Abstract:Nonspherical particles can uniquely probe soft system dynamics. We show that laser tweezers stably trap thin coinlike microdisks in 3D with an edge-on orientation. Scattered light forms a streak that we track using a fast camera to measure the disk's angular displacement. Linearly polarized tweezers rotationally trap a birefringent disk, and we measure its harmonically bound Brownian rotation over 5 decades in time. Near a surface, the disk exhibits a translational-orientational switchback oscillation. Circularly polarized tweezers rotate the disk and streak, yielding a colloidal lighthouse.

Abstract:Cellular functions are enabled by cascades of transient biological events. Imaging and tracking the dynamics of these events have proven to be a powerful means of understanding the principles of cellular processes. These studies have typically focused on translational dynamics. By contrast, investigations of rotational dynamics have been scarce, despite emerging evidence that rotational dynamics are an inherent feature of many cellular processes and may also provide valuable clues to understanding those cell functions. Such studies have been impeded by the limited availability of suitable rotational imaging probes. This has recently changed thanks to the advances in the development of anisotropic particles for rotational imaging. In this review, we will summarize current techniques for imaging rotation using particle probes that are anisotropic in shape or optical properties. We will highlight two studies that demonstrate how these techniques can be applied to explore important facets of cellular functions.

Abstract:Single-particle tracking is a powerful tool for understanding protein dynamics and characterizing microenvironments. As the motion of unconstrained nanoscale particles is governed by Brownian diffusion, deviations from this behavior are biophysically insightful. However, the stochastic nature of particle movement and the presence of localization error pose a challenge for the robust classification of non-Brownian motion. Here, we present , a versatile tool for classifying track behaviors and extracting key parameters for particles undergoing Brownian, confined, or directed motion. Our tool quickly and accurately estimates motion parameters from individual tracks and determines their likely motion state. Further, our tool can analyze populations of tracks and determine the most likely number of motion states. We determine the working range of our approach on simulated tracks and demonstrate its application for characterizing particle motion in cells and for biosensing applications. Our tool is implemented as a stand-alone software package, making it simple to analyze tracking data.

Abstract:We employ a novel framework for information-optimal microscopy to design a family of point spread functions (PSFs), the Tetrapod PSFs, which enable high-precision localization of nanoscale emitters in three dimensions over customizable axial (z) ranges of up to 20 μm with a high numerical aperture objective lens. To illustrate, we perform flow profiling in a microfluidic channel and show scan-free tracking of single quantum-dot-labeled phospholipid molecules on the surface of living, thick mammalian cells.

Abstract:Objective-type total internal reflection fluorescence technique and Gaussian fit single particle tracking algorithm have been used to study the constrained Brownian motion characteristics of single fluorescent microspheres of 40 nm diameter in real-time. Three-dimensional tracking has been realized from the two-dimensional images by utilizing the exponential decay of the fluorescence intensity excited by the evanescent field. Trajectory analyses have shown that single fluorescent beads diffused in a constrained Brownian mode with a diffusion coefficient a little smaller than theoretical estimates in the plane-parallel direction.

Abstract:Correlated translation-orientation tracking of single particles can provide important information for understanding the dynamics of live systems and their interaction with the probes. However, full six-dimensional (6D) motion tracking has yet to be achieved. Here, we developed synchronized 3D translation and 3D rotation tracking of single diamond particles based on nitrogen-vacancy center sensing. We first performed 6D tracking of diamond particles attached to a giant plasma membrane vesicle to demonstrate the method. Quantitative analysis of diamond particles' motion allowed elimination of the geometric effect and revealed the net rotation on the vesicle. 6D tracking was then applied to measure live cell dynamics. Motion characteristics of nanodiamonds on cell membranes under various controlled physiological conditions suggest that the nanodiamonds' rotation is associated with cell metabolic activities. Our technique extends the toolbox of single particle tracking and provides a unique solution to problems where correlated analysis of translation and rotation is critical.

