Abstract:γ/δ T cells have stimulated a lot of interest because of their unique features in antigen recognition and cytotoxicities to many autologous and/or allogeneic tumor cells. We have developed a novel method to selectively expand larger amounts of human tumor–infiltrating γ/δ T lymphocytes (γ/δ TILs) ex vivo by immobilized pan– anti–TCRγ/δ monoclonal antibody in the presence of exogenous IL–2. The expanded γ/δ TILs mainly expressed CD45RO and HLA–DR molecules and did not express CD4. CD8+ γ/δ TILs accounted for 19% of γ/δ TILs. The expression of CD25 molecule on expanded γ/δ T cells was inducible and downregulated following a time course. The Vδ1 and Vδ2 subsets amount to 37 and 58%, respectively. The expanded γ/δ TILs show an IL–2–dependent proliferation, MHC class I–unrestricted and TCRγ/δ–related cytotoxicities to two MHC class I+ and two MHC class I+ allogeneic tumor cell lines in vitro.

Abstract:Ag-dependent T cell activation requires multiple transmembrane signals including activation of Ag-specific T cell receptor in combination with signals delivered through cytokine receptors. IL-12 is a heterodimeric cytokine involved in the regulation of NK and T lymphocyte responses. In examining the role of IL-12 in T cell activation, we found a direct relationship between Ag stimulation and IL-12-induced proliferation. Unlike IL-2, which induced proliferation of CTL either in the presence or absence of a CD3/TCR co-signal, IL-12 mediated proliferation of CTL only when the cells were recently co-stimulated with alloantigen or solid-phase anti-CD3 antibody. After culture in the absence of alloantigen or anti-CD3 for 7 to 14 days, these CTL lost the ability to proliferate to IL-12 alone. Under these conditions, however, IL-12 synergized with low-dose IL-2 to induce CTL proliferation. Restimulation with alloantigen or solid-phase anti-CD3 restored the ability of the CTL to proliferate to IL-12 alone. Not all Ag signals resulted in IL-2 independent proliferation to IL-12. For example, CTL with specificity for influenza matrix peptide proliferated best when co-cultured with peptide Ag presented on self MHC and a combination of IL-2 and IL-12. This evidence suggests that IL-12 may be useful in expanding an Ag-specific T cell population, as the culture of CTL with IL-12 and low-dose IL-2 leads to proliferation only in response to an Ag co-signal.

Abstract:Enzyme-linked immune absorbent spot (Elispot) is a quantitative method for measuring relevant parameters of T-cell activation. The sensitivity of Elispot allows the detection of low-frequency antigen-specific T-cells that secrete cytokines and effector molecules, such as granzyme B and perforin. Cytotoxic T-cell (CTL) studies have taken advantage with this high-throughput technology by providing insights of quantity and immune kinetics. Accuracy, sensitivity, reproducibility, and robustness of Elispot resulted in a wide range of applications in research as well as in diagnostic field. Actually, CTL monitoring by Elispot is a gold standard for the evaluation of antigen-specific T-cell immunity in clinical trials and vaccine candidates where the ability to detect rare antigen-specific T-cells is of relevance for immune diagnostic. The most utilized Elispot assay is the Interferon-gamma (IFN-γ) test, a marker for CD8+ CTL activation, but Elispot can be also used to distinguish different subsets of activated T-cells by using other cytokines such as T-helper (Th) 1 type cells (characterized by the production of IFN-γ, IL-2, IL-6, IL-12, IL-21 and TNF-α), Th2 (producing cytokines like IL-4, IL-5, IL-10 and IL-13), and Th17 (IL-17) cells.The reliability of Elispot generated data, by the evaluation of T-cell frequency recognizing individual antigen/peptide, is the core of this method currently applied widely to investigate specific immune responses in cancer, infections, allergies, and autoimmune diseases. The Elispot Assay is competing with other methods measuring single-cell cytokine production, e.g., intracellular cytokine by FACS or Milteny cytokine secretion assay. Other types of lymphocyte frequency and function assays include limiting dilution assay (LDA), cytotoxic T-cell assay (CTL), and tetramer staining. Compared with respect to sensitivity the Elispot Assay is outranking other methods to define frequency of antigen-specific lymphocytes. The method described herein would like to offer helpful and clear protocols for researchers that apply Elispot. IFN-γ and Perforin Elispot assays will be described.

