Abstract:Zinc is one of trace elements that play essential roles in several cell functions, and is unquestionably important to the normal health and function of the central nervous system. Growing evidence suggests that Zn2+ can become a pathogenic agent in certain neurological disease states, such as ischemia, seizures, and trauma. The main role of the Zn2+ may serve as an endogenous neuromodulator in the brain. In the present study, we used the electrophysiology method to investigate the effects of Zn2+ on the excitability of hippocampus CA1 region. Our results have demonstrated that the Zn2+ activates the Wistar rat hippocampal CA1 region network by significantly enhancing the spike rate of the spontaneous firing. In addition, Zn2+ can increase the intrinsic membrane excitability by enhancing the firing rate and half-width of the evoked action potential. Meanwhile, our results also indicate that Zn2+ can effectively inhibit voltage-dependent potassium currents (both transient outward potassium currents and delayed rectifier potassium currents). On the other hand, Zn2+ also inhibits excitatory neurotransmitter release by decreasing the inter-event interval and the total charge transfer of the excitatory postsynaptic currents. The present results, in combination with other works, suggest that Zn2+ can influence neuronal excitability, intrinsic membrane excitability and synaptic transmission in the hippocampus CA1 neurons by multiple mechanisms.

Abstract:In the central nervous system(CNS),zinc has a modulatory role of the ligand-gated ion channels. This divalent cation is released from presynaptic vesicles into the synaptic cleft upon neuronal activity and acts as a modulator of many ligand-gated ion channels.The NMDA-type glutamate receptors are inhibited by zinc,whereas non-NMDA type glutamate receptors appear to be differently affected.Furthermore,inhibitory transmission mediated via GABA receptors could be suppressed by zinc;however,zinc may bidirectionally modulate glycinergic transmission in a dose-dependent way.In pathological conditions,zinc effect also contributes to the excitotoxicity-induced neuronal death.Taken together,this review highlights the putative roles for zinc involved in the synaptic transmissions mediated by the ligand-gated ion channels in the CNS. Furthermore,the physiological and pathological roles of the zinc modulation are also discussed in this review.

Abstract:Objective To investigate the effect of zinc deficiency on hippocampal zinc content and long term potentiation (LTP) in the mouse. Methods CD-1 mice at 3 weeks of age were fed with zinc-deficient diet (0.85mg/kg) for 5 weeks. Autometallography (AMG) was carried out to analyze the zinc content in the hippocampus. A concentric bipolar stimulating electrode was placed in the orientation of the mossy fiber lamellae of the right dentate gyrus (DG) and a glass capillary recording electrode was placed ipsilaterally in the stratum pyramidal of the CA3 region of the hippocampus. LTP was evoked by a train of high frequency stimulations (HFS), and the changes of population spike (PS) and field excitatory postsynaptic potential (f-EPSP) were recorded, in order to analyze the effect of LTP under zinc deficiency. Results AMG staining showed that treatment with zinc-deficient diet reduced the level of zinc in hippocampal CA1, CA3 and dentate gyrus ( P0.05-0.01). Electrophysiological in vivo recordings showed that zinc deficiency resulted in impairments of mossy fiber LTP ( P0.01). Conclusion Zinc deficiency results in a decreased level of free zinc ions in the hippocampus, which is associated with the inhibition of the formation of LTP.

Abstract:In the nervous system, the trace metal ion zinc is required for normal mammalian brain development and physiology. Zinc homeostasis is essential for the control of physiological and pathophysiological brain functions. Synapses, the junctions between neurons, are the center of the brain’s information transmission. Zinc deficiency or excess leads to neurological disorders. However, it is still unclear whether and how zinc ion regulates synapse formation. Here, we investigated the effect of zinc on synapse formation in a cultured neuron system, and found that synapse formation and synaptic transmission were regulated by zinc ions. Finally, we identified that PTPRM is the key gene involved in synapse formation regulated by zinc ions. This study provides a new perspective to understanding the regulation of brain function by zinc ion.

Abstract:Zinc, a suspected potentiator of learning and memory, is shown to affect exocytotic release and storage in neurotransmitter-containing vesicles. Structural and size analysis of the vesicular dense core and halo using transmission electron microscopy was combined with single-cell amperometry to study the vesicle size changes induced after zinc treatment and to compare these changes to theoretical predictions based on the concept of partial release as opposed to full quantal release. This powerful combined analytical approach establishes the existence of an unsuspected strong link between vesicle structure and exocytotic dynamics which can be used to explain the mechanism of regulation of synaptic plasticity by Zn 2+ through modulation of neurotransmitter release.

