Abstract:Apelin is a peptide which has been seen to act as an endogenous ligand of the orphan G-protein-coupled APJ(putative receptor protein related to the angiotensin receptor AT1).Apelin/APJ system displays several roles in the cardiovascular system.Among the total,the roles in cardiovascular system of apelin/APJ system can be summarized to 3 aspects,first,apelin/APJ system can relax vessels and decrease the blood pressure through anti-angiotensin Ⅱactions;second,apelin/APJ system showed strong positive inotropic activity;third,the expression of apelin in blood increased in the early stage of heart failure and decreased in the late stage of heart failure,therefore,someone inferred that apelin/APJ system was a effective compensative manner to inhibit heart failure.Apelin/APJ system may involved in the secretion of insulin and promote the proliferation of retina blood capillary.All these results indicated that apelin/APJ system might have certain pathophysiological roles in cardiovascular diseases,such as hypertension,heart failure and type 2 diabetes mellitus.

Abstract:Apelin is a recently isolated novel endogenous ligand for the angiotensin-like 1 receptor (APJ). Initial experiments in animal models indicate that the cardiovascular system is the main target of the apelin-APJ system. Apelin plays an opposite role to the renin-angiotensin-aldosterone system as a compensatory mechanism. It is reduced in patients with heart failure, also of ischemic origin. However, only animal studies concern the role of the apelin-APJ system in myocardial ischemia. Less is known about the function of this adipokine in an acute phase of myocardial infarction in human. The apelin-APJ system could perhaps be involved in myocardial protection during acute myocardial ischemia. In the current review we have summarized recent data concerning the role of apelin in acute myocardial infarction and heart failure induced by ischemia.

Abstract:Apelin is an endogenous ligand of G protein-coupled receptor APJ. It is extensively expressed in many tissues such as heart, liver, and kidney, especially in lung tissue. A growing body of evidence suggests that apelin/APJ system is closely related to the development of respiratory diseases. Therefore, in this review, we focus on the role of apelin/APJ system in respiratory diseases, including pulmonary arterial hypertension (PAH), pulmonary embolism (PE), acute lung injury (ALI)/acute respiratory distress syndrome (ARDS), obstructive sleep apnoea syndrome (OSAS), non-small cell lung cancer (NSCLC), pulmonary edema, asthma, and chronic obstructive pulmonary diseases. In detail, apelin/APJ system attenuates PAH by activating AMPK-KLF2-eNOS-NO signaling and miR424/503-FGF axis. Also, apelin protects against ALI/ARDS by reducing mitochondrial ROS-triggered oxidative damage, mitochondria apoptosis, and inflammatory responses induced by the activation of NF-κB and NLRP3 inflammasome. Apelin/APJ system also prevents the occurrence of pulmonary edema via activating AKT-NOS3-NO pathway. Moreover, apelin/APJ system accelerates NSCLC cells' proliferation and migration via triggering ERK1/2-cyclin D1 and PAK1-cofilin signaling, respectively. Additionally, apelin/APJ system may act as a predictor in the development of OSAS and PE. Considering the pleiotropic actions of apelin/APJ system, targeting apelin/APJ system may be a potent therapeutic avenue for respiratory diseases.

Abstract:To the Editor : Pathological cardiac hypertrophy is commonly stimulated by stress or induced by cardiovascular diseases, such as hypertension and myocardial infarction. A sustained pathological hypertrophic response can lead to declines in diastolic and systolic functions and eventually develop heart failure. Angiotensin II (Ang II) can mediate cardiac hypertrophy through the angiotensin II type 1 receptor (AT1R). [ 1 ] Angiotensin receptor-like 1 (APJ), a member of the seven-transmembrane G protein-coupled receptor (GPCR) family, has high homology with AT1R in amino acid sequence (identity: 27.56%, similarity: 74.28%) [Supplementary Figure 1A, https://links.lww.com/CM9/B887 ]. Apelin is the endogenous ligand of APJ. Published studies report that the Apelin/APJ system participates in various physiological processes of myocardial cells. In the previous study, we demonstrated that the Apelin-13/APJ system promotes the development of cardiac hypertrophy. [ 2 ] Therefore, both Ang II/AT1R and Apelin/APJ may contribute to the development of cardiac hypertrophy.