Abstract:BACKGROUND:Tuberculosis (TB) is an infectious disease and its mortality rate ranks first. Latent tuberculosis infection (LTBI) means that a patient is infected with Mycobacterium tuberculosis, but has no relative clinical symptoms. It has been estimated that approximately 10% of patients with LTBI would develop into active tuberculosis. Therefore, it was urgent to search for more efficient biomarkers to discriminate LTBI from healthy population. METHODS:The Luminex assay was employed to detect the quantity of cytokines secreted by mononuclear cells from peripheral blood stimulated with the ESAT6 protein among TB, LTBI and healthy controls. The cytokine profile was analyzed by principal components analysis and the receiver operating characteristic curve analysis. RESULTS:The principal components analysis indicated that LTBI and TB were clearly separated from healthy controls, and that LTBI was also successfully differentiated from healthy controls. The cytokine profiling method to distinguish LTBI from healthy controls has a sensitivity and specificity of 100%. Nine potential biomarkers, including IL-23, IL-21, HGF, Bngf, IL-27, IL-31, IL-1β, IL-22 and IL-18, were identified, and these cytokines were considered as a potential cytokine complex for more effectively discriminating LTBI from healthy controls. CONCLUSION:IL-23, IL-21, HGF, Bngf, IL-27, IL-31, IL-1β, IL-22 and IL-18 were demonstrated to be the potential cytokine complex for the assessment between LTBI and healthy controls.
99Mtc-Labeled Mutant Interleukin-2 As a Promising SPECT Probe for in Vivo Imaging of Tumor-Infiltrating T Cells

Abstract:161 Objectives: T cell infiltration of tumor lesions is a known prognostic factor in several tumor types and is used as treatment mechanism in some of these tumor types. Tumor infiltrating T cells express the high affinity interleukin-2 (IL2) receptor (CD25) on their surface. These T cells could therefore be visualized by molecular imaging with a radiolabeled IL2 for this receptor. For this purpose, [18F]FB-IL2 was designed as a PET imaging probe for T cell imaging, but strictly limited by its low target-to-background ratios and toxicity, which was resulted from direct binding of IL-2 to αβγ IL-2Rs on lung endothelial cells. Interaction of IL-2 with lung endothelial cells can be abrogated by blocking IL2 and IL-2Rα interaction. Here we developed a novel T cell imaging probe based on mutant Interleukin-2 (mIL2), which reduced binding to IL-2Rα and increased binding affinity for IL-2Rβ and may lead to higher target-to-background ratios and less toxicity in vivo than IL2. Methods: Mutant Interleukin-2 was expressed in Sf9 cells and purified by Ni-NTA, then concentrated using spin concentrators and purified with an FPLC Superdex 75 sizing column. The bioactivity of purified mIL2 was detected with the CTLL2 cells which were dependent upon IL-2 for growth. IL2/IL-2Rα interaction was detected by ELISA. Then, mIL2 was radiolabeled with 99mTc-HYNIC-GGGGK conjugated to the LPETG tag through Sortase A. The pharmacokinetic data of 99mTc-HYNIC-mIL2 was obtained by ex vivo biodistribution experiments. Finally, the tumor infiltrating T cells detecting capability of 99mTc-HYNIC-mIL2 in MC38 subcutaneous tumor-bearing mice was evaluated by in vivo NanoScan SPECT/CT imaging. Results: mIL2 showed bioactivity with CTLL2 cells whereas lower CD25 binding compared with IL2. The 99mTc-labeling procedure was done within 2 h, with a yield of 70%. The radiochemical purity was greater than 98%. As shown in figure 1, the tumor-infiltrating T cells located in the MC38 tumor margin was clearly visualized by 99mTc-HYNIC-mIL2 NanoScan SPECT/CT imaging with low background except relatively high kidney uptake at all detected time points (0.5 and 2 h p.i.). Specificity of 99mTc-HYNIC-mIL2 to T cells was demonstrated by competitive blocking of tumor uptake with excess mIL2. Biodistribution results showed that the tumor uptake of 99mTc-HYNIC-mIL2 at 0.5 and 2 h p.i. was 2.59 ± 0.37 %ID/g and 2.19 ± 0.16 %ID/g, respectively. Conclusions: 99mTc-HYNIC-mIL2 is a promising SPECT radiotracer for the noninvasive imaging of tumor infiltrating T cells, which showed high target-to-background ratios. This novel tracer possesses enormous potential for evaluating and guiding immunotherapy in clinical practice, and its further investigation is still in progress.

