Abstract:It provides a theoretical basis for the artificial promotion of the reproduction and regeneration of Liriodendron chinense through exploring the effects of light intensity and seeding density on the seed germination and seedling growth of this rare plant. Controlled experiments were conducted by setting different seeding densities and shading intensities to observe and detect the seed germination and seedling growth under different experimental conditions. The data were analyzed by SPSS and the model was fitted to calculate the seed germination rate, seedling emergence rate and seedling growth parameters of Liriodendron chinense, which were used to analyze the effects of seeding density and light intensity on the seed germination and seedling growth of Liriodendron chinense. Light has a significant impact on the seed germination and seedling growth of Liriodendron chinense (P P > 0.05). High density promotes the seedling height (the seedling height showed a rising trend with the increase of seeding density, reaching the highest at 500 seeds/m2. “3-stitch + 300 seeds/m2” combination is an appropriate condition for Liriodendron chinense reproduction since it can realize higher germination rate and better seedling growth potential. The seedling height and basal diameter growth curve of Liriodendron chinense fits well to the Logistic equation (R2 ≥ 0.977). Light has a significant impact on the seed germination and seedling growth of Liriodendron chinense, and appropriate shading treatment can increase the seed germination rate and seedling emergence rate of Liriodendron chinense. Seeding density has little influence on the seed germination rate of Liriodendron chinense, but high density will promote seedling height.

Abstract:Inheritable variation induced by plant grafting is a universal phenomenon; however, its mechanism has always been a controversial subject. In recent years, research on horizontal transfer of genetic materials between stock and scion has made great progress. The latest studies have found that genetic information in grafted plants is transported through plasmodesma and phloem to modulate the growth and development of the scion plants. Furthermore, non-coding RNAs could induce epigenetic modification, which facilitates plants to adapt to the grafting-caused stress. This article focuses on the review of recent advances in the induction and maintenance of the graft-induced genetic variation and could be helpful in thoroughly understanding the mechanism.

Abstract:Expanding the southern range of herbaceous peony (Paeonia lactiflora Pall.) is a meaningful and worthwhile horticultural endeavor in the Northern Hemisphere. However, high temperatures in winter seriously hinder the bud dormancy release and flowering of peony in the more southern areas of subtropical and tropical regions. Resource introduction and hybridization can contribute to creating new cultivars with high adaptability in a warmer winter climate. In this study, three representative cultivars of P. lactiflora were screened for flowering capabilities and their annual growth cycles were observed to provide information needed for hybridization. Among these three cultivars, 'Hang Baishao' is the best adapted cultivar for southern growing regions and is unique in its ability to thrive in southern areas of N 30 degrees 00'. Pollen viability of 'Hang Baishao' was 55.60% based on five measuring methods, which makes it an excellent male parent in hybridization. Hybrid plants among these three cultivars grew well, but all of their flower buds aborted. Additionally, the ability of three growth regulators that advance the flowering of 'Hang Baishao' to promote an indoor cultivation strategy for improving peony application as a potted or cut-flower plant was tested. 5-azacytidine could impact the growth of 'Hang Baishao' and induce dwarfism and small flowers but not advance the flowering time. Gibberellin A(3) promoted the sprouting and growth significantly, but all plants eventually withered. Chilling at 0-4 degrees C for four weeks and irrigation with 300 mg/L humic acid was the optimal combination used to hasten flowering and ensure flowering quality simultaneously. These results can lay the foundation for future studies on the chilling requirement trait, bud dormancy release and key functional gene exploration of herbaceous peony. Additionally, this study can also provide guidance for expanding the range of economically important plants with the winter dormancy trait to the low-latitude regions.