Abstract:Zinc is one of the most abundant metal ions in the central nervous system (CNS), where it plays a crucial role in both physiological and pathological brain functions. Zinc promotes antioxidant effects, neurogenesis, and immune system responses. From neonatal brain development to the preservation and control of adult brain function, zinc is a vital homeostatic component of the CNS. Molecularly, zinc regulates gene expression with transcription factors and activates dozens of enzymes involved in neuronal metabolism. During development and in adulthood, zinc acts as a regulator of synaptic activity and neuronal plasticity at the cellular level. There are several neurological diseases that may be affected by changes in zinc status, and these include stroke, neurodegenerative diseases, traumatic brain injuries, and depression. Accordingly, zinc deficiency may result in declines in cognition and learning and an increase in oxidative stress, while zinc accumulation may lead to neurotoxicity and neuronal cell death. In this review, we explore the mechanisms of brain zinc balance, the role of zinc in neurological diseases, and strategies affecting zinc for the prevention and treatment of these diseases.

Abstract:In addition to the essential structural and catalytic functions of zinc, evolution has adopted synaptic zinc as a neuromodulator. In the brain, synaptic zinc is released from primarily glutamatergic neurons, notably in the neocortex, hippocampus, amygdala, and auditory brainstem. In these brain areas, synaptic zinc is essential for neuronal and sensory processing fine-tuning. But what niche does zinc fill in neural signaling that other neuromodulators do not? Here we discuss the evolutionary history of zinc as a signaling agent and its eventual adoption as an essential neuromodulator in the mammalian brain. We then attempt to describe the roles that zinc has carved out of the vast and diverse landscape of neuromodulators.

Abstract:Zinc, an essential micronutrient for life, was first discovered in 1869 and later found to be indispensable for the normal development of plants and for the normal growth of rats and birds. Zinc plays an important role in many physiological and pathological processes in normal mammalian brain development, especially in the development of the central nervous system. Zinc deficiency can lead to neurodegenerative diseases, mental abnormalities, sleep disorders, tumors, vascular diseases, and other pathological conditions, which can cause cognitive impairment and premature aging. This study aimed to review the important effects of zinc and zinc-associated proteins in cognitive impairment and aging, to reveal its molecular mechanism, and to highlight potential interventions for zinc-associated aging and cognitive impairments.

Abstract:Zinc is released from glutamatergic (zincergic) neuron terminals in the brain, followed by the increase in Zn2+ concentration in the intracellular (cytosol) compartment as well as that in the extracellular compartment. Intracellular Zn2+ concentration mainly increases through calcium-permeable channels and serves as Zn2+ signal as well as extracellular Zn2+ concentration. Hippocampal Zn2+ signaling may participate in synaptic plasticity such as long-term potentiation and cognitive function. On the other hand, subclinical zinc deficiency is common in the old who might be more susceptible to depression. Zinc deficiency causes abnormal glucocorticoid secretion and increases depression-like behavior in animals. Neuropsychological symptoms are observed prior to the decrease in Zn2+ signal in the hippocampus under zinc deficiency. This paper summarizes that hippocampal Zn2+ signaling serves to maintain healthy brain and that glucocorticoid signaling, which is responsive to zinc homeostasis in the living body, is linked to the pathophysiology of depression.

Abstract:Zinc (Zn2+) is the second most abundant necessary trace element in the human body, exerting a critical role in many physiological processes such as cellular proliferation, transcription, apoptosis, growth, immunity, and wound healing. It is an essential catalyst ion for many enzymes and transcription factors. The maintenance of Zn2+ homeostasis is essential for the central nervous system, in which Zn2+ is abundantly distributed and accumulates in presynaptic vesicles. Synaptic Zn2+ is necessary for neural transmission, playing a pivotal role in neurogenesis, cognition, memory, and learning. Emerging data suggest that disruption of Zn2+ homeostasis is associated with several central nervous system disorders including Alzheimer's disease, depression, Parkinson's disease, multiple sclerosis, schizophrenia, epilepsy, and traumatic brain injury. Here, we reviewed the correlation between Zn2+ and these central nervous system disorders. The potential mechanisms were also included. We hope that this review can provide new clues for the prevention and treatment of nervous system disorders.