Abstract:Apelin is the endogenous ligand of a G protein-coupled receptor called APJ, its propeptide is an X-linked and coded peptide with 77 amino acids. Apelin/apelin receptor system is widely distributed in the body and is involved in various physiological and pathological regulations. It has been demonstrated that apelin is associated with the occurrence and development of many diseases, including angiocardiopathy, pulmonary arterial hypertension, nephropathy, cancer, endocrine disease, and so on. Currently, studies on apelin in nervous system have been developed gradually. This article focuses on apelin and summarizes the distribution and pathophysiological roles of apelin in nervous system, as well as in the occurrence and development of diseases associated with nervous system.

Abstract:Apelin has been recognized as an adipokine that plays an important role in regulating energy metabolism and is credited with antiobesity and antidiabetic properties. This study was designed to examine the effect of exogenous apelin on obesity-associated cardiac dysfunction. Oral glucose tolerance test, echocardiography, cardiomyocyte contractile and intracellular Ca2+ properties were assessed in adult C57BL/6J mice fed – low or a – high-fat diet for 24weeks followed by apelin treatment (100nmol/kg, i.p. for 2weeks). High-fat diet resulted in increased left ventricular diastolic and systolic diameters, and wall thickness, compromised fractional shortening, impaired cardiomyocyte mechanics (peak-shortening, maximal velocity of shortening/relengthening, and duration of shortening and relengthening) and compromised intracellular Ca2+ handling, all of which were reconciled by apelin. Apelin treatment also reversed high fat diet-induced changes in intracellular Ca2+ regulatory proteins, ER stress, and autophagy. In addition, microRNAs (miR) -133a, miR-208 and miR-1 which were elevated following high-fat feeding were attenuated by apelin treatment. In cultured cardiomyocytes apelin reconciled palmitic acid-induced cardiomyocyte contractile anomalies. Collectively, these data depict a pivotal role of apelin in obesity-associated cardiac contractile dysfunction, suggesting a therapeutic potential of apelin in the management of cardiac dysfunction associated with obesity.

Abstract:Vascular disease is a common problem with high mortality all over the world. Apelin-13, a key subtype of apelin, takes part in many physiological and pathological responses via regulating many target genes and target molecules or participating in many signaling pathways. More and more studies have demonstrated that apelin-13 is implicated in the onset and progression of vascular disease in recent years. It has been shown that apelin-13 could ameliorate vascular disease by inhibiting inflammation, restraining apoptosis , suppressing oxidative stress , and facilitating autophagy. In this article, we sum up the progress of apelin-13 in the occurrence and development of vascular disease and offer some insightful views about the treatment and prevention strategies of vascular disease.

Apelin Antagonizes Myocardial Impairment In Sepsis

Abstract:Background: Apelin is a cardiovascular peptide with multiple functions regulating homeostasis of the circulatory system and is the endogenous ligand of angiotensin II receptor like-1 (AGTRL1). Apelin has anti-inflammatory and inhibitory effects on release of inflammatory mediators. We aimed to analyze whether apelin antagonizes myocardial impairment in sepsis by attenuating inflammatory responses.Methods and Results: Male rats underwent sepsis by cecal ligation and puncture (CLP) after receiving low- or high-dose apelin for 3 days. Twenty hours later, rats with sepsis showed severe disturbance of hemodynamic features. Reverse transcription-polymerase chain reaction revealed decreased mRNA levels of apelin and AGTRL1 in myocardia of rats with sepsis. Enzyme immune assay detected a lower level of apelin in plasma and myocardia. Western blot analysis revealed decreased level of myocardial AGTRL1 protein. Low- and high-dose apelin administration ameliorated disorders of cardiac function: increased mean arterial blood pressure, attenuated heart rate, elevated +LVdp/dt(max) and LVdp/dt(max), and lowered left ventricular end-diastolic pressure. Rats treated with low- or high-dose apelin showed lower content of plasma monocyte chemoattractant protein land interleukin 8. In cultured rat peritoneal macrophages, apelin directly inhibited the production of monocyte chemoattractant protein 1 and interleukin-8 induced by lipopolysaccharide.Conclusions: These results suggest that apelin antagonizes cardiac impairment in sepsis by attenuating inflammatory responses and might be a promising therapeutic target for severe sepsis and septic shock. Therapeutic potential of apelin and Elabela in cardiovascular disease

