Abstract:A general and clean method for the one-pot synthesis of thioamides was investigated using a combination of experiments and quantum chemical calculations. Ester, amide, and elemental sulfur were employed as starting materials and ethylene diamine tetraacetic acid (EDTA) as catalyst. Both alkyl and aryl thioamides with various functional groups were obtained in moderate to good yields. Neither transition metals nor external oxidants were required in this reaction. The catalytic performance of EDTA originates from a stepwise, proton transfer process in which EDTA transports a proton from the benzyl carbon to a sulfur atom. Such a process is concerted by discrete charge transfer such that the associated barrier gets lowered.

Abstract:We report a new thiourea-Bronsted acid cooperative catalytic system for the enantioselective cyanosilylation of aldehydes with yields up to 90% and enamioselectivities up to 88%. The addition of an achiral acid was found to be crucial for high asymmetric induction. Mechanistic investigations using a combination of NMR, ESI-MS, and density functional theory computations (including solvent corrections) at the M06/6-31G(d,p) level of theory suggest that the key catalytic species results from the cooperative interaction of bifunctional thioureas and an achiral acid that form well-defined chiral hydrogen-bonding environments.

Abstract:Cooperative hydrogen atom transfer and chiral hydrogen-bonding catalysis as a new platform for the asymmetric synthesis of azaarene derivatives is reported. By using a tetrabutylammonium decatungstate as the photocatalyst and a chiral phosphoric acid as the hydrogen-bonding catalyst, transformations of a variety of commercially available hydrocarbons and silanes with diverse α-branched 2-vinylazaarenes could efficiently experience a tandem radical conjugate addition and enantioselective protonation process, providing a convenient and fully atom economical approach to access a range of valuable enantioenriched α-tertiary azaarenes in high yields with good to excellent enantioselectivities (up to 93% ee). Through the direct use of tert-butyl methylcarbamate as the feedstock, this method enables a highly practical and concise synthesis of the enantiomerically pure medicinal molecule pheniramine (Avil).

Abstract:Asymmetric ion pair catalysis presents a powerful strategy for the construction of chiral molecules. However, the ion‐pair interactions are weakly directional and result in difficultly controlling the conformational constraint for high stereo‐ inductions. Based on the hydrogen bonding interactions, we have successfully designed and synthesized a new family of bifunctional ion pair catalysts derived from chiral amino acids via simple operations. With these chiral ammonium salts and phosphonium salts in hand, the enantioselective construction of C—C and C—X bonds was realized in our lab.

Abstract:A newly designed chiral iminophosphorane‐thiourea catalyst was developed to afford α‐acyloxythioesters through enantioselective acyl‐transfer/protonation of α‐acyloxy‐β‐ketosulfides. The desired reaction proceeded in good yields with high enantioselectivities using a chiral iminophosphorane‐thiourea bearing a pyrrolidine scaffold.

Abstract:A series of novel bifunctional phosphine-sulfonamide organic catalysts were designed and readily prepared from natural amino acids, and they were utilized to promote enantioselective aza-Morita-Baylis-Hillman (MBH) reactions. L-threonine-derived phosphine-sulfonamide 9b was found to be the most efficient catalyst, affording the desired aza-MBH adducts in high yields and with excellent enantioselectivities.

Abstract:A series of bifunctional phosphine-thiourea organic catalysts based on natural amino acid scaffolds were designed and prepared. L-threonine-derived bifunctional phosphine catalysts were found to be very efficient in promoting asymmetric Morita-Baylis-Hillman (MBH) reaction of acrylates with aromatic aldehydes, affording the desired MBH adducts with up to 90% ee. To gain mechanistic insights into the reaction, the effects of adding various additives on the MBH reaction were investigated. We propose that the hydrogen bonding interactions between the thiourea moiety of the catalyst and the enolate intermediate are crucial for the stereochemical outcome of the reaction. The method described in this report may provide a practical solution to the enantioselective MBH reaction of simple acrylates.

Abstract:A very efficient, chiral phase-transfer catalyst, (S)-2Db, was prepared by taking advantage of the combinatorial approach from the readily available (S)-1,1'-binaphthyl-2,2'-dicarboxylic acid. This catalyst exhibited high catalytic performance (0.01-0.1mol%) in the asymmetric alkylation of N-(diphenylmethylene)glycine tert-butyl ester and N-(p-chlorophenyl-methylene)alanine tert-butyl ester relative to other chiral phase-transfer catalysts in current use. This has created a general and highly practical procedure for the enantioselective synthesis of structurally diverse natural and unnatural alpha-alkyl-alpha-amino acids as well as alpha,alpha-dialkyl-alpha-amino acids. A similar simplified catalyst, (S)-2Fb, is also applicable to the direct asymmetric aldol reaction between glycine Schiff base and aldehydes with moderate syn selectivity and high enantioselectivity.

Abstract:Chiral thiourea, with a double hydrogen-bonding motif, emerged as attractive structural templates for asymmetric catalysis. Despite the significance, synthesis of enantioenriched thiourea predominantly relies on the nucleophilic addition of a chiral amine to isothiocyanate; catalytic synthesis of NH-free thiourea is highly desirable, albeit a formidable challenge. We herein describe NHCs-catalyzed desymmetrizative amidation of axially biaryl dialdehydes, providing structurally diversified axially chiral thiourea and imides. Sequential kinetic resolution improves the enantioenrichment of the desymmetrization product, dramatically expanding the range of applicable substrates. This strategy features a broad subtract scope and extremely excellent enantioselectivity. NHCs-catalyzed desymmetrizative amidation of axially prochiral biaryl dialdehydes provides modular platforms for synthesizing challenging axially chiral thiourea, imines, and derivatives.