Abstract:<span class="a-plus-plus abstract-section id-a-sec1"><h3 class="a-plus-plus">Background</h3><p class="a-plus-plus">Zinc (Zn) has a diverse role in many biological processes, such as growth, immunity, anti-oxidation system, homeostatic, and repairing. It acts as a regulatory and structural catalyst ion for activities of various proteins, enzymes, and signal transcription factors, as well as cell proliferation, differentiation, and survival. The Zn ion is essential for neuronal signaling and is mainly distributed within presynaptic vesicles. Zn modulates neuronal plasticity and synaptic activity in both neonatal and adult stages. Alterations in brain Zn status results in a dozen neurological diseases including impaired brain development. Numerous researchers are working on neurogenesis, however, there is a paucity of knowledge about neurogenesis, especially in neurogenesis in adults.</p></span><span class="a-plus-plus abstract-section id-a-sec2"><h3 class="a-plus-plus">Summary</h3><p class="a-plus-plus">Neurogenesis is a multifactorial process and is regulated by many metal ions (e.g. Fe, Cu, Zn, etc.). Among them, Zn has an essential role in neurogenesis. At the molecular level, Zn controls cell cycle, apoptosis, and binding of DNA and several proteins including transcriptional and translational factors. Zn is needed for protein folding and function and Zn acts as an anti-apoptotic agent; organelle stabilizer; and an anti-inflammatory agent. Zn deficiency results in aging, neurodegenerative disease, immune deficiency, abnormal growth, cancer, and other symptoms. Prenatal deficiency of Zn results in developmental disorders in humans and animals.</p></span><span class="a-plus-plus abstract-section id-a-sec3"><h3 class="a-plus-plus">Conclusion</h3><p class="a-plus-plus">Both in vitro and in vivo studies have shown an association between Zn deficiency and increased risk of neurological disorders. This article reviews the existing knowledge on the role of Zn and its importance in neurogenesis.</p></span>

Abstract:Abstract The subgranular zone of the dentate gyrus is a subregion of the hippocampus that has two uniquely defining features; it is one of the most active sites of adult neurogenesis as well as the location where the highest concentrations of synaptic zinc are found, the mossy fiber terminals. Therefore, we sought to investigate the idea that vesicular zinc plays a role as a modulator of hippocampal adult neurogenesis. Here, we used ZnT3−/− mice, which are depleted of synaptic-vesicle zinc, to test the effect of targeted deletion of this transporter on adult neurogenesis. We found that this manipulation reduced progenitor cell turnover as well as led to a marked defect in the maturation of newborn cells that survive in the DG toward a neuronal phenotype. We also investigated the effects of zinc (ZnCl2), n-acetyl cysteine (NAC), and ZnCl2 plus 2NAC (ZN) supplement on adult hippocampal neurogenesis. Compared with ZnCl2 or NAC, administration of ZN resulted in an increase in proliferation of progenitor cells and neuroblast. ZN also rescued the ZnT3 loss-associated reduction of neurogenesis via elevation of insulin-like growth factor-1 and ERK/CREB activation. Together, these findings reveal that ZnT3 plays a highly important role in maintaining adult hippocampal neurogenesis and supplementation by ZN has a beneficial effect on hippocampal neurogenesis, as well as providing a therapeutic target for enhanced neuroprotection and repair after injury as demonstrated by its ability to prevent aging-dependent cognitive decline in ZnT3−/− mice. Therefore, the present study suggests that ZnT3 and vesicular zinc are essential for adult hippocampal neurogenesis.

Abstract:We applied electrochemical techniques with nano-tip electrodes to show that micromolar concentrations of zinc not only trigger changes in the dynamics of exocytosis, but also vesicle content in a model cell line. The vesicle catecholamine content in PC12 cells is significantly decreased after 100 μm zinc treatment, but, catecholamine release during exocytosis remains nearly the same. This contrasts with the number of molecules stored in the exocytosis vesicles, which decreases, and we find that the amount of catecholamine released from zinc-treated cells reaches nearly 100 % content expelled. Further investigation shows that zinc slows down exocytotic release. Our results provide the missing link between zinc and the regulation of neurotransmitter release processes, which might be important in memory formation and storage.
Chronic Zinc Exposure Decreases the Surface Expression of NR2A-Containing NMDA Receptors in Cultured Hippocampal Neurons

Abstract:Background Zinc distributes widely in the central nervous system, especially in the hippocampus, amygdala and cortex. The dynamic balance of zinc is critical for neuronal functions. Zinc modulates the activity of N-methyl-D-aspartate receptors (NMDARs) through the direct inhibition and various intracellular signaling pathways. Abnormal NMDAR activities have been implicated in the aetiology of many brain diseases. Sustained zinc accumulation in the extracellular fluid is known to link to pathological conditions. However, the mechanism linking this chronic zinc exposure and NMDAR dysfunction is poorly understood. Methodology/Principal Findings We reported that chronic zinc exposure reduced the numbers of NR1 and NR2A clusters in cultured hippocampal pyramidal neurons. Whole-cell and synaptic NR2A-mediated currents also decreased. By contrast, zinc did not affect NR2B, suggesting that chronic zinc exposure specifically influences NR2A-containg NMDARs. Surface biotinylation indicated that zinc exposure attenuated the membrane expression of NR1 and NR2A, which might arise from to the dissociation of the NR2A-PSD-95-Src complex. Conclusions Chronic zinc exposure perturbs the interaction of NR2A to PSD-95 and causes the disorder of NMDARs in hippocampal neurons, suggesting a novel action of zinc distinct from its acute effects on NMDAR activity.
Effects of Zinc Deficiency on Zinc Ion and Zinc Transporter 4 Expression in Hippocampal Neurons

