Abstract:The present study discloses the synthesis of aryl/vinyl carboxylic acids from Csp(2)-bound halides (Cl, Br, I) in a carbonylative path by using silyl formate (from CO2 and hydrosilane) as an instant CO-surrogate. Hydrosilane provides hydride for reduction and its oxidation product silanol serves as a coupling partner. Mono-, di-, and tri-carboxylic acids were obtained from the corresponding aryl/vinyl halides.

Abstract:A cobalt-catalyzed asymmetric reduction of beta,beta-dialkyl alpha,beta-unsaturated esters with polymethylhydrosiloxane (PMHS) was reported to deliver the corresponding esters containing a chiral trialkyl carbon center at beta-position with up to 97 % yield and 98 % ee. The chiral tridentate ligand oxazoline iminopyridine (OIP) could perform well for the asymmetric reduction instead of chiral bidentate ligands. This operationally simple protocol shows a broad scope of substrates using one equivalent of readily available PMHS as a cheap and easy-to-handle reductive reagent.

Abstract:Cooperative bimetallic catalysis is a fundamental approach in modern synthetic chemistry. We report bimetallic cooperative catalysis for the direct decarbonylative heteroarylation of ubiquitous carboxylic acids via acyl C-O/C-H coupling. This novel catalytic system exploits the cooperative action of a copper catalyst and a palladium catalyst in decarbonylation, which enables highly chemoselective synthesis of important heterobiaryl motifs through the coupling of carboxylic acids with heteroarenes in the absence of prefunctionalization or directing groups. This cooperative decarbonylative method uses common carboxylic acids and shows a remarkably broad substrate scope (>70 examples), including late-stage modification of pharmaceuticals and streamlined synthesis of bioactive agents. Extensive mechanistic and computational studies were conducted to gain insight into the mechanism of the reaction. The key step involves intersection of the two catalytic cycles via transmetallation of the copper-aryl species with the palladium(II) intermediate generated by oxidative addition/decarbonylation.

Abstract:We report a one-pot synthesis of aryl carboxylic acids utilizing HCOOH as a CO surrogate with low Pd-catalyst loading. This operationally simple and scalable method does not require use of a high-pressure reactor, two-chamber reaction vessel, phosphine ligand, or base and proceeds in a relatively short amount of time at ambient temperature. Notably, halides, including iodo and bromo groups, and nitro groups remain intact under these mild reaction conditions. This methodology has been successfully applied to synthesizing 13 C-labeled aryl carboxylic acids with satisfactory yields.

Abstract:Aryl carboxylic acids are among the most abundant substrates in chemical synthesis and represent a perfect example of a traceless directing group that is central to many processes in the preparation of pharmaceuticals, natural products and polymers. Herein, we describe a highly selective method for the direct step-down reduction of carboxylic acids to arenes, proceeding via well-defined Pd(0)/(ii) catalytic cycle. The method shows a remarkably broad substrate scope, enabling to direct the classical acyl reduction towards selective decarbonylation by a redox-neutral mechanism. The utility of this reaction is highlighted in the direct defunctionalization of pharmaceuticals and natural products, and further emphasized in a range of traceless processes using removable carboxylic acids under mild, redox-neutral conditions orthogonal to protodecarboxylation. Extensive DFT computations were conducted to demonstrate preferred selectivity for the reversible oxidative addition and indicated that a versatile hydrogen atom transfer (HAT) pathway is operable.

Abstract:A metal-free multicomponent strategy has been developed for the synthesis of various α-hydroxyamides via carbonylation of aldehydes/ketones and amines enabled by the difluorocarbene reagent TMSCF2Br (TMS = trimethylsilyl). TMS-protecting group derived from TMSCF2Br plays a crucial role for the tunability of reaction pathways from intramolecular cyclization to intermolecular hydrolysis. The synthetic utility had been demonstrated by the late-stage modification of several drug-related molecules and the highly selective synthesis of 18O-labeled α-hydroxyamides from 18O-labeled water.

Abstract:Starting from diverse alkene-tethered aryl halides and silyl formate formed in situ from CO2 and PMHS (polymethylhydrosiloxane), the carbo-carboxylation of alkenes has been achieved using an easily accessible Pd/monophosphine ligand combination as the catalyst. This protocol provides competent access to construct a series of 2,3-dihydrobenzofurans, chromans, isochroman, indanes and tetralins bearing a β-acid moiety in moderate to excellent yield. Additionally, the example of domino carbo-carbonylation reactions has been demonstrated using diene-tethered aryl iodide as a starting material, forming an oxygen-containing bicyclic product in good yield. It is speculated that the carbo-carboxylation reaction of alkenes proceeds via a carbo-silyloxycarbonylation pathway.

