Abstract:Drug-impaired driving has surpassed alcohol-impaired driving become a major cause of death in car accidents in US. The emergence of new psychoactive drugs and the recreational marijuana legalization in more and more countries and regions have made the situation even more complex. Conventional drug detection methods, such as chromatography-mass spectrometry, were of high selective and accuracy, however, the tedious analytical process makes it difficult to meet the needs for rapid screening of drug on the roadside. Oral fluid testing has become a significant direction for roadside drug testing because of its convenient and non-invasive operation. This review summarizes the currently available techniques for identification/quantification of drugs in oral fluid, and we conclude the review with outlining the roles and applications of testing for drugs in oral fluid, describing the difficulties associated with this form of testing and illustrating where efforts should be directed to answer the most pressing open question. Abstract:Bioanalytical methods are used to create a protocol that can identify and quantify bioactive molecules and their metabolites in human and animal samples. For determining the medication and its metabolites present in human/animal body, the bioanalytical method is efficient. One of the prominent bioanalytical roles is creating new techniques, validating existing processes, and analyzing samples. Most importantly, a compound can be calculated using many methods and can also be detected using various analytical methods. In specific biological samples, the drugs can be evaluated by different extraction procedures, such as solid-phase extraction, liquid extraction, protein precipitation etc.., All the steps in methods were investigated to know how environment, sample, or techniques influence the matrix assessment up to the study's time. The analytical methods, including high-pressure (HPLC), liquid chromatography combined with dual-mass spectroscopy (LCMS-MS), and Ultra-performance extraction (UPLC), can be used to obtain the in-depth analysis of medications. All have their strengths and weaknesses. Currently, bioanalysis is usually done by HPLC-MS and GCMS-MS. Linearity, repeatability, Accuracy, precision, selectivity, recovery and consistency are essential criteria. In this review we proposed to give importance of bioanalytical system production and validation parameters to aid in drug quality, purity, safety and efficacy.
Analytical Quality by Design Oriented Development of the UPLC Method for Analysing Multiple Pharmaceutical Process Intermediates: A Case Study of Compound Danshen Dripping Pills

Abstract:This work presented the integration of the Analytical Quality by Design concept in developing a UPLC method for the quantification of saponins and phenolic acids in two Compound Danshen Dripping Pills intermediates of a concentrated extract and a supernatant. Prior knowledge and systematic risk analysis were conducted to identify potential critical method parameters (CMPs) that affected critical quality attributes (CQAs). A definitive screening design was employed to investigate the quantitative relationship among the intermediate type, CPPs, and CQAs. Intermediate type was added in the experimental design table as a categorical variable. After the models were built, the method operable design region (MODR) was calculated with an exhaustive search-Monte Carlo method. The CPP combinations with a 95 % or higher probability of reaching the preset standard were collected in MODR. The MODR robustness test was conducted with a Plackett-Burman design. The separation of analytes was achieved with a Waters Acquity HSS T3 (100 x 2.1 mm, 1.8 mu m) column at 30 degrees C, using 0.05 % phosphoric acid and acetonitrile as the mobile phases in gradient elution at a flow rate of 0.40 mL/min. The elution was monitored at UV detection wavelengths of 280 nm and 203 nm. The optimized method was also validated. Good robustness, accuracy, precision, stability and repeatability, and linear regression (R > 0.9997) were obtained. All recoveries of the 9 compounds ranged from 97.8 % to 101.7 %, with an RSD of less than 3.49 %. Nine batches of Danshensu, protocatechualdehyde, rosmarinic acid, salvianolic acid B, salvianolic acid A, notoginsenoside R1, ginsenoside Rg1, ginsenoside Re, and ginsenoside Rb1 were analysed. Danshensu was found to be the compound with the highest content.

