity of degraded chitosan decreased. Using the above analysis, CS degradation by the combination of sonolysis with the H2O2/Vc redox reaction was proposed to be due to mechanical effects along with HO center dot attack on the beta-1,4-glucoside linkages of glucosamine units. These results suggest that the sonolysis/H2O2/Vc combined technique is promisingly suitable for large-scale manufacture of chitooligosaccharide.

Abstract:A new water soluble and biodegradable crosslinker of chitosan oligosaccharide-graft-acrylic acid(CSO-g-AA) was synthesized through the acrylyl reaction of amino groups in the chitosan oligosaccharide(CSO) chains with acrylic acid(AA) catalyzed by 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimidehydro-chloride(EDC) and N-hydroxysuccinimide(NHS).Polyacrylamide(PAM) hydrogels were prepared by copolymerization of acrylamide and CSO-g-AA.CSO-g-AA was structurally characterized by Fourier transform infrared spectroscopy(FT-IR) and H-1 nuclear magnetic resonance(1H-NMR) spectroscopy.The content of unsaturated double bond in CSO-g-AA was quantitatively analysed by titration analysis using potassium bromate(KBrO) and potassium bromide(KBr).The results show that the grafting ratio of unsaturated double bond in CSO-g-AA is about 4%.At a proper temperature,pH value and concentration of enzyme,the PAM hydrogels can be degraded to water soluble polyacrylamide chains in 72 h under the catalysis of snailase.
Random Attacking Model for Enzymatic Hydrolysis of Chitosan

Abstract:Assuming the degradation of polysaccharide chains of chitosan is solely caused by endochitosanase which will randomly cleavage the β-1, 4 glycosidic bond of chitosan, a mathematical model named Random Attacking Model for chitosan hydrolysis was proposed. The model is based on Michaelis-Menten kinetics considering noncompetitive inhibition by end product - D-glucosamine. The model parameters were correlated by a Powell optimization program from the experimental data of enzymatic hydrolysis of chitosan. The good agreement between the experimental data and the calculated results indicated that the model was capable to simulate the enzymatic hydrolysis of soluble chitosan by chitosanase from Trichoderma reesei ATCC56764 for various initial chitosan concentration and chitosanase activity.
Study on preparation of chitosan oligosaccharide by ultrasonic-enzyme synergistic method and its antioxidant activity

Abstract:Chitosan oligosaccharide is the degradation product of chitosan,which has small molecular weight,good water- soluble and a higher oavailability. In this experiment chitosan was degraded by pectinase under ultrasonic wave.The orthogonal experiments were conducted to obtain the optimal hydrolysis conditions,The results showed that the optimal technological condition was as follows: pectinase addition 1600 U /g,enzymolysis temperature 50 ℃,enzymolysis time under ultrasonic wave 60 min and ultrasonic power 120 W. In these conditions,the concentration of reducing sugar was determined to 1. 96 mg /mL,and the effect was better than the preparation effect by ultrasonic method and enzymatic hydrolysis respectively. The antioxidative activities of chitosan oligosaccharideprepared by this method were studied. The results showed that chitosan oligosaccharidedemonstrated significant scavenging effects on DPPH and hydroxyl radical.

Abstract:This study evaluated the degradation kinetics and structural characteristics of chitin suspension (CS) with a combination of ultrasound and chitinase. Compared with the enzymolysis, the degradation degree of sonoenzymolysis was enhanced to the maximum by 27.93 % at an intensity of 25 W/mL for 20 min. According to degradation kinetics, ultrasound intensified molecular collision rate between chitinase and substrate, thereby increasing the degradation degree. What’s more, combined with chitinase, ultrasound intensified the rate of the breakage of glycosidic bond and viscosity-average molecular weight (Mv) decrease, but no obvious change in acetylation degree (DA). Additionally, the intra- or inter-hydrogen bindings were weakened by ultrasound during sonoenzymolysis, leading to a slight decrease in crystalline index and a more ordered structure, which increased the accessibility of the substrate to enzyme. In conclusion, combination of chitinase and ultrasound could enhance the hydrolysis of CS while without changing its primary structure.