Abstract:Apelin and Elabela (Ela) are peptides encoded by APLN and APELA, respectively, which act on their receptor APJ and play crucial roles in the body. Recent research has shown that they not only have important effects on the endocrine system, but also promote vascular development and maintain the homeostasis of myocardial cells. From a molecular biology perspective, we explored the roles of Ela and apelin in the cardiovascular system and summarized the mechanisms of apelin-APJ signaling in the progression of myocardial infarction, ischemia-reperfusion injury, atherosclerosis, pulmonary arterial hypertension, preeclampsia, and congenital heart disease. Evidences indicated that apelin and Ela play important roles in cardiovascular diseases, and there are many studies focused on developing apelin, Ela, and their analogues for clinical treatments. However, the literature on the therapeutic potential of apelin, Ela and their analogues and other APJ agonists in the cardiovascular system is still limited. This review summarized the regulatory pathways of apelin/ELA-APJ axis in cardiovascular function and cardiovascular-related diseases, and the therapeutic effects of their analogues in cardiovascular diseases were also included.
Apelin and APJ Orchestrate Complex Tissue-Specific Control of Cardiomyocyte Hypertrophy and Contractility in the Hypertrophy-Heart Failure Transition.

Abstract:The G protein-coupled receptor APJ is a promising therapeutic target for heart failure. Constitutive deletion of APJ in the mouse is protective against the hypertrophy-heart failure transition via elimination of ligand-independent, β-arrestin-dependent stretch transduction. However, the cellular origin of this stretch transduction and the details of its interaction with apelin signaling remain unknown. We generated mice with conditional elimination of APJ in the endothelium (APJendo-/-) and myocardium (APJmyo-/-). No baseline difference was observed in left ventricular function in APJendo-/-, APJmyo-/-, or control (APJendo+/+, APJmyo+/+) mice. After exposure to transaortic constriction, APJendo-/- mice displayed decreased left ventricular systolic function and increased wall thickness, whereas APJmyo-/- mice were protected. At the cellular level, carbon fiber stretch of freshly isolated single cardiomyocytes demonstrated decreased contractile responses to stretch in APJ-/- cardiomyocytes compared with APJ+/+ cardiomyocytes. Ca2+ transients did not change with stretch in either APJ-/- or APJ+/+ cardiomyocytes. Application of apelin to APJ+/+ cardiomyocytes resulted in decreased Ca2+ transients. Furthermore, hearts of mice treated with apelin exhibited decreased phosphorylation in cardiac troponin I NH2-terminal residues (Ser22 and Ser23) consistent with increased Ca2+ sensitivity. These data establish that APJ stretch transduction is mediated specifically by myocardial APJ, that APJ is necessary for stretch-induced increases in contractility, and that apelin opposes APJ's stretch-mediated hypertrophy signaling by lowering Ca2+ transients while maintaining contractility through myofilament Ca2+ sensitization. These findings underscore apelin's unique potential as a therapeutic agent that can simultaneously support cardiac function and protect against the hypertrophy-heart failure transition. NEW & NOTEWORTHY These data address fundamental gaps in our understanding of apelin-APJ signaling in heart failure by localizing APJ's ligand-independent stretch sensing to the myocardium, identifying a novel mechanism of apelin-APJ inotropy via myofilament Ca2+ sensitization, and identifying potential mitigating effects of apelin in APJ stretch-induced hypertrophic signaling.

Abstract:Hypertension is the leading risk factor for global mortality and morbidity and those with hypertension are more likely to develop severe symptoms in cardiovascular and cerebrovascular system, which is closely related to abnormal renin-angiotensin system and elabela/apelin-apelin receptor (APJ) axis. The elabela/apelin-APJ axis exerts essential roles in regulating blood pressure levels, vascular tone, and cardiovascular dysfunction in hypertension by counterbalancing the action of the angiotensin II/angiotensin II type 1 receptor axis and enhancing the endothelial nitric oxide (NO) synthase/NO signaling. Furthermore, the elabela/apelin-APJ axis demonstrates beneficial effects in cardiovascular physiology and pathophysiology, including angiogenesis, cellular proliferation, fibrosis, apoptosis, oxidative stress, and cardiovascular remodeling and dysfunction during hypertension. More importantly, effects of the elabela/apelin-APJ axis on vascular tone may depend upon blood vessel type or various pathological conditions. Intriguingly, the broad distribution of elabela/apelin and alternative isoforms implicates its distinct functions in diverse cardiac and vascular cells and tissue types. Finally, both loss-of-function and gain-of-function approaches have defined critical roles of the elabela/apelin-APJ axis in reducing the development and severity of hypertensive diseases. Thus, targeting the elabela/apelin-APJ axis has emerged as a pre-warning biomarker and a novel therapeutic approach against progression of hypertension, and an increased understanding of cardiovascular actions of the elabela/apelin-APJ axis will help to develop effective interventions for hypertension. In this review, we focus on the physiology and biochemistry, diverse actions, and underlying mechanisms of the elabela/apelin-APJ axis, highlighting its role in hypertension and hypertensive cardiovascular injury and dysfunction, with a view to provide a prospective strategy for hypertensive disease therapy.