Abstract:Objective To investigate the effects of zinc deficiency on the zinc ion and zinc transporter 4 expression in hippocampal neurons.Method CD-1 mice were fed with zinc deficient diets or normal diets after weaning.The hippocampi were taken after five weeks.Autometallography,histochemistry and Western blotting were used for the detection of zinc ion content,and qualitative and quantitative detection of zinc transporter 4 expressions in the hippocampal neurons in both groups.Results The zinc ion contents were decreased in various areas of the hippocampus.And the expression of zinc transporter 4 was downregulated after zinc deficiency for 5 weeks.Conclusion The zinc deficiency might result in a decrease of zinc ion and a down-regulation expression of zinc transporter 4 in hippocampus,which may affect the homeostasis of zinc and finally affect mice's learning and memory function.

Abstract:Dietary zinc deficiency may affect zinc homeostasis in the brain and lead to reductions of neurogenesis and neuronal survival. However, the mechanisms responsible for the effects of zinc deficiency on hippocampal neurogenesis and neuronal death remain obscure. In the present study, young CD-1 mice were fed with zinc-deficient diet (0.85 ppm) for 5 weeks. The vesicular zinc was reduced at CA1 and CA3 regions of the hippocampus in zinc-deficient mice. The significant decreased zinc ions was associated with a reduction in proliferating cells labeled with bromo-deoxyuridine (BrdU) and immature neurons labeled with doublecortin (DCX) immunoreactivity in the dentate gyrus of the hippocampus. The processes of DCX-positive neurons were shortened, and flexuously went through into the granular cell layer in zinc-deficient hippocampus. There was also a conspicuous increase in the number of TUNEL-positive cells in the hippocampus after zinc-deficient diet treatment. Meanwhile, the apoptosis proteins, including Fas, Fas ligand (FasL), apoptosis inducing factor (AIF), and caspase-3, were significantly activated in zinc-deficient mouse hippocampus. These data suggest that chronic treatment with zinc-deficient diet results in reduction in hippocampal neurogenesis and increases neuronal apoptosis, indicating that zinc deficiency is associated with destroying structural plasticity in the hippocampus.

Abstract:Zinc ions are colocalized with glutamatergic synaptic vesicles in vertebrate photoreceptors and may act as a diffusible molecular switch regulating neurotransmitter signaling at two distinct sites in the outer retina. In the dark, extracellular zinc acts presynaptically at rods and cones to minimize the depletion of tonically released glutamate, and selectively reduces GABA-mediated depolarization in horizontal cells, accelerating the response kinetics of the second-order cells. The discovery of zinc ions in photoreceptors provides a mechanism for gain control, kinetics modulation, and the balance of rod vs cone output at the first synapse in the visual system.

Abstract:Zinc is an essential trace element presenting in particularly high concentration in the brain. In some regions, e.g. lateral amygdala, subiculum and hippocampus, rapidly-exchangeable zinc may transiently reach even up to 600 μM. To explore the possible roles of high-concentration Zn2+ in regulating the blood-brain barrier (BBB), we investigated the effects of Zn2+ on the functions and structures of the tight junction (TJ) with an in vitro model of a Madin-Darby canine kidney (MDCK) cell monolayer. The experimental results indicated that high concentrations (>200 μM) of Zn2+ can affect the TJ integrity in a polarized manner. Basolateral addition of Zn2+ led to reversible TJ opening with pore paths of r ∼ 2 nm or more depending on Zn2+ concentration. The efflux/influx ratios of different sized probes were found to be ∼4.6 for FD4 (MW 4000) and ∼1.8 for Eu-DTPA (MW 560), suggesting that the Zn2+-induced paracelluar channels favour efflux especially for macromolecules. Further mechanistic studies revealed that the elevated intracellular Zn2+ taken from the basolateral side can increase phosphorylation of glycogen synthase kinase (GSK) 3β, primarily due to the inhibition of calcineurin (CaN), thus resulting in the elevation of the snail transcriptional repressors. Subsequently, Zn2+ can cause the down-regulation of claudin-1, breakage of occludin and ZO-1 rings, and collapse of basolateral F-actin structures. These overall factors result in the formation of a trumpet-like paracellular channel, which allows asymmetric solute permeation. The ERK1/2 and JNK1/2 pathways may also be involved in the Zn2+-induced TJ opening process, while the activation of matrix metalloproteinase was not observed. Our results may suggest a potential role of zinc in regulation of BBB permeability associated with brain clearance of metabolites through the glymphatic system.