Abstract:Chondroitin sulfate (CS) is widely known for its various biological activities which are closely related to the sulfate substitution and the molecular weight. Effective degradation methods without striping sulfate groups are in a need. In the present study, a photocatalytic degradation method using H2O2 and TiO2 has been developed and it could decrease the average molecular weight of CS into 5 kDa within 6 h. The chemical composition of CS before and after degradation were compared by FT-IR, NMR, etc., and no removement of sulfate group was observed. Then the identification of the oligosaccharides in the degradation product by mass spectroscopy revealed that glucuronic acid or its derivative, arabinuronic acid, was at most of the reducing ends, and the depolymerization mechanism was proposed. Furthermore, the absorption of CS in rats was enhanced by the degradation while the excertion profile of the degradation product was similar to that of CS.

Abstract:High-molecular-weight chitosan has limited applications due to unsatisfactory solubility and hydrophilicity. Discharge plasma coupled with peracetic acid (PAA) oxidation ("plasma+PAA") realized fast depolymerization of high-molecular-weight chitosan in this study. The molecular weight of chitosan rapidly declined to 81.1 kDa from initial 682.5 kDa within 60 s of "plasma+PAA" treatment, and its reaction rate constant was 12-fold higher than single plasma oxidation. Compared with 1O2, ∙CH3, CH3O2·, and O2∙-, CH3CO2∙ and CH3CO3∙ played decisive roles in the chitosan depolymerization in the plasma+PAA system through mechanisms of radical adduct formation. The attacks of CH3CO2∙ and CH3CO3∙ destroyed the β-(1,4) glycosidic bonds and hydrogen bonds of chitosan, leading to generation of low-molecular-weight chitosan; the main chain structure of chitosan was not changed during the depolymerization process. Furthermore, the generated low-molecular-weight chitosan exhibited greater antioxidant activities than original chitosan. Overall, this study revealed the radical adduct formation mechanisms of CH3CO2∙ and CH3CO3∙ for chitosan decomposition, providing an alternative for fast depolymerization of high-molecular-weight chitosan.

Radiation Degradation of Chitosan in the Presence of H2o2

Abstract:In order to obtain the chitosan oligomers, chitosan was irradiated in the solid state with and without H2O2 as a radiation degradation sensitizer, respectively. At room temperature, the viscosity average-molecular weight (M-eta) of chitosan was decreased from 1.6 x 10(6) to 2.2 x 10(5) at an absorbed dose of 72 kGy without H2O2, and decreased to 2.7 x 10(4) at 2 kGy in the presence of an appropriate H2O2 content. In addition, the radiation degradation rate of chitosan containing 38.2wt% H2O2 is 59 times higher than that in the solid state without H2O2. FT-IR analysis suggests that there is no obvious change in the chemical structure of irradiated chitosan with and without H2O2 at a dose below 20 kGy, compared with unirradiated chitosan. On the other hand, the degree of deacetylation (DD) of irradiated chitosan in the studied dose range changed slightly, while DD of irradiated chitosan with H2O2 increased significantly. The XRD pattern indicates that the irradiated chitosan with H2O2 has more perfect crystalline structure than unirradiated chitosan. Therefore, it could be expected that irradiation of chitosan using H2O2 as a sensitizer would be a very effective method to prepare low molecular weight chitosan, because of its feasibility and benignancy to environment.

