Methods and Applications in Fluorescence

Abstract:Fluorescent samples typically emit isotropically in all directions. Large lenses and other optical components are needed to capture a significant fraction of the emission, and complex confocal microscopes are required for high resolution focal-plane imaging. It is known that Bessel beams have remarkable properties of being able to travel over long distances, over 1000 times the wavelength, without diverging, and hence are called non-diffracting beams. In previous reports the Bessel beams were formed by an incident light source, typically with plane-wave illumination on a circular aperture. It was not known if Bessel beams could form from fluorescent light sources. We demonstrate transformation of the emission from fluorescent polystyrene spheres (FPS) into non-diverging beams which propagate up to 130 mm (13 cm) along the optical axis with a constant diameter. This is accomplished using a planar metal film, with no nanoscale features in the X-Y plane, using surface plasmon-coupled emission. Using samples which contain many FPS in the field-of-view, we demonstrate that an independent Bessel beam can be generated from any location on the metal film. The extremely long non-diffracted propagation distances, and self-healing properties of Bessel beams, offer new opportunities in fluorescence sensing and imaging.
Annular Oblique Epi-Illumination Design for Ultra-Large-scale and High-Resolution Fluorescence Microscopy

Abstract:Biological processes are inherently multi-scale, and supramolecular complexes at the cellular scale. However, in their current form these modalities are limited by a highly constrained field of view (FOV) and field-dependent imaging resolution. New-generation photographic instruments with ultra-large-scale and high-resolution imaging capabilities are strongly demanded in life science and medicine research, ranging from neuroscience to oncology. In practice, the size and quality of images from above instruments are severely limited by several factors related to the illumination. In this work, we present an efficient illumination design for ultra-large-scale and high-resolution fluorescence microscopy to be used on samples of any thickness, yielding strikingly clearer images. Compared with traditional epi-illumination device of fluorescence microscopy, the annular light of our design whose radius is large enough is incident on the sample directly from outside of the objective, rather than through the objective lens, which effectively improves the utility of off-axis illumination. Our design simplifies the overall system and reduces aberrations induced from dichroic mirror because the dichroic mirror is not required any more to separate the illumination path and imaging path. Meanwhile, more uniform illumination can be provided due to the symmetry of annular illumination. No need to consider the lighting angle, the proposed design is especially suitable for fluorescence microscopy because the direction of emitting light is spherical, regardless of the direction of excitation light. The apparatus is simple, robust and inexpensive, making it broadly useful to biological and clinical researchers alike.
Monte Carlo simulation for transmission of photon in scintillation crystal

Abstract:In order to find out an optimized method for constructing scintillation crystal detector, a simulation study for light collecting efficiency of scintillation crystal BGO(bismuth germinate) is carried out by means of Monte Carlo method. The simulation results show that the maximum light output (about 59.1% photon is collected) can be obtained when its incident surface is a coarse surface, the other surfaces are polished surfaces and all its surfaces are coated with high reflecting material. Refractive index of the couplant plays an important role for obtaining high light output.

Abstract:The scintillator ray -light converter plays an important role in digital radiography system and affects the spatial resolution of the radiographic apparently due to its fluorescence dispersion effect.The Point Spread Function(PSF) is usually used to describe the fluorescence dispersion effect of the scintillator and presently the Monte Carlo(MC) method is still the most accurate method to calculate it.An MC computational model is used to calculate PSFs of different LSO and ST -401 scintillators.Based on the calculation,the effect to the PSF of the scattered ray from the interaction between the incident ray and the scintillator is analyzed.The variation of the PSF with the energy of X/γ- ray and the thickness of the scintillator is discussed and explained in theory.
Comparative Study of NaI,BGO,LSO,GSO and YSO Scintillators Using Monte Carlo Simulations

Abstract:Currently,the majority of emission computerized tomography detector uses the scintillation crystal coupled with photomultiplier tube for detecting γ-rays.Improved positioning,timing and energy resolution will be realized if light output can be improved further.The choice of the reflector,coupling material and optical condition of the crystals are the dominated factors affecting the light signal from the scintillator.In order to obtain optimal method to build the scintillation block,the simulation study of the light collection from NaI,BGO,LSO,GSO,YSO scintillator is carried out by Monte Carlo method.The simulation results have identified as:the crystal with all surfaces polished except the incident face roughened and covered with high reflectivity reflector will give the best light output.Among the above crystals,YSO scintillator shows the best light output performance.The geometric size of the crystal also plays an important role to provide high light output.