Abstract:Abstract Interleukin 2 (IL 2) is known to have multiple immunoenhancing activities that are related to its ability to promote the proliferation and the expression of effector functions of human T lymphocytes. We investigated the potential of IL 2 to induce human T lymphocyte migration. Unstimulated T cells did not respond to IL 2, but T cells exposed to dextran or phytohemagglutinin did respond to IL 2 concentrations from 0.01 to 10.0 U/ml, with significantly increased migration. This activity could be specifically blocked with anti-Tac antibody. Analysis of T lymphocyte subsets revealed that OKT4+ but not OKT8+ lymphocytes responded to IL 2 in the chemotaxis assay. Checkerboard analysis demonstrated that the IL 2-induced chemoattractant activity was predominantly chemotactic rather than chemokinetic in nature. The activity of IL 2 was compared with that of another chemoattractant lymphokine, lymphocyte chemoattractant factor, which was found to stimulate lymphocyte migration without prior exposure to mitogen, and which was not inhibited by anti-Tac. Our data suggest that the lymphocyte migratory response to IL 2 is under the control of the inducible receptor recognized by anti-Tac in a manner similar to the proliferative response to IL 2, but differs from proliferation in its OKT4+ cell specificity.

Synergistic effects of IL-2 and IL-12 on antitumor activity of human PBMC in vitro

Abstract:Objective To examine the in vitro antitumor activity of normal human peripheral blood mono-nuclear cells(PBMC) induced by interleukin-2(IL-2) and IL-12.Methods MTT and flow cytometry assay were used to detect the cytotoxic effects of IL-2 or/and IL-12 on 3 cell lines.Results PBMC induced by IL-2 and IL-12,had significantly different cytotoxic effects on three cell lines(P<0.05).A good correlation was observed between the cytotoxicity by MTT and flow cytometry(r=0.997,P <0.05).Conclusion The cytotoxic synergy by IL-2 and IL-12 against 3 tumor cell lines in vitro is significantly differents.The different antitumor effects maybe associated with different immune characteristics of tumor cells.

Comparative dose-responses of recombinant human IL-2 and IL-7 on STAT5 phosphorylation in CD4+FOXP3- cells versus regulatory T cells: a whole blood perspective

Abstract:Interleukin(IL)-2 and IL-7 are cytokines with important functions related to CD4(+) lymphocyte proliferation, differentiation and survival. Depending on doses, they theoretically activate regulatory (Treg) and/or effector T cells (Teff) and thus may be indicated with different therapeutic objectives. In this study we assessed ex vivo the differential dose-responses of CD4(+) T cell subsets (Treg versus CD4(+)FOXP3(-) cells) to recombinant human (rh) IL-2 and rhIL-7. Fresh whole blood from healthy donors was stimulated with increasing doses of cytokines. By using a novel flow cytometry procedure of intracellular signaling pathway staining (e.g., detection of STAT5 phosphorylation; a pivotal marker of cytokine-induced activation; in combination with intracellular FOXP3 staining), we were able to specifically measure Treg and CD4(+)FOXP3(-) cell responses in the same tube. Half maximal effective concentrations (EC50) were calculated. We observed a dose-response effect on Treg and CD4(+)FOXP3(-) cells for both cytokines. Interestingly, low doses of hIL-2 preferentially activated Treg (EC50 Treg = 0.15 pg/ml versus CD4(+)FOXP3(-) cells = 750 pg/ml - p < 0.0001) whereas low doses of rhIL-7 preferentially induced CD4(+)FOXP3(-) cell activation (EC50 Treg = 25 pg/ml and CD4(+)FOXP3(-) cells = 2.5 pg/ml - p < 0.0001). To our knowledge, this work is the first to show differential dose-response effects on CD4(+)FOXP3(-) cells versus Treg of rhIL-7 and rhIL-2 in one ex vivo whole blood single tube assay including two intracellular stainings (i.e., pSTAT5 and FOXP3). Beyond the confirmation of the dose-dependent differential effects of IL-2 versus IL-7 on CD4(+)FOXP3(-) cells/Treg, our results illustrate the value of this approach for monitoring drugs' activities by flow cytometry in daily clinical practice.