Sepsis can lead to life-threatening organ dysfunction and is one of the main causes of death for patients in the intensive care unit. 1,2 Fluid resuscitation and vasopressors are the cornerstones for sepsis treatment. The major causes for circulatory failure and failed interventions for septic shock are microvascular leakage, irresponsiveness of arteries to vasopressors and myocardial injury. Despite the implementation of organ support measures like fluid resuscitation, vasoactive drugs, inotropic agents, mechanical ventilation and hemodialysis, the overall prognosis of sepsis has not shown improvement. 3 An effective strategy for the prevention and treatment of sepsis is lacking. Figure 1 There is a wide range of physiological effects caused by the activation of APJ receptors in both the central and peripheral nervous systems. (eg vasodilatory, vasoconstrictive, angiogenic, and possibly antithrombotic; increases myocyte conduction velocity, arrhythmogenic, and decreases myocardial hypertrophy and fibrosis; enhances renal blood flow and diuresis, and reduces fibrosis; inhibits the release of vasopressin from the hypothalamus and reduces water intake). Also, the Apelin/APJ system has a variety of metabolic effects. It increases muscle glucose uptake and usage. It also improves insulin sensitivity. Figure 2 Potential impacts of the apelin/APJ system in human sepsis induced organ disfunction. Both apelin and ELA reduce organs inflammation, improve hemodynamics (eg, improvement of inotropy, reduction of pre- and post-load as well as vascular permeability). Also, apelin reduce the inflammation in the brain, lung, kidney and liver. Furthermore, it prevents pulmonary edema and fibrosis, reduce blood brain barrier permeability, and enhance diuresis. Abstract:Apelin, the endogenous ligand of the G protein-coupled APJ receptor, is a peptide mediator with emerging regulatory actions in the heart. We aimed to determine whether the endogenous apelin/APJ system is an intrinsic protective pathway in ischemic/reperfusion injury. A Langendorff model of perfused isolated rat hearts and primary cultured myocardial cells from neonatal rats were used. Cardiac function was monitored and apelin/APJ expression was determined by real-time PCR and Western blot analysis. In rats under I/R, cardiac function was significantly decreased as compared with controls, and APJ was over-expressed at both the mRNA and protein levels (by 7-fold and 35%, respectively, both p<0.01). However, pre-administration of apelin (30pmol/L) greatly ameliorated the reduced heart function. To gain mechanistic insight into the cardio-protective effects of apelin/APJ, cultured cardiomyocytes were treated with apelin (30pmol/L), and those under hypoxia/re-oxygenation showed H/R-induced apoptosis and up-regulated apelin/APJ mRNA expression by 6-fold and 7-fold, respectively (both p<0.01). And lactate dehydrogenase leakage was greatly increased as well. Meanwhile, apoptosis, the generation of reactive oxygen species and malonaldehyde content as well as lactate dehydrogenase leakage were inhibited by apelin. Furthermore, apelin enhanced superoxide dismutase activity and phosphorylation of extracellular signal-regulated kinase 1/2 and Akt after hypoxia/re-oxygenation. In conclusion, apelin/APJ has protective effects in ischemic heart disease and might constitute an important therapy target.

Abstract:Objectives Apelin is a recently-discovered cardiovascular bioactive polypeptide with multiple biological effects, which is the endogenous ligand of the orphan G protein-coupled receptor-APJ receptor and may cause vasodilation and lower blood pressure levels. This study focused on the effects of Apelin on the phosphodiesterase 1 (PDE1) expression and oxidative stress levels in mouse kidney fibroblast cells. Methods The mouse kidney fibroblast cells were primary cultured using the routine method. The angiotensin II (Ang II; 100 nmol/L) was used to stimulate the cells for 1 h in the presence and absence of Aplein (100 nmol/L), PDE1 inhibitor vinpocetine (10 μmol/L) and extracellular regulated protein kinase (ERK) inhibitor PD98059 (10 mmol/L). The specific fluorescent probe-dihydropyridinum (DHE) staining and colorimetric method were used for the determination of superoxide generation and malondialdehyde (MDA) content in cells, respectively. Results In the cultured mouse kidney fibroblast cells, exposure to Ang II (100 nmol/L) obviously promoted protein expressions of PDE1A and PDE1B (n = 5-6; P P P P P Conclusions Treatment withApelin significantly prevents the oxidative stress formation in cultured mouse kidney fibroblast cells in response to Ang II through the modulation of PDE1-ERK1/2 signalling pathway, suggesting the potential anti-oxidative stress effect of Apelin. The enhanced activities and/or expression of Apelin may have potential therapeutic benefits on renal diseases associated with oxidative stress. Apelin activates l-arginine/nitric oxide synthase/nitric oxide pathway in rat aortas

