Abstract:Pseudostellaria heterophylla has been becoming a popular research topic because of its functionally active components. The immunomodulatory activity of P. heterophylla peptide (PPH) derived from protein hydrolysate and the molecular mechanism underlying its immunomodulatory effect were investigated in this study. Immunomodulatory PPH achieved the highest stimulation index of 1.53 at a concentration of 100 mu g mL(-1) for 48 h in spleen lymphocytes and promoted the secretions of tumor necrosis factor-alpha, interferon-gamma, and interleukin-10. Moreover, PPH could elevate the intracellular Ca2+ concentration, calcineurin activity and nuclear factor of activated T cell (NFAT) c1 mRNA expression. Meanwhile these effects could be diminished by the treatment of verapamil and cyclosporin A, suggesting that PPH may activate spleen lymphocytes via the Ca2+/CaN/NFATc1/IFN-gamma signaling pathway. These findings demonstrate that the P. heterophylla peptide has the potential to be utilized as a nutraceutical supplement to strengthen the immune system in the human body.

Abstract:Pseudostellaria heterophylla protein hydrolysate has been reported to be a potential nutraceutical supplement to strengthen the immune system. A specific peptide from P. heterophylla protein hydrolysate with proliferation promotion effect on spleen lymphocytes was purified by RP-HPLC. The amino acid sequence of the peptide was determined to be RGPPP, with molecular weight of 522.29 Da. 100 μg/mL of RP-5 achieved the highest stimulation index of 1.27 on spleen lymphocytes and activated RAW264.7 cells resulting in up-regulating tumor necrosis factor-α (TNF-α) secretion to 827.24 pg/mL and elevating pinocytosis rate to 123.81%. Results of western-blot and siRNA experiments indicated that RP-5 activated nuclear factor-κB (NF-κB) signaling pathway through combining to TLR2 expressed on RAW264.7 cells surface. Furthermore, TNF-α secretion increased by RP-5 could be decreased to 401.68 pg/mL by pyrrolidine dithiocarbamate or BAY11-7082. These findings further the research progress of peptide with immunomodulatory activity and its action mechanism.

Abstract:In this study, a bioactive peptide YGPSSYGYG (YG-9) with immunomodulatory activity was isolated and identified from Pseudostellaria heterophylla protein hydrolysate. The highest proliferation index of mouse spleen lymphocytes reached 1.19 in the presence of 50 μg/mL YG-9. YG-9 could activate RAW264.7 cells by promoting the secretions of NO, the pinocytosis activity, and the productions of ROS and TNF-α. Moreover, YG-9 enhanced the expressions of TLR2 and TLR4 in RAW264.7 cells. TNF-α secretions induced by YG-9 were reduced in TLR2 and TLR4 siRNAs knockdown cells, and this suggested that macrophage activation of YG-9 was through TLR2 and TLR4. Furthermore, YG-9 promoted the translocation of NF-κB through the acceleration of IκB-α phosphorylation and degradation. Also, TNF-α secretions promoted by YG-9 were inhibited by NF-κB-specific inhibitors pyrrolidine dithiocarbamate and BAY11-7082. Altogether, these results suggested YG-9 activated RAW264.7 cells via the TLRs/NF-κB/TNF-α signaling pathway.
The regulatory effects of Pseudostellaria heterophylla polysaccharide on immune function and gut flora in immunosuppressed mice

Abstract:We discovered for the first time that the polysaccharides of Pseudostellaria heterophylla can significantly enhance the immune function of immunosuppressed mice. Pseudostellaria heterophylla polysaccharide might be a promising natural functional foods for reducing chemotherapy‐induced immunosuppression. Polysaccharide (PF40) has been recognized as a main bioactive substances in Pseudostellaria heterophylla (Miq.). The current study explored the potential protective effects of PF40 on immune system in mice with cyclophosphamide‐induced immunosuppression. The mice were intragastric administered PF40 at the dosage of 100, 200 or 400 mg/kg once daily for 30 days, On the 24th and 25th day, the additional intraperitoneal injection of PF40 (50 mg/kg) were administered. The results showed that PF40 enhanced the cell‐mediated immunity via improvements in macrophage phagocytosis, splenocyte proliferation, NK cell activity and delayed type hypersensitivity. Equally, it improves humoral immunity through promoting the formation of serum hemolysin. Moreover, PF40 maintain the immune balance of splenic lymphocytes and altered the intestinal physiological status in Cyp‐induced mice. PF40 regulates the intestinal microbiota by restoring the relative abundance of Odoribacter and Mucispirillum and reducing the relative abundance of Sporosarcina, Yaniella, and Jeotgalicoccus in Cyp‐intervened mice. The findings suggested that PF40 might be a promising natural functional foods for reducing chemotherapy‐induced immunosuppression.

