Abstract:A series of novel carbazole aminothiazoles as a new type of antimicrobial agents were designed, synthesized and characterized by H-1 NMR, C-13 NMR, IR, MS and HRMS spectra. Some of the carbazole aminothiazoles exhibited good antimicrobial activities; particularly, heptyl-derived carbazole aminothiazole 4f could effectively inhibit the growth of MRSA with an MIC value of 4 mu g mL(-1), which was superior to the reference drugs Chloromycin and Norfloxacin. Moreover, cytotoxicity investigation indicated that bioactive compound 4f did not exhibit cytotoxicity to Hep-2 cells within its MIC against MRSA. Preliminary interactive investigation revealed that compound 4f could effectively intercalate into calf thymus DNA to form 4f-DNA complexes which might block DNA replication and thus exert antimicrobial activities. In addition, the binding behavior of compound 4f to DNA revealed that compound 4f could interact with DNA by hydrogen bonds and electrostatic interactions.

Abstract:Aryl diamines were condensed with (R)-glycidyl butyrate to form (R,R)-l,4-bis-[(2-hydroxyl-3-butyryloxy)-propylamino] benzene derivatives (3a and 3b), which were cyclized with carbonyldiimidazole to get the bis-oxazolidinones(4a and 4b), the latter were hydrolyzed into (R,R)-l,4-bis-(5-hydroxymethyl-2-oxo-oxazolidin-3-yl)benzene derivatives (5a and 5b). Compounds 5a and 5b were converted via 6a, 6b and 7a, 76 to (S,S)-1,4-bis-(5-acetamidomethyl-2-oxo-oxazolidinyl-3)benzene derivatives (8a and 8b). The structures of these new compounds were confirmed by HNMR and MS. The in vitro antibacterial activities on 20 bacteria were tested and the result showed that compd 5b had better activity than linezolid against Staphylococcus epidermidis, Staphylococcus enteridis and Staphylococcus nonhemolyticus.

Abstract:A series of novel 11-[3-[(arylcarbamoyl)oxy]propylamino]-11-deoxy-6-O-methyl-3-oxoerythromycin A 11-N,12-O-cyclic carbamate derivatives (6a–h) were designed, synthesized and evaluated for their antibacterial activities in vitro. Most of these compounds had significant antibacterial activity against two groups of pathogens of Methicillin-sensitive Staphylococcus aureus (MIC50=0.031–2 μg ml−1) except 6g and Methicillin-sensitive S. epidermidis (MIC50=0.031–0.5 μg ml−1). MIC90 of 6d against Methicillin-resistant S. epidermidis was at least 16-fold better than that of erythromycin (EMA), azithromycin (AZM) and ABT-773. 6d and 6e had more potent antibacterial activity against S. pneumoniae than EMA, AZM and ABT-773. In particular, compounds 6d and 6e also showed relatively potent activity against Haemophilus influenzae and Streptococcus hemolyticus.

Abstract:A series of novel aminopyrimidinyl benzimidazoles as potentially antimicrobial agents were designed, synthesized and characterized by IR, NMR and HRMS spectra. The biological evaluation in vitro revealed that some of the target compounds exerted good antibacterial and antifungal activity in comparison with the reference drugs. Noticeably, compound 7d could effectively inhibit the growth of A. flavus, E. coli DH52 and MRSA with MIC values of 1, 1 and 8 μg/mL, respectively. Further studies revealed that pyrimidine derivative 7d could exhibit bactericidal mode of action against both Gram positive (S. aureus and MRSA) and Gram negative (P. aeruginosa) bacteria. The active molecule 7d showed low cell toxicity and did not obviously trigger the development of resistance in bacteria even after 16 passages. Furthermore, compound 7d was able to beneficially regulate reactive oxygen species (ROS) generation for an excellent safety profile. Molecular docking study revealed that compound 7d could bind with DNA gyrase by the formation of hydrogen bonds. The preliminary exploration for antimicrobial mechanism disclosed that compound 7d could effectively intercalate into calf thymus DNA to form a steady supramolecular complex, which might further block DNA replication to exert the powerful bioactivities. The binding investigation of compound 7d with human serum albumins (HSA) revealed that this molecule could be effectively transported by HSA.