Abstract:We propose a novel physical mechanism for breaking the diffraction barrier in the far field. Termed fluorescence emission difference microscopy (FED), our approach is based on the intensity difference between two differently acquired images. When fluorescence saturation is applied, the resolving ability of FED can be further enhanced. A detailed theoretical analysis and a series of simulation tests are performed. The validity of FED in practical use is demonstrated by experiments on fluorescent nanoparticles and biological cells in which a spatial resolution of <λ/4 is achieved. Featuring the potential to realize a high imaging speed, this approach may be widely applied in nanoscale investigations.

Abstract:Bombardment of materials by high-energy particles (e.g., electrons, nuclei, X- and $\gamma$-ray photons) often leads to light emission, known generally as scintillation. Scintillation is ubiquitous and enjoys widespread applications in many areas such as medical imaging, X-ray non-destructive inspection, night vision, electron microscopy, and high-energy particle detectors. A large body of research focuses on finding new materials optimized for brighter, faster, and more controlled scintillation. Here, we develop a fundamentally different approach based on integrating nanophotonic structures into scintillators to enhance their emission. To start, we develop a unified and ab initio theory of nanophotonic scintillators that accounts for the key aspects of scintillation: the energy loss by high-energy particles, as well as the light emission by non-equilibrium electrons in arbitrary nanostructured optical systems. This theoretical framework allows us, for the first time, to experimentally demonstrate nearly an order-of-magnitude enhancement of scintillation, in both electron-induced, and X-ray-induced scintillation. Our theory also allows the discovery of structures that could eventually achieve several orders-of-magnitude scintillation enhancement. The framework and results shown here should enable the development of a new class of brighter, faster, and higher-resolution scintillators with tailored and optimized performances - with many potential applications where scintillators are used.

Abstract:<p>Scintillators convert high‐energy radiation into visible luminescence and have been widely applied in various significant fields such as nuclear fusion monitoring, high‐energy particle physics, medical imaging, and geological exploration. The scalable fabrication of scintillating fiber with the desirable combination of uniform size, superior radiation blocking ability, and excellent thermal/radiation stability is a major challenge. Here, a template solidification strategy for fabrication of full‐inorganic and efficient scintillating fiber is presented. The strategy involves the direct solidification of high‐density scintillator melt in a softened silica tube, allowing for the continuous production of small and uniform scintillating fiber. The fabricated scintillating fiber can effectively attenuate radiation up to ≈118 times higher than what conventional silica fiber can. In addition, it exhibits good compatibility with commercial fiber. Using this scintillating fiber, a radiation fiber detector is constructed and its application for online radiation monitoring is demonstrated.</p>

Abstract:Abstract High‐resolution X‐ray imaging is increasingly required for medical diagnosis and large‐area detection. However, the issues of scattering and optical crosstalk are limiting the spatial resolution of the indirect X‐ray imaging. In this study, a feasible and efficient strategy is proposed to in situ synthesize flexible Cs 3 Cu 2 I 5 :2%In + @paper as a superior scintillator film, which can be scaled up to an ultra‐large area of 4800 cm 2 . The as‐obtained Cs 3 Cu 2 I 5 :2%In + @paper performs a fascinating photoluminescence quantum efficiency up to 88.14%, a steady state light yield of 70169 photons/MeV, and spatial resolution of 15 lp mm −1 . Moreover, the suppressed physical scattering and optical crosstalk of the corresponding film are demonstrated. Accordingly, this work explores a feasible fabrication of customizable scintillation films with large area for high‐resolution X‐ray detection.