Abstract:Kenaf hybrid H318,red stem,divided leaf,which was bred and selected from several decades of kenaf crosses,is the most promising hybrid used in production at present.The average fiber yield of H318 was 5460kg/hm2,30.2% and 64.2% higher than that of CK KB2 and"Hongying135" respectively in comparison of kenaf hybrids in 2003,4411.65kg/hm2,20.75% higher than that of CK "KB2"innationalkenafregionaltrialat7locationsin3years(2004-2006),thehighestamongnewlines and hybrids.The average dry stalk yield of H318 was 22.06t/hm2,20.5% higher than that of CK at 3 sites.Its average fiber strength was 327N(1g fiber,length 30cm),fineness 266m/g.Under the artificial inoculation,the anthracnose disease index was 17.8,the rate of died top stems 2.4%.The main advantages of kenaf hybrid H318 were large plants,more available plants at final harvest,high ratio of bark to stalk,good fiber quality,high resistance to the anthracnose disease and wide adaptability.

Abstract:Kenaf hybrid H368 ( originally named H368) was selected and bred from the progeny of“991 ×LC0301A”, which would be a textile and multipurpose new kenaf variety with high yield , disease and lodging resistance , high quality and wide adaptability.It was non -early -flowering , red stem and lobed leaf.Its average height was 426.07cm, stem diameter was 1.93 cm, fresh bark thickness was 1.42 mm, small and short plant rate was 18.85%, the retting rate to dry bast was 62.96%.In national region-al trials, the average fiber yield of Kenaf hybrid H 368 was 4456.20kg/hm2 , 25.32%higher than the CK“Fuhong 951” from 2009 to 2010.Its average fiber and stem yields ranked the first place among the con-ventional varieties.In 2010, in the productive trials , its average fiber yield was 4079.19 kg/hm2 , 23.65%higher than the CK.The disease index was 14.00 after 15 days and 12.73 after 45 days, which could be concluded that this new kenaf variety was high resistant to the anthracnose disease .Its average fiber strength was 315 N, fiber fineness was 354 m/g, all the traits were better than those of CK.
Agrobacterium-Mediated Genetic Transformation of Embryogenic Callus in a Liriodendron Hybrid (L. Chinense × L. Tulipifera)

Abstract:A highly efficient genetic transformation system of Liriodendron hybrid embryogenic calli through Agrobacterium -mediated genetic transformation was established and optimized. The Agrobacterium tumefaciens strain EHA105, harboring the plasmid pBI121, which contained the ß -glucuronidase ( GUS ) gene and neomycin phosphotransferase II ( npt II) gene under the control of the CaMV35S promoter, was used for transformation. Embryogenic calli were used as the starting explant to study several factors affecting the Agrobacterium -mediated genetic transformation of the Liriodendron hybrid, including the effects of various media, selection by different Geneticin (G418) concentrations, pre-culture period, Agrobacterium optical density, infection duration, co-cultivation period, and delayed selection. Transformed embryogenic calli were obtained through selection on medium containing 90 mg L −1 G418. Plant regeneration was achieved and selected via somatic embryogenesis on medium containing 15 mg L −1 G418. The optimal conditions included a pre-culture time of 2 days, a co-culture time of 3 days, an optimal infection time of 10 min, and a delayed selection time of 7 days. These conditions, combined with an OD 600 value of 0.6, remarkably enhanced the transformation rate. The results of GUS chemical tissue staining, polymerase chain reaction (PCR), and southern blot analysis demonstrated that the GUS gene was successfully expressed and integrated into the Liriodendron hybrid genome. A transformation efficiency of 60.7% was achieved for the regenerated callus clumps. Transgenic plantlets were obtained in 5 months, and the PCR analysis showed that 97.5% of plants from the tested G418-resistant lines were PCR positive. The study of the Liriodendron hybrid reported here will facilitate the insertion of functional genes into the Liriodendron hybrid via Agrobacterium -mediated transformation.