Abstract:The aim of this study was to present a novel analytical quality by design (AQbD) approach for developing an HPLC method to analyze herbal extracts. In this approach, critical method attributes (CMAs) and critical method parameters (CMPs) of the analytical method were determined using the same data collected from screening experiments. The HPLC-ELSD method for separation and quantification of sugars in Codonopsis Radix extract (CRE) samples and Astragali Radix extract (ARE) samples was developed as an example method with a novel AQbD approach. Potential CMAs and potential CMPs were found with Analytical Target Profile. After the screening experiments, the retention time of the D-glucose peak of CRE samples, the signal-to-noise ratio of the D-glucose peak of CRE samples, and retention time of the sucrose peak in ARE samples were considered CMAs. The initial and final composition of the mobile phase, flow rate, and column temperature were found to be CMPs using a standard partial regression coefficient method. The probability-based design space was calculated using a Monte-Carlo simulation method and verified by experiments. The optimized method was validated to be accurate and precise, and then it was applied in the analysis of CRE and ARE samples. The present AQbD approach is efficient and suitable for analysis objects with complex compositions.

Abstract:INTRODUCTION:Charged aerosol detection (CAD) has the merits of high sensitivity, high universality and response uniformity. The strategy that combines the quantitative analysis of multi-components by single marker (QAMS) with CAD has certain advantages for the quantification of multi-components. However, relevant research was limited.OBJECTIVES:To comprehensively investigate the crucial factors that affect the performance of the HPLC-CAD-QAMS approach and validate the credibility and feasibility of the method.METHODOLOGY:Multiple components of Qishen Yiqi dripping pills (QSYQ) were assayed using the high-performance liquid chromatography (HPLC)-CAD-QAMS approach. Some factors that affect the sensitivity and accuracy of the approach were sufficiently studied. After the method verification, principal component analysis (PCA) was applied to evaluate the quality consistency of three types of samples: normal samples, expired samples and negative samples.RESULTS:A HPLC-CAD-QAMS method was successfully developed for the multi-component determination of QSYQ. First, chromatographic conditions were optimised by a definitive screening design, and the optimised ranges of operating parameters were obtained with a Monte Carlo simulation method. Next, a new method to select the internal reference standards was successfully introduced based on the heatmap of Pearson correlation coefficients of the response factors. Then, the multi-point method was selected to calculate the relative correction factors, and a robustness test was conducted with Plackett-Burman design. Finally, the PCA was proved to be effective for the quality consistency evaluation of different samples.CONCLUSION:The developed HPLC-CAD-QAMS method can be a reliable and superior means for the multi-component quantitative analysis of QSYQ.

Abstract:Pharmaceutical analysis plays a significant role in pharmaceutical formulation quality assurance and control. Due to the pharmaceutical industries' rapid expansion and the production of pharmaceuticals all over the world, there is a greater need for novel analytical procedures in this sector. Establishing the identification, purity, physical properties, and potency of medications as well as the medication's bioavailability and stability is the goal of analytical method development. A few new drug applications were recently given regulatory flexibility by the Food and Drug Administration for an analytical method based on quality by design. With Quality by design, product design and development are performed methodically. Analytical methodologies have similar opportunities for implementing Quality by design as production procedures do. It consequently enhances formulation design, development efficiency, and capacity. The underpinnings of the QbD approach have been explored in this article due to their use in the creation and validation of analytical procedures. Additionally, a summary of experimental studies reporting the application of the QbD methodology to method development is included.