Abstract:This perspective considers light propagation in metal halide scintillators as the focus. By correlating the reported spatial resolution with light propagation in various forms, this perspective clarifies and deepens the understanding of untouched yet fundamental issues, namely, material‐form‐associated light propagation in metal halide scintillators, and provides the design rules for their future development. The ever‐growing need to inspect matter with hyperfine structures requires a revolution in current scintillation detectors, and the innovation of scintillators is revived with luminescent metal halides entering the scene. Notably, for any scintillator, two fundamental issues arise: Which kind of material is suitable and in what form should the material exist? The answer to the former question involves the sequence of certain atoms into specific crystal structures that facilitate the conversion of X‐ray into light, whereas the answer to the latter involves assembling these crystallites into particular material forms that can guide light propagation toward its corresponding pixel detector. Despite their equal importance, efforts are overwhelmingly devoted to improving the X‐ray‐to‐light conversion, while the material‐form‐associated light propagation, which determines the optical signal collected for X‐ray imaging, is largely overlooked. This perspective critically correlates the reported spatial resolution with the light‐propagation behavior in each form of metal halides, combing the designing rules for their future development.

New fiber read-out design for the large area scintillator detectors: providing good amplitude and time resolutions

Abstract:Most of the time-of-flight systems as well as the fast interaction trigger detectors have large surfaces and the principal requirements for the above mentioned detectors is a good time resolution of the order 100-200ps. The easiest solution here is to split the large surface into the small tiles with a read-out directly attached to the photo-sensor. This solution is expensive because the number of the channels grows proportional to the surface. Although if the coverage of the whole surface by the sensors is not sufficient, one can obtain non uniform response, which is not acceptable if uniformity is required. Our suggestion is based on the usage of clear fibres mounted perpendicular on the surface of the scintillator as a matrix providing uniform surface reduction and keeping most of the benefices of the small tiles with the photo-sensors. In this work the design and some analytic estimations of the light collection efficiency for direct and reflected photons will be presented. Also some simple estimations are presented for the time-spread and for the light pulse wave form dependent on the lateral sizes and the thickness of the scintillator.

Abstract:Scintillators, which can convert high-energy ionizing radiation into visible light, have been serving as the core component in radiation detectors for more than a century of history. To address the increasing application demands along with the concern on nuclear security, various strategies have been proposed to develop a next-generation scintillator with a high performance in past decades, among which the novel approach via structure control has received great interest recently due to its high feasibility and efficiency. Herein, the concept of "structure engineering" is proposed for the exploration of this type of scintillators. Via internal or external structure design with size ranging from micro size to macro size, this promising strategy cannot only improve scintillator performance, typically radiation stopping power and light yield, but also extend its functionality for specific applications such as radiation imaging and therapy, opening up a new range of material candidates. The research and development of various types of structured scintillators are reviewed. The current state-of-the-art progresses on structure design, fabrication techniques, and the corresponding applications are discussed. Furthermore, an outlook focusing on the current challenges and future development is proposed.

Abstract:<p>Scintillators are widely used in ionization radiation detection. The light extraction efficiency and thus the effective light output of scintillators are limited by the total internal reflection due to their high refractive indices. Furthermore, scintillators have an approximate Lambertian profile for the spatial distribution of the light emission, which is detrimental to the collection of light. In this paper, hollow microlens arrays (HMLAs) composed of periodic arrangement of individual hollow shell hemispherical microlenses are proposed to achieve the directional light extraction enhancement of scintillators. The optical properties of HMLAs can be conveniently adjusted by varying the refractive index and the thickness of the shell layer. Simulation results reveal that the refractive index and the thickness of the shell of the individual hollow microlenses are the key parameters to achieve the control of the light output of scintillators. The measurement results of the prepared HMLAs-coated LYSO scintillator are consistent with the prediction of the numerical simulations. This scheme of directional light extraction enhancement by HMLAs is also applicable to other light-emitting devices that have the requirement of directional emission.</p>