Abstract:The aim of this study was to investigate the molecular mechanism underlying the immunomodulatory effect of Ganoderma atrum polysaccharide (PSG-1) in spleen lymphocytes. Our results showed that PSG-1 increased the intracellular Ca2+ concentration and calcineurin (CaN) activity. Moreover, PSG-1 was found to elevate nuclear factor of activated T cells (NFAT) activity, but this effect could be diminished by the treatment of CaN inhibitors (cyclosporin A and FK506). PSG-1-induced interleukin (IL)-2 production was also inhibited by cyclosporin A and FK506. In addition, PSG-1 was found to significantly enhance protein kinase C (PKC) activity. PKC was involved in induction of NFAT activity by PSG-1, as evidenced by abrogation of NFAT activity by PKC inhibitor calphostin C, which significantly decreased PSG-1-induced IL-2 production. On the basis of these results, we concluded that PSG-1 may induce activation of spleen lymphocytes at least in part via the Ca2+/CaN/NFAT/IL-2 signaling pathway and the PKC/NFAT/IL-2 signaling pathway cooperatively regulated PSG-1-induced activation of spleen lymphocytes.
Immunopotentiation on Murine Spleen Lymphocytes Induced by Polysaccharide Fraction of Panax Ginseng Via Upregulating Calcineurin Activity

Abstract:Calcineurin (CN), a unique Ca2+/calmodulin (CaM)-dependent serine/threonine protein phosphatase, plays a pivotal role in the activation and proliferation of T lymphocytes. Based on the effective molecular screening model established in our laboratory, we found that a part of polysaccharides from the stem and leaves of Panax ginseng, termed PGP-SL, could activate CN activity. Subsequently, we investigated whether PGP-SL also has immunological competence on murine spleen lymphocytes. In the present study, we demonstrated that PGP-SL could significantly promote in vitro spleen lymphocyte proliferation in the absence of either concanavalin A or LPS in a concentration-dependent manner at concentrations ranging from 100 to 500 microg/ml (p<0.001). In addition, the proliferation of cyclosporin A (CsA)-treated spleen lymphocytes was also significantly promoted in the same pattern (p<0.001); the production of IL-2 was elevated and the effect appeared as early as 24 h after PGP-SL treatment. The results of RT-PCR also indicated that the IL-2 mRNA level was markedly enhanced, particularly at PGP-SL concentrations of 300 and 500 microg/ml, and Fura-2/AM fluorescence probe analysis showed that PGP-SL could dramatically increase the intracellular free calcium concentration of spleen lymphocytes, i.e. [Ca2+]i was significantly increased by approximately 181 and 107% at 300 and 500 microg/ml of PGP-SL, respectively. However, this effect could be totally inhibited by verapamil treatment. Taking our results together, we suggest that PGP-SL exhibits immunopotentiation effects on murine spleen lymphocytes by the Ca2+-CN-NFAT-IL-2 signaling pathway.