Abstract:Three-dimensional (3D) tracking of surface-tethered single particles reveals the dynamics of the molecular tether. However, most 3D tracking techniques lack precision, especially in the axial direction, for measuring the dynamics of biomolecules with a spatial scale of several nanometers. Here, we present a plasmonic imaging technique that can track the motion of similar to 100 tethered particles in 3D simultaneously with sub-nanometer axial precision and single-digit nanometer lateral precision at millisecond time resolution. By tracking the 3D coordinates of a tethered particle with high spatial resolution, we are able to determine the dynamics of single short DNA and study its interaction with enzymes. We further show that the particle motion pattern can be used to identify specific and nonspecific interactions in immunoassays. We anticipate that our 3D tracking technique can contribute to the understanding of molecular dynamics and interactions at the single-molecule level.

Abstract:In real time and space, four-dimensional electron microscopy (4D EM) has enabled observation of transient structures and morphologies of inorganic and organic materials. We have extended 4D EM to include liquid cells without the time resolution being limited by the response of the detector. Our approach permits the imaging of the motion and morphological dynamics of a single, same particle on nanometer and ultrashort time scales. As a first application, we studied the rotational dynamics of gold nanoparticles in aqueous solution. A full transition from the conventional diffusive rotation to superdiffusive rotation and further to a ballistic rotation was observed with increasing asymmetry of the nanoparticle morphology. We explored the underlying physics both experimentally and theoretically according to the morphological asymmetry of the nanoparticles.

Abstract:Here we demonstrated a simple yet powerful method, planar illumination microscopy, to directly track the rotational and translational diffusion dynamics of individual anisotropic nanoparticles in solution and living cells. By illuminating gold nanorods (GNRs) with two orthogonal sheets of light and resolving the polarized scattering signal with a birefringent crystal, we readily achieved three-dimensional angular resolving capability for single GNRs in noisy surroundings. The rotational dynamics of individual GNRs dispersed in glycerol/water mixtures with different chemical modification were tracked, and the measured rotational diffusion coefficient was well fitted to a previously reported theoretical model (Torre, J. G. d. l.; Martinez, M. C. L. Macromolecules 1987, 20, 661-666; Tirado, M. M.; Torre, J. G. d. l. J. Chem. Phys. 1980, 73, 1986-1993). In addition, the translational and rotational movements of individual GNRs transported by kinesin motor protein on microtubules inside living cells were directly imaged. Compared to its motion in free solution, a GNR attached to motor-protein did not rotate significantly while moving forward. Our method can be further generalized to allow determination of three-dimensional orientation of single dipoles using many different illumination modes.

Abstract:Real time visualization and tracking of colloidal particles with 3D resolution is essential for probing the local structure and dynamics in complex fluids. Although tracking translational motion of spherical colloids is well-known, accessing rotational dynamics of such particles remains a great challenge. Here, we report a novel approach of using fluorescently labeled raspberry-like colloids with an optical anisotropy to concurrently track translational and rotational dynamics in 3 dimensions. The raspberry-like particles are coated by a silica layer of adjustable thickness, which allows tuning the surface roughness. The synthesis and applicability of the proposed method is demonstrated by two types of probes: rough and smoothened. The accuracy of measuring Mean Squared (Angular) Displacements are also demonstrated by using these 2 probes dispersed in 2 different solvents. The presented 3D trackable colloids offer a high potential for wide range of applications and studies, such as probing crystallization dynamics, phase transitions and the effect of surface roughness on diffusion.

Abstract:Monitoring chemical reactions in solutions at the scale of individual entities is challenging: single particle detection requires small confocal volumes which are hardly compatible with Brownian motion, particularly when long integration times are necessary. Here, we propose a real-time (10 Hz) holography-based nm-precision 3D tracking of single moving nanoparticles. Using this localization, the confocal collection volume is dynamically adjusted to follow the moving nanoparticle and allow continuous spectroscopic monitoring. This concept is applied to the study galvanic exchange in freely-moving collo{ï}dal silver nanoparticles with gold ions generated in-situ. While the Brownian trajectory reveals particle size, spectral shifts dynamically reveal composition changes and transformation kinetics at the single object level, pointing at different transformation kinetics for free and tethered particles.