Abstract:Two kinds of chitosans, namely N-acetylated and N-deacetylated chitosan were prepared by the modified processes. They can dissolve in both acid and alkali solution. 13C NMR was used to study the basic solution of chitosan, and XRD, FT-IR and SEM were used to study the structure of N-acetylated and N-deacetylated chitosan. The result from X-ray diffraction showed that a transformation of crystal structure occurred during the N-acetylation or N-deacetylation process with the decrease of crystallinity and expansion of crystal lattices. FT-IR spectra revealed that the intermolecular and intramolecular hydrogen bonds were destroyed by both treatments and a looser structure was observed by the SEM. The lower crystallinity, the decreased intermolecular interactions, the more disordered and looser structure were easy for the permeation of LiOH/urea aqueous solution and coordinated with the breakage of intermolecular and intramolecular hydrogen bond by LiOH at low temperature, the prepared chitosans dissolved in LiOH/urea/H2O mixture.

Abstract:In situ H2O2 generation systems are efficient for H2O2-dependent biocatalytic oxidation reactions. Here, we report that lytic polysaccharide monooxygenases (LPMOs), copper-dependent polysaccharide monooxygenases, can efficiently supply H2O2 in situ to dye-decolorizing peroxidases (DyPs) using substrate gallic acid (GA) for chitosan functionalization. The maximum grafting ratio induced by the cascade reaction was significantly higher than that achieved by a reaction with initial exogenous H2O2. The maximum grafting ratio was obtained with 12 g/L GA, 5.6 mg/L DyP, 20-30 mg/L LPMO, and pH 4.5-5.0. UV-vis, Fourier transform infrared (FT-IR), and nuclear magnetic resonance (1H NMR) spectroscopy confirmed GA grafting onto chitosan. X-ray diffraction (XRD) analysis and thermogravimetric analysis (TGA) indicated that GA-chitosan conjugates had lower thermal stability and crystallinity than chitosan. The GA-chitosan conjugates had significantly higher antioxidant activity than chitosan. This study supplies a green and high-efficiency approach to achieve an enzymatic cascade reaction for chitosan functionalization and has potential applications in H2O2-dependent biocatalytic oxidation reactions.

High-efficiency and energy-saving preparetion for Chitosan oligosaccharide with integrated reaction and separation by CSMB technology

Abstract:To work out the technical problems of the preparation methods and equipments of chitosan oligosaccharide,the paper creatively proposed the integrated reaction and separation technology which is integrated electrochemical oxidation degradation of chitosan with continuous preparative chromatography separation and extraction of chitosan oligosaccharide.It can be used to obtain chitosan oligosaccharide under high-efficiency,energy-saving and green production.With the numerical simulation and a large number of experiments,it is shown that 120 minutes is the best coupling point between the reaction and the separation.Based on them, an integrated reaction and separation equipment,Coupled/cascade Simulated Moving Bed was developed.The results of practical tests showed that the purity of the product can reach to more than 98%,its energy-savings can be up to 25%and the emissions of elution solvent can be significantly reduced.The method and equipment has a high performance and potential application.
Oxidative degradation of chitosan by Fe-MCM-41 heterogeneous Fenton-like system

Abstract:We herein disclosed an efficient multiphase Fenton-like catalytic system for oxidative degradation of chitosan. By utilizing Fe-MCM-41, featured with regular mesoporous structure and high specific surface area, the activation efficiency of H 2 O 2 was significantly enhanced and the chitosan degradation efficiency proved by 28.1% higher than the conventional system using H 2 O 2 alone. Under optimized conditions (5 g/L chitosan, 0.5 g/L Fe-MCM-41, 0.16 mol/L CH 3 COOH, 0.86 mol/L H 2 O 2 , 50 °C, 140 min), the viscosity reduction rate of chitosan reached an impressive 98.2%. Among the catalysts tested, Fe-MCM-41 with a loading factor of x = 0.12 demonstrated optimal degradation performance. After four recycles, the degradation efficiency maintained > 93.6%, demonstrating its excellent stability and recyclability for potential industrial applications. Kinetic studies provided further elucidation of the reaction mechanism, which indicated that the degradation process of chitosan followed a first-order kinetic model, with an apparent activation energy (E a ) of 48.91 kJ/mol. This novel and efficient strategy for chitosan degradation, addressed the challenges of catalyst recovery and secondary pollution typically associated with traditional Fenton systems, and posed broad application potential for polysaccharide material processing.