Abstract:Aim of the study: The objectives of this study were to evaluate the in vivo antitumor potential of the triterpenoid fraction from the rhizomes of Astilbe chinensis (Saxifragaceae) (Saxifragaceae) (ATF) and to elucidate its immunological mechanisms by determining its effects on the growth of mouse transplanted tumors and the immune response in naive and tumor-bearing mice.Materials and methods: The mice inoculated with mouse tumor cell lines were treated per os with ATF at the doses of 20,40,60 mg/kg for 10 days. The effects of ATF on the growth of transplantable tumor, splenocyte proliferation, the activity of natural killer (NK) cells, and production of interleukin-2 (IL-2) from splenocytes in tumor-bearing mice were measured. Meanwhile, the effects of ATF on 2,4-dinitrofluorobenzene (DNFB)-induced delayed type hypersensitivity (DTH) reaction and the sheep red blood cell (SRBC)-induced antibody response in naive mice were also studied.Results: ATF could not only significantly inhibit the growth of mice transplantable tumor, but also remarkably increase splenocytes proliferation, NK cells activity, and the level of IL-2 secreted by splenocytes in tumor-bearing mice, promote the DTH reaction and enhance anti-SRBC antibody level in naive mice, which indicated that the ATF could improve both specific and non-specific cellular and humoral immune response.Conclusions: The antitumor activity of ATF might be achieved by improving immune response, and ATF could act as antitumor agent with immunomodulatory activity. Abstract:Radix Pseudostellariae, the root of Pseudostellaria heterophylla (Miq.) Pax, has historically been used as medicine food homology plant for thousand years in China. Herein, PHP, a water soluble polysaccharides from Radix Pseudostellariae, was isolated and identified by HPGPC, GC–MS, FT-IR, and scanning electron microscopy. Results showed that PHP (Mw = 1.07 × 104 kDa) mainly consisted of rhamnose, galacturonic acid, glucose, galactose, arabinose, fucose in a molar ratio of 0.055:0.208:0.306:0.055:0.753:0.594:0.014. It contained nine types of linkages, including → 6)-Galp(A)-(1→, →6)-Galp-(1→, →3)-Arap-(1→, Galp-(1→, →3)-Rhap-(1→, Glup-(1→, →3)-Araf-(1→, →2,3,4)-Manp-(1 → and → 3,4)-Fucp-(1→. Morphology analysis showed that PHP took flakes with irregular size and properties. It significantly activated TLR4 in vivo. In DSS induced mouse-acute-colitis molds, PHP90-3a improved and the colon length was on ulcerative in mice. the production of IL-1β and TNF-α were decreased. The intestinal mucosa was more complete, there were much more goblet cells, and the infiltration of lymphocytes and neutrophils was significantly decreased. The content of SCFA increased, whereas acetic acid, propionic acid and butyric acid increased. 16S RNA sequencing and OTUs cluster analysis revealed that Bacteroides and Pseudomonas were the dominant intestinal flora. The analysis of community thermogram showed that the abundance ratio of Pseudomonas and Bacteroides (F/B) was significantly lower than the dominant group and significantly inhibited the levels of Verrucomicrobia and Akkermansia. Altogether, the data suggested that the anti-inflammatory activity might be implicated with the activation of TLR4 signal pathways, reducing IL-1β and TNF-α, and the regulation of mucosal immunity and the composition of intestinal flora.

Abstract:Our previous study has demonstrated that Pseudostellaria heterophylla protein hydrolysate (PPH) has immunomodulatory activity on murine spleen lymphocytes. The aim of this study was to investigate the excitation of PPH in RAW264.7 macrophage cells and the protective effect in cyclophosphamide (CTX)-treated mice. The results showed PPH of 50 μg/mL could stimulate macrophages resulting in significant promotions of nitric oxide (NO) production, endocytosis and reactive oxygen species formation. Meanwhile, enzyme-linked immunosorbent assay (ELISA) revealed that the levels of tumor necrosis factor-α and interleukin-10 were significantly upregulated by PPH. Furthermore, 50 mg/kg per day PPH restored the T lymphocyte proliferation and natural killer cell activity, and increased NO production and pinocytosis of peritoneal macrophages in CTX-treated mice. These findings indicate PPH plays a crucial role in RAW264.7 macrophage cells activation and in the protection against immunosuppression in CTX-treated mice and could be used as a potential immunostimulant agent.

Abstract:Our previous study has demonstrated that Ganoderma atrum polysaccharide (PSG-1) has immunomodulatory activity on spleen lymphocytes. However, how PSG-1 exerts its effect on purified lymphocytes is still obscure. Thus, this study aimed to investigate the immunomodulatory activity of PSG-1 on purified T lymphocytes and further elucidate the underlying mechanism based on RNA sequencing (RNA-seq). Our results showed that PSG-1 promoted T lymphocytes proliferation and increased the production of IL-2, IFN-γ, and IL-12. Meanwhile, RNA-seq analysis found 394 differentially expressed genes. KEGG pathway analysis identified 20 significant canonical pathways and seven biological functions. Furthermore, PSG-1 elevated intracellular Ca2+ concentration and calcineurin (CaN) activity and raised the p-ERK, p-JNK, and p-p38 expression levels. T lymphocytes proliferation and the production of IL-2, IFN-γ, and IL-12 were decreased by the inhibitors of calcium channel and mitogen-activated protein kinases (MAPKs). These results indicated that PSG-1 possesses immunomodulatory activity on purified T lymphocytes, in which Ca2+/CaN and MAPK pathways play essential roles.

