Abstract:To tackle the devastating microbial infections for the public health, a continuous search for effective and safe nanobiocides based on their prominent nanoscale effects has been extensively explored during past decades. In this study, a green electrolysis method was employed to synthesize silver-doped molybdenum sulfide (Ag@MoS2) composite materials. The obtained nanocomposites exhibited a sheet-like structure with a large specific surface area, which contributed to the efficient loading and refined distribution of silver nanoparticles. G(-)E. coli and G(+)S. aureus were used as model bacteria for the antibacterial test, which revealed enhanced antibacterial activity of produced nanocomposites with an identified destructive effect on preformed biofilms. It was found that within 72 hour incubation, 20 mu g mL(-1) Ag@MoS2 was sufficient to inhibit the growth of E. coli and S. aureus without visible colony formation, pointing to a desirable long-term antibacterial activity. Further a mechanistic antibiosis study of Ag@MoS2 indicated the involvement of a generation of reactive oxygen species. Notably, owing to the well-distributed silver nanoparticles on the nontoxic MoS2 nanosheet, the cytotoxicity evaluation results revealed that produced nanocomposites exhibited negligible toxicity to mammalian cells, and thereby held promising potential for biomedical applications.

Abstract:Bacterial infections have evolved as a life-threatening problem afflicting people due to the abuse of antibiotics and emergence of drug-resistant bacteria. Thus developing novel antibacterial materials is an urgent need. Herein, chitosan-MoS2 (CS-MoS2) nanocomposite was synthesized through thiol ligand functionalization and chemical modification to achieve more efficient bactericidal activity. CS-MoS2 exhibited synergistic remarkable bactericidal capability against Gram-positive Staphylococcus aureus (S. aureus) and Gram-negative Escherichia coli (E.coli), where bacterial viabilities were significantly reduced. After treatment with 10 μg/mL of CS-MoS2, 100% and 98.1% of S. aureus and E.coli cells were killed respectively. A mechanism study revealed that the positively charged CS-MoS2 could interact with cell membrane, then cause damage to the membrane and cellular constituents by generation of reactive oxygen species (ROS) and glutathione (GSH) oxidation, finally inhibit bacterial growth. The CS-MoS2 could be an attractive antibacterial agent with improved efficiency and provide more strategies for application of MoS2.

Abstract:In this study, catechol-conjugated chitosan (CSS) was synthesized to prepare silver nanoparticles in aqueous solution as both a reducing and stabilizing agent. The entire reaction process complied with the principles of green chemistry. The stability and physicochemical properties of CSS-coated silver nanoparticles (CSS–Ag NPs) were well characterized by a series of techniques. A narrow diameter distribution and excellent stability were observed in the target CSS–Ag NPs. Determination of the minimum inhibitory concentration (MIC) and the disk diffusion test were applied to evaluate the antibacterial activities with respect to quantity and quality, respectively. The cytotoxic effects on HepG2 cells were also evaluated with a series of assays. The resulting CSS–Ag NPs combined the unique properties of chitosan and silver nanoparticles, showing effective antibacterial activities and low cytotoxicity. The effective toxic concentration of CSS–Ag NPs towards E. coli and S. aureus is too low to damage HepG2 cells. The results demonstrated that CSS–Ag NPs could be a potential candidate for use in biological and pharmaceutical areas to prevent infections caused by microorganisms.
Facile Synthesis, Antibacterial Mechanisms and Cytocompatibility of Ag–MnFe2O4 Magnetic Nanoparticles

