≪i>pmaiaa4</i> Mediates Abiotic Stress Tolerance and the Mechanism of Its Interaction with <i>pmaarf3</i> in <i>pinus Massoniana</i>

Abstract:Auxin is crucial for plants' ability to withstand abiotic stress. Aux/IAA genes are stimulated by drought and other abiotic stimuli, and some of these genes are crucial signaling elements in the auxin system in plants. However, there are few studies on the regulatory function of Aux/IAA genes in regulating the drought stress tolerance of Pinus massoniana. In this study, it was preliminarily explored that the PamIAA4 gene of P. massoniana plays a regulatory role in abiotic stress. The effects of AUX/IAA and ARF genes on Pinus massoniana were investigated under drought and hormone treatments. Gene cloning, functional testing of transgenic tobacco, and gene interaction were then carried out. Exogenous auxin irrigation significantly decreased and increased the expression of PmaIAA4 and PmaARF3 genes, respectively, in Pinus massoniana seedlings. The auxin content of transgenic tobacco with the PmaIAA4 gene was remarkably lower compared to the control group, but the proline content was increased. Therefore, overexpression of PmaIAA4 gene in Pinus massoniana can improve drought resistance. To further verify the interaction between PmaIAA4 gene and PmaARF3 gene, bioinformatics analysis and yeast two-hybrid experiments were carried out on these two genes. There were certain interactions between the PB1 structural region of PmaARF3 gene and PmaIAA4 gene. Our study provides new insights into the regulatory function of AUX/IAA and ARF genes in Pinus massoniana, which have great application prospects.