Abstract:Novel C6-carbamate ketolides with C2-fluorination and C9-oximation have been synthesized. The best compounds in this series displayed MIC values of 0.03-0.12 microg/mL against streptococci containing erm and mef resistance determinants and 2-4 microg/mL against Haemophilus influenzae. Several compounds also showed measurable activity against erm(B)-containing enterococci with MIC values of 2-8 microg/mL. In vivo activity was adversely affected by fluorination, possibly as a result of increased serum protein binding.

Abstract:Novel clarithromycin derivatives with C-4″ elongated arylalkyl groups were designed, synthesized and evaluated to probe the effect of different lengths of their C-4″ side chains on the activity against resistant bacterial strains. These derivatives had excellent activity against erythromycin-susceptible Streptococcus pneumoniae, Streptococcus aureus or Streptococcus pyogenes and some of them exhibited greatly improved activity against erythromycin-resistant strains. Compounds 18 and 16, which had the C-4″ elongated arylalkyl groups with eight atoms from the 4″-oxygen atom to the terminal benzene ring, were the most effective against S. pneumoniae expressing the erm gene and the erm and mef genes. In contrast, the most potent compounds 3, 5, 9, 17 and 18 against S. pneumoniae expressing the mef gene had C-4″ elongated arylalkyl groups with three to eight atoms between the 4″-oxygen atom and the terminal aromatic ring.

Abstract:A new series of dimers of de(N-methyl) erythromycin derivatives, joined by a -COCH2- or -CH2CH2- linker, was synthesized and their in vitro antiproliferative activity against three human cancer cell lines was evaluated. Structure–activity relationships indicated that transformation of the linker -COCH2- into -CH2CH2- led to an increase in the potency of the compound; removal of cladinose and conversion of the carbonyl group at the C9 position to 9(S)-hydroxyl- or 9(S)-amino-containing groups resulted in a marked decrease in activity. Among the compounds synthesized, compounds 3d and 3e, both had C9 oxime groups and showed strong inhibitory activity against the three cell lines. Flow cytometry studies on HT-1080 cells indicated that the mechanism underlying the dimers' antiproliferative activity involved inducing cell cycle arrest in the G0/G1 phase. It was discovered that the percentage of HT-1080 cells arrested depended on the concentration of the erythromycin derivative dimer and on the length of time of treatment.

Abstract:A series of novel 1-(acetyl,carboxamide,carbothioamide)substituted-5-(substituted-phenyl)-3-methy-4,5-dihydropyrazole derivatives have been synthesized and characterized by 1H NMR, IR, ESI-MS, and elemental analysis. Compounds 6h and 6q were further characterized by single crystal X-ray structural analysis. All of the compounds have been screened for their antibacterial potential in vitro against Bacillus subtilis ATCC 6633, Escherichia coli ATCC 35218, Pseudomonas fluorescens ATCC 13525, and Staphylococcus aureus ATCC 6538. Among the tested compounds, some of them display significant activity against the tested strains, and compounds 5ac and 6h show potent activity with a minimum inhibitory concentration value of 1.562 μg mL–1 against B. subtilis ATCC 6633, which is comparable to the positive control penicillin. Structure–effect relationships are also discussed based on the experimental data.

Abstract:Objective: The most comprehensive collection of imine was synthesized via a simple, efficient, economical, and rapid method via Schiff base formation. Methods: (( E )-1,5-dimethyl-2-phenyl-4-(((2-phenylimidazo[1,2- a ]pyridine-3-yl)methylene)amino)-1,2-dihydro-3 H -pyrazol-3-one and their derivatives are synthesized using 4-aminoantipyrine and some substituted 2-phenylimidazo[1,2- a ]pyridine-3-carbaldehyde derivatives ( SB01 ) to ( SB10 ). Results and Discussion: The newly synthesized molecules were confirmed using various analytical techniques MS, FT-IR, 1 H, and 13 C NMR spectrometric analyses. Newly synthesized imidazo[1,2- a ]pyridine compounds undergo their biological evaluation with Gram-positive and Gram-negative bacteria as well as antifungal. Conclusions: ( SB10 ), ( SB07 ), ( SB05 ), and ( SB01 ) molecules show good, moderate, and excellent results against tested drugs.