Abstract:75% ethanol extracts of Radix Pseudostellariae could improve spleen-deficiency and immunologic function from Yixing(Jiangsu), Tuorong (Fujian), Guangde(Anhui), Zhonggou(Shandong), Linmu(Shandong), Shanghai, Langxi(Anhui) and Xuanzhou (Anhui). They could decrease spleen-deficienly ratio, increase body weight, anus temperature, thymus and spleen indexes, and prolong swimming time in 15 degrees C water and survival time under anoxic circumstances on resperpine-induced mice. They could also inhibit delayed hypersensitivity induced by prednisolone on mice. Radix Pseudostellariae from Xuanzhou was most effective on spleen-deficiency mice.

Abstract:Our study aimed to investigate the immune-enhancing mechanism of the pentadecapeptide (RVAPEEHPVEGRYLV) from Cyclina sinensis (SCSP) in a cyclophosphamide (CTX)-induced murine model of immunosuppression. Our results showed that SCSP treatment significantly increased mouse body weight, immune organ indices, and the production of serum IL-6, IL-1β, and tumor necrosis factor (TNF)-α in CTX-treated mice. In addition, SCSP treatment enhanced the proliferation of splenic lymphocytes and peritoneal macrophages, as well as phagocytosis of the latter in a dose-dependent manner. Moreover, SCSP elevated the phosphorylation levels of p38, ERK, JNK, PI3K and Akt, and up-regulated IKKα, IKKβ, p50 NF-κB and p65 NF-κB protein levels, while down-regulating IκBα protein levels. Our results indicate that SCSP has immune-enhancing activities, and that it can activate the MAPK/NF-κB and PI3K/Akt pathways to enhance immunity in CTX-induced immunosuppressed mice.

Abstract:Biologically active peptides released by proteins are important in regulating immunity. The purpose of this study was to isolate and purify an immunologically active peptide from Hericium erinaceus (H. erinaceus) and to explore its effect on cytokine secretion and differentiation of macrophages. An active peptide with an amino acid sequence, Lys-Ser-Pro-Leu-Tyr (KSPLY) was obtained from H. erinaceus protein by ultrafiltration combined with multistage chromatography separation and identification technology. Subsequently, it was confirmed that the synthetic peptide KSPLY had a good immunomodulatory activity at a concentration of 100 μmol/L and could promote the secretion of NO, IL-1β, IL-6 and TNF-α by macrophages. The effects of KSPLY on M1 macrophages and M2 macrophages were also studied. Results showed that KSPLY inhibited the secretion of NO and IL-6 by M1 macrophages and promoted the tendency of M2 macrophages to transform to M1 macrophages. Therefore, it can be concluded that KSPLY is an effective immunomodulatory peptide that may be beneficial in cancer treatment and human health improvement.

Abstract:Polysaccharopeptide (PSP) is well known for its immunoregulatory effects. In the present study, the effect of PSP on white blood cell (WBC) count, T lymphocyte subsets, B lymphocytes, Th1/Th2 balance and negative immune regulators was investigated using an immunosuppressed mouse model. The results demonstrated that the WBC count and the absolute number of CD3(+)CD4(+) T cells, CD3(+)CD8(+) T cells and CD3(-)CD19(+) B cells in the peripheral blood were increased in PSP-treated groups as compared with the cyclophosphamide (Cy) group. In addition, PSP reduced interleukin (IL)-4 and GATA binding protein 3 (GATA-3) mRNA relative expression levels and elevated the ratios of IL-2/IL-4 and the transcription factors, T-box-containing protein/GATA-3. The relative mRNA expression levels of the forkhead/winged-helix transcription factor box protein 3 (Foxp3), programmed death-1 (PD-1) and IL-10 were also downregulated by PSP. These observations indicate that the immunoregulatory effects of PSP are associated with restoration of WBC number, improving the absolute number of T lymphocyte subsets and B lymphocytes, inducing the Th1/Th2 response and downregulating the negative immune regulators, Foxp3, PD-1 and IL-10.
The Structural Characterization and Immunomodulatory Activity of Polysaccharides from Pleurotus abieticola Fruiting Bodies