Abstract:For estimating agronomic traits of the new asparagus hybrid Zhefeng 121 F1, we carried out the cultivar comparative test in Xinchang county of Zhejiang province from 2008 to 2011, and took two cultivars Grande F1 and UC157 F2 as control. We measured nine agronomic traits in plastic greenhouse from 2009 to 2011. The results showed that the average marketable spear yields of Zhefeng 121 F1, Grande F1 and UC157 F2 were 25 459.52, 22 775.13 and 16 292.13 kg/hm2 in the three years, respectively, and the yield of Zhefeng 121 F1 increased by 11.79% and 56.53%compared with the control cultivars Grande F1 and UC157 F2. The marketable spear yield of Zhefeng 121 F1 in the third harvesting year (2011) was extremely significantly increased by 24.49% (p=0.01) compared with cultivar Grande F1. We measured the contents of flavonoid, rutin, total phenols and saponins of the young spears in the second harvesting year (2010), and the results showed that the four contents of Zhefeng 121 F1 were significantly higher than those of Grande F1, with the increasing rate of 15.61%-32.19%. Additionally, both Zhefeng 121 F1 and Grande F1 were moderately susceptible to stem blight, but UC157 F2 was highly susceptible.

Abstract:This paper studied the early growth performance and the site condition of Slash pine × Honduras Caribbean pine F2 hybrid from Australia that was planted in Xijiang forest farm,Yinzhan forest farm,Tongmen forest farm,Dashudun in Jintan town of Qingxin county and Longping forest farm in Guangdong province.The results indicated that the Slash pine × Honduras Caribbean pine F2 hybrid could grow well in Guangdong province.The mean height,diameter breast-height at the age of 3 years were 20.26% and 27.27% respectively higher than Pinus elliottii at the same age.Slope position and direction,soil conditions have important effects in early growth of Slash pine × Honduras Caribbean pine F2 hybrid.

Abstract:Stand density affects the potentially superior productivity of forest ecosystems directly by regulating the light and nutrient availability of trees. Understanding how stand density influences the growth and development of trees is crucial for supporting forest management in the context of climate change. We focused on Liriodendron chinense in experimental plantations created in 2003, with planting densities ranging from 277 to 10,000 trees per hectare at six plots. The leaf structure and photosynthetic capacity of L. chinense changed significantly under different stand densities, which had a negative impact on their biomass (leaf mass) and nutrient (total carbohydrate content) accumulation. Transcriptional differences were observed among samples from plots with different planting densities. The expression of 1784 genes was negatively dependent on stand density, participating mainly in the biological processes of "circadian rhythm", "carbon metabolism", and "amino acid biosynthesis". Furthermore, we identified a photosynthesis-related module and constructed a gene regulatory network to discover that the transcription factors of MYB and bHLH may have important roles in the transcriptional regulation of photosynthesis biosynthesis by activating or repressing the expression of petA (Litul.15G096200), psbE (Litul.10G033900), and petD (Litul.17G061600) at different stand densities. Our study quantified the impact of stand density on tree growth at physiological and molecular levels. Our observations provide theoretical support for plantation establishment of L. chinense.

Study on the Graft Compatibility Between ‘jingganghongnuo’ and Other Litchi Cultivars

Abstract:Litchi (Litchi chinensis Sonn.), is one of the most important tropical and subtropical fruit crop in the Sapindaceae family. Grafting is widely used for litchi propagation, but not all cultivars are compatible. In this study, Jingganghongnuo', a new litchi cultivar, was grafted on the rootstock of cultivar 'Jingfeng', 'Heiye', 'Huaizhi', 'Songjiaxiang', 'Guilin', 'Xuehuaizi', 'Zhuangyuanhong', 'Liuyuexue', 'Baitangying', 'Wuye', 'Shuangjianyuhebao', 'Wanpu', 'Zaopu' to determine their compatibility. The survival rate was determined 6 months after grafting. Significant difference in chlorophyll contents was detected between compatible combinations and incompatible combinations 6 months after grafting. Compatible combinations showed dark green leaf and smooth graft joint, while, the incompatible ones had yellow leaf. Histological assay indicated that the compatible combinations developed vascular bundle bridge from the wound tissues, while, there was few vascular bundle bridge connected in incompatible combinations. Arbuscular Mycorrhizal Fungi Effectively Enhances the Growth of Gleditsia Sinensis Lam. Seedlings under Greenhouse Conditions