Abstract:A series of novel 2-(benzo[d]thiazol-2-yl)-1-(3-methyl-5-substituted-phenyl-4,5-dihydropyrazol-1-yl)ethanone derivatives were synthesized and characterized by NMR, ESI-MS. Biological activity tests results show that compounds 4a, 4b, 4d and 4g displayed activity with MIC of 1.562 mu g/mL against B. subtilis ATCC 6633, compound 4d displayed significant activity with MIC of 1.562 mu g/mL against S. aureus ATCC 6538. Compound 4a with potent antibacterial activities strongly inhibited S. aureus DNA gyrase and B. subtilis DNA gyrase with IC(50s) of 0.5 mu g/ml against S. aureus DNA gyrase and B. subtilis DNA gyrase.

Abstract:The biosynthesis of fatty acids constitutes a critical metabolic pathway in bacterial organisms. Prior investigations have highlighted the synthesis of antimicrobial compounds anchored in the benzodioxepin scaffold, noted for their pronounced antibacterial properties. Leveraging this foundational knowledge, the current research endeavors to meticulously engineer and synthesize a series of eight innovative benzodioxepin amide-biphenyl derivatives. This achievement was realized through the sophisticated optimization of synthetic methodologies. The scope of this study extends to a rigorous evaluation of the antibacterial prowess and biocompatibility of the aforementioned novel derivatives. Notably, Compound E4 emerged as a supremely potent antimicrobial agent. A detailed elucidation of the crystalline architecture of Compound E4 was conducted, alongside a thorough docking study to explore its interactions with the FabH enzyme.

Abstract:Thirty-one 3-aryl-4-alkylaminofuran-2(5H)-one derivatives were prepared and tested for their antimicrobial activities. 3-(4-Bromophenyl)-4-(2-(4-nitrophenyl)hydrazinyl)furan-2(5H)-one (4a11) showed the most potent antibacterial activity with MIC50 of 0.42μg/mL against Staphylococcus aureus ATCC 25923 and was the most potent inhibitor against tyrosyl-tRNA synthetase from S. aureus with IC50 of 0.12±0.04μM.

Abstract:Nine 2-arylthiazolidine-4-carboxylic acid derivatives and nine 3-tert-butoxycarbonyl-2-arylthiazolidine-4-carboxylic acid derivatives were synthesized to screen for their antibacterial activities. Compounds 5, 14–18 were first reported. Their chemical structures were clearly determined by 1H NMR, 13C NMR, ESI mass spectra and elemental analyses, coupled with one selected single-crystal structure. All the compounds were assayed for antibacterial activities against two Gram-positive bacterial strains (Bacillus subtilis ATCC 6633 and Staphylococcus aureus ATCC 6538) and two Gram-negative bacterial strains (Escherichia coli ATCC 35218 and Pseudomonas aeruginosa ATCC 13525) by MTT method. Most of the 3-tert-butoxycarbonyl-2-arylthiazolidine-4-carboxylic acid derivatives exhibited better antibacterial activities against the four bacterial strains than relative 2-arylthiazolidine-4-carboxylic acid derivatives. Compound (2RS,4R)-3-(tert-butoxycarbonyl)-2-(5-fluoro-2-hydroxyphenyl)thiazolidine-4-carboxylic acid (14) showed powerful antibacterial activities against P. aeruginosa with IC50 value of 0.195μg/mL, which was superior to the positive controls Penicillin G and Kanamycin B, respectively. On the basis of the biological results, structure–activity relationships were discussed.

Abstract:Infectious diseases pose a major challenge to human health, and there is an urgent need to develop new antimicrobial agents with excellent antibacterial activity. A series of novel triazolo[4,3-a]pyrazine derivatives were synthesized and their structures were characterized using various techniques, such as melting point, 1H and 13C nuclear magnetic resonance spectroscopy, mass spectrometry, and elemental analysis. All the synthesized compounds were evaluated for in vitro antibacterial activity using the microbroth dilution method. Among all the tested compounds, some showed moderate to good antibacterial activities against both Gram-positive Staphylococcus aureus and Gram-negative Escherichia coli strains. In particular, compound 2e exhibited superior antibacterial activities (MICs: 32 μg/mL against Staphylococcus aureus and 16 μg/mL against Escherichia coli), which was comparable to the first-line antibacterial agent ampicillin. In addition, the structure–activity relationship of the triazolo[4,3-a]pyrazine derivatives was preliminarily investigated.

