Abstract:A convenient and facile method for Michael addition of methylene active compounds to chalcones has been developed by using 1,8-diazabicyclo[5.4.0]undec-7-ene-derived task-specific ionic liquids as reaction medium in the absence of any catalyst. This protocol could afford the Michael adducts in good to excellent yields in a short time, the workup is very simple, and the ionic liquid could be reused six times without significant loss of activity.

Abstract:A tandem Michael addition-cyclization of nitroalkenes with 1,3-dicarbonyl compounds was developed using phase transfer catalyst (PTC), allowing for the synthesis of polysubstituted-[4,5]-dihydrofuran in high yields. A wide range of substrates were demonstrated by this one-step process. Meanwhile, nitro group was substituted to form corresponding nitrite ion detected in the aqueous phase providing a reasonable pathway for denitrating poisonous and explosive nitro-containing compounds. The proposed mechanism was also supported by our DFT calculations.

Abstract:The development of Michael addition reactions to conjugated cyclopropenes is a challenge in organic synthesis due to the fleeting and reactive nature of such strained Michael acceptor systems. Herein, the development of a photochemical approach towards such conjugated cyclopropenes is reported that serves as a strategic entry point to densely functionalized cyclopropanes in a diastereoselective fashion. The process involves the light-mediated generation of transient cyclopropenyl α,β-unsaturated esters from vinyl diazo esters, followed by an organic base catalyzed nucleophilic addition of N-heterocycles to directly access β-N-heterocyclic cyclopropanoic esters. With this synergistic approach, various trisubstituted cyclopropanes bearing N-heteroaryl and N-heterocyclic rings such as indole, pyrrole, benzimidazole, isatin, pyridinone and quinolinone were accessed efficiently in good yield and decent to good diastereoselectivities. Further, β-indolyl cyclopropanoic acids have been synthesized and were successfully evaluated as FABP-4 inhibitors. Theoretical calculations have been performed to elucidate the mechanism which was further supported by experimental findings.

Abstract:A highly stereoselective tandem Michael addition-Wittig reaction of (3-carboxy-2-oxopropylidene)triphenylphosphorane and alpha,beta-unsaturated aldehydes has been developed by employing the combined catalysis of a newly designed bulky chiral secondary amine 1g, LiClO(4), and DABCO. The multifunctional 6-carboxycyclohex-2-en-1-ones were generally obtained in excellent diastereo- and enantioselectivities (dr up to >50:1, 86-99% ee).

Abstract:A novel and efficient copper-catalyzed Michael addition of acrylic derivatives with primary alcohols in the presence of base has been developed. The protocol uses readily available acrylic derivatives and primary alcohols as the starting materials, inexpensive CuCl2 as the catalyst, and the corresponding addition products were obtained in moderate to excellent yields.

Abstract:N-acylation of l-prolinol (2) with enoyl chloride provided l-cinnamoylprolinol (3a) and l-crotonylprolinol (3b), respectively. The attachment of (3a) or (3b) to Merrifield resin and the utilization of such a system for asymmetric Michael additions of Grignard reagents to α,β-unsaturated carbonyl compounds were studied. The stereoselectivities have been increased compared with those observed for the same reactions in solution. On the other hand, it has been proved by the FT-IR difference spectrum of the Merrifield resin and polymer-supported chiral auxiliary that l-prolinol derivatives have been coupled to the resin.

Abstract:Ferric chloride hexahydrate was shown to be an efficient catalyst for the conjugate addition of 2,3-allenoates with alkyl-, aryl-, or vinyl-Grignard reagents to synthesize polysubstituted beta,gamma-unsaturated alkenoates with high regio- and stereoselectivity. The in situ formed magnesium dienolate may readily react with different electrophilic reagents under different reaction conditions with or without a catalyst to construct an allylic quaternary carbon at the alpha-position of the ester group and a stereocontrol able retention of the carbon-carbon double bond.