Abstract:The Chinese honey locust tree Gleditsia sinensis Lam. (Fabaceae) is a precious ecological and economic tree species that has wide-ranging usage. However, knowledge regarding seedling cultivation (especially the use of arbuscular mycorrhizal fungi (AMF)) is scarce, which limits the developent of Gleditsia plantations. A pot experiment was carried out under greenhouse conditions to estimate the effects of three AMF strains (Funneliformis mosseae 1, Funneliformis mosseae 2, and Diversispora tortuosa) on the growth, photosynthetic rate, and nutrient content of G. sinensis seedlings. Results showed that the growth parameters (seedling height, basal diameter, dry biomass) of the seedlings were significantly increased by each of the three AMF strains, associated with high root colonization rates (greater than 75%). Chlorophyll concentrations and photosynthetic rates were also increased by AMF, and phosphorus (P), and potassium (K) content in the three organs (leaf, stem, and root), and nitrogen (N) content in the leaf and stem of arbuscular mycorrhizal (AM) seedlings were significantly higher than in non-AM seedlings. Mycorrhizal dependency of the AM seedlings was greater than 350%, and significantly correlated with the increased P and K content in all three organs and increased N content in the leaf and stem. Positive effects of F. mosseae on growth and the nutrient content of seedlings were higher than those of D. tortuosa, but no significant different effects on G. sinensis seedlings were observed between the two strains of F. mosseae. Hence, growth of G. sinensis seedlings was effectively enhanced by AMF, with F. mosseae being more suitable for the inoculation of G. sinensis seedlings. These results indicate that arbuscular mycorrhization is beneficial for the growth of young G. sinensis plants. Further research is needed to determine whether the effects can be reproduced in a forest situation.
Effect of Grafting on the Growth and Flowering of Sprays Chrysanthemums

Abstract:To study the effect of grafting on the growth and flowering of spray chrysanthemum, Artemisia annua was used as the rootstock and eight spray chrysanthemum varieties 'Zhongshan Jinyang' ('ZSJY'), 'Zhongshan Xinmolian' ('ZSXML'), 'Zhongshan Xuerong' ('ZSXR'), 'Zhongshan Yinggui' ('ZSYG'), 'Zhongshan Zaohong' ('ZSZH'), 'Zhongshan Zhiying' ('ZSZY'), 'Zhongshan Zisongguo' ('ZSZSG'), and 'Zhongshan Ziwei' ('ZSZW') were used as the scions. The plant survival ratio, growth index (crown width, crown circumference, and biomass), and flowering characteristics (total inflorescences per plant, flower diameter, fresh and dry weight of single flowers, and flowering period) were studied after grafting. We found that for variety 'ZSYG', the grafted plants showed a 58.4% increase in crown width, a 68.7% increase in crown circumference, a 61.8% increase in the dry weight of the scion, a 108.4% increase in the dry weight of root, a 72.3% increase in the total dry weight, a 28.3% increase in root/shoot, and a 135.2% increase in the total inflorescences per plant compared to those in the own-rooted plants. Grafting can not only promote plant growth and increase plant biomass, and the total inflorescences per plant of spray chrysanthemum but also lengthen the ornamental period of spray chrysanthemum by advancing flowering and delaying the final flowering period. The budding period can be advanced by 13 d, the final flowering period can be delayed by 4 d, and the ornamental period can be increased by 10 d. Therefore, grafting facilitates the growth of spray chrysanthemums, advances flowering, and prolongs the ornamental period.

Abstract:Larch species are widely distributed in the northern hemisphere where permafrost and seasonal frozen soil exist. This species with heterophyllous shoots has been intensively planted in northeast Asia as well as in northeast China as the principal afforestation species for restoring agricultural lands to forests from 1999. Although approximately 15 species exist in the northern hemisphere and they are easy to hybridize. Among them, Japanese larch grows the fastest and was exported to Europe as a breeding species from early 20s. Although Japanese larch is tolerant to cold, it suffered from various biological stresses. After nearly 40 years of vigorous breeding effort, hybrid larch F1 (Dahurian larch × Japanese one) was developed with simple propagation methods. With the use of free-air CO2 enriched (FACE) systems, we revealed growth responses of the F1 and its parent larches to environmental conditions. From experiments, F1 showed high responses to elevated CO2 and O3 but not so much to N loading. As future perspectives for larch plantations as an important eco-environmental resource, we expect to afforest F1 seedlings infected with ectomycorrhizae (e.g., Suillus sp.) for efficient afforestation at nutrient-poor sites and at the same time for the production of delicious mushrooms.

