Determination of Five Main Free Amino Acids in Kangfuxin Liquid by HPLC

Abstract:Objective To establish a quantitative method with dinitrofluorobenzene DNFB pro-column derivtization for determining the five main free amino acids in kangfuxin liquid by HPLC.Methods Contents of glycine,proline,alanine,valine,and leucine were determined in kangfuxin liquid by HPLC using DNFB as the derivatization reagent.The analysis was performed on Elite-AAK C18 chromatographic column(4.6 mm×250 mm,5 μm)at 27 ℃,using 0.05 mol·L-1 sodium acetate solution(containing 1% DMF,pH6.8) and 50% acetonitrile with gradient elution as the mobile phase at a flow rate of 1.2 mL·min-1.The detection wavelength was 360 nm,and the injection volume was 20 μL.Results The linear range of glycine was 2.25-18.06 μg,and the proline1.81-14.53 μg,the alanine 3.73-29.8 μg,the valine1.76-14.12 μg,and the leucine 2.06-16.48 μg,with the correlations coefficient of 0.999 9.The average recoveries of the glycine,proline,alanine,valine,and leucine were 98.53%,99.60%,100.26%,96.90%,99.91% with the average RSD being 1.99%,1.68%,1.85%,1.69%,1.80%(n=9),respectively.There was great difference in the content of five main free amino acids from four different manufacturers.Conclusion This method is convenient,accurate and steady.