Abstract:A panoply of new tools for tracking single particles and molecules has led to an explosion of experimental data, leading to novel insights into physical properties of living matter governing cellular development and function, health and disease. In this Perspective, we present tools to investigate the dynamics and mechanics of living systems from the molecular to cellular scale via single-particle techniques. In particular, we focus on methods to measure, interpret, and analyse complex data sets that are associated with forces, materials properties, transport, and emergent organisation phenomena within biological and soft-matter systems. Current approaches, challenges, and existing solutions in the associated fields are outlined in order to support the growing community of researchers at the interface of physics and the life sciences. Each section focuses not only on the general physical principles and the potential for understanding living matter, but also on details of practical data extraction and analysis, discussing limitations, interpretation, and comparison across different experimental realisations and theoretical frameworks. Particularly relevant results are introduced as examples. While this Perspective describes living matter from a physical perspective, highlighting experimental and theoretical physics techniques relevant for such systems, it is also meant to serve as a solid starting point for researchers in the life sciences interested in the implementation of biophysical methods.
Reveal Heterogeneous Motion States in Single Nanoparticle Trajectory Using Its Own History

Abstract:Single particle tracking (SPT) has long been utilized for investigation of complex system dynamics such as nanoparticle-cell interaction, however, the analysis of individual particle motions is always a difficult issue. Existing methods treat each data point or fragment on the recorded trajectory as an isolated "atom" and determine their relationship based on externally predefined models or physical states, which inevitably lead to oversimplification of the associated spatiotemporal complexity. Herein, inspired by the historical analysis in social science, we propose a modeless preprocessing framework for SPT analysis based on the "history" of the particle. This new strategy consists of 3 steps: (1) assign a "history" to each data point and construct successive overlapped historical vectors; (2) perform unsupervised clustering in the vector space to find their relative differences; (3) project differences back to the trajectory by coloring each point accordingly for visualization. As a result, the inner heterogeneity of the particle motion self-emerges as a colored trajectory, exhibiting a global picture of the local state transitions and providing valuable information for further model-based analysis. Since the complexity issues at various spatiotemporal scales have attracted increasing attention, and individual objects such as single molecules, cells, vehicles and even stars in the universe could all be treated as "single particles", this presuppositionless data preprocessing approach could help the investigations of many complex systems in fundamental research.
A high order accurate space-time trajectory reconstruction technique for quantitative particle trafficking analysis

Abstract:The study of moving particles (e.g. molecules, virus, vesicles, organelles, or whole cells) is crucial to decipher a plethora of cellular mechanisms within physiological and pathological conditions. Powerful live-imaging approaches enable life scientists to capture particle movements at different scale from cells to single molecules, that are collected in a series of frames. However, although these events can be captured, an accurate quantitative analysis of live-imaging experiments still remains a challenge. Two main approaches are currently used to study particle kinematics: kymographs, which are graphical representation of spatial motion over time, and single particle tracking (SPT) followed by linear linking. Both kymograph and SPT apply a space-time approximation in quantifying particle kinematics, considering the velocity constant either over several frames or between consecutive frames, respectively. Thus, both approaches intrinsically limit the analysis of complex motions with rapid changes in velocity. Therefore, we design, implement and validate a novel reconstruction algorithm aiming at supporting tracking particle trafficking analysis with mathematical foundations. Our method is based on polynomial reconstruction of 4D (3D+time) particle trajectories, enabling to assess particle instantaneous velocity and acceleration, at any time, over the entire trajectory. Here, the new algorithm is compared to state-of-the-art SPT followed by linear linking, demonstrating an increased accuracy in quantifying particle kinematics. Our approach is directly derived from the governing equations of motion, thus it arises from physical principles and, as such, it is a versatile and reliable numerical method for accurate particle kinematics analysis which can be applied to any live-imaging experiment where the space-time coordinates can be retrieved.

Interrogating the Ballistic Regime in Liquids with Rotational Optical Tweezers

Abstract:Accessing the ballistic regime of single particles in liquids remains an experimental challenge that shrouds our understanding of the particle-liquid interactions on exceedingly short time scales. We demonstrate the ballistic measurements of rotational probes to observe these interactions in the rotational regime within microscopic systems. This study uses sensitive high-bandwidth measurements of polarisation from light scattered by orientation-locked birefringent probes trapped within rotational optical tweezers. The particle-liquid interactions in the ballistic regime are decoupled from the optical potential allowing direct studies of single-particle rotational dynamics. This enabled us to determine the dissipation of rotational inertia and observe and validate rotational hydrodynamic effects in a previously inaccessible parameter space. Furthermore, the fast angular velocity thermalisation time enables calibration-free viscometry using less than 50ms of data. This methodology will provide a unique way of studying rotational hydrodynamic effects and enable ultra-fast microrheometry in systems out-of-equilibrium.