Abstract:A series of new 2-phenyl-quinoline-4-carboxylic acid derivatives was synthesized starting from aniline, 2-nitrobenzaldehyde, pyruvic acid followed by Doebner reaction, amidation, reduction, acylation and amination. All of the newly-synthesized compounds were characterized by ¹H-NMR, (13)C-NMR and HRMS. The antibacterial activities of these compounds against Gram-negative (Escherichia coli, Pseudomonas aeruginosa) and Gram-positive bacteria (Staphylococcus aureus, Bacillus subtilis), as well as one strain of methicillin-resistant Staphylococcus aureus (MRSA) bacteria were evaluated by the agar diffusion method (zone of inhibition) and a broth dilution method (minimum inhibitory concentration (MIC)), and their structure-activity relationships were obtained and discussed. The results revealed that some compounds displayed good antibacterial activity against Staphylococcus aureus, and Compounds 5a₄ and 5a₇ showed the best inhibition with an MIC value of 64 μg/mL against Staphylococcus aureus and with an MIC value of 128 μg/mL against Escherichia coli, respectively. The results of the MTT assay illustrated the low cytotoxicity of Compound 5a4.

Abstract:A series of novel botanical active component 4-chromanone derivatives incorporating carboxamide and 1,3,4-thiadiazole thioether moieties were synthesized and evaluated for their in vitro antibacterial and activities antifungal against Xanthomonas axonopodis pv. citri ( Xac ), Xanthomonas oryzae pv. oryzicolaby ( Xoc ) and Mucor bainieri ( M. bainieri ), Mucor fragilis ( M. fragilis ), Trichoderma atroviride ( T. atroviride ), respectively. Antibacterial screening results suggested that the target compounds showed moderate to good antibacterial activities against Xac and Xoc , to the contrary, had lower inhibitory effects on M. fragilis , M. bainieri , and T. atroviride . Among the target compounds, compound 6-chloro- N -(5-(methylthio)-1,3,4-thiadiazol-2-yl)-4-oxochromane-2-carboxamide ( 6f ) had the best inhibition rates against Xac (85.15% and 72.41%, respectivelty) and Xoc (95.14% and 82.09%) at 200 and 100 μg/mL, which were superior to Bismerthiazol and Thiodiazole copper. Abstract:Eleven new 1-{5-[4-(benzyloxy)phenyl]-3-methyl-4,5-dihydropyrazol-1-yl} oxime ester derivatives were synthesized and characterized by elemental analysis, HRMS, H-1 NMR data. All the compounds were screened for their antibacterial potential in vitro against Bacillus subtilis, Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa. The results indicate that compounds 8c and 8f possess potent activity with the minimum inhibitory concentrations(MIC) of 1.562-3.125 mu g/mL against all the four bacteria. Compounds 8c, 8e and 8f show moderate inhibition against the DNA gyrase(IC50 = 1.9-2.5 mu g/mL). On the basis of the biological activities, structure-activity relationship was discussed.

Biological activity tests results show that compound 3h displayed activity with MIC of 0.39, 0.78, 1.562 mu g/mL against B. subtilis ATCC 6633, S. aureus ATCC 6538, P. fluorescens ATCC 13525 and could strongly inhibit S. aureus DNA gyrase and B. subtilis DNA gyrase with IC50s of 0.25 and 0.18 mu g/mL respectively.

Abstract:We report here on the structure-activity relationships of hybrids combining 3-descladinosyl clarithromycin with quinolones linked by extended diamine connectors. Several hybrids, exemplified by 23Bc, 23Be, 23Bf, 26Be, and 30Bc, not only restored potency against inducibly resistant pathogens but also exhibited significantly enhanced activities against constitutively resistant strains of Staphylococcus pneumoniae and Staphylococcus pyogenes, which express high-level resistance independent of clarithromycin or erythromycin induction. Additionally, the novel hybrids showed susceptibility against Gram-negative Haemophilus influenzae. Notably, hybrid 23Be demonstrated dual modes of action by inhibiting both protein synthesis and DNA replication in vitro and in vivo. Given these promising characteristics, 23Be emerges as a potential candidate for the treatment of community-acquired bacterial pneumonia.
