TNNI3 Expression and Prognostic Value and Correlation with Tumor-Infiltrating Immune Cells in LIHC: A Bioinformatics Analysis

Abstract:Background: TNNI3 encodes a protein related to tumorigenesis and development, although its biological function remains unclear. There are no studies examining the relationship between TNNI3 gene expression and liver hepatocellular carcinoma (LIHC) occurrence and development. Methods: We analyzed the expression of TNNI3 through TIMER and UALCAN online debate in different types of human cancer and LIHC. Using UALCAN gene expression data, we examined the relationship between TNNI3 gene expression and clinicopathological factors. After searching the KM-Plotter database, we evaluated the association between TNNI3 gene expression and clinical outcome. TNNI3 genetic alterations were analyzed using the cBioPortal database. Subsequently, to verify the enrichment of the TNNI3 proteins and the TNNI3 genes, we used the "Express Analysis" module of Metascape, and GeneMANIA was used. For the purpose of assessing the correlation between TNNI3 and tumor immune cell infiltration, we used the TIMER website. Results: Our results highlighted that TNNI3 was differentially expressed in different tumors. Among LIHC patients, its expression is high, closely correlated with poor prognosis. LIHC expression of TNNI3 is related to the clinical stage and pathological type. The TNNI3 gene is highly expressed in LIHC, but very lowly expressed in adjacent normal tissues. Meanwhile, TNNI3 is co-expressed with many genes, and its biological function is regulated by each other . Furthermore, TNNI3 showed significant association with the expression of multiple immune markers. Conclusions: TNNI3 was more highly expressed in LIHC. Lower levels of TNNI3 suggest patients have a favorable prognosis. TNNI3 was associated with the level of immune infiltration as well. TNNI3 might contribute to immunity regulation and affect LIHC prognosis according to all research results. In short, TNNI3 might be a potential target for LIHC.