Abstract:Abstract High concentrations of interleukin 2 (IL 2) were shown to produce a delayed but pronounced proliferation of purified resting T cells in the apparent absence of other activation signals. Because these stimulatory effects of IL 2 occurred in the absence of detectable Tac+ cells, the possibility that IL 2 might be initially interacting with an IL 2 binding protein distinct from the Tac protein was studied. Chemical cross-linking studies with 125I-IL 2 revealed the presence of an IL 2 binding protein distinct from the Tac protein on the surface of these unstimulated T cells. This second IL 2 receptor has an estimated molecular size of 70,000 daltons, lacks reactivity with the anti-Tac antibody, and appears to be identical to the p70 protein recently proposed as a component of the high affinity IL 2 receptor. Scatchard analysis of IL 2 binding assays performed with the unactivated T cells revealed approximately 600 to 700 p70 sites per cell and an apparent Kd of 340 pM. These data indicate that the p70 protein present on resting T cells binds IL 2 with an intermediate affinity compared with the previously recognized high and low affinity forms of the receptor and may account for the high concentration of IL 2 needed to induce resting T cell proliferation. To investigate the early biologic consequences of IL 2 binding to the p70 protein, potential changes in the expression of genes involved in T cell activation were examined. Northern blotting revealed the rapid induction of c-myc, c-myb, and Tac mRNA after stimulation of resting T cells with a high concentration of IL 2. The anti-Tac antibody did not inhibit IL 2 induced expression of these genes, suggesting that the p70 protein rather than the Tac antigen or the high affinity IL 2 receptor complex mediated this signal. However, in contrast to these early activation events, the anti-Tac antibody significantly inhibited IL 2 induced T cell proliferation. This finding implicates the high affinity form of the IL 2 receptor in the proliferative response of the IL 2 activated T cells. Thus these data support a two step model for the induction of resting T cell proliferation by high doses of IL 2 involving the initial generation of an activation or "competence" signal through the p70 protein and a subsequent proliferation or "progression" signal through the high affinity form of the receptor.

Abstract:BACKGROUND:The Mycobacterium tuberculosis (Mtb)-specific T-cell interferon gamma release assays (IGRAs) are useful in detecting Mtb infection but perform poorly at distinguishing active tuberculosis disease (ATB) and latent tuberculosis infection (LTBI). This study is aimed at evaluating additional cytokines as biomarkers besides interferon-gamma (IFN-γ) to improve the identification of ATB and LTBI.METHODOLOGY/PRINCIPAL FINDINGS:Sixty-six patients with ATB, 73 household contacts (HHC) of ATB patients and 76 healthy controls (HC) were recruited to undergo QuantiFERON TB GOLD in-tube assay (QFT) and the enzyme-linked immunosorbent assay (ELISA) where the release of IFN-γ, IFN-γ inducible protein 10 (IP-10), Interleukin 2 (IL-2) and Tumor Necrosis Factor-α (TNF-α) was determined in the whole blood with or without antigen-stimulation. The positive rates of the QFT, IP-10 and IL-2 tests were 86.4%, 89.4% and 86.4% for the ATB group with no difference between them (p>0.05). However, QFT in combination with IP-10 and IL-2 significantly increased the detection rate to 95.5% in the ATB group (p = 0.03) and the indeterminate rate of all samples decreased from 2.3% (5/215) to 0.4% (1/215). The un-stimulated level of IP-10 was significantly higher in the HHC than the ATB and HC groups. The IP-10 responses were strongly associated with extended Mtb exposure time and the degree of smear-positivity of the index cases. The IL-2/IFN-γ ratio in the antigen-stimulated plasma could discriminate LTBI from ATB with a sensitivity of 77.2% and a specificity of 87.2%.CONCLUSION:The increased Mtb-specific antigen-stimulated expression of IP-10 and IL-2 may be useful for detecting both ATB and LTBI. Combining the QFT with IP-10 and IL-2 could increase the detection accuracy of active TB over the QFT alone.

Abstract:To study ralationship beteen interteukin 2 (JL-2) and carcinoma open up a new field in tumor immunity. In this study, the method of modified rapid detection of IL-2 activity with mouse thymocyte was applied. IL-2 activity of peripheral blood was detected in 30 malignant lymphoma patients with 30 normal person. Patients received assay before and after COP scheme chemotherapy. The results show that pretherapeutic IL-2 activity value is 6.51±1.18 u/ml, and posttherapeutic .increase to 13.78±2.69 u/ml, but control's, 26.40±14.79 u/ml. To compare each other, the differences are highly significant (P0.01). The posttherapeu utic values and their increase degrees; of patients with stage Ⅲ-Ⅳ, partial response, and dead or tumor-bearing are lower than those with stage Ⅰ-Ⅱ (P0.02, P0.05), complete response (P0.05), and free (P0.01). There are no statistical significant (P0.05) in the values of patients with different courses, pathological types, and positive or negative extranodal involvm-e.nts. In the discussion, we have suggested that 1. decrease of IL-2 activity is a probable feature of all patients, with carcinoma; 2. tumor load perhaps plays a major role to suppress IL-2 activity; 3. chemotherapy may increase IL-2 activity with decrement of the dose of tumor load; 4. it is difficult to get rid of suppression to IL-2 activity with exacerbation of malignant lymphoma patients; 5. IL-2 activity assay may be able to become a index to predict prognosis of patients with carcinoma.