Abstract:Bioanalysis is an essential part in drug discovery and development. Bioanalysis is related to the analysis of analytes (drugs, metabolites, biomarkers) in biological samples and it involves several steps from sample collection to sample analysis and data reporting. The first step is sample collection from clinical or preclinical studies; then sending the samples to laboratory for analysis. Second step is sample preparation and it is very important step in bioanalysis. In order to reach reliable results, a robust and stable sample preparation method should be applied. The role of sample preparation is to remove interferences from sample matrix and improve analytical system performance. Sample preparation is often labour intensive and time consuming. This guideline defines key elements necessary for the validation of bioanalytical methods. The guideline focuses on the validation of the bioanalytical methods generating quantitative concentration data used for pharmacokinetic and toxicokinetic parameter determinations. Guidance and criteria are given on the application of these validated methods in the routine analysis of study samples from animal and human studies. Measurement of drug concentrations in biological matrices (such as serum, plasma, blood, urine, and saliva) is an important aspect of medicinal product development. It is therefore paramount that the applied bioanalytical methods used are well characterised, fully validated and documented to a satisfactory standard in order to yield reliable results. This review provides an overview of bioanalytical method development and validation and main principles of method validation stages discussed. Abstract:IntroductionThe concept of quality by design (QbD) has been widely accepted and applied in the pharmaceutical manufacturing industry. There are still two key issues to be addressed in the implementation of QbD for herbal drugs. The first issue is the quality variation of herbal raw materials and the second issue is the difficulty in defining the acceptable ranges of critical quality attributes (CQAs). ObjectiveTo propose a feedforward control strategy and a method for defining the acceptable ranges of CQAs for the two issues. MethodsIn the case study of the ethanol precipitation process of Danshen (Radix Salvia miltiorrhiza) injection, regression models linking input material attributes and process parameters to CQAs were built first and an optimisation model for calculating the best process parameters according to the input materials was established. Then, the feasible material space was defined and the acceptable ranges of CQAs for the previous process were determined. ResultsIn the case study, satisfactory regression models were built with cross-validated regression coefficients (Q(2)) all above 91 %. The feedforward control strategy was applied successfully to compensate the quality variation of the input materials, which was able to control the CQAs in the 90-110 % ranges of the desired values. In addition, the feasible material space for the ethanol precipitation process was built successfully, which showed the acceptable ranges of the CQAs for the concentration process. ConclusionThe proposed methodology can help to promote the implementation of QbD for herbal drugs. Copyright (c) 2013 John Wiley & Sons, Ltd. Abstract:IntroductionThe batch-to-batch quality consistency of herbal drugs has always been an important issue.ObjectivesTo propose a methodology for batch-to-batch quality control based on HPLC-MS fingerprints and process knowledgebase.MethodsThe extraction process of Compound E-jiao Oral Liquid was taken as a case study. After establishing the HPLC-MS fingerprint analysis method, the fingerprints of the extract solutions produced under normal and abnormal operation conditions were obtained. Multivariate statistical models were built for fault detection and a discriminant analysis model was built using the probabilistic discriminant partial-least-squares method for fault diagnosis.ResultsBased on multivariate statistical analysis, process knowledge was acquired and the cause-effect relationship between process deviations and quality defects was revealed. The quality defects were detected successfully by multivariate statistical control charts and the type of process deviations were diagnosed correctly by discriminant analysis.ConclusionThis work has demonstrated the benefits of combining HPLC-MS fingerprints, process knowledge and multivariate analysis for the quality control of herbal drugs. Optimization and Validation of the Analytical Method to Detect Common Illicit Drugs in Wastewater

Abstract:Illicit drugs have been recognized as a group of emerging pollutants.Based on previous research,procedures to pre-treat wastewater including SPE column,pH of water samples,and processes of washing,acidifying and re-dissolving were compared.It was found that the best recovery was achieved when wastewater was adjusted to pH =2 and Oasis MCX column was used.It was not necessary to flush the loaded column or to acidify the eluate prior to evaporation using nitrogen steam.After evaporation,it is recommended to redissolve the evaporated samples using 200 μL acetonitrile and 100 μL acetonitrile + 100 μL 5 mmol· L-1 ammonium acetate in Milli-Q water for HILIC-UPLC-MS/MS and C18-UPLC-MS/MS,respectively.After comparing retention times,limit of detection,limit of quantification,recoveries,and matrix effects of HILIC and C18 methods,C18-UPLC-MS/MS was chosen.In fluent and effluent samples from 12 wastewater treatment plants in Beijing were analyzed using the optimized method for validation.The validated methods set a firm foundation to apply the sewage-based epidemiology to monitor illicit drug abuse in China.