Abstract:A solution of partially N-deacetylated chitosan in aqueous lithium hydroxide (LiOH)/urea was prepared successfully through a freeze-thawing process and the dissolution behavior was studied. The results indicated that chitosan can directly dissolve in LiOH/urea aqueous solution. LiOH mainly contributed to the breakage of intramolecular and intermolecular hydrogen bonds in chitosan. Urea, LiOH, and chitosan formed inclusion compound (IC) with urea as the IC host, and the LiOH-chitosan complex as the guest. Aqueous 4.8 wt% LiOH/8.0 wt% urea was verified to be the optimal solvent for chitosan. The results of rheology and viscosity characterizations revealed that chitosan/4.8 wt% LiOH/8.0 wt% urea aqueous solution was pseudoplastic fluid, and was more stable than the solution of chitosan in acetic acid at ambient temperature.

Abstract:A new scheme was proposed for synthesizing poly(ethylene glycol)-g-chitosan (PEG-g-CS), where methoxy poly(ethylene glycol) iodide (MPEG-I) (Mn 2000) was used for N-substitution of triphenylmethyl chitosan (TPM-CS) in organic medium. The graft copolymers were obtained by subsequent removal of protecting groups with dichloroacetic acid. By varying PEG-I/TPM-CS feed ratio, the grafting levels (GL) of PEG can be adjusted. The chitosan derivatives were characterized by FTIR, 1H NMR, 13C NMR and DSC. All the copolymers were soluble in water over wide pH range. Furthermore, organosolubility of the hybrids in DMF and DMSO was also achieved when the DS value more than 24%. The lysozyme degradation rate of the copolymers in aqueous neutral medium decreased with the increase of GL value.

Abstract:Chitosan samples were irradiated by 60Co γ-rays in the presence of hydrogen peroxide with radiation dose from 10 kGy to 100 kGy. The degradation was monitored by gel permeation chromatography (GPC), revealing the existence of a synergetic effect on the degradation. Structures of the degraded products were characterized with Fourier-transform infrared spectra (FT-IR), ultraviolet–visible spectral (UV–vis) analysis, and X-ray diffraction (XRD). Results showed that the crystallinity of chitosan decreases with degradation, and the crystalline state of water-soluble chitosan is entirely different from that of water-insoluble chitosan. An elemental analysis method was employed to investigate changes in the element content of chitosan after degradation. Mechanism of chitosan radiation degradation with and without hydrogen peroxide was also discussed.
Superadsorption of LiOH Solution on Chitosan As a New Type of Solvent for Chitosan by Freezing/blasting

Abstract:The adsorption behavior and mechanism of chitosan in aqueous LiOH solution was studied systemically. The results showed that the adsorption of chitosan was mainly due to the breakage of its hydrogen bonds, which were destroyed by the reaction of LiOH with the acetyl and the hydroxyl groups of chitosan. Low temperature also played a crucial role in the adsorption of chitosan. The adsorption of chitosan decreased with increased DD. The adsorption ratio of LiOH to chitosan (nLiOH/nCS) increased linearly while the adsorption ratio of water to chitosan (n(H2O)/n(CS)) decreased with the increased DD. All chitosans reached their maximal swelling degree when the concentration of LiOH was 4.8 wt%. Chitosan was stable in LiOH aqueous solution. The LiOH solution may be a potential favorable solvent for chitosan.