Abstract:Polysaccharides obtained from mushrooms have been reported to possess immunomodulatory properties. In this study, a water-soluble polysaccharide was purified from the fruiting bodies of Pleurotus abieticola, entitled PAPS1. After its composition and structural analysis, the immunomodulatory activity was investigated in immunosuppressed mice induced by cyclophosphamide (CTX) at a dosage of 70 mg/kg by intraperitoneal injection for 7 days. After 28 days of intragastric administration, PAPS1 alleviated cyclophosphamide (CTX)-induced histopathological damage and increased the expressions of splenic CD4, CD8, CD56 and IgM in the serums of immunosuppressed mice. PAPS1 suppressed the oxidative stress indicated by preventing the increases in ROS and MDA levels. According to the intestinal microflora analysis, PAPS1 regulated 11 bacteria at the gene level, including Helicobacter and Paraprevotella, which are related to immunity and oxidative capacity. Compared with CTX-treated mice, significant increases in immune-related cytokines, such as interleukin (IL)-2, IL-6 and IL-12 in the serums of mice treated with PAPS1, were observed. Finally, PAPS1 can strongly increase the expression of nuclear factor erythroid 2-related factor 2 (Nrf2) and its downstream proteins. In conclusion, PAPS1-boosted immunity may be related to its suppression on oxidative stress via enhancing the activity of Nrf2 signaling. Thus, PAPS1 can be investigated as a candidate for immunomodulatory therapy.

Abstract:Bletilla striata (Thunb.) Reichb.f., is a traditional Chinese medicine, and the Bletilla striata polysaccharide (BSP) is one of the principal components extracted from Bletilla striata with various biological activities. Previous studies have shown that many natural polysaccharides have significant immunomodulatory activities. However, as a plant polysaccharide, the research of BSP on immunomodulatory activities is limited. In this study, we aim to investigate the immunomodulatory effect of BSP in vivo and further explore its underlying mechanism in vitro. In vivo, a cyclophosphamide (CTX)-induced immunosuppression mice mode was established by intraperitoneal injection of CTX, and the immune-enhancing effect of BSP (25, 50 and 100mg/kg) on immunosuppressed mice were evaluated. The result indicated that BSP could significantly improve the immune organ index and the content of immunoglobulin, TNF-alpha and IL-4 in serum. It was also found that BSP could clearly ameliorate the spleen damage induced by CTX. Meanwhile, the result showed that BSP could not only improve the proliferation of splenocytes, but also activate the lactate dehydrogenase (LDH) and acid phosphatase (ACP) in mouse spleen tissue. In vitro, potential mechanism was further revealed in macrophages. The result supported that BSP could activate macrophages with high phagocytic ability, and induce macrophages to secrete cytokines. Finally, it revealed that activation of NF-kappa B and MAPK signalling pathway should be the underlying mechanism of the immunoenhancment of BSP.
Immunomodulatory effects of Nigella sativa seed polysaccharides by gut microbial and proteomic technologies

Abstract:Cyclophosphamide (CTX) was used to establish the immunosuppressive mice model. The immune organ viscera index, phagocytes vitality, the levels of cytokines in serum, the oxidative stress resistance, proteomics and intestinal flora in mice were investigated to evaluate the effect of immune regulation of Nigella sativa seed polysaccharide (NSSP). The results showed that the high-dose NSSP group could significantly increase the thymus and spleen index. The levels of ACP, LDH, T-AOC, SOD, IL-2, IL-4 and IL-6 were significantly increased and the levels of TNF-α and MDA were reduced. All evidences indicated that NSSP could improve the immune effects of the immunosuppressed mice. Proteomics investigation showed that NSSP could improve the immune by regulating the differential proteins of PI3K and PTEN, and regulating the metabolism-related pathways such as autoimmune diseases and PI3K-Akt signaling pathway. of Gut microbes analysis showed that NSSP could exert immunomodulatory effects by improving the structure of the intestinal flora, increasing the diversity of the flora, and regulating metabolic pathways such as lipid metabolism, polysaccharide synthesis and signal transduction by the prediction of flora metabolic functions. In addition, NSSP could regulate intestinal environment by regulating the content of short chain fatty acids.