Abstract:Magnetic MnFe2O4 nanoparticles containing 0, 1 and 3 at% silver, respectively were synthesized by one-pot sol-gel method for antibacterial applications in biomedical fields. Material characterizations indicate that MnFe2O4 begins crystallization at 134 degrees C and oxidation at 450 degrees C, the grain size and agglomeration degree increase with the silver content and silver exists as metallic state for the particles. The saturation magnetization decreases with the sintering temperature and slightly increases with the silver content, with the maximum of 50.0 emu/g obtained. Antibacterial tests by plate counting and PI-Hoechst 33342 staining suggest that the antibacterial activity of Ag-MnFe2O4 nanoparticles is silver content-dependent. Silver ions concentration measurement, beta-galactosidase activity assay and transmission electron microscopic observation show that the antibacterial activity is dominated by the actions of the released silver ions, rather than the membrane cell impairment or reactive oxygen species-induced oxidative stress mechanism. MC3T3-E1 cell test demonstrates the best cytocompatibility of the nanoparticles with 3 at.% silver, which is likely related to the reduced cell endocytosis of the aggregated particles. The combination of magnetism, antibacterial activity and biocompatibility would make Ag-MnFe(2)O(4 )nanoparticles a potential multi-functional material in various biomedical applications.

Abstract:In this paper, a simple method has been used to synthesize silver-molybdenum disulfide nanoparticles (Ag-MoS2NPs) with photothermal and physical inhibition of bacteria. The MoS2nanocarriers were obtained by an environment-friendly liquid-phase separation method. The coated Ag NPs with high antibacterial activity and fluorescence property endowed the synthesized Ag-MoS2NPs with the ability to be traced. The Ag-MoS2NPs had photothermal properties with a maximum photothermal increase of [Formula: see text]C. The results of the dual photothermal–chemical antibacterial activity using Ag-MoS2NP carriers showed that Ag-MoS2NPs had broad-spectrum inhibition activity with MIC values up to [Formula: see text][Formula: see text][Formula: see text]g/mL against Escherichia coli. The Ag-MoS2NPs were also used to destroy cell integrity by disrupting cell membranes and cell walls to achieve the inhibition of cell survival. The toxicity test in mice demonstrated the nontoxicity of Ag-MoS2NPs. Ag-MoS2NPs were studied for dual photothermal–chemical antibacterial activity, and the material reduced the amount of Ag NPs used, resulting in lower biotoxicity, hence providing a possible avenue for the clinical application of Ag NPs.

Abstract:In this work, we demonstrate a convenient, efficient, and environmentally benign strategy to achieving antimicrobial and antiadhesive purposes using a silver-zwitterion nanocomposite. The synthesis of the nanocomposite relies on loading zwitterionic polymer brushes with Ag+ precursor ions, followed by their in situ reduction to Ag nanoparticle by ultraviolet (UV) irradiation. Both poly(sulfobetaine methacrylate) (pSBMA) and poly(carboxybetaine methacrylate) (pCBMA) have been studied as matrices for the embedding of silver. Well-dispersed silver nanoparticles are embedded into pCBMA matrices. The obtained pCBMA-silver hybrid (CB-Ag) is capable of killing bacteria upon contact and releasing dead bacteria under wet conditions. Results suggest the feasibility of using this nanocomposite system as a robust and reliable antimicrobial and antiadhesive platform for the prevention of microbial colonization.

Abstract:Silver nanoparticles (AgNPs) have been used as a broad-spectrum antimicrobial agent, whose toxicity originates from the localized release of Ag+ ions. However, the residual AgNPs core could generate potential risk to humans and waste of noble metals. Herein, we infused the cysteine-modified molybdenum disulfide with minimum Ag+ ions and coated with a layer of cationic polyelectrolyte to construct an efficient and benign antimicrobial depot. The system exhibited much enhanced broad-spectrum antibacterial activity compared with an equivalent amount of silver nitrate, owing to its increasing accessibility of released Ag+ to the cell walls of microorganisms. More importantly, the antibacterial system could be successfully applied to treat wound infection, while retaining high antibacterial activities, exhibiting negligible biotoxicity and avoiding the waste of Ag.