Abstract:Through variable-temperature H-1 and C-13 NMR experiments and density functional calculations, the kinetics and mechanism of Michael type addition were investigated using alkynyl carbene A as the substrate. The two conformers of substrate A were distinguished from the H-1 and C-13 NMR spectra, and the calculated results showed that the syn-conformer is more stable than the anti- by 6.5 kJ mol(-1) with the activation barrier between these two conformers as 62.5 kJ mol(-1). The full reaction mechanism of Michael type addition was first presented to us, which differs from the previous solely based on the kinetic studies. It contains three elementary steps (see the scheme): (1) Formation of C-8-N-2 bond via transition state TS1. (2) Conformation conversion from In-1 to In-2, which is very important but ignored before. (3) Intramolecular proton transfer via transition state TS2 forming the product. The first step is rate determining with an activation barrier of 73.0 kJ mol(-1), very close to the experimental value of 89.6 kJ mol(-1). The product P is dominant over P' in population contrary to the situation of tautomer B' over B, which is caused by larger activation barriers to P' and the less stabilities of structures related to B' from the first transition state to the product.[GRAPHICS](c) 2006 Elsevier B.V. Abstract:A number of upper rim-functionalized calix[4]thiourea cyclohexanediamine derivatives have been designed, synthesized and used as catalysts for enantioselective Michael addition reactions between nitroolefins and acetylacetone. The optimal catalyst 2 with a mono-thiourea group exhibited good performance in the presence of water/toluene (v/v =1:2). Under the optimal reaction conditions, high yields of up to 99% and moderate to good enantioselectivities up to 94% ee were achieved. Detailed experiments clearly showed that the upper rim-functionalized hydrophobic calixarene scaffold played an important role in cooperation with the catalytic center to the good reactivities and enantioselectivities.

Abstract:Recently the hydrogen-bond activated reactions have attracted much attention.1 Takemoto2 reported a highly enantioselective Michael addition of manolate to nitroolefins catalyzed by a bifunctional organocatalyst with tertiary amine and thiourea moiety. As we known,stereoselective conjugate additions of thiols are interesting due to the standpoint of biological and synthetic importance, however, only very limited good results have been obtained except for the works of Shibasaki3, Kanemasa4 and Deng5 et al.In this letter, we report an efficient catalytic asymmetric Michael reactions of thiols to a,a-unsaturated carbonyl compounds promoted by bifunctional organocatalysts. A series of organocatalysts with chiral amine and thiourea structures were designed and synthesized and have been successfully applied in the conjugated additions of thiols to a,a-unsaturated imides and enones.The reactions got quantitative yields and the ee values were up to 84%. It is noteworthy that the a-asymmetric protonation (up to 43% ee) also could be achieved.The Michael addition between aromatic thiols and a,a-unsaturated carbonyl compounds isdescribed as follows:Works to further increase the enantioselectivity is under investigation in our laboratory.

Abstract:Bifunctional secondary amine-thiourea organocatalysts were successfully applied in the stereocontrolled synthesis of barbiturate-fused tetrahydropyrano scaffolds. Compared with typically used tertiary amine-thiourea organocatalysts, the developed catalysts exhibited excellent catalytic performance in the domino Michael-Michael reaction between N, N'-dimethylbarbituric acid and Morita-Baylis-Hillman acetates of nitroalkenes to yield pharmaceutically important heterocycles in good yields with excellent enantioselectivities. Moreover, this catalytic protocol can also be applied to synthesize biologically active pyranocoumarin compounds.

Abstract:It is one of the most attractive types of asymmetric synthesis that chiral commpounts are generated under the influence of chiral catalysts. In recent years catalytic asymmetric Michael addition by chiral metal complexes has been recognized as an efficient method for obtaining asymmetric products. In this work we reported that using potassium thiazolidine-2-thione-4-carboxylate [(R)-TTCA (.) K] as the chiral catalyst, the addition reaction of cyclohexanone with acrylonitrile was carried out and optically active (S)-(+)-2-cyanoethyl cyclohexanone 1 ([alpha](D)(20) = +2.8degrees, e. e. 19%) and hydrolysis product of 1, (S)-(-)-2-oxocyclohexanepropionic acid 2 ([alpha](D)(20) = -2.95degrees, e. e. 19%), were obtained. Enantiomeric excess of compound 1 was determined by resolution of (R,S)-2-oxocyclohexanepropionic acid with (-)-quinine. Compounds 1 and 2 were easily racemized at a long time for laying aside and in the presence of dil H2SO4 or dil base. When (S)-(+)-2-cyanoethyl cyclohexanone reacts with ethylene glycol in the presence of toluene sulfonic acid as the catalyst the racemic 2-ethylendioxy cyclohexyl propionitrile was obtained. The mechanism of the reaction of cyclohexanone with acrylonitrile would be a complicated catalysis process.