Abstract:Ethnopharmacological relevance: Polygonatum sibiricum, known as "Huangjing" in Chinese traditional medicine, of which functions include invigorating Qi and nourishing Yin, tonifying spleen and kidney, which are considered to replenish energy, and strengthen immunity. However, both the active components and mechanism of the immune-enhancing effect of Polygonatum sibiricum have not been clarified. Aim of the study: To evaluate the immunoregulation effects of PSE30 (Polygonatum sibiricum ethanol 30) and PSE75 (Polygonatum sibiricum ethanol 75). The gut microbial and activation of RAW264.7 cells were also evaluated for exploring the mechanism of PSE75. Materials and methods: Female ICR mice were randomly divided into different groups, which were pretreated with 0.9% saline, Yupingfeng granules, different dosage of PSE30 or PSE75. And the immunosuppressed mice model was constructed using cyclophosphamide. And the total duration of the experiment was 15 d. After that, the serum Immunoglobulins G (IgG) and Immunoglobulins M (IgM) antibody, regular blood testing, assessment of natural killer cell activity, and histological observation of spleen in immunosuppressed mice were measured to evaluate the immunoregulation effects of PSE30 and PSE75. Besides, effects of PSE75 on gut microbial were evaluated using 16s rRNA sequence. And the mRNA expression and cytokine secretion of RAW264.7 cell were evaluated to analyze the immunoregulation mechanism of PSE75. Results: The content of serum IgG, IgM was significantly elevated by PSE75 (P＜0.05, P＜0.001). The NK cells killing activity in splenocytes against K562 cells induced by PSE30, and PSE75 was pronounced higher than that of the model group (P < 0.05). Both mRNA expression of Th1 molecular markers including interleukin (IL)-2, interferon (IFN)-γ, and signal transducers and activators of transcription (STAT) 4, and Th2 molecular markers including IL-4 in splenocytes were markedly enhanced by PSE30, and PSE75 (P < 0.05, P < 0.01, or P < 0.001). Besides, the result of 16s rRNA sequence indicated that PSE75 could recover the gut microbial community disturbed by cyclophosphamide. PSE75 could markedly promote the secretion of IL-6, IL-10, and IL-12 p40 from RAW264.7 cell (P＜0.01, or P＜0.001). Conclusions: PSE75 was proved to be a more promising immunomodulation agent, of which may enhance the immunity of immunosuppressed mice by improving gut microbial and activating macrophages. And PSE75 could be developed as a good immune booster in the future.

Abstract:Neuropeptide S (NPS), a newly identified neuropeptide, is involved in many physiological and pathological activities through the NPS receptor (NPSR). Recently, the NPS and NPSR have been detected in peripheral systems of pigs including immune tissues, suggesting that NPS may play an important role in the regulation of immune function. The aim of this study was to demonstrate the presence and function of NPS and NPSR in splenic lymphocytes (SPLs) and pulmonary alveolar macrophages (PAMs) of pigs. By RT-PCR, the expression of NPS and NPSR mRNA was detected in the SPLs and PAMs. NPS immunoreactivity was observed in the membrane and cytoplasm of both SPLs and PAMs. We found that NPS could stimulate the proliferation of SPLs, when NPS was added at concentrations of 0.01, 0.1, 1, 10, 100 and 1000 nM alone or in combination with PHA/LPS in vitro. In macrophages from bronchoalveolar lavage (BAL) fluid of pigs, various doses of NPS (0.01, 0.1, 1, 10, 100 and 1000 nM) up-regulated the phagocytosis of PAMs in comparison to controls. In PAMs, NPS could induce the production of the pro-inflammatory cytokines IL-1β, IL-6 and TNF-α. Taken together, all data suggest that NPS is capable of inducing phagocytosis of non-opsonized E. coli. NPS might act as potent neuroimmunomodulatory factors and affects the maintenance of immune homeostasis.