Abstract:The selective, transition metal‐free hydrosilylation of CO2 to CH2(OSiEt3)2 has been achieved under mild conditions and in high isolated yields (up to 90%) using Et3SiH and the simple, easily prepared borohydride catalyst Li+[HB(C6F5)3]−. The resulting CO2‐derived bis(silyl)acetal product – whose mechanism of formation has been interrogated through detailed computational and experimental studies – can be rapidly valorized through the facile synthesis of N‐heterocyclic carbenes, via their corresponding imidazolium salts. By using relatively inexpensive, isotopically enriched 13CO2 this protocol can be exploited to prepare NHC isotopologues that are selectively 13C labelled at the key, ligating C2 position. This provides an electronically responsive 13C NMR spectroscopic handle with dramatically enhanced sensitivity which can directly benefit reactivity studies in both organo‐ and organometallic catalysis, where NHC use is ubiquitous.

Abstract:The development of facile and convenient atom-economical methods for preparation of organosulfur compounds from CS2 is a challenging endeavor. Herein, a one-pot, environmentally friendly access to S-aryl/alkyl dithiocarbamates has been demonstrated by the three-component coupling involving aryl/alkyl thiols, CS2 and amines in the presence of a common base K2CO3. The transformation process can proceed in H2O-DMAc (3:1) mixed solvent without requiring any catalysts or extensive prefunctionalization of reactants. The protocol is operationally simple and affords dithiocarbamates with a various moieties (including aryl, aliphatic, heteroaryl and alkenyl) in good yields.

Abstract:The palladium-catalyzed cycloaminocarbonylation of 2-(aminomethyl)aryl tosylates with CO has been established, by which a variety of salicylaldehyde derived 2-(aminomethyl)aryl tosylates may be cyclocarbonylated in the presence of CO, to afford the corresponding substituted isoindolinones in moderate to excellent yields. Furthermore, the method is also effective for the synthesis of isoindoline-1,3-diones and 2-alkyl-1H-benzo[e]isoindol-3(2H)-ones from 2-(N-alkylcarbamoyl)aryl tosylates and 1-(aminomethyl)naphthalene-2-yl tosylates, respectively.

Abstract:A novel approach to 2-substituted-2-(dimethoxyphosphoryl)-pyrrolidines 7a-7o and 9a-9r has been developed, which features a TMSOTf-mediated one-pot intramolecular cyclization and phosphonylation of substituted tert-butyl 4-oxobutylcarbamates. The major advantages of this method include simple operation under mild reaction conditions, the use of cheap Lewis acid, and good to excellent yields with high diastereoselectivities (dr up to 99:1).

Abstract:Carbonylation coupling reaction has emerged as a powerful and versatile strategy for the construction of carbonyl‐containing compounds in modern synthetic chemistry over the past years. Carbon dioxide, a renewable one carbon molecule, has become one of the most attractive and promising alternative carbonyl source due to its highly abundance, nontoxicity and stability in comparison with CO in recent years. However, in most cases, a two chamber technique was generally necessary to allow the CO‐producing and CO‐consuming processes to perform successfully because of the complexities and incompatibility of reaction conditions, when carbon dioxide was utilized as carbonyl source. Herein, a practical one‐pot protocol using carbon dioxide as the carbonyl source for the palladium‐catalyzed carbonylative Sonogashira coupling has been established, providing an expedient and practical route to a wide range of functionalized alkynones and indoxyls under mild reaction conditions. By finding a suitable catalytic system, the method allowed the CO‐generating and CO‐consuming processes to proceed in one pot, wherein carbon monoxide was generated in situ from the reduction of carbon dioxide in the absence of any fluoride reagents. Simple and safe operation, readily available substrates, good functional group tolerance and mild reaction conditions are the features of the method.

Abstract:An efficient method for the direct preparation of high synthetic valuable α-hydroxycarbonyls is described. The simple and readily available I2 or NBS was used as catalyst. DMSO acts as the oxidant, oxygen source, and solvent. A diverse range of tertiary Csp(3)-H bonds as well as more challenging secondary Csp(3)-H bonds could be hydroxylated in this transformation. The reaction is mild, less toxic and easy to perform.

Abstract:Carbon dioxide (CO2), as a one-carbon synthon, has many advantages such as abundant, non-toxic, clean and so on. So the reactions using CO2 as a one-carbon synthon have been widely concerned in organic chemistry. Transition-metalcatalyzed reaction of unsaturated hydrocarbons with CO2 to produce carboxylic acid is one of the most commonly-used method to convert CO2, and organometallic reagents can be added to the reaction as reducing agent. This kind of reaction can be realized by the strategy of tandem reaction. In the reaction, the unsaturated hydrocarbons react with transition-metal catalysts and organometallic reagents to generate new organometallic reagents in situ first, and then complete carboxylation with CO2. Common organometallic reagents such as organozinc reagents, Grignard reagents, and organoaluminum reagents can all achieve this kind of carboxylation reaction. In this review, reactions are divided according to the type of unsaturated hydrocarbons, and each type can also be divided into hydrocarboxylation and carbocarboxylation. This review would introduce reaction according to this classification.