Abstract:In this study, silver nanoparticles (Ag NPs) coated with catechol-conjugated chitosan (CSS) were prepared using green methods. Interestingly, we uncovered that CSS-coated Ag NPs (CSS-Ag NPs) exhibited a higher toxicity against gram-negative Escherichia coli (E. coli) bacteria than against grampositive Staphylococcus aureus (S. aureus) bacteria. The differences revealed that the CSS-Ag NPs killed gram bacteria with distinct, species-specific mechanisms. The aim of this study is to further investigate these underlying mechanisms through a series of analyses. The ultrastructure and morphology of the bacteria before and after treatment with CSS-Ag NPs were observed. The results demonstrated the CSS-Ag NPs killed gram-positive bacteria through a disorganization of the cell wall and leakage of cytoplasmic content. In contrast, the primary mechanism of action on gram-negative bacteria was a change in membrane permeability, induced by adsorption of CSS-Ag NPs. The species-specific mechanisms are caused by structural differences in the cell walls of gram bacteria. Gram-positive bacteria are protected from CSS-Ag NPs by a thicker cell wall, while gram-negatives are more easily killed due to an interaction between a special outer membrane and the nanoparticles. Our study offers an in-depth understanding of the antibacterial behaviors of CSS-Ag NPs and provides insights into ultimately optimizing the design of Ag NPs for treatment of bacterial infections.
Preferential Facet of Nanocrystalline Silver Embedded in Polyethylene Oxide Nanocomposite and Its Antibiotic Behaviors

Abstract:In this article, a magnetron sputtering (MS) method is reported to prepare a nanocomposite of embedded silver nanoparticles in simultaneously polymerized, nonfouling polyethylene oxide (PEO) film. X-ray diffraction (XRD) shows that the silver nanoparticles had tunable growth in the preferential (111) facet, and transmission electron microscopy (TEM) reveals that diameters of silver could also be controllable in the range of 5-10 nm by varying the processing parameters, such as the working pressure and the target-to-substrate distance. The antibacterial activities tests demonstrate that nanocrystalline silver in the Ag/PEO composite fabricated by this method has a markedly efficient effect.

Abstract:There is a significant need for magnetite-silver nanocomposites that exhibit durable and recyclable antimicrobial activity. In this study, magnetic iron oxide nanoparticles (Fe3O4 NPs) coated with ethylenediamine-modified chitosan/polyacrylic acid copolymeric layer (Fe3O4@ECS/PAA) were fabricated. Subsequently, directly deposited silver (Ag) NPs procedure was carried out to form the antibacterial heterodimers of Fe3O4@ECS/PAA-Ag NPs. The composition and morphology of the resultant nanostructures were confirmed by FT-IR, XRD, TEM and TGA. The overall length of the heterodimers was approximately 45 nm, in which the mean diameter of Fe3O4@ECS/PAA NPs reached up to 35 nm, and that of Ag NPs was around 15 nm. The mass fraction of silver NPs in the nanocomposites was about 63.1%. The obtained Fe3O4@ECS/PAA NPs exhibited good colloidal stability, and excellent response to additional magnetic field, making the NPs easy to recover after antibacterial tests. In particular, the Fe3O4@ECS/PAA-Ag NPs retained nearly 100% biocidal efficiency (106–107 CFU/mg nanoparticles) for both Gram-negative bacteria E. coli and Gram-positive bacteria S. aureus throughout ten cycles without washing with any solvents or water, exhibiting potent and durable antibacterial activity.

Abstract:Bacterial infections have posed a large threat on human health. It is necessary to look for various strategies for inhibiting the growth of pathogenic bacteria. Silver-based nanomaterials have become good candidate for designing antibacterial agents. Different from the synthesis of silver nanoparticles (AgNPs) using additional ligand and reducing agent, our simpler strategy constructs carbon-silver nanocomposites (C-Ag NCs) using polysaccharide-derived CDs as reductant and stabilizer. The facile C-Ag NCs possess superior antibacterial activity with MBC (minimum bactericidal concentration) of 1.3 μg/mL Ag after incubation for 6 h. The designed C-Ag NCs with MBC of 1.3 μg/mL Ag can kill a wide range of bacteria (including Staphylococcus aureus, Listeria monocytogenes, Escherichia coli, Salmonella typhimurium, Vibrio parahemolyticus, and Shigella castellani). We believe that the simple hybrid C-Ag NCs provide alternative strategies for design of antimicrobial agents in biomedicine and food industry.