Abstract:To locate functional domains of the interleukin-2 (IL-2) protein, a cDNA clone encoding biologically active human IL-2 was mutagenized using synthetic oligonucleotides to incorporate defined amino acid substitutions and deletions in the mature protein. The IL-2 analogs were then produced in Escherichia coli and assayed for the ability to induce proliferation of IL-2-dependent cells and the ability to compete for binding to the IL-2 receptor. Our analysis of over 50 different mutations demonstrated that the integrity of at least three regions of the IL-2 molecule is required for full biological activity: the NH2 terminus (residues 1-20), the COOH terminus (residues 121-133), and 2 of the 3 cysteine residues (58 and 105). Deletion of the NH2-terminal 20 amino acids or the COOH-terminal 10 amino acids resulted in the loss of greater than 99% of bioactivity and binding. Amino acid substitutions at specific positions in these regions also resulted in proteins which retained less than 1% activity. The NH2 terminus and an adjacent internal region were recognized by neutralizing anti-IL-2 antibodies. In combination with the results from epitope competition analysis with neutralizing antibodies, these data are consistent with the IL-2 protein being folded such that the NH2 terminus, the COOH terminus, and the internal 30- to 60-region are juxtaposed to form the binding site recognized by the IL-2 receptor.

Abstract:Objective To investigate the improvements of 3(4,5dimethylthiazole2yl)2,5diphenyltetrazolium bromide(MTT) assay and apply the improved MTT assay to test the lymphokine activated killer (LAK) cells' activity.Methods The study was done through the detection of the absorbance of resolvent dimethyl sulfoxide and the comparison of different culture times of the target cells.Results The best measurement degree was 510?nm and the best culture time was 4 hours.Conclusion Improved MTT assay is better than MTT assay.
A New Reliable Bioassay for Determining the Biological Activity of Human Interleukin-12 by Using Human NK Cell Line NKG Cells.

Abstract:A specific and accurate bioassay for determining the biological activity of human interleukin-12 (hIL-12), an important typical Th1 cytokine in both innate and adaptive immunity, is extensively required for biomedical and clinical study. In this paper, we used a new established NK cell line NKG cells as the responder to hIL-12 stimulation by detecting their IFN-γ production. It was found that NKG cells produced high level of IFN-γ when simulated by hIL-12, and the dose–response curve became the best Sigmoid curve (R2=0.9977, p<0.0001) when stimulated for 24h. The intra-assay CVs (<10%) and inter-assay CVs (<5%) demonstrated that the bioassay was precise and reproducible. Furthermore, no obvious cross-effects of other cytokines such as IL-2 and IL-18 was observed on the bioassay. Addition of the neutralizing anti-hIL-12 antibody to the bioassay significantly inhibited the IFN-γ production in a dose dependent manner, indicating that the bioactivity was actually mediated by hIL-12. The bioassay by using NKG cells was also suitable for determining the biological activity of recombinant hIL-12 in the form of purified product or culture supernatant by CHO-hIL-12 cell line. In conclusion, a reliable hIL-12 bioassay for determining its biological activity was established by using NKG cells as a responder and measuring their production of IFN-γ.
The co-stimulation of anti-CD28 and IL-2 enhances the sensitivity of ELISPOT assays for detection of neoantigen-specific T cells in PBMC

Abstract:Neoantigen-based cancer immunotherapies hold the promise of being a truly personalized, effective treatment for diverse cancer types. ELISPOT assays, as a powerful experimental technique, can verify the existence of antigen specific T cells to support basic clinical research and monitor clinical trials. However, despite the high sensitivity of ELISPOT assays, detecting immune responses of neoantigen specific T cells in a patient or healthy donor's PBMCs is still extremely difficult, since the frequency of these T cells can be very low. We developed a novel experimental method, by co-stimulation of T cells with anti-CD28 and IL-2 at the beginning of ELISPOT, to further increase the sensitivity of ELISPOT and mitigate the challenge introduced by low frequency T cells. Under the optimal concentration of 1 μg/ml for anti-CD28 and 1 U/ml for IL-2, an 11.7-fold increase of T cell response against CMV peptide was observed by using our method, and it outperforms other cytokine stimulation alternatives (5-10 folds). We also showed that this method can be effectively applied to detect neoantigen-specific T cells in healthy donors' and a melanoma patient's PBMCs. To the best of our knowledge, this is the first report that the co-stimulation of anti-CD28 and IL-2 is able to significantly improve the sensitivity of ELISPOT assays, indicating that anti-CD28 and IL-2 signaling can act in synergy to lower the T cell activation threshold and trigger more neoantigen-specific T cells.