Abstract:Botanical drug products have batch-to-batch quality variability due to botanical raw materials and the current manufacturing process. The rational evaluation and control of product quality consistency are essential to ensure the efficacy and safety. Chromatographic fingerprinting is an important and widely used tool to characterize the chemical composition of botanical drug products. Multivariate statistical analysis has showed its efficacy and applicability in the quality evaluation of many kinds of industrial products. In this paper, the combined use of multivariate statistical analysis and chromatographic fingerprinting is presented here to evaluate batch-to-batch quality consistency of botanical drug products. A typical botanical drug product in China, Shenmai injection, was selected as the example to demonstrate the feasibility of this approach. The high-performance liquid chromatographic fingerprint data of historical batches were collected from a traditional Chinese medicine manufacturing factory. Characteristic peaks were weighted by their variability among production batches. A principal component analysis model was established after outliers were modified or removed. Multivariate (Hotelling T 2 and DModX) control charts were finally successfully applied to evaluate the quality consistency. The results suggest useful applications for a combination of multivariate statistical analysis with chromatographic fingerprinting in batch-to-batch quality consistency evaluation for the manufacture of botanical drug products.

Abstract:Near-infrared (NIR) spectroscopy is one of the most successful pharmaceutical process analytical tools. For botanical drug products (BDPs), many studies have applied NIR spectroscopy for rapid analysis of botanical raw materials, extracts and formulations. However, the real-time process analysis reported for BDPs was still mainly conducted on lab- or pilot- scale equipment, where some essential conditions of the materials and process parameters can be easily controlled for NIR spectral measurement. Due to the chemical and physical characteristics of the commercial production of BDPs, it is challenging to develop in-line NIR methods with enough robustness for industrial-scale application. This is the first reported case study of the development and validation of the in-line NIR method for the commercial production of BDPs, taking Compound Danshen Dripping Pill (CDDP) as an example. An in-line NIR method was developed for simultaneous measurement of the three critical quality attributes, i.e. the relative density, the moisture content and the content of 3,4-dihydroxyphenyl lactic acid (danshensu, a key active compound), during the extract concentration process. The NIR spectra and sample collection lasted for three years (67 production batches) to cover the variability of raw materials and process conditions. NIR calibration models were established respectively, with determination coefficients (r2) of 0.9905, root mean square errors of prediction (RMSEP) of 0.004 for the relative density in the range of 1.042-1.184; r2 of 0.9870, RMSEP of 1.1% for the moisture content in the range of 50.8-83.0%; r2 of 0.9870, RMSEP of 0.461 mg/g for danshensu in the range of 2.563-8.869 mg/g. Then, all the method validation parameters (accuracy, precision, range, specificity, linearity, robustness, detection and quantitation limits) were discussed according to the characteristics of the commercial production of BDPs. The NIR method development and validation strategy proposed may also be applied in the future for the commercial production of other BDPs.