High-Efficiency Photodynamic Antibacterial Activity of NH2-MIL-101(Fe)@MoS2/ZnO Ternary Composites

Abstract:Bacterial infections are a serious threat to human health, and the development of effective antibacterial agents represents a critical solution. In this study, NH2-MIL-101(Fe)@MoS2/ZnO ternary nanocomposites are successfully prepared by a facile wet-chemistry procedure, where MoS2 nanosheets are grown onto the MIL-101 scaffold forming a flower-like morphology with ZnO nanoparticles deposited onto the surface. The ternary composites exhibit a remarkable sterilization performance under visible light irradiation toward both Gram-negative and Gram-positive bacteria, eliminating 98.6% of Escherichia coli and 90% of Staphylococcus aureus after exposure to visible light for 30 min, a performance markedly better than that with NH2-MIL-101(Fe)@MoS2 binary composites and even more so than MoS2 nanosheets alone. This is ascribed to the unique electronic band structure of the composites, where the separation of the photogenerated carriers is likely facilitated by the S-scheme mechanism in the NH2-MIL-101(Fe)@MoS2 binary composites and further enhanced by the formation of a p-n heterojunction between MoS2 and ZnO in the ternary composites. This interfacial charge transfer boosts the effective production of superoxide radicals by the reduction of oxygen, and the disproportionation reaction with water leads to the formation of hydroxy radicals, as attested in spectroscopic and microscopic measurements. Results from this study highlight the significance of structural engineering of nanocomposites in the manipulation of the electronic band structure and hence the photodynamic activity.
Facile synthesis of silver doped manganese oxide nanocomposite with superior photocatalytic and antimicrobial activity under visible spectrum

Abstract:Water pollution and antimicrobial resistance (AMR) have become two global threats; 80% of diseases and 50% of child deaths are due to poor water quality. In this study, hydrothermal processing was employed to manufacture manganese oxide nanorods. Silver dopant was deposited on the surface of manganese oxide. XRD diffractogram confirmed the facile synthesis of Ag/Mn2O3 nanocomposite. XPS survey analysis demonstrated silver content of 9.43 atom %. Photocatalytic measurements demonstrated the outstanding efficiency of the Ag-Mn2O3 compared to virgin oxide particles under visible radiation. Degradation efficiencies Mn2O3 and Ag/Mn2O3 on methyl orange (MO) dye was found to be 53% and 85% under visible spectrum. Silver dopant was found to decrease the binding energy of valence electrons; this action could support electron-hole pair generation under visible spectrum and could promote catalytic performance. Ag/Mn2O3 NPs demonstrated most effective performance (95% removal efficiency) at pH 3; this could be ascribed to the electrostatic attraction between positively charged catalyst and the negatively charged MO. Ag/Mn2O3 demonstrated enhanced antibacterial activity against Gram-positive Staphylococcus aureus (S. aureus) (19 mm ZOI), and Gram-negative Escherichia coli (E. coli) (22 mm ZOI) respectively; the developed nanocomposite demonstrated advanced anti-film activity with inhibition percentage of 95.5% against E. coli followed by 89.5% against S. aureus.