Abstract:Oxidation has been profitably utilized to improve some properties of chitosan. However, only solvent-based oxidation procedures have been proposed so far, which are hardly feasible at industrial scale in an economic way because of product recovery cost. In this study, a solvent-free, rapid, and effective oxidation method is proposed. It is based on direct solid-state reaction between chitosan and oxidant powder in a mechanochemical reactor. Results prove that by short high energy ball milling (< 3 h) it is possible to achieve diverse physicochemical modifications employing different reagents. Apart from polysaccharidic chain shortening, persulfate provokes high amorphization and induces formation of ketonic groups; percarbonate heightens deacetylation degree, preserving in part crystallinity; calcium peroxide merely deprotonates amino groups and increases amorphization degree. Adsorption tests with the azo-dye reactive red 2 show that adsorption capacity of chitosan co-milled with persulfate (974 mg/g milled product), which is the best performing adsorbent, is twice that of pristine chitosan, while adsorption rate is outstandingly boosted (125 times).
Preparation and Structure of Chitosan Swellable in Alkali Solution by Ultrasonic Treatment

Abstract:By treating chitin with ultrasonic in hot concentrated NaOH solution for about 30-120min, chitosan swellable highly in alkali was prepared. The swelling behavior and the structure of the prepared chitosan were invested. It was shown that the prepared chitosan can swell highly in NaOH solution by freeze-thawing treatment, with the particles turns into hydrogel. The swelling behavior showed that with the increase of NaOH concentration, the swelling degree of chitosan increases, until a concentration of 8.0 wt%, and ultrasonic treatment time has less significant influence on the swelling behavior. SEM showed that ultrasonic treatment makes the structure of chitin enough looser for the permeation of NaOH and H2O, resulting in the swelling of chitosan in alkali solution.

Abstract:Chitosan dissolved in CH_3COOH/NaCl aqueous solution was irradiated at various conditions,and the influence of H_2O_2,N_2O,isopropanol,pH and molecular weight of chitosan on the radiation degradation of chitosan in aqueous solutions was investigated.In acidic solutions(pH=3),the degradation of chitosan in aqueous solutions might be attributed to the interaction with both ·H and ·OH radicals.The degradation of chitosan was slightly faster in the presence of H_2O_2 or N_2O,because the concentrations of ·OH radicals increased slightly.The degradation of chitosan was inhibited obviously after adding isopropanol,due to the decrease of both ·H and ·OH radicals in the presence of isopropanol.With the pH of the solution approaching neutral,the concentration of ·H decreased and the ·OH radicals increased rapidly with the presence of H_2O_2 or N_2O.The chain scission of chitosan was promoted significantly after adding H_2O_2 or saturated with N_2O.In addition,it was found that the rate constant of radiation degradation of chitosan increased with the decrease of molecular weight of chitosan.UV and FTIR analyses of radiation-degraded chitosan manifested that except for carbonyl groups generated at the ends of molecular chains,no obvious change in the main chain structure of radiation-degraded chitosan,comparing with unirradiated chitosan.Therefore,gamma irradiation can be expected as an effective way to control the molecular weight of chitosan without obvious change in its backbone structure.
Preparation of low molecular weight chondroitin sulfate from different sources by H2O2/ascorbic acid degradation and its degradation mechanism

Abstract:Low molecular weight chondroitin sulfate (LMCS) has attention for enhanced bioavailability and bioactivity compared to native CS. We optimized H2O2/ ascorbic acid (Vc) degradation conditions to prepare LMCS from chicken, bovine, and shark cartilages. Degradation kinetics models and chemical composition data of LMCS showed the GlcA residues of chondroitin-4-sulfate (CSA) may be preferentially attacked. Nuclear magnetic resonance (NMR) spectroscopy and high-performance liquid chromatography-electrospray mass spectrometry (HPLC-MS) indicated that the CH of GlcA in CS was broken through a hydrogen abstraction reaction to break the β-(1 → 3) bond and form the hexendioic acid product. Standard density functional theory (DFT) calculations indicated that the energy required for the hydrogen abstraction from the C1-H bond in GlcA was lower than that of GalNAc. Molecular dynamics (MD) showed that CSA was more likely to interact with hydroxyl radicals (·OH) than non-sulfated chondroitin (CSO) and chondroitin-6-sulfate (CSC). These results provide guidance for producing LMCS.