Abstract:Ademetionine 1,4-Butanedisulfonate (SAMe) is widely used as a prescription drug to treat cholestasis associated with liver disease, and is also used to treat depression, Alzheimer's disease and other diseases. Currently, the main way to produce SAMe is by fermentation of S-adenosylmethionine synthetase（SAMS）. However, during the fermentation process, host cells produce contaminants unrelated to the treatment. Among them, host cell protein (HCP), which is co-purified with the drug, is the main impurity in the production process. HCP-induced antigenic reactions can lead to allergies or other adverse reactions and affect drug quality, efficacy, and safety. Enzyme-linked immunosorbent assay (ELISA) and mass spectrometry (MS) are currently used as the main analytical methods for measuring HCP. The ELISA method may ignore some HCPs with low immunogenicity, while mass spectrometry is primarily employed for the characterization of proteomes. Orthogonal methods can more effectively evaluate impurities in drugs. In this study, ELISA was initially utilized to quantify the content of Saccharomyces cerevisiae host protein in 11 batches of SAMe active pharmaceutical ingredient (API) from two suppliers, then the above APIs were qualitatively analyzed by MS method. Following the identification of surrogate peptide of SAMS, an Ultra Performance Liquid Chromatography-Tandem Mass Spectrometry（UPLC-MS/MS）method was established and validated, the SAMS protein residues in 11 batches of samples were subsequently determined. The results demonstrated that the levels of Saccharomyces cerevisiae host protein and SAMS in the samples were both low, with values below 20 ppm and 1.19 ppm, respectively. The qualitative results also indicate that the types of peptide residues in the samples are more diverse than those of protein residues. Furthermore, residues of proteins and peptides can be detected in all samples, which underscores the importance of evaluating the HCP in API. This study employs a range of complementary detection methods to conduct a comprehensive and sensitive evaluation of total HCP and specific proteins in drugs, thereby guiding more informed assessments of the risks posed by HCP impurities in raw materials during pharmaceutical processes.

Abstract:The role of biomarkers in drug discovery and development has gained precedence over the years. As biomarkers become integrated into drug development and clinical trials, quality assurance and, in particular, assay validation become essential with the need to establish standardized guidelines for analytic methods used in biomarker measurements. New biomarkers can revolutionize both the development and use of therapeutics but are contingent on the establishment of a concrete validation process that addresses technology integration and method validation as well as regulatory pathways for efficient biomarker development. This perspective focuses on the general principles of the biomarker validation process with an emphasis on assay validation and the collaborative efforts undertaken by various sectors to promote the standardization of this procedure for efficient biomarker development.

Abstract:OBJECTIVE To introduce approaches and techonologies for validating the stable expression of drug transporters in vitro,and provide suggestions for application in cellular models.METHODS Review related literatures about the recent research progress on stably expressing transporters in transfected cells.RESULTS Real-time polymerase chain reaction and Western blot were classical methods respectively for detecting the expression of messenager ribonucleic acid(mRNA) and protein.CONCLUSION The combination of real time-PCR,Western blot with fluorescence detection techonology have promising applications in verifying the stable expression of transporter in different levels including transcription,protein expression and functional activity.

Abstract:Precise measurement of drug concentration in pharmaceutical research is critical, especially for anti-viral drugs like boceprevir, elvitegravir, indinavir, and saquinavir that combat viral infections. It is well-known that analytical techniques play an imperative role in identifying and characterizing active pharmaceutical ingredients in biological samples and drug formulations. Moreover, precise drug assessment directly influences safety, stability, and efficacy while providing in-depth insight into drug pharmacokinetics. Other than this, analytical techniques also aid in identifying impurities, deteriorated products, and potential pollutants. Thus, reliable analytical methods have become crucial for addressing challenges imposed by complex drug formulations. The most commonly used analytical technique is UV spectrophotometry, which does not have the high sensitivity to detect complex drug formulations. In contrast, Liquid Chromatography-Mass Spectrometry/Mass Spectrometry (LC-MS/MS) merges two analytical techniques, chromatography and mass spectrometry, to accurately quantify biological samples. Furthermore, Ultra-Performance Liquid Chromatography (UPLC) provides enhanced resolution, faster analysis in short duration, and low solvent consumption in contrast to HPLC. This comprehensive review aims to critically assess each analytical approach's accuracy, applicability, selectivity, and limitation to provide valuable insights for researchers and analysts. Understanding the weaknesses and strengths of these analytical techniques will enable the researchers to select the suitable analytical method based on their needs and requirements for quality assessment, precise drug quantification, and optimal therapeutic efficiency. Eventually, this review intends to advance pharmaceutical research and development, specifically for anti-viral drugs, by ensuring the effective and secure administration of therapies.