Abstract:Porous Fe3O4 shell/silver core nanocomposites featuring sustainable and recyclable antibacterial activity have been successfully prepared via a facile one-pot hydrothermal method. The unique structural feature of the Ag@Fe3O4 nanocomposites with Ag embedded in porous Fe3O4 shell endows them with the ability of sustained-release of silver ions. Their antimicrobial activity studies were investigated on both Gram negative Escherichia coli and Gram positive Bacillus subtilis, which demonstrate that the nanocomposites are highly toxic to microorganisms and exhibit sustainable antibacterial activity. Besides, the Ag@Fe3O4 nanocomposites can be separated easily from the medium by a small magnet, which provided an effective way to eliminate the residual nanosilver from the surroundings. We finally demonstrate that the recovered nanocomposites exhibit recyclable antibacterial activity, acting as an ideal long-acting antibacterial agent.

Abstract:The mounting threat of antibiotic-resistant bacterial infections has made it imperative to develop innovative antibacterial strategies. Here we propose a novel antibacterial nanoplatform of silver nanoparticles-decorated and mesoporous silica coated single-walled carbon nanotubes constructed via a N-[3-(trimethoxysilyl)propyl]ethylene diamine (TSD)-mediated method (SWCNTs@mSiO 2 -TSD@Ag). In this system, the outer mesoporous silica shells are able to improve the dispersibility of SWCNTs, which will increase their contact area with bacteria cell walls. Meanwhile, the large number of mesopores in silica layers act as microreactors for in situ synthesis of Ag NPs with controlled small size and uniform distribution, which induces an enhanced antibacterial activity. Compared with TSD modified mesoporous silica coated single-walled carbon nanotubes (SWCNTs@mSiO 2 -TSD) and commercial Ag NPs, this combination nanosystem of SWCNTs@mSiO 2 -TSD@Ag exhibits much stronger antibacterial performance against multi-drug-resistant bacteria Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) in vitro through damaging the bacterial cell membranes and a fast release of silver ions. Furthermore, the in vivo rat skin infection model verifies that SWCNTs@mSiO 2 -TSD@Ag have remarkably improved abilities of bacterial clearance, wound healing promoting as well as outstanding biocompatibility. Therefore, this novel nanoplatform indicates promising potentials as a safe and powerful tool for the treatment of clinical drug-resistant infections.

Abstract:Bacterial infection is a serious public health issue because this may cause bacterial pneumonia and bacteria-infected sepsis. The conventional antibiotic therapy can cause bacterial resistance and other adverse effects. Herein, we designed a novel photoresponsive hybrid, in which MoS2 nanosheets were doped with copper ions (MoS2@Cu2+). This system can effectively kill bacteria under 660 nm visible light irradiation due to enhanced photocatalytic and photothermal performances. The underlying mechanism is that when doping copper ions in MoS2, the photogenerated electrons can be rapidly transferred from the conduction band of MoS2 to copper ions, consequently reducing the electron-hole recombination, thereby enhancing the yield of radical oxygen species (ROS). Besides the intrinsic photothermal performance of MoS2, the Cu2+ ion can absorb photons and transfer the photoenergy into heat due to the d-d transition of electrons, resulting in enhanced photothermal properties of MoS2@Cu2+ compared with pristine MoS2. Therefore, owing to the synergistic action of hyperthermia, ROS and released Cu2+, this system exhibited highly effective antibacterial efficacy when exposed to 660 nm light for 20 min, i.e., 99.64% against Staphylococcus aureus. The cytotoxicity test showed that the synthesized MoS2@Cu2+ had good biocompatibility. This system will be a potential platform for environmental disinfection, healing of bacteria-infected wounds and other rapid bacteria-killing fields.

Abstract:Uniform and water-soluble Ag@reduced graphene oxide (Ag@rGO) nanocomposites can be prepared by a facile approach in the absence of additional reductants, which display much better antibacterial properties than that of pure silver nanoparticles synthesized by microwave irradiation, and an equivalent antibacterial effect in comparison with that of the general antibacterial drug ampicillin. Their skin irritation tests with the use of rat models are taken in order to explore the toxicity of this nanocomposite, which confirm that no oedema or erythema appears on the injured rat skin after exposure to the as-prepared Ag@rGO nanocomposites.