Abstract:Scintillators are materials that convert ionizing radiation in the form of particles or photons into low-energy photons in the ultraviolet to near-infrared spectral range. This work reviews efforts to use colloidal nanocrystals as scintillator materials. To date, research on colloidal nanocrystals as scintillators has focused on doped phosphor systems, quantum dots and related structures, and perovskite-based nanocrystals. Among various material classes and forms, colloidal semiconductor nanocrystals stand out thanks to their appealing fluorescence properties, yet understanding of their radioluminescence and cathodoluminescence is incomplete. This review discusses fundamental limits and material design challenges toward achieving high brightness, fast speed, and durable scintillator performance with nanocrystal scintillators. First, this review describes the basic principles and efficiency limitations of scintillation, particularly the large influence of multiple exciton generation in many nanocrystal species, as well as performance metrics. Second, methods for measurement are described. Third, we review the results of the main classes of nanocrystal scintillators, including quantum dots and related particles, perovskites, and doped-phosphor particles. Fourth, scintillation imaging is discussed in terms of the relevant performance metrics and results obtained using nanocrystal materials. Finally, we note the strengths and weaknesses of nanocrystal scintillators and discuss potential areas of further development.
Influences of scintillation volume on determination of various nuclides by liquid scintillation counting

Abstract:Scintillation volume,as well as quench,is an important factor in change of counting efficiency for liquid scintillation counting (LSC).Various representative radionuclides have been determined in two liquid scintillation counters,with the discrimination between the influence of scintillation volume and that of quench.The influence of scintillation volume is very little on the counting efficiency of α particles or high energy β particles.The wall effect resulting from small scintillation volume changes the shape of LSC spectrum,but has little influence on the counting efficiency.The influence of scintillation volume is remarkable on the counting efficiency of γ rays or low energy β particles.The quench with the same level has greater influence on the counting efficiency of the sample with larger scintillation volume.When a quench correction curve is used to calculate the counting efficiency or a curve of "optimum PSA-quench index" is used to set the parameter of α/β discrimination,in order to minimize the measurement uncertainty,the sample to be determined should have the same scintillation volume in the vial with the same size as the quenching standards,no matter what quench index is applied.

Abstract:The efficient production of high-quality scintillators with long radioluminescence afterglow is crucial for high-performance X-ray luminescence extension imaging. However, scaling-up the synthesis of ligand-free scintillators to fabricate large-area X-ray imaging screens for industrial applications remains a challenge. In this study, we report an efficient method to synthesize ligand-free, lanthanide-doped microscintillators by a one-pot reaction via the concentrated hydrothermal method. The as-synthesized microscintillators exhibit prolonged persistent radioluminescence for up to 30 days after X-ray exposure and remain high stability in air or water for more than 18 months without deterioration. Monte Carlo simulations indicate that the size effect is responsible for the excellent afterglow performance of the microscintillators. We employ these high-quality lanthanide-doped microscintillators to fabricate a large-area X-ray imaging detector using a blade-coating method, a spatial resolution of 24.9 lp/mm for X-ray imaging. Our study offers a solution for scaling-up the synthesis of low-cost microscintillators for practical applications.

Abstract:The congruent glass composite scintillator with good processing capability and high scintillation light yield is constructed, which solves the problem of poor preparation and processing properties of scintillation single crystal and ceramic. The constructed glass composite scintillator is further verified in applications such as X‐ray imaging and real‐time radiation detection of fibers. These findings not only represent a novel design concept for the development of glass composite but also suggest a great step for expanding the scope of scintillators. The scintillator is the most crucial component in the high‐energy ray detection system. The available scintillators suffer from the insurmountable drawbacks including poor shaping ability for single crystal and ceramic and low efficiency for glass. Here, a glass composite scintillator is proposed from a congruent crystallization system which possesses both excellent processability and high scintillating yield. It can be fabricated into diverse shapes and sizes including the bulk and tiny fiber. Benefitting from the unique compositing combination with a high crystallization ratio, the glass composite exhibits a giant scintillating light yield of ≈26,000 photons per MeV. The practical application for X‐ray imaging is demonstrated and a high spatial resolution of 12 l p mm−1 is achieved. Furthermore, the fiber derived detector is built and the remote and micro‐area detection is realized. These findings not only represent a novel design concept for the development of glass composite but also suggest a great step for expanding the scope of scintillators.