Abstract:The development of the pharmaceuticals brought a revolution in human health. These pharmaceuticals would serve their intent only if they are free from impurities and are administered in an appropriate amount. To make drugs serve their purpose various chemical and instrumental methods were developed at regular intervals which are involved in the estimation of drugs. These pharmaceuticals may develop impurities at various stages of their development, transportation and storage which makes the pharmaceutical risky to be administered thus they must be detected and quantitated. For this analytical instrumentation and methods play an important role. This review highlights the role of the analytical instrumentation and the analytical methods in assessing the quality of the drugs. The review highlights a variety of analytical techniques such as titrimetric, chromatographic, spectroscopic, electrophoretic, and electrochemical and their corresponding methods that have been applied in the analysis of pharmaceuticals.

Development and optimization of SPE-HPLC-UV/ELSD for simultaneous determination of nine bioactive components in Shenqi Fuzheng Injection based on Quality by Design principles

Abstract:A method combining solid phase extraction, high performance liquid chromatography, and ultraviolet/evaporative light scattering detection (SPE-HPLC-UV/ELSD) was developed according to Quality by Design (QbD) principles and used to assay nine bioactive compounds within a botanical drug, Shenqi Fuzheng Injection. Risk assessment and a Plackett-Burman design were utilized to evaluate the impact of 11 factors on the resolutions and signal-to-noise of chromatographic peaks. Multiple regression and Pareto ranking analysis indicated that the sorbent mass, sample volume, flow rate, column temperature, evaporator temperature, and gas flow rate were statistically significant (p < 0.05) in this procedure. Furthermore, a Box-Behnken design combined with response surface analysis was employed to study the relationships between the quality of SPE-HPLC-UV/ELSD analysis and four significant factors, i.e., flow rate, column temperature, evaporator temperature, and gas flow rate. An analytical design space of SPE-HPLC-UV/ELSD was then constructed by calculated Monte Carlo probability. In the presented approach, the operating parameters of sample preparation, chromatographic separation, and compound detection were investigated simultaneously. Eight terms of method validation, i.e., system-suitability tests, method robustness/ruggedness, sensitivity, precision, repeatability, linearity, accuracy, and stability, were accomplished at a selected working point. These results revealed that the QbD principles were suitable in the development of analytical procedures for samples in complex matrices. Meanwhile, the analytical quality and method robustness were validated by the analytical design space. The presented strategy provides a tutorial on the development of a robust QbD-compliant quantitative method for samples in complex matrices.

Abstract:Automation of analytical sample preparation and testing is always attractive due to its high efficiency and continuous operation, but it can also be very challenging due to the operational complexity and method development. Studies focused on method development in automated systems are extremely rare and little has been published on the subject. In this paper, we investigate several unique and advanced aspects of this subject. The example product is selected containing two APIs to be tested by a single method to be productive. The product strengths of the two APIs are significantly different, making development of a single method much more challenging. In addition, we use a "green chemistry" approach, avoiding any organic solvent in the dissolving media in order to achieve an environmentally friendly solution. The selection and optimization of several key operational parameters are then discussed. The successful method is achieved using a new strategy called "Automated Online Standard (AO-STD)" methodology. This novel approach is compared to the manual operation procedure. The proposed approach can substantially reduce the method transfer time, generally considered the biggest obstacle to practical deployment of automated systems. The automated system applied in this paper is most suitable for content uniformity testing, but is also applicable for potency assays, blend uniformity testing, and degradation product tests.

Abstract:The aim of this work was to analyze the current development of China's pharmaceutical analysis discipline and its related fields. The analysis showed that considerable progress had been made in basic research as well as applied research in the analytical area. However,compared with other areas in drug discovery and development,basic research is still weak. A key turning point has been reached where the role of analysis is changing from"service and support"to"innovation and leadership". Thus,it is essential to strengthen interdisciplinary relations,develop new methods,and produce related technologies for pharmaceutical ingredient and activity analysis. This would both solve the scientific and technical problems of pharmaceutical sciences and pharmacology,as well as enable us to establish a new and effective system for pharmaceutical analysis science in China.