Abstract:Protoplast monokaryons prepared from Lentinula edodes,strains 939 and 135,were intercrossed using symmetric and asymmetric hybridization methods.Following test cultivations at different locations within Zhejiang Province,'Shenxiang 16'(S605) was selected from 79 hybrid offspring on the basis of improved characteristics including higher disease resistance compared to parental strain 135,improved fruit body morphology(shorter stipe) compared with parental strain 939,and enhanced fruit body yields compared with both parents.The strain is now popular among growers in Zhejiang Province due to easier management of the fruiting stage,shorter times between flushes,and higher yields with a higher ratio of good quality fruit bodies.

Abstract:Ninteen primary selected half-sib elite families of Chinese fir were tested on seedling growing in Jiande City , Zhejiang Province .The results showed that seed germination potential , seedling disease resistance and height increment of elite families were 20.6 %, 82.1 % and 33.6 % higher than those of control family respectively .Seedling height increment and growth rhythm of each family were of significant difference , and field germination percentage , disease resistance and cold resistance of each family seedlings were of obvious difference too .Therefore , some different management measures should be taken on families or family categories in seedling growing .

Abstract:In order to study grafting methods in Xanthoceras Sorbifolia and to improve survival rate of grafting,effects of grafting time and methods on survival quality of grafting were researched,at different seasons in a year,by bud with xylem grafting in spring,T-shape bud grafting in summer,bud with xylem grafting in autumn,and bark grafting in spring.The results show that bud with xylem grafting is the best grafting method in March or April,the survival rate is 93.6%,seedling height is 170.5 cm,and diameter at ground is 10.8 mm.The relationship between stock diameter and survival rate of grafting show that stock diameter more than 0.4 cm is beneficial to increasing survival rate of grafting.

Abstract:The root is the main organ of a plant for absorbing resources and whose spatial distribution characteristics play an important role in the survival of seedlings after afforestation. Chinese fir (Cunninghamia lanceolata) is one of China's most important plantation species. To clarify the effects of propagation methods on root growth and spatial distribution characteristics of Chinese fir trees, sampled trees cultivated by seed germination, tissue culture, and asexual cutting of Chinese fir were taken as the research objects. The root morphology, geometric configuration, and spatial distribution characteristics of different trees were analyzed. The influence of geometric root morphology on its spatial distribution pattern was explored by correlation analysis, and the resource acquisition characteristics reflected by the roots of Chinese fir trees with different propagation methods are discussed. The main results showed that the root mean diameter (1.56 mm, 0.95 mm, and 0.97 mm), root volume (2.98 m3, 10.25 m3, and 4.07 m3), root tip count (397, 522, and 440), main root branch angle (61°, 50° and 32°) and other geometric configurations of Chinese fir under seed germination, tissue culture and rooted cutting respectively, were significantly different, which resulted in different distribution characteristics of roots in space. Chinese fir seed germination had noticeable axial roots, and the growth advantage was obvious in the vertical direction. A fishtail branch structure (TI = 0.87) was constructed. The shallow root distribution of tissue culture and rooted cutting was obvious, and belonged to the fork branch structure (TI = 0.71 and 0.74, respectively). There was a tradeoff in the spatial growth of the root system of Chinese fir trees with different propagation methods to absorb nutrients from heterogeneous soil patches. A negative correlation was present between the root system and root amplitude. There was an opposite spatial growth trend of Chinese fir trees with different propagation methods in the vertical or horizontal direction. In conclusion, selecting suitable propagation methods to cultivate Chinese fir trees is beneficial to root development and the "ideal" configuration formation of resource acquisition to improve the survival rate of Chinese fir afforestation.
Progeny performance and selection of superior trees within families in Larix olgensis