Abstract:Apelin was recently found to be an inotropic polypeptide in isolated rat hearts, and intravenous injection of apelin can induce a transient decrease in blood pressure. To illustrate the mechanism of apelin-induced vasodilation, we observed the in vitro effects of apelin on the l-arginine (l-Arg)/nitric oxide (NO) pathway in the incubated, isolated rat aorta. Apelin stimulated vascular NO2− product and NOS activation in a concentration- and time-dependent manner. Compared with no apelin treatment, incubation with apelin (10−9, 10−8, and 10−7mol/L) increased NO2− product by 33%, 46%, and 69% (all p<0.01), respectively, and Ca2+-dependent constitutive NOS (cNOS) activity by 200%, 460%, and 550% (all p<0.01), respectively. However, Ca2+-independent NOS (iNOS) activity was not significantly altered (p>0.05). Apelin incubation (10−9, 10−8, and 10−7mol/L) increased l-Arg uptake by 130%, 180%, and 240% (all p<0.01), respectively. The mRNA level of cationic amino acid transporters, CAT-1 and CAT-2B, in rat aortic tissues treated with 10−7mol/L apelin was increased by 110% and 128%, respectively (both p<0.01). Incubation with 10−7mol/L apelin elevated eNOS mRNA and protein levels, by 53% (p<0.05) and 319% (p<0.01), respectively. Collectively, these results demonstrate that apelin directly activated the vascular l-Arg/NOS/NO pathway, which could be one of the important mechanisms of apelin-regulated vascular function.

Abstract:Objective: The Apelin/APJ system has recently been implicated in pathologies of hypertension. However, little was elucidated upon effects of Apelin on vascular adventitial remodeling progression. Here, we examined regulatory roles of Apelin in pressure overload-induced adventitial remodeling and fibrosis in hypertensive rats. Design and Method: The male Sprague–Dawley rats were performed with transverse aortic constriction (TAC). The rats with TAC were randomized to daily deliver either pyroglutamyl Apelin-13 (50 &mgr;g/kg) or saline for 4 weeks. Results: Histomorphometric analysis by HE and Masson trichrome staining revealed increased medial and adventitial thicknesses, especially in the adventitia, in ascending aortas in rats with TAC when compared with the sham-operated rats. Downregulation of APJ receptor and elevations in systolic blood pressure, phosphorylated mTOR and ERK1/2 levels were observed in hypertensive rats with TAC. Pressure overload-mediated by TAC led to marked increases in heart weight (HW), HW/body weight ratio, malonyldialdehyde (MDA) contents and aortic fibrosis in the hypertensive rats with enhanced activation of NADPH oxidase activity. The pressure overload-mediated pathological adventitial remodeling was strikingly rescued by Apelin-13 supplementation, associated with attenuation of aortic fibrosis and reduced mRNA expression of TGF-ß1 and collagen I. In cultured rat adventitial fibroblasts, exposure with Ang II (100 nmol L−1) resulted in obvious increases in MDA, phosphorylated levels of mTOR and ERK1/2 and mRNA levels of TGF-ß1, collagen I and fibronectin 1, which were dramatically prevented by Apelin-13 (100 nmol L−1) or mTOR inhibitor rapamycin (10 &mgr;mol L−1). However, Apelin-13 had no effect on collagen III levels. Conclusions: Our results demonstrate the importance of Apelin-13 in amelioration of aortic oxidative stress, adventitial remodeling and fibrosis in hypertensive rats with TAC via modulation of the mTOR/ERK signaling, thus indicating potential therapeutic strategies by enhancing Apelin/APJ action for preventing hypertensive vascular adventitial remodeling and fibrosis and pressure overload-associated cardiovascular disorders.