Abstract:Larix olgensis is one of the most important timber species in Northeast China. Although L. olgensis seed orchards have been established for many years and some progeny tests have been conducted, studies of progeny performance and the selection of superior families are still lacking. These tests are important for upgrading and improvement of seed orchards. Here, we estimate the genetic variation for major economic traits between families to provide selection materials for the establishment of a second-generation seed orchard. This study examined 71 half-sib families and 2 control families of L. olgensis in the Xiyang forest seed orchard in Yongji County, Northeast China. We measured tree height, diameter at breast height, volume, branch angle, stem straightness degree and crown width of 4-year-old progeny. Genetic parameters for the studied traits were determined, and comprehensive analyses of multiple traits and breeding values were used for selection of superior families and individual trees within families. The results show that all these traits are significantly different (P < 0.01) among families where family variance was greater than environmental variance. The phenotypic and genotypic coefficients of variation ranged from 9.01% to 78.22% and from 1.81% to 39.20%, respectively. The heritability values of families and individual trees ranged from 0.663 to 0.959 and 0.034 to 0.983, respectively. There was a significant positive correlation between investigated traits. Using the multiple-traits comprehensive evaluation method, seven families and 56 superior individual trees were selected. Selection of superior parents using breeding values for timber yield, stem quality and crown width as a proxy for planting density again resulted in the selection of seven superior families. We determined that the comprehensive multiple-traits evaluation approach is effective for selecting superior families and individual trees for the establishment of advanced second generation seed orchards. Families selected based on breeding value may be used as preferred parents for future hybridization breeding.

Abstract:Abstract The effects of mutual grafting on growth and moisture content in post-grafting generations of two varieties cherry tomato seedlings (yellow cherry tomato and red cherry tomato) under stress of Cd were studied in a pot experiment involving the following: un-grafted yellow cherry tomato and red cherry tomato (Yellow CK and Red CK), yellow cherry tomato as rootstock and red cherry tomato as scion (Yellow Rootstock, Red Scion), yellow cherry tomato as scion and red cherry tomato as rootstock (Yellow Scion, Red Rootstock). Mutual grafting increased plant height, stem weight of post-grafting of yellow cherry tomato; however, stem diameter, root and leaf weight and moisture content were no significantly effects compared with Yellow CK. As for red cherry tomato seedlings, mutual grafting increased root length of post-grafting; plant height and moisture content of post-grafting of Red Rootstock were increased, while in Red Scion, the these parameters decreased or showed no significant effects. These results imply that mutual grafting may improve or no effect in growth and moisture content of post-grafting generations.
Transcriptome Changes Between Compatible and Incompatible Graft Combination of Litchi Chinensis by Digital Gene Expression Profile.

Abstract:Plant grafting has been practiced widely in horticulture and proved as a useful tool in science. However, the mechanisms of graft healing or graft incompatibility remain poorly understood. In this study, Litchi chinensis cv. ‘Jingganghongnuo’ homograft (‘J/J’) and ‘Jingganghongnuo’/‘zhuangyuanhong’ heterograft (‘J/Z’) as compatible and incompatible combination, respectively, was used to study transcriptional changes between incompatible and compatible graft during graft union formation. Anatomical observation indicated that three stages (2 h, 14 d and 21 d after grafting) were critical for graft union formation and selected for high-throughput sequencing. Results indicated 6060 DEGs were differentially expressed in the compatible combination and 5267 DEGs exhibiting in the incompatible one. KEGG pathway enrichment analysis revealed that DEGs were involved in metabolism, wound response, phenylpropanoid biosynthesis and plant hormone signal transduction. The expression of 9 DEGs annotated in auxin pathway was up-regulated in compatible combination than that in incompatible combination. The IAA concentration confirmed that the IAA might promote the graft compatibility. In addition, 13 DEGs related to lignin biosynthesis were differentially expressed during graft healing process. Overall, our results provide abundant sequence resources for studying mechanisms underlying graft compatibility and establish a platform for further studies of litchi and other evergreen fruit trees.
